Description
本明細書に記載されているのは、TNFに結合して、TNF媒介のシグナル伝達を抑制する核酸アプタマー(抗TNFアプタマー)であり、予想通りに、炎症を軽減する。本明細書に使用される核酸アプタマーは、特定の標的分子(TNF、例えばヒトTNF)に対する結合活性を有する核酸分子(DNAまたはRNA)を指す。前記アプタマーは、TNF分子に結合することでTNF媒介のシグナル伝達を阻害できる。本開示の抗TNFアプタマーは、線状または環状の形式で、RNA、DNA(例えば、一本鎖DNA)、修飾された核酸、またはこれらの混合物であってもよい。前記抗TNFアプタマーは、非自然発生分子(例えば、天然遺伝子に存在しないヌクレオチド配列を含むか、または自然界に存在しない修飾されたヌクレオチドを含む)であってもよい。あるいは、またはさらに、前記抗TNFアプタマーは、機能性ペプチドをコードするヌクレオチド配列を含まなくてもよい。
本明細書に記載の一つまたは複数の抗TNFアプタマー(単量体または多量体、例えば、二量体)またはそのPEG共役物は、薬学上許容できる担体(賦形剤)と混合して、対象疾患を治療するための医薬組成物を形成してもよい。「許容できる」とは、担体が必ず組成物の活性成分と相溶し(好ましくは、活性成分を安定させられる)、治療対象に有害ではないことを意味する。緩衝剤を含む薬学上許容できる賦形剤(担体)は、当技術分野で周知のものである。例えば、Remington:The Science and Practice of Pharmacy 20th Ed.(2000)Lippincott Williams and Wilkins,Ed.K.E.Hooverを参照する。
TNFは、ほぼ全種類の炎症関連疾患で役割を担っており、また、TNF分泌の調節不全は、例えば、関節リウマチ、乾癬、強直性脊椎炎、炎症性腸疾患、神経変性疾患、急性肺損傷(または急性肺不全)、急性肝損傷、成人呼吸窮迫症候群、および癌などの疾患を引き起こす。したがって、TNF媒介のシグナル伝達の調節および/またはTNFの存在/レベルの検出は、TNF媒介の疾患を効果的に治療または診断できる。

化学物質およびオリゴヌクレオチド.
すべての化学物質は、Sigma−Aldrich(St.Louis、MO、USA)から購入され、オリゴヌクレオチドは、Integrated DNA technologies(Coralville、IA、USA)によって合成された。aptTNFαの配列は5’−GCGCCACTACAGGGGAGCTGCCATTCGAATAGGTGGGCCGC−3’(配列番号1)であった。
ヒトTNFα指向性アプタマーは、ニトロセルロースフィルターSELEXによって同定された。合成した一本鎖DNAライブラリーは、80個のヌクレオチド長の一本鎖DNAで構成され、40ヌクレオチドのランダム配列にプライマー配列が隣接する5’−ACGCTCGGATGCCACTACAG[N]40CTCATGGACGTGCTGGTGAC(配列番号2)であり、ここで、N=A、T、G、C。一回目のSELEXラウンドでは、1015分子のssDNAライブラリーを組換えヒトTNFαタンパク質(R&D Systems、Minneapolis、MN、USA)と一緒に培養した。TNFαタンパク質に結合したssDNAは、ニトロセルロースフィルターで収集し、非結合のssDNAは、繰り返して洗浄することで、除去した。TNFαに結合したssDNAsを加熱して溶出し、ネガティブセレクションのためにアルブミンと培養し、ニトロセルロースフィルターに通した。フロースルーを収集し、PCRによって増幅した。SELEXは10ラウンド繰り返された。TNFαに結合したssDNAおよびアルブミンに結合したssDNAの両方は、次世代シーケンシング(Illumina MiSeq System、Illumina、San Diego、CA)を行った。出力読み取りはFASTApatmer(Alam et al., Mol.Ther.Nucleic Acids.2015;4:e230)によってクラスター化され、アルブミン結合グループに現れたクラスターで差し引かれた。次に、残りのクラスターで最も高い読み取り値を示した代表的な配列を、Mfoldを使用して構造解析を行った。これらの切り捨てられた派生物は、予測された二次構造に従って設計した。
過剰量のアミン標識のaptTNFαを、二官能性N−ヒドロキシルスクシンイミドポリエチレングリコール(NHS−PEG−NHS、分子量20kDa、Polysciences Inc.、Warrington、PA)と一緒に、重炭酸ナトリウム緩衝剤(pH8.3)において37℃で18時間培養した。PEG化二量体aptTNFα(aptTNFα−PEG)は非変性ポリアクリルアミドゲル電気泳動によって精製され、その濃度はNanodrop分光光度計(Thermo Scientific、Hudson、NH、USA)によって測定した。
ヒトTNFαタンパク質(0、8.75、17.5、35、70、140nM、R&D Systems)を、aptTNFα(50nM)と一緒に、37℃で1時間培養した。さらに、マウスTNFαタンパク質(140nM)またはBSA(140nM)を、aptTNFα(50nM)またはaptTNFα−PEG(50nM)と培養した。タンパク質結合アプタマーをニトロセルロースフィルターによって収集し、加熱して溶出した。溶出されたアプタマーの量は、定量的PCR(LightCycler 480 system、Roche Applied Science、Mannheim、Germany)によって定量化された。解離定数(Kd)は、式Y=Amax×X/(Kd+X)を使用して、GraphPad Prism 5(GraphPad Software、San Diego、CA)によって算出した。タンパク質結合アプタマー(ヒトTNFαタンパク質およびマウスTNFαタンパク質)の相対量は、BSAを参照として使用して倍率変化として表された(1倍)。
マウスは国立実験動物センター(台北、台湾)から購入した。すべての動物実験は、中央研究院の動物施設の指導に従って行われた。6週齢のBalb/c雄マウスにLPS(10mg/kg、気管内)を投与してALIを誘導した。IRDye(登録商標)800CW標識のaptTNFα(Integrated DNA technologies)は、LPS投与の1時間後に静脈内注射された。aptTNFαから放出された蛍光シグナルを、Xenogen IVIS Imaging System 200シリーズ(Caliper Life Sciences、Alameda)によって、アプタマー投与後の2時間、4時間、7時間、10時間、および24時間後に、それぞれ検出した(Cakarova et al.,Am J Respir Crit Care Med.2009;180:521−32)。さらに、IRDye(登録商標)800CW標識のaptTNFαで処理された群のマウスは、アプタマー投与後の4時間後に犠牲された。心臓、肝臓、脾臓、肺臓、腎臓、膀胱を含む重要な臓器を収集し、aptTNFαから放出された蛍光シグナルも検出した。血液をサンプリングし、LDH、AST、およびALTのレベルをFuji Dri−Chem 4000i(Fujifilm、Tokyo、Japan)によって検出した。
HEK293細胞を、10%FBS(Gibco)を含むDMEM(Gibco BRL、Life Technologies、Grand Island、NY、USA)で培養し、NF−κBレポーター(pGL4.32、Promega、Madison、WI、USA)でトランスフェクトした。トランスフェクトした後の24時間後、抗生物質耐性クローンを選択するために、細胞をハイグロマイシンで処理し。NF−κBレポーターを発現するHEK293細胞を96ウェルプレートそれぞれ、2000個の細胞)に播種し、一晩培養した。TNFαタンパク質(5ng)と、aptTNFα(50、500nM)、aptTNFα−PEG(10、50nM)、またはanti−ヒトTNFα抗体(10、50nM、R&D Systems)とをそれぞれ独立したウェルに加えた。4時間または24時間の培養の後、ルシフェラーゼ活性は、製造元のプロトコルに従ってルシフェラーゼアッセイシステム(Promega)によって測定した。データは、陰性対照として処理なしの群(活性0%)および陽性対照としてTNFα処理ありの群(活性100%)を使用して、相対的なルシフェラーゼ活性として表された。
ALIを誘導するために、6週齢のBalb/c雄マウスをLPS(10mg/kg)で気管内処理した。LPS処理の1時間後、aptTNFα(1600μg/kg)またはaptTNFα−PEG(32、320μg/kg)を、気管内または静脈内投与した。血中酸素飽和度は、MouseMonitorTM S plusパルスオキシメータモジュール(Indus Instrumeヌクレオチド、Webster、TX、USA)による処理の24時間後に記録した。ある研究群では、マウスを犠牲にした。肺の重量を測定し、組織をRNAおよびタンパク質の抽出と免疫化学染色に供した。もう一つの群では、気管支肺胞洗浄液(BALF)を収集した。BALFにおける総細胞数をカウントし、BALFにおける総タンパク質濃度をNanodrop分光光度計で定量し、BALFにおけるミエロペルオキシダーゼ(MPO)活性を、製造元のプロトコルに従ってMPO蛍光活性アッセイキット(BioVision)で測定した。
RNAはTrizol(Invitrogen)によってマウスの肝組織から抽出され、cDNAは製造元のプロトコルに従って、ランダムヘキサマー(Invitrogen)を使用してSuperScript III逆転写酵素(Invitrogen)によって合成された。定量的PCRは、LightCycler480システム(Roche Applied Science、Mannheim、Germany)で実行された。qPCRで使用されるマウスcDNAに対するプライマー配列は、以下のように示す:il−1β、5’−agttgacggaccccaaaag−3’(順方向)(配列番号3)および5’−agctggatgctctcatcagg−3’(逆方向)(配列番号4);il−6、5’−gctaccaaactggatataatcagga−3’(順方向)(配列番号5)および5’−ccaggtagctatggtactccagaa−3’(逆方向)(配列番号6);cxcl2、5’−aatcatccaaaagatactgaacaaag−3’(順方向)(配列番号7)および5’−ttctctttggttcttccgttg−3’(逆方向)(配列番号8);actb、5’−ctaaggccaaccgtgaaaag−3’(順方向)(配列番号9)および5’−accagaggcatacagggaca−3’(逆方向)(配列番号10);CCL2、5’−catccacgtgttggctca−3’(順方向)(配列番号11)および5’−gatcatcttgctggtgaatgagt−3’(逆方向);5’−IL17(配列番号12)、5’−cagggagagcttcatctgtgt−3’(順方向)(配列番号13)および5’−gctgagctttgagggatgat−3’(逆方向)(配列番号14);IL23、5’−tccctactaggactcagccaac−3’(順方向)(配列番号15)および5’−agaactcaggctgggcatc−3’(逆方向)(配列番号16);CCND1、5’−tttctttccagagtcatcaagtgt−3’(順方向)(配列番号17)および5’−tgactccagaagggcttcaa−3’(逆方向)(配列番号18);およびPCNA、5’−ctagccatgggcgtgaac−3’(順方向)(配列番号19)および5’−gaatactagtgctaaggtgtctgcatt−3’(逆方向)(配列番号20)。
ウエスタンブロットに使用された一次抗体は、以下のように示す:抗GAPDH(Santa Cruz)および抗PCNA(Cell Signaling Technology、Beverly、MA、USA)。ヘマトキシリンおよびエオシン染色(H&E)は、中央研究院の生物医科学研究所の病理学核心室によって実施された。肺損傷スコアは、動物急性肺損傷研究グループ(Matute−Bello et al.,Am.J.Respir Cell Mol.Biol.2011;44:725−38)によって設計されたスコアシステムに従って計算された。免疫組織化学染色には、抗Ly6G(clone 1A8、Biolegend、San Diego、CA、USA)抗体を1:100で希釈して使用した。ImmPRESS HRP抗ラットIgG、マウス吸着(ペルオキシダーゼ)ポリマー検出キット(Vectors laboratories、Burlingame、CA、USA)を使用してシグナルを増幅し、DABペルオキシダーゼ(HRP)基質キット(Vectors laboratories)を使用して発色させた。
結果は平均値±平均値の標準誤差として示され、P値はスチューデントのt検定によって計算された。両側P値が0.05未満であることを統計的に有意であると定義した。
AptTNFαが高親和性でTNFαに結合し、生体内でTNFαを標的とする
TNFα指向性アプタマー(aptTNFまたはaptTNFα、図2A)は、ニトロセルロースフィルターSELEXによって選択され、FASTAptamerによって分析され、Mfoldを使用して予測された二次構造に基づいて最適化された。aptTNFαおよびヒトTNFαの解離定数(Kd)は、8nMである(図2B、左側)。データは、さらに、aptTNFαもマウスTNFαに結合することを示したため(図2B、右側)、その後、ALIマウスモデルを使用してaptTNFαの生体内結合効果を調査した。
次に、aptTNFαまたはその誘導体がTNFα媒介のシグナル伝達を効果的に阻害するかどうかを研究した。生理活性TNFαが生理的条件において三量体形態で存在するため、2つのaptTNFα単量体(aptTNFα−PEG、図3A)の間にポリエチレングリコール(PEG)リンカーを追加して二量体aptTNFαを合成することで、aptTNFαの潜在的な拮抗作用を増加した。データは、二量体aptTNFα−PEGがヒトおよびマウスTNFαタンパク質に対しても特異的な結合活性を有することを示した(図3B)。
次に、LPS誘発ALIマウスモデルを使用してaptTNFαおよびaptTNFα−PEGの生体内効果を研究した。データは、酸素飽和度の有意な低下に示されるように、LPS治療が呼吸窮迫症を誘発したことを示した(図4A)。LPS処理群における肺の湿重量の増加は、LPS誘発損傷の時に肺胞毛細血管膜の透過性が増強したことを示唆した(図4B)。LPS誘発ALI群の組織学的検査では、肺胞腔には肺胞中隔の肥厚と、赤血球、好中球、およびフィブリン鎖の蓄積の現象に、肺損傷スコアの増加を伴うことを示した(図4C〜4D)。BALFのさらなる分析は、総タンパク質レベルの増加、総細胞数の増加、ミエロペルオキシダーゼ(MPO)活性の増強、および炎症促進性サイトカイン/ケモカイン(il−1β、il−6、およびcxcl2)発現の上方制御を示した(図4E〜4J)。これらの表現型および分子の結果は、予想されているALIに反応してサイトカインおよびケモカインによって調整された組織反応カスケードに符合した。
使用した材料方法は、以下に記載する動物実験を除いて、実施例1と同じであった。
ALFを誘発するために、6週齢のBalb/c雄マウスにD−ガラクトサミン(D−GalN、100mg/kg、腹腔内)およびヒトTNFα(40μg/kg、静脈内)を注射した。次に、静脈内でN−アセチルシステイン(NAC、600mg/kg)、aptTNFα(1600μg/kg)、またはaptTNFα−PEG(3.2、32、320μg/kg)を与える処理をして、処理の6時間後に血液を採取した(Saito et al.,Hepatology.2010;51:246−54;Sass et al., Cytokine.2002;19:115−20)。血液をASTおよびALT分析に使用した(Fuji Dri−Chem 4000i)。処理の6時間または24時間後に、マウスを犠牲にした。RNA、タンパク質抽出、および免疫化学染色のために、肝組織を収集した。
AptTNFαがTNFα媒介の急性肝不全(ALF)を軽減し、早期的な肝再生を促進する
劇症性結果を有するALF患者の場合、肝移植前に現在利用可能な治療法は、N−アセチルシステイン(NAC)の全身注入である(Bernal et al.,N.Engl.J.Med.2013;369:2525−34)。ALFはTNFαが媒介されるため、その後、ALFにおけるaptTNFαおよびaptTNFα−PEGの役割を研究し、D−ガラクトサミン(D−GalN)/TNFα−誘発のマウスALFモデルを使用してそれらの効果をNACと比較した。
生体内分布分析
内因性マウスのTNF誘導について、急性肝不全を誘発するために、6週齢のBalb/c雄マウスにD−GalN(650mg/kg)およびLPS(10μg/kg)を腹腔内注射した。処理の6時間後にマウスを犠牲し、血清および肝臓組織を収集した。生体内分布分析のために、D−GalNおよびLPS処理の0.5時間後に、IRDye(登録商標)800CW標識のaptTNF(Integrated DNA technologies)を静脈注射し、aptTNFから放出された蛍光シグナルをXenogen IVIS Imaging System 200シリーズ(Caliper Life Sciences、Alameda)によって検出した。
生体内でTNFαをモニタリングするための診断剤として機能するAptTNFα
肝組織におけるTNFα濃度が高い患者は、抗TNFα療法に対してより良い反応を示す可能性がある。しかしながら、TNFα濃度の常規的な予測マーカーまたは生体内でTNFαを検出する診断ツールは、未だに存在していない。反応がないものに対する抗TNFα療法の望ましくない副作用を減らすために、生体内でTNFαをモニタリングするための診断剤としてのaptTNFαの実現可能性を研究した。マウスの内因性TNFα分泌および急性肝損傷を誘発するために、マウスにLPSおよびD−GalNを注射した。LPSおよびD−GalN注射の30分後に蛍光標識のaptTNFαを投与した。LPSおよびD−GalNを注射していないが、蛍光標識のaptTNFαを投与したマウスを陰性対照として使用した。陰性対照群と比較して、LPSおよびD−GalN注射群は、AptTNFαが肝組織に有意に蓄積され(図9A〜9C)、ただし、両群で大量のaptTNFαが腎臓濾過から膀胱に***された。

Claims (28)
- GCGCCACTACAGGGGAGCTGCCATTCGAATAGGTGGGCCGC(配列番号1)と少なくとも85%同一の核酸配列を含む、ヒト腫瘍壊死因子α(TNFα)に結合できる核酸アプタマー。
- GCGCCACTACAGGGGAGCTGCCATTCGAATAGGTGGGCCGC(配列番号1)と少なくとも90%同一の核酸配列を含む、請求項1に記載の核酸アプタマー。
- GCGCCACTACAGGGGAGCTGCCATTCGAATAGGTGGGCCGC(配列番号1)と少なくとも95%同一の核酸配列を含む、請求項2に記載の核酸アプタマー。
- GCGCCACTACAGGGGAGCTGCCATTCGAATAGGTGGGCCGC(配列番号1)の核酸配列を含む、請求項3に記載の核酸アプタマー。
- 40〜100個のヌクレオチドを含む、請求項1〜4のいずれか一項に記載の核酸アプタマー。
- GCGCCACTACAGGGGAGCTGCCATTCGAATAGGTGGGCCGC(配列番号1)の核酸配列からなる、請求項4に記載の核酸アプタマー。

- 有効量の請求項1〜10のいずれか一項に記載の核酸アプタマーを必要のある対象に投与することを含む、対象におけるTNFα活性を抑制する方法。
- 前記対象は、TNFαが媒介される疾患に罹患している、罹患の疑いがある、もしくはその危険性があるヒト患者である、請求項13に記載の方法。
- 前記TNFαが媒介される疾患は、関節リウマチ、乾癬、クローン病、急性肝損傷、急性肺損傷、急性呼吸窮迫症候群、眼球乾燥症候群、全身性炎症反応症候群(SIRS)関連脳疾患、喘息、ブドウ膜炎、急性膵炎、急性糸球体損傷、急性腎不全、ANCA関連血管炎、または急性脳疾患である、請求項14に記載の方法。
- 前記対象は、TNFα拮抗剤に関与する治療法を受けた、もしくは受けている、請求項13〜15のいずれか一項に記載の方法。
- 有効量の請求項1〜10のいずれか一項に記載の核酸アプタマーを必要のある対象に投与することを含む、肝損傷を緩和または肝再生を促進する方法。
- 前記対象は、肝疾患関連の肝損傷を有する、請求項17に記載の方法。
- 前記肝疾患は、肝炎、肝硬変症、肝線維症、脂肪性肝疾患、肝癌、または急性肝損傷である、請求項18に記載の方法。
- 前記対象は、前記疾患の急性期にある、請求項17〜19のいずれか一項に記載の方法。
- 前記核酸アプタマーの量は、前記対象における血清アスパラギン酸トランスアミナーゼ(AST)レベル、血清アラニントランスアミナーゼ(ALT)レベル、またはその両者を十分に低下させる量である、請求項13〜20のいずれか一項に記載の方法。
- 前記核酸アプタマーの量は、前記対象における肝臓への好中球浸潤を十分に低下させる量である、請求項13〜20のいずれか一項に記載の方法。
- 前記核酸アプタマーは、気管内で投与する、請求項13〜22のいずれか一項に記載の方法。
- 前記核酸アプタマーは、吸入または皮下注射で投与する、請求項13〜22のいずれか一項に記載の方法。
- TNFαを含んでいる可能性がある生体試料に、請求項11に記載の核酸アプタマーを接触させ、試料におけるTNFαと前記核酸アプタマーとの結合を検査することを含む、試料における腫瘍壊死因子α(TNFα)の存在を検出する方法。
- 有効量の請求項11に記載の核酸アプタマーを必要のある対象に投与し、前記検出可能標識からのシグナルにより、前記核酸アプタマーの位置を検出することを含む、生体内で腫瘍壊死因子α(TNFα)をモニタリングする方法。
- 前記対象は、肝疾患に罹患している、もしくは罹患の疑いがあるヒト患者である、請求項26に記載の方法。
- 前記検出工程は、ヒト患者の肝臓における前記検出可能標識からのシグナルのレベルを測定することで行われる、請求項27に記載の方法。
