さらなる態様では、本発明は、上記で定義した抗体または断片を使用してヒトPLA2−GIBを検出するための方法に関する。この方法は、成熟分泌型(sPLA2−GIB)、プロ型(プロ型sPLA2−GIB)、および/または両方の型のヒトPLA2−GIBタンパク質の存在を検出するために使用または実施することができる。そのような方法または抗体は、試料中のPLA2−GIBの存在または量を検出または投与(または定量化)するため、およびそのプロ型および成熟分泌型の比率を評価するために使用され得る。当該方法は、PLA2−GIBの活性に関連する対象の病態を検出または監視するのに特に有用である。
1−ヒト組換えプロ型sPLA2−GIBおよびsPLA2−GIBタンパク質の産生
ヒトのプロ型sPLA2−GIBおよびsPLA2−GIBを、公開された手順(Singer等、2002;Rouault等、2007)に従って産生した。
最初の一連の免疫を、組換えヒトsPLA2および組換えヒトプロ型sPLA2−GIBでマウスを免疫することにより、行った。2回目の一連の免疫を、ヒトPLA2−GIBプロペプチドに対応する18個のアミノ酸ペプチド(配列NH2−DSGISPRAVWQFRKMIKC−COOH、配列番号62)でマウスを免疫し、その後、成熟活性sPLA2−GIBの最初の11個のアミノ酸(配列番号1のアミノ酸残基16〜33位を参照)で免疫することにより、行った。mAbを、プロテインA親和性により精製し、定量化した。mAbのIg類を、製造元の指示に従って市販のマウスmAbアイソタイピングキットで決定した。
異なるモノクローナル抗体(mAb)によるsPLA2−GIB酵素活性の阻害を、公開された手順(Rouault等、2007)に従って試験した。要するに、組換えヒトsPLA2−GIB(0.1nM)について、ハイブリドーマ細胞培養上清(データを示さない)の量を増やしながら、またはヒトsPLA2−GIBを標的とする精製ウサギポリクローナル免疫血清もしくは精製モノクローナル抗体の量を増やしながら(0.1、0.3、1、および3μg)、37℃で30分間インキュベートした。その後、[3H]−オレイン酸で放射標識し加圧滅菌した大腸菌の膜をsPLA2−GIB活性緩衝液のリン脂質基質として用いて、sPLA2−GIB酵素活性を測定した。300μLの停止緩衝液を加えて反応を停止後、混合物を10000gで5分間遠心分離し、放出された[3H]−オレイン酸を含む上清を数えた。
マイクロプレートウェルを、通常、一晩4℃、pH7.5のPBS中で100ngの組換えヒトsPLA2−GIBまたはプロ型sPLA2−GIBにより、コーティングした。試料ウェルを0.05%のTween(登録商標)20を含むPBSで3回洗浄した。最終洗浄後、試料ウェルをPBS緩衝液中で1%ウシ血清アルブミン(BSA)を含むブロッキング溶液により室温で60分間処理した。0.05%のTween(登録商標)20を含むPBSで洗浄後、ヒトのプロ型sPLA2−GIBまたはsPLA2−GIBに対して、mAbを(10ng/mLから最大3μg/mLまで)増量しながら抗原がコーティングされたウェルに加え、室温で120分間インキュベートした。0.05%のTween(登録商標)20を含むPBSで洗浄した後、HRP共役ヤギ抗マウスIgGまたはIgAポリクローナル抗体試薬でmAbの結合を検出し、比色基質を使用して現像した。溶液中の抗原濃度に対するシグナルの依存性を、Prism(登録商標)ソフトウェア(Graph Pad)を使用した4パラメーターフィット分析で分析し、EC50として報告した。

mAbsの可変領域を、古典的な手順に従って配列化した。要するに、ハイブリドーマ細胞から高純度のRNAを抽出し、高忠実度の逆転写酵素を使用して相補的なDNA鎖を合成した。高忠実度PCRを使用して生成されたPCR産物と、抗体重鎖および軽鎖をコードするcDNAを特異的にターゲットする縮重プライマーを直接配列化(二重鎖配列化)した。cDNA配列を分析し、組み立てた。ペプチド配列を、抗体の構造に応じて翻訳し検証した。
上記のmAbを使用することにより、プロ型sPLA2−GIB、sPLA2−GIBまたは両方の型に特化したサンドイッチELISAを構築した。プロ型sPLA2−GIB検出のため、#8G11mAbを捕捉抗体として使用し、#1C11mAbを曝露mAbとして使用した。sPLA2−GIB検出のため、#14G9mAbを捕捉抗体として使用し、#1C11mAbを曝露mAbとして使用した。両方の型を検出するために、#7A10mAbを捕捉抗体として使用し、#1C11mAbを曝露mAbとして使用した。マイクロプレートの性質、ウェルの最終容量、インキュベートの時間と温度、ストレプトアビジン−HRPの性質と濃度、アッセイ緩衝液の組成、添加した界面活性剤の性質と濃度などのさまざまなパラメータを検討して、アッセイを最適化した。典型的なアッセイ条件は次のとおりである。すなわち、#8G11または#14G9がコーティングされたマイクロプレートを、組換えヒトプロ型sPLA2−GIBまたはsPLA2−GIB標準(アッセイ緩衝液中のタンパク質濃度が様々である)と1時間インキュベートして、較正曲線を作成した。EDTA血漿試料を希釈緩衝液でそれぞれのウェルに希釈(5〜50倍)し、ELISAプレートを室温で1時間インキュベートした。吸引後、ウェルを0.05%のTween(登録商標)20を含むPBSで3回洗浄し、ビオチン化#1C11共役体を室温で30分間ウェルに添加した。0.05%のTween(登録商標)20を含むPBSで洗浄した後、ストレプトアビジン−HRP共役体により室温で15分間処理することにより、mAbの結合を検出した。TMBを加え、反応を停止し、マイクロプレートリーダーで450nmでの吸光度を決定した。Prism(登録商標)ソフトウェア(Graph Pad)を使用して、4パラメーターフィット分析でシグナルを分析した。
年齢、性別、ボディマス指数(Bioreclamation IVT社)が既知の明らかに健康な99人のドナーからなる商業コホートのEDTA血漿試料を、上記で開発した条件によるプロ型sPLA2−GIB、sPLA2−GIB、および両方の型(総PLA2−GIB)を検出する3つの異なるELISAアッセイで試験した。パラメータ間の相関を、ノンパラメトリック検定(R. Spearman)を使用して評価した。
(抗sPLA2−GIB抗体および抗プロ型sPLA2−GIB抗体の生成)
モノクローナル抗体を、組換えヒトプロ型sPLA2−GIB、組換えヒトsPLA2−GIBまたはヒトsPLA2−GIBプロペプチドに対応する18アミノ酸ペプチドおよびその後の成熟活性sPLA2−GIBの最初の11アミノ酸でマウスを免疫することによって産生した。各サブクローンの生成に使用した抗原を表1に示す。
異なるmAbのsPLA2−GIB酵素活性を阻害する能力について、精製モノクローナル抗体を増量させながら、放射標識された膜を組み換えヒトsPLA2−GIBでインキュベートすることによって試験した。結果を、試験した最高濃度のmAbで観察された酵素活性の阻害率として表し、表1に示す。
異なるmAbについて、間接的ELISA法において、ヒトのsPLA2−GIBまたはプロ型sPLA2−GIBによりマイクロプレートをコーティングし、結合したmAbをHRP共役ヤギ抗マウスIgGポリクローナル抗体試薬により検出することによって、比較した。典型的なシグナル曲線の例を図2に示し、それらのプロファイルから推定したEC50を表1に示す。

#14G9、#1C11、#8G11、および#7A10mAbのエピトープマッピングを競合ELISAで評価した。ビオチン化mAbである#14G9、#1C11、#8G11または#7A10を、過剰な非ビオチン化mAbの存在下で、sPLA2−GIBまたはプロ型sPLA2−GIBをコーティングしたプレートに添加した。その後、結合したmAbをストレプトアビジン−HRP共役体で検出した。結果を、自己競合を差し引いた後のシグナルのパーセンテージとして表し、0%は最大競合、100%は競合が無いことを表す(表4)。
本発明の抗体の軽鎖および重鎖の可変領域のアミノ酸および核酸配列を決定し、配列表に提示している。
プロ型sPLA2−GIB、sPLA2−GIBまたは両方の型に特化したサンドイッチELISAを、適合するmAbのペアを用いて開発した(表5を参照)。チモーゲン不活性型酵素を特異的に捕捉するため#8G11mAbを使用し、成熟型酵素を捕捉するため成熟型sPLA2−GIBに非常に特異なmAbとして#14G9サブクローンを使用し、両方の型を捕捉するため#7A10mAbを使用した。高親和性#1C11サブクローンのビオチン化共役体を使用して、捕捉した抗原を検出した。異なるアッセイ条件を最適化し、3つの異なるサンドイッチELISAの分析性能を評価した。それぞれ、プロ型sPLA2−GIB、sPLA2−GIBおよび両方の型に特化したものである。これら3つの異なるアッセイの分析性能の概要を表5に示す。
上記のELISAサンドイッチ法を使用して、25〜59歳の臨床的に適切な年齢範囲内の50人の明らかに健康な男性および49人の明らかに健康な女性からの合計99試料で、年齢、性別、およびボディマス指数(BMI)によるプロ型sPLA2−GIBおよびsPLA2−GIB分布を調べた。この集団の平均(標準偏差SD)年齢は41.5(9.8)歳で、平均(SD)BMIは30.5(7.9)であった。
CD4T細胞中でのIL−7誘導リン酸化STAT5の核移行に対するsPLA2−IB注入の阻害効果を生体内で相殺する#14G9mAbの能力について試験し、マウスに最初、1mgの#14G9mAbを注入し、その後、100μgのsPLA2−GIBを注入した。処置されたマウスの脾細胞から単離されたCD4T細胞におけるリン酸化STAT5の核移行を、その後、共焦点顕微鏡によって分析した。
ヒト化型#14G9の抗原結合性(Fab)断片をその抗原と一緒に共結晶化し、sPLA2−GIBおよびその共結晶のX線回折データをソレイユ シンクロトロン(サン・トーバン、フランス)のPROXIMA−1ビームラインで収集した。結晶は、非対称ユニットごとに2つのsPLA2−GIB/Fab複合体を持つP21空間グループ(a=71.9Å、b=83.8Å、c=103.9Å;α=β=99.58°)に属する。sPLA2−GIB/Fab複合体構造の解明により、複合体における非共有相互作用に関与する重要な残基の同定およびエピトープの定義が可能になった。図5に示すように、sPLA2−GIBにおいて、W3、R6、およびK7は、抗原結合性軽鎖との相互作用に関与し、K10、C77、Y75、G79およびS80は、抗原結合性重鎖との相互作用に関与する。

- 配列番号19の前記軽鎖可変領域のCDR−L1、CDR−L2、およびCDR−L3のそれぞれから成る、または本質的に成るCDR−L1、CDR−L2、および/もしくはCDR−L3を含む軽鎖可変領域、並びに配列番号21の前記重鎖可変境域のCDR−H1、CDR−H2、およびCDR−H3のそれぞれから成る、または本質的に成るCDR−H1、CDR−H2、および/もしくはCDR−H3を含む重鎖可変領域を含む、請求項11に記載のモノクローナル抗体またはその断片。
- 配列番号23を含む軽鎖可変領域および配列番号25を含む重鎖可変領域を含むモノクローナル抗体7A10の分泌および/またはプロ型ヒトPLA2−GIBタンパク質への結合を競合阻害する、請求項11に記載のモノクローナル抗体またはその断片。
- 配列番号23の前記軽鎖可変領域のCDR−L1、CDR−L2、およびCDR−L3のそれぞれから成る、または本質的に成るCDR−L1、CDR−L2、および/もしくはCDR−L3を含む軽鎖可変領域、並びに配列番号25の前記重鎖可変境域のCDR−H1、CDR−H2、およびCDR−H3のそれぞれから成る、または本質的に成るCDR−H1、CDR−H2、および/もしくはCDR−H3を含む重鎖可変領域を含む、請求項13に記載のモノクローナル抗体またはその断片。
- 請求項1〜14のいずれか一項に記載の抗体を含む組成物。
- 請求項1〜14のいずれか一項に記載の抗体、その軽鎖もしくは重鎖、またはその可変ドメインをコードする核酸。
- 試料中のヒトPLA2−GIBタンパク質を検出するための方法であって、
(i)前記試料を請求項1〜14のいずれか一項に記載のモノクローナル抗体と接触させ、
(ii)工程(i)で形成された抗体−抗原複合体の存在を検出する、方法。 - 前記試料を反復して分割し、前記成熟分泌型(sPLA2−GIB)、前記プロ型(プロ型PLA2−GIB)、および/または前記ヒトPLA2−GIBタンパク質の両方の型を別々に検出する、請求項17に記載の方法。
- 前記成熟分泌型(sPLA2−GIB)、前記プロ型(プロ型PLA2−GIB)、および/または前記ヒトPLA2−GIBタンパク質全体の量を投与する、または監視する、請求項17または18に記載の方法。
- 免疫反応もしくは抗体−抗原複合体を実行、検出または定量化するための、請求項1〜14のいずれか一項に記載の抗体の少なくとも1つまたはその断片、および試薬を含むキット。
