しかし、この分野での数十年にわたる研究の後、現在、異なるサブタイプのインフルエンザウイルスを中和する能力を評価するためにわずかな抗体のみが臨床試験中であるが(例えば、Crucell Holland((US2012/0276115、US2014/0065156、US8470327、US2014/0120113、EP2731967、US8691223、US2013/0243792、US2014/0065165、WO2008/028946およびWO2010/130636);Osaka University(US2011/0319600、EP2380976、US2012/0058124、US2012/0058124)、Celltrion(US2013/0004505、EP2545074;WO2014/158001);Vanderbilt University(US2013/0289246)、SeaLane Biotechnologies(US2012/0128671)、Trellis Bioscience,Inc.(US2012/0020971 EP2582721);Visterra,Inc.(US2013/0302349);Burnham Institute/Dana Farber(US2014/011982、EP2222701、WO2010/027818);Temasek(US8444986、US8574581、US 8637644、US8637645、US8383121、US8540996、US8574830、US8540995);HUMABS Biosciences/Institute for Research in Biomedicine(US8871207、US8685402、EP2313433);MedImmune(WO2015/051010);AIMM Therapeutics(WO2013/081463、EP12798020)および Genentech(US2014/0161822)により開発中の抗体を参照のこと)、インフルエンザAウイルスの感染を広く中和もしくは阻害、またはこのウイルスのさまざまなサブタイプにより生じる疾病を弱める市販の抗体はまだない。したがって、インフルエンザウイルス感染を防止または治療するために使用できるインフルエンザAウイルスの複数のサブタイプを中和する新しい抗体を特定するためのニーズが、当技術分野において依然としてある。
(a) 完全ヒトモノクローナル抗体である;
(b) 表面プラズモン共鳴で測定した場合、インフルエンザAグループ2HAに10−8M未満の解離定数(KD)で結合する;
(c) 75分を超える解離半減期(t1/2)を示す;
(d) H3N2株およびH7N9株から選択されたインフルエンザAグループ2ウイルスの、200nM未満および500nM未満のIC50でのそれぞれの中和を示す;
(e) インフルエンザウイルス感染細胞の、約150nM未満のEC50での補体媒介性溶解を示す;
(f) レポーターバイオアッセイを用いたウイルス感染した標的細胞の、約0.9nM未満のEC50での抗体依存性細胞媒介性細胞傷害を示す;
(g) ヒト末梢血単核細胞(PBMC)の存在下でのウイルス感染した標的細胞の、約0.180nM未満のEC50での抗体依存性細胞媒介性細胞傷害を示す;
(h) ウイルス攻撃後、24、48、72、または96時間で投与した場合のインフルエンザ感染動物の生存率の増加を示す;
(i) 感染後96時間でオセルタミビルと組み合わせて投与した場合のインフルエンザ感染動物の生存率の増加を示す;または
(j) 抗体またはその抗原結合性断片は、表1に記載された重鎖可変領域(HCVR)配列のいずれか一つに含まれる三つの重鎖相補性決定領域(CDR)(HCDR1、HCDR2、およびHCDR3)、および表1に記載された軽鎖可変領域(LCVR)配列のいずれか一つに含まれる三つの軽鎖CDR(LCDR1、LCDR2、およびLCDR3)を含む、のうちの二つ以上を有する。
(a) 配列番号4または20からなる群から選択されるアミノ酸配列を有するHCDR1ドメイン;
(b) 配列番号6または22からなる群から選択されるアミノ酸配列を有するHCDR2ドメイン;
(c) 配列番号8または24からなる群から選択されるアミノ酸配列を有するHCDR3ドメイン;
(d) 配列番号12または28からなる群から選択されるアミノ酸配列を有するLCDR1ドメイン;
(e) 配列番号14または30からなる群から選択されるアミノ酸配列を有するLCDR2ドメイン;および
(f) 配列番号16または32からなる群から選択されるアミノ酸配列を有するLCDR3ドメイン;
「インフルエンザヘマグルチニン」という用語はまた「インフルエンザHA」とも呼ばれ、ウイルスの宿主細胞への付着(α−2,3シアル酸またはα−2,6シアル酸へのHA1結合を介して)および侵入(構造変化による)を媒介する、インフルエンザウイルス粒子表面に見られる三量体糖タンパク質である。HAは、二つの構造ドメイン、受容体結合部位を含む球状頭部ドメイン(高頻度の抗原性変異の影響を受けやすい)およびステム領域(インフルエンザウイルスのさまざまな株の間でより保存される)からなる。インフルエンザHAは、前駆体(HA0)として合成され、タンパク質分解処理を受けて、二つのサブユニット(HA1およびHA2)を生成し、これらは互いに関連付けられ、ステム/球状頭部構造を形成する。ウイルスHAは、ウイルス(18のサブタイプは二つのグループに分類可能)で最も変化しやすい抗原であるが、ステム(HA2)は各グループ内で高度に保存されている。
インフルエンザは、オルトミクソウイルス科のRNAウイルス(インフルエンザウイルス)によって引き起こされる感染疾患である。インフルエンザウイルスは、コアタンパク質に基づいて、三つの属A、BおよびCに分類され、さらに、ウイルスエンベロープ糖タンパク質ヘマグルチニン(HA)およびノイラミニダーゼ(NA)によって決定されるサブタイプに分割される。インフルエンザAウイルスは、哺乳類および鳥類のさまざまな種に感染するが、B型およびC型の感染は主にヒトに限定されている。A型およびB型のみが、懸念されるヒト疾患を引き起こす。
別段の具体的な記載がない限り、本明細書で使用する場合、「抗体」という用語は、二つの免疫グロブリン重鎖および二つの免疫グロブリン軽鎖を含む抗体分子(すなわち、「完全抗体分子」)ならびにその抗原結合性断片を包含すると理解される。本明細書で使用される場合、抗体の「抗原結合性部分」、抗体の「抗原結合性断片」などという用語は、抗原に特異的に結合して複合体を形成する、任意の天然に存在する、酵素的に得ることができる、合成の、または遺伝子的に操作されたポリペプチドまたは糖タンパク質を含む。本明細書で使用する場合、抗体の「抗原結合性断片」または「抗体断片」という用語は、本明細書で使用する場合、インフルエンザAグループ2HAに特異的に結合する能力を保有する抗体の一つまたは複数の断片を指す。抗体断片は、Fab断片、F(ab’)2断片、Fv断片、dAb断片、CDRを含有する断片、または単離されたCDRを含みうる。特定の実施形態では、「抗原結合性断片」という用語は、多重特異性抗原結合分子のポリペプチド断片を指す。抗体の抗原結合性断片は、例えば、抗体可変ドメインおよび(任意に)定常ドメインをコードするDNAの操作および発現を伴うタンパク質消化技術または組換え遺伝子操作技術などの任意の適切な標準的技術を用いて、完全抗体分子から得られうる。このようなDNAは公知であり、かつ/または例えば、市販の供給源、DNAライブラリ(例えば、ファージ抗体ライブラリを含む)から容易に入手可能であるか、もしくは合成されうる。DNAを配列決定し、化学的にまたは分子生物学技法を使用することによって操作して、例えば、一つまたは複数の可変ドメインおよび/もしくは定常ドメインを好適な構成に配置するか、またはコドンを導入する、システイン残基を作成する、アミノ酸を修飾、付加、もしくは欠失させるなどが可能である。
トランスジェニックマウスにおいてヒト抗体を生成するための方法は、当該技術分野で知られている。インフルエンザAグループ2HAに特異的に結合するヒト抗体を作製するために、任意のこのような公知の方法が、本発明の状況において使用されうる。以下のいずれか一つを含む免疫原を使用して、インフルエンザAグループ2HAに対する抗体を生成することができる。特定の実施形態では、本発明の抗体は、全長で天然のインフルエンザAグループ2HA(例えば、GenBank登録番号ACF41911.1、またはAAR02640.1を参照)、または弱毒化ウイルスもしくは不活性化ウイルス、またはタンパク質もしくはその断片をコードするDNAを用いて免疫したマウスからから得られる。あるいは、インフルエンザAグループ2HAタンパク質またはその断片は、標準的な生化学技術を使用して、産生され、改変され、免疫原として使用されうる。一実施形態では、免疫原は、組換えによって産生されたインフルエンザAグループ2HAタンパク質またはその断片であってもよい。本発明の特定の実施形態では、免疫原はインフルエンザウイルスワクチンであってもよい。特定の実施形態では、一つまたは複数の追加免疫注射が投与されてもよい。特定の実施形態では、追加免疫注射は、一つまたは複数のインフルエンザウイルス株、またはこれらの株に由来するヘマグルチニン、例えば、A/Hong Kong/08/1968(H3N2)、次いでA/Hong Kong/05/1972−PR8−X36(H3N2)、次いで再度A/Hong Kong/08/1968(H3N2)を含んでもよい。すべてのマウスを、A/Wisconsin/67/X−161/2005(H3N2)およびA/chicken/Netherlands/01/2003(H7N7)からのHAをコードするDNAの1:1混合物を用いて追加免疫した。特定の実施形態では、追加免疫注射は、インフルエンザ株の1:1混合物、または株に由来するヘマグルチニンの1:1混合物、またはHAをコードするDNAを含有しうる。特定の実施形態では、免疫原は、大腸菌、または中国のハムスターの卵巣(CHO)細胞などの他の真核細胞もしくは哺乳類細胞、またはインフルエンザウイルス自体で発現した組換えインフルエンザHAペプチドでありうる。
本発明の抗インフルエンザAグループ2HA抗体および抗体断片は、説明された抗体のアミノ酸配列とは異なるがインフルエンザHAに結合する能力を保有するアミノ酸配列を有するタンパク質を包含する。このようなバリアント抗体および抗体断片は、親配列と比較してアミノ酸の一つまたは複数の付加、欠失、または置換を含むが、説明された抗体の生物学的活性とは本質的に等価である生物学的活性を呈する。同様に、本発明の抗体コードDNA配列は、開示される配列と比較してヌクレオチドの一つまたは複数の付加、欠失、または置換を含むが、本発明の抗体または抗体断片と本質的に生物学的に等価である抗体または抗体断片をコードする配列を包含する。
本発明の特定の実施形態によると、例えば、中性pHと比較して酸性pHで、FcRn受容体への抗体結合を増強または減少させる一つまたは複数の突然変異を含むFcドメインを含む、抗インフルエンザAグループ2HA抗体が提供される。例えば、本発明は、FcドメインのCH2またはCH3領域に変異を含む抗インフルエンザHA抗体を含み、変異は、酸性環境において(例えば、pHが約5.5〜約6.0の範囲のエンドソームにおいて)FcRnに対するFcドメインの親和性を増加させる。そのような突然変異は、動物に投与されたときに抗体の血清半減期の増加をもたらしうる。このようなFc修飾の非限定的な例には、例えば、250位(例えば、EまたはQ)、250位および428位(例えば、LまたはF)、252位(例えば、L/Y/F/WまたはT)、254位(例えば、SまたはT)、および256位(例えば、S/R/Q/E/DまたはT)における修飾、または428位および/もしくは433位(例えば、H/L/R/S/P/QまたはK)および/もしくは434位(例えば、A、W、H、F、またはY[N434A、N434W、N434H、N434F、またはN434Y])における修飾、あるいは250位および/もしくは428位における修飾、あるいは307位もしくは308位(例えば、308F、V308F)、および434位における修飾が含まれる。一実施形態では、修飾は、428L(例えば、M428L)および434S(例えば、N434S)修飾、428L、259I(例えば、V259I)、および308F(例えば、V308F)修飾、433K(例えば、H433K)および434(例えば、434Y)修飾、252、254、および256(例えば、252Y、254T、および256E)修飾、250Qおよび428L修飾(例えば、T250QおよびM428L)、ならびに307および/または308修飾(例えば、308Fおよび/または308P)を含む。さらに別の実施形態では、修飾は、265A(例えば、D265A)および/または297A(例えば、N297A)修飾を含む。
一般に、本発明の抗体は、インフルエンザAグループ2HAに結合することにより機能する。例えば、本発明は、インフルエンザAグループ2HAに(例えば、25oCまたは37oCで)、Biacore測定器での表面プラズモン共鳴、またはリアルタイムバイオレイヤー干渉計に基づくバイオセンサー(Octet HTXアッセイ)によって測定される、10nM未満のKDで結合する抗体および抗原結合性断片を含む。特定の実施形態では、抗体またはその抗原結合性断片は、インフルエンザAグループ2HAに、例えば、本明細書に記載のアッセイフォーマット、または実質的に類似したアッセイを使用して、表面プラズモン共鳴によって測定した、約5nM未満、約2nM未満、約1nM未満、約500pM未満、250pM未満、または100pM未満のKDで結合する。

本発明は、インフルエンザAグループ2HA分子の一つまたは複数のドメイン内に見られる一つまたは複数のアミノ酸と相互作用する、抗インフルエンザAグループ2HA抗体を含む。抗体が結合するエピトープは、インフルエンザHA分子内に位置する3つ以上(例えば、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20以上)のアミノ酸の単一の連続配列からなりうる(例えば、ドメイン内の直線状エピトープ)。あるいは、エピトープは、インフルエンザAグループ2HA分子内に位置する複数の非連続アミノ酸(またはアミノ酸配列)からなりうる(例えば、立体構造エピトープ)。
本発明は、インフルエンザウイルス感染を治療するためのトキソイドまたは抗ウイルス薬などの治療用部分とコンジュゲートした、ヒト抗インフルエンザAグループ2 HAモノクローナル抗体(「免疫コンジュゲート」)を包含する。本明細書で使用される場合、「免疫コンジュゲート」という用語は、放射性薬剤、サイトカイン、インターフェロン、標的もしくはレポーター部分、酵素、ペプチドもしくはタンパク質、または治療剤と化学的または生物学的に連結する抗体を指す。抗体は、その標的に結合できる限り、分子に沿った任意の場所で、放射性薬剤、サイトカイン、インターフェロン、標的またはレポーター部分、酵素、ペプチドまたは治療剤に連結されてもよい。免疫コンジュゲートの例には、抗体薬物コンジュゲートおよび抗体−毒素融合タンパク質が含まれる。一実施形態では、薬剤はインフルエンザ−HAに対する第二の異なる抗体であってもよい。特定の実施形態では、抗体は、ウイルス感染細胞に特異的な薬剤とコンジュゲートされてもよい。抗インフルエンザ−HA抗体にコンジュゲートされうる治療用部分のタイプは、治療される状態および達成される望ましい治療効果を考慮する。免疫コンジュゲートを形成するための適切な薬剤の例は、当該技術分野で公知である;例えば、WO05/103081を参照のこと。
本発明の抗体は、単一特異性、二重特異性、または多重特異性であってもよい。多重特異性抗体は、一つの標的ポリペプチドの異なるエピトープに対して特異的であってもよく、または二つ以上の標的ポリペプチドに特異的な抗原結合ドメインを含有してもよい。例えば、Tutt et al.,1991,J.Immunol.147:60−69;Kufer et al.,2004,Trends Biotechnol.22:238−244を参照のこと。
本発明は、本発明の抗インフルエンザAグループ2HA抗体またはその抗原結合性断片を含む治療用組成物を提供する。本発明による治療用組成物は、移行、送達、耐性などの改善をもたらすために製剤に組み込まれる、適切な担体、賦形剤、および他の薬剤と共に投与される。多数の適切な製剤は、全ての薬学的化学において公知の処方集、Remington’s Pharmaceutical Sciences,Mack Publishing Company,Easton,PAにおいて見出されうる。これらの製剤としては、例えば、粉末、ペースト、軟膏、ゼリー、ワックス、油、脂質、小胞を含有する脂質(カチオン性またはアニオン性)(LIPOFECTIN(商標)など)、DNA共役体、無水吸収ペースト、水中油および油中水乳剤、乳剤カルボワックス(様々な分子量のポリエチレングリコール)、半固体ゲル、ならびにカルボワックスを含有する半固体混合物が挙げられる。Powell et al.“Compendium of excipients for parenteral formulations”PDA(1998)J Pharm Sci Technol 52:238−311も参照されたい。
本発明の抗体は、インフルエンザウイルス感染に関連する疾患または障害または状態の治療、および/または防止、ならびに/またはかかる疾患、障害または状態に関連する少なくとも一つの症状を改善するために有用である。
併用療法は、本発明の抗インフルエンザAグループ2HA抗体、および本発明の抗体または本発明の抗体の生物学的に活性な断片と有利に組み合わせられうる、任意の追加的治療剤を含みうる。本発明の抗体は、インフルエンザウイルスを治療するために使用される一つまたは複数の薬物または薬剤(例えば、抗ウイルス剤)と相乗的に組み合わせられうる。
特定の実施形態によると、本発明の抗インフルエンザAグループ2HA抗体(または抗インフルエンザAグループ2HA抗体と本明細書で言及される追加の治療上活性な薬剤のいずれかとの組合わせを含む医薬組成物)の単回投与を、それを必要とする対象に投与することができる。本発明の特定の実施形態によると、抗インフルエンザAグループ2HA抗体(または抗インフルエンザAグループ2HA抗体と本明細書で言及される追加の治療上活性な薬剤のいずれかとの組合わせを含む医薬組成物)の複数回用量を、対象に定義された時間コースにわたって投与することができる。本発明の本態様による方法は、本発明の抗インフルエンザAグループ2HA抗体の複数回用量を対象に逐次的に投与することを含む。本明細書で使用される場合、「逐次的に投与する」とは、抗インフルエンザAグループ2HA抗体の各用量が、例えば所定の間隔(例えば、時間、日、週または月)をあけた異なる日など、異なる時点で対象に投与されることを意味する。本発明は、抗インフルエンザAグループ2HA抗体の単一の初回用量、次いで抗インフルエンザAグループ2HA抗体の一つまたは複数の二次用量を、および任意選択で、その後、抗インフルエンザAグループ2HA抗体の一つまたは複数の三次用量を、患者に逐次的に投与することを含む方法を含む。
本発明の抗インフルエンザAグループ2HA抗体は、例えば、診断上の目的のために、試料中のインフルエンザAグループ2HAを検出および/または測定するために使用されうる。いくつかの実施形態は、ウイルス感染などの疾患または障害を検出するアッセイにおける、本発明の一つまたは複数の抗体の使用を企図している。インフルエンザAグループ2HAの例示的な診断アッセイは、例えば、患者から得られた試料を、本発明の抗インフルエンザAグループ2HA抗体と接触させることを含み、抗インフルエンザAグループ2HA抗体は、検出可能な標識またはレポーター分子で標識されるか、またはインフルエンザAグループ2HAを患者試料から選択的に単離するための捕捉リガンドとして使用される。あるいは、標識されていない抗インフルエンザAグループ2HA抗体は、それ自体が検出可能に標識された二次抗体との組み合わせで、診断用途に使用することができる。検出可能な標識またはレポーター分子は、3H、14C、32P、35S、または125Iなどの放射性同位体、フルオレセインイソチオシアネートもしくはローダミンなどの蛍光部分もしくは化学発光部分、またはアルカリホスファターゼ、β−ガラクトシダーゼ、セイヨウワサビペルオキシダーゼ、もしくはルシフェラーゼなどの酵素でありうる。試料中のインフルエンザAグループ2HAを検出または測定するために使用することのできる具体的な例示的アッセイには、酵素結合免疫吸着検定法(ELISA)、ラジオイムノアッセイ(RIA)、および蛍光標識細胞分取(FACS)が含まれる。
以下の実施例は、本発明の方法および組成物をどのように作成し使用するかを当業者へ完全に開示するために提供されており、発明者がその発明として見なすものの範囲を限定することを意図していない。使用した数字(例えば、量、温度等)に対する正確さを確保するために努力がなされているが、一部の実験誤差および偏差を考慮に入れるべきである。別途示さない限り、部は重量部であり、分子量は平均分子量であり、温度は摂氏であり、室温は約25oCであり、圧力は大気圧または大気圧に近い。
インフルエンザAグループ2HAに対するヒト抗体が、ヒトイムノグロブリン重鎖可変領域およびカッパ軽鎖可変領域をコードするDNAを含むVELOCIMMUNE(登録商標)マウスで生成された。マウスはA/Hong Kong/08/1968(H3N2)に続いてA/Hong Kong/05/1972−PR8−X36(H3N2)を用いて免疫し、次いでA/Hong Kong/08/1968(H3N2)を用いて再度免疫された。すべてのマウスを、A/Wisconsin/67/X−161/2005(H3N2)およびA/chicken/Netherlands/01/2003(H7N7)からのHAをコードするDNAの1:1混合物を用いて追加免疫した。抗体の免疫応答を、インフルエンザAグループ2HA特異的イムノアッセイによってモニタリングした。所望の免疫応答が達成されたとき、脾細胞を収集し、マウス骨髄腫細胞と融合させて、それらの生存率を維持し、ハイブリドーマ細胞株を形成した。ハイブリドーマ細胞株をスクリーニングおよび選択して、インフルエンザAグループ2HA特異的抗体を産生する細胞株を識別した。この技術と上記のさまざまな免疫原を使用して、数種のキメラ抗体(すなわち、ヒト可変ドメインとマウス定常ドメインを保有する抗体)が取得された。

ヒトモノクローナル抗インフルエンザAグループ2HA抗体の結合親和性および動力学的定数は、抗原捕捉フォーマットを用いた表面プラズモン共鳴によって決定された。測定はBiacore測定器で行われた。このフォーマットでは、Biacore高密度センサ表面が、モノクローナルマウス抗ヒトFc抗体とのアミンカップリングによって誘導体化され、ヒトFc定常領域で発現した抗インフルエンザAグループ2HA抗体を捕捉した。全てのHAタンパク質の捕捉されたmAbそれぞれに対する関連付けを、標準的な方法を使用してモニタリングした。この実験で利用されたフォールドン(foldon)タンパク質は、以下の表3に示すように、BEI Resources またはInfluenza Reagent Resource(IRR)から得た。結合解離平衡定数(KD)および解離半減期(t1/2)を、動力学的速度定数から以下のように計算した:
H1H14611N2は、グループ2、H3 HAに高親和性で結合する(表3)。平衡解離定数(KD値)は、A/Hiroshima/52/2005のnM範囲が低かった(H3N2;KD=8.22E−09)。H1H14611N2は、グループ1株に結合しなかった。
例示的モノクローナル抗体(mAb)H1H14611N2およびH1H14612N2は、二つの異なるフォーマットを使用して、インビトロで、マイクロ中和アッセイのために選択され、抗体の幅および効力を評価した。
モノクローナル抗体をマイクロ中和アッセイで試験し、幅および効力を評価した。簡潔に述べると、メイディン・ダービー・イヌ腎臓(MDCK)細胞を、96ウェルプレートに6.0×103細胞/ウェルで播種した。ウイルス希釈剤のみをバックグラウンド対照ウェルに添加した。細胞を、5%のCO2、37℃で4〜5時間インキュベートした。モノクローナル抗体を、ウイルス希釈剤で最終濃度の4倍に希釈した。抗体を二連で1:3に希釈した。ウイルスを氷上で解凍し、適切な所定濃度まで希釈した。希釈ウイルスを希釈したmAbに添加した。mAb−ウイルス混合物を、MDCK細胞に直ちに移し、5%のCO2、37℃で72時間インキュベートした。ウイルス対照および非感染細胞対照ウェルも含まれた。4日目に、プレートを1200RPMで3分間遠心分離した。細胞を、100μLのCelTiter−Glo基質を使用して溶解し、発光(Victor X3、PerkinElmer)を使用してATP放出を測定した。非感染対照と比較して生存率を判定した。生存率値を、非線形4PL回帰を用いて分析して、IC50値を決定した(GraphPad Prism)。
簡潔に述べると、メイディン・ダービー・イヌ腎臓(MDCK)細胞を播種し、一晩インキュベートして、翌日80〜100%の培養密度を得た。モノクローナル抗体をウイルス感染培地(VIM)中で、50μg/mLまで希釈し、三連または四連で1:2に希釈した。H5N1 A/Vietnam/1203/2004またはH1N1 A/California/07/2009をVIMで希釈し、希釈抗体に添加し、1時間インキュベートした。その後、試料をMDCKに移し、48時間インキュベートした。インキュベーション後、50μLの上清を新しい96ウェルプレートに移した。希釈したシチメンチョウまたはウマ赤血球を上清に加え、30分間または60分間、室温でインキュベートした。血球凝集力価は、血球凝集を完全に阻害する最後の希釈の逆数として記録された。
結果は、H1H14611N2およびH1H14612N2が、マイクロ中和アッセイにおいて、多数の多様なグループ2インフルエンザAウイルス分離株を中和したことを示した。三つの異なる株のインフルエンザの中和に関して、H1H14611N2およびH1H14612N2のIC50(nM)(A/Aichi/02/1968−PR8−X31(H3N2)、A/Philippines/01/1982(H3N2)およびA/Shanghai/01/2013−PR8(H7N9))が表4に示されており、A/Aichi/02/1968−PR8−X31(H3N2)、A/Phillipines/02/1982(H3N2)、およびA/Shanghai/01/2013(H7N9)を含む、株の用量反応曲線も、それぞれ、図1A、BおよびCに示されている(三角形で示されるH1H14611N2、逆三角形で示されるH1H14612N2、ひし形で示される無関連hIgG1)。血球凝集の読み出しを使用したインフルエンザ株A/Victoria/316/2011(H3N2)に対する追加の中和試験の結果を表5に示す。表5に示されるのは、RBC血球凝集が検出されなかったmAb(nM)の最低濃度である。また、これらの研究には負のhIgG1対照が含まれた。

例示的モノクローナル抗体(mAb)H1H14611N2およびH1H14612N2は、補体依存性細胞傷害(CDC)アッセイのために、インビトロで選択された。簡潔に述べると、アッセイの30時間前に、感染多重度(MOI)が3で、メイディン・ダービー・イヌ腎臓(MDCK)細胞を、A/Aichi/02/1968−PR8−X31(H3N2)に感染させた。標的細胞を、指定された濃度で、指定したmAbと組み合わせた。正常なヒト血清補体(NHSC)を、CDCアッセイ培地中の最終濃度(15%)で三回調製した。CytoTox−Glo(Promega)を使用して、細胞毒性の割合を測定した。洗剤で溶解した対照および血清バックグラウンド対照を使用して、最大溶解値およびバックグラウンド溶解値を決定した。図2に示すのは、H1H14611N2(三角形)、H1H14612N2(逆三角形)、とともに対照としての無関連なhIgG1(ひし形)である。特異的溶解の割合を、((mAb+NHSCによる溶解)−(NHSCのみによる溶解))/((洗剤の最大溶解)−(NHSCのみによる溶解))×100として計算した。
H1H14611N2およびH1H14612N2は、補体保存ヒト血清の存在下で、A/Aichi/02/1968−PR8−X31(H3N2)に感染したMDCKをCDCアッセイで試験した場合、ウイルス感染細胞を特異的に溶解する能力において用量依存性増加を示した。三つのうち一つの代表的なCDCアッセイの結果を図2に示す。この実験では、無関連/陰性のhIgG1対照も使用された。H1H14611N2およびH1H14612N2は、細胞溶解を媒介するのに有効であり、それぞれ最大溶解は78.3%および86.3%、EC50は140nMおよび126nMであった。
例示的モノクローナル抗体、H1H14611N2およびH1H14612N2は、二つの抗体依存性細胞媒介性細胞傷害アッセイにおいて、HA装飾細胞を特異的に溶解する能力を試験するために選択された。
A/Wisconsin/67/200(H3N2)過剰発現3T3細胞を用いたADCC−Gloレポーターバイオアッセイ(Promega)を使用したFcγRIIIA活性化アッセイを最初に使用して、HA装飾細胞を特異的に溶解するH1H14611N2およびH1H14612N2抗体の能力を試験した。標的細胞を、指定された濃度で、指定したmAbと組み合わせた。次いで、標的細胞およびエフェクター細胞を、5:1のE:T比で組み合わせた。図3に示すのは、H1H14611N2(三角形)、H1H14612N2(逆三角形)、とともに対照としての無関連なhIgG1(ひし形)を用いたこのアッセイの結果である。
次いで二つの例示的な抗グループ2HA抗体、H1H14611N2およびH1H14612N2を、ヒトドナーPBMCを用いたADCCアッセイで試験した。PBMCを、ヒトドナーバフィーコートから単離し、使用前に10〜12時間、IL−2(5ng/mL)で刺激した。A/Wisconsin/67/2005(H3N2)過剰発現3T3標的細胞を、指示された濃度で指示されたmAbと組み合わせた。次いで、標的細胞およびエフェクター細胞を、30:1のE:T比で組み合わせた。CytoTox−Glo(Promega)を使用して、細胞毒性の割合を測定した。図4に示すのは、H1H14611N2(丸)、H1H14612N2(逆三角形)、とともに対照としての無関連なhIgG1(ひし形)を用いたこのアッセイの結果である。
H1H14611N2およびH1H14612N2は、両方のADCCアッセイで、HA装飾細胞を特異的に溶解する能力において用量依存性増加を示した。
ヒトにおけるインフルエンザウイルス感染の治療または防止のための改善された標準治療法に対するまだ対処されていない実質的なニーズがある。現在、アダマンタンおよびノイラミニダーゼ阻害剤(NAI)の二つのクラスの薬剤のみが利用可能である。アダマンタン(アマンタジンおよびリマンタジン)は、薬剤耐性株の急速な出現と関連付けられており、インフルエンザ治療にはもはや推奨されていない。オセルタミビルのようなNAI(TAMIFLU(登録商標))は、インフルエンザの治療および防止のための最先端の薬剤であるが、有効性の範囲は限定される:NAIは、抗ウイルス剤が、症状発症から48時間内に投与された場合、治療状況において発熱および疾病症状の持続時間を約一日短縮させることが示されているが、48時間以降に投与された場合、有効性の臨床的証拠はほとんどない。
Claims (30)
- グループ2インフルエンザAヘマグルチニン(HA)に特異的に結合する、単離された組換え抗体またはその抗原結合性断片であって、前記抗体が、以下の特徴:
(a) 完全ヒトモノクローナル抗体である;
(b) 表面プラズモン共鳴で測定した場合、インフルエンザAグループ2HAに10−8M未満の解離定数(KD)で結合する;
(c) 75分を超える解離半減期(t1/2)を示す;
(d) H3N2株およびH7N9株から選択されたインフルエンザAグループ2ウイルスの、200nM未満および500nM未満のIC50でのそれぞれの中和を示す;
(e) インフルエンザウイルス感染細胞の、約150nM未満のEC50での補体媒介性溶解を示す;
(f) レポーターバイオアッセイを用いたウイルス感染した標的細胞の、約0.9nM未満のEC50での抗体依存性細胞媒介性細胞傷害を示す;
(g) ヒト末梢血単核細胞(PBMC)の存在下でのウイルス感染した標的細胞の、約0.180nM未満のEC50での抗体依存性細胞媒介性細胞傷害を示す;
(h) ウイルス攻撃後、24、48、72、または96時間で投与した場合のインフルエンザ感染動物の生存率の増加を示す;
(i) 感染後96時間でオセルタミビルと組み合わせて投与した場合のインフルエンザ感染動物の生存率の増加を示す;または
(j) 前記抗体またはその抗原結合性断片が、
配列番号2または18に記載されるアミノ酸配列を含む、重鎖可変領域(HCVR)内に含有される、三つの重鎖相補性決定領域(CDR)(HCDR1、HCDR2、およびHCDR3)、および
配列番号10または26に記載されるアミノ酸配列を含む、軽鎖可変領域(LCVR)内に含有される、三つの軽鎖CDR(LCDR1、LCDR2、およびLCDR3)を含む、のうちの二つ以上を有する、グループ2インフルエンザAヘマグルチニン(HA)に特異的に結合する、単離された組換え抗体またはその抗原結合性断片。 - インフルエンザウイルスに感染した哺乳類に、感染後3日目に開始して、約7〜15mg/kgの単回静脈内用量として投与した場合、感染後3日目に開始して、感染後7日目まで継続して、約2mg/kgの用量で一日二回を5日間投与する、オセルタミビルの経口投与を受けたインフルエンザウイルス感染哺乳類よりも、高い程度で前記感染から前記哺乳類を保護する、請求項1に記載の単離された抗体。
- 感染後96時間で約15mg/kgの単回投与として、インフルエンザウイルスに感染した哺乳類に投与した場合、前記哺乳類が約100%の生存率を有する、請求項1に記載の単離された抗体。
- 約15mg/kgの単回投与として、インフルエンザウイルスに感染した哺乳類に投与した場合、前記哺乳類が約100%の生存率を有する、請求項1に記載の単離された抗体。
- 前記インフルエンザウイルスに感染した哺乳類に投与する場合、感染後96時間でオセルタミビルを投与すると、前記哺乳類における相加的保護効果を示す、請求項1に記載の単離された抗体。
- インフルエンザウイルスに感染した哺乳類に、オセルタミビルを約2mg/kgの用量で一日二回、5日間の経口投与と組み合わせて、約7mg/kg〜約15mg/kgの範囲の単回静脈内用量として投与する場合に、哺乳類において相加的保護効果を示す、請求項1に記載の単離された抗体。
- 配列番号2および18からなる群から選択されるアミノ酸配列を有するHCVRを含む、請求項1に記載の単離された抗体またはその抗原結合性断片。
- 配列番号10または26からなる群から選択されるアミノ酸配列を有するLCVRを含む、請求項1に記載の単離された抗体またはその抗原結合性断片。
- (a) 配列番号4および20からなる群から選択されるアミノ酸配列を有するHCDR1ドメイン;
(b) 配列番号6および22からなる群から選択されるアミノ酸配列を有するHCDR2ドメイン;
(c) 配列番号8および24からなる群から選択されるアミノ酸配列を有するHCDR3ドメイン;
(d) 配列番号12および28からなる群から選択されるアミノ酸配列を有するLCDR1ドメイン;
(e) 配列番号14および30からなる群から選択されるアミノ酸配列を有するLCDR2ドメイン;
(f) 配列番号16および32からなる群から選択されるアミノ酸配列を有するLCDR3ドメインを含む、請求項1に記載の単離された抗体またはその抗原結合性断片。 - (a) 配列番号4のHCDR1、
(b) 配列番号6のHCDR2、
(c) 配列番号8のHCDR3、
(d) 配列番号12のLCDR1、
(e) 配列番号14のLCDR2、および
(f) 配列番号16のLCDR3を含む、請求項9に記載の単離された抗体またはその抗原結合性断片。 - (a) 配列番号20のHCDR1、
(b) 配列番号22のHCDR2、
(c) 配列番号24のHCDR3、
(d) 配列番号28のLCDR1、
(e) 配列番号30のLCDR2、および
(f) 配列番号32のLCDR3を含む、請求項9に記載の単離された抗体またはその抗原結合性断片。 - 配列番号2/10および18/26からなる群から選択されるHCVR/LCVRアミノ酸配列対を含む、請求項1に記載の単離された抗体または抗原結合性断片。
- 配列番号2および18からなる群から選択されるアミノ酸配列を含むHCVRの前記CDRと、
配列番号10および26からなる群から選択されるアミノ酸配列を含むLCVRの前記CDRと、を含む、インフルエンザHAへの結合をめぐって抗体または抗原結合性断片と競合する単離された抗体またはその抗原結合性断片。 - 配列番号2および18からなる群から選択されるアミノ酸配列を含むHCVRの前記CDRと、
配列番号10および26からなる群から選択されるアミノ酸配列を含むLCVRの前記CDRと、を含む、抗体または抗原結合性断片と同じエピトープに結合する単離された抗体またはその抗原結合性断片。 - 前記抗体が、インフルエンザウイルスの宿主細胞への付着および/または侵入を防止する、請求項1に記載の単離された抗体。
- 請求項1に記載のインフルエンザHAに結合する単離された抗体またはその抗原結合性断片、および薬学的に許容可能な担体または希釈剤を含む、医薬組成物。
- 請求項1に記載の抗体のHCVRまたはLCVRをコードするポリヌクレオチド配列を含む、単離されたポリヌクレオチド分子。
- 請求項17に記載の前記ポリヌクレオチドを含むベクター。
- 請求項18に記載の前記ベクターを発現する細胞。
- それを必要とする対象におけるインフルエンザ感染の少なくとも一つの症状を防止、治療または改善する方法であって、それを必要とする前記対象に、請求項1に記載の抗体もしくは抗原結合性断片、または前記抗体もしくはその抗原結合性断片を含む医薬組成物を投与することを含む、方法。
- 前記少なくとも一つの症状が、熱、咳、体の痛み、鼻漏、息切れ、肺炎および気管支炎からなる群から選択される、請求項20に記載の方法。
- 前記医薬組成物が、それを必要とする前記対象に予防的に投与される、請求項20に記載の方法。
- 前記医薬組成物が、免疫不全個体、高齢者(65歳以上)、医療従事者、および病歴または基礎疾患を有する人からなる群から選択される対象に予防的に投与される、請求項22に記載の方法。
- 前記医薬組成物が、第二の治療剤と組み合わせて投与される、請求項20に記載の方法。
- 前記第二の治療剤が、抗ウイルス薬、抗炎症薬、グループ1またはグループ2のインフルエンザHAに特異的に結合する異なる抗体、インフルエンザ用ワクチン、栄養補助食品、およびインフルエンザ感染を治療するための他の任意の緩和療法からなる群から選択される、請求項24に記載の方法。
- 前記第二の治療剤が、前記抗体またはその抗原結合性断片として異なる投与経路を介して投与される、請求項25に記載の方法。
- 前記第二の治療剤が経口投与される、請求項26に記載の方法。
- 前記抗ウイルス薬がオセルタミビルである、請求項25に記載の方法。
- 前記オセルタミビルが、前記抗体の投与前に、同時に、または投与後に投与される、請求項28に記載の方法。
- 前記医薬組成物が、皮下、静脈内、皮内、筋肉内、鼻腔内、または経口投与される、請求項20に記載の方法。
