た透明で鮮やかな青色溶液を、さらに5分間激しく撹拌し、次いで蒸留水で100ミリリ
ットルの最終容積にした。

淡青色の不溶性銅−亜鉛−アンモニウム錯体を形成させ、これを激しく撹拌しながら懸濁
状態に保った。銅−亜鉛−アンモニウム錯体を可溶化するために、4.0グラムの亜リン
酸を約0.5gの分割量で徐々に加えた。銅−亜鉛−アンモニウム錯体を完全に可溶化し
、かつ溶液のpHを調節するように十分な酸を加えなければならない。得られた透明な淡
青色溶液を、さらに5分間激しく撹拌し、次いで蒸留水で100ミリリットルの最終容積
にした。

いて、銅および銅/亜鉛組成物を希釈するために使用した。美容目的および/または室温
での保存のためにこのゲルを使用することになる場合には、抗菌防腐剤を加えなければな
らない。これらの防腐剤としては、例えば、マンデル酸、グリセロール、ソルビン酸カリ
ウム、フェノキシエタノール、安息香酸ナトリウムを挙げることができる。有効な組合せ
は、ソルビン酸カリウム(KS)と安息香酸ナトリウム(NaB)である。10.0%重
量/容積のKSおよびNaBの別々の保存溶液を調製し、2.0ミリリットルの各生成物
を、激しく混合しながら上記のキサンタンガム−アロエベラ溶液に加え、次いで室温の蒸
留水を加えて200ミリリットルの最終容積にした。酸可溶性銅−アンモニウムまたは銅
−亜鉛−アンモニウム錯体組成物を、例えば、このゲル中に1:100容量/容量に希釈
した。

eus)株(S1)を、トリプトン−大豆寒天(TSA)上で培養し、単一コロニーをR
PMI−1640培地中で37℃にて一晩培養した。S1(S.アウレウス(S.aur
eus))培養物を、新鮮なRPMI−1640培地で1:10に希釈し、0.2ミリリ
ットルを、TSAを含有する9センチメートルのペトリ皿上に広げ、乾燥させた。プロピ

TCC 11828(P.アクネス(P.acnes))を、1%グルコース含有脳心臓

トルのサンプルを、BHI寒天を含有する9センチメートルのペトリ皿上に広げ、乾燥さ
せた。

eus))の増殖阻害において、組成物Cu#1が硫酸銅よりも約4倍効力が高いことが
明らかである。この結果から、酸可溶性銅−アンモニウム錯体の組成物は、驚くべきこと
に、硫酸銅単独よりも抗菌性が高いことがわかる。
増殖に対する、硫酸亜鉛単独の効果とゲルに溶解した組成物Cu#1と併用した硫酸亜鉛
の効果を比較する。硫酸亜鉛単独は、試験したいずれの濃度(50〜200マイクログラ
ム/ミリリットル)でも、S1(S.アウレウス(S.aureus))の増殖に対して
効果を示さなかった。これに対して、組成物Cu#1(100マイクログラム/ミリリッ
トル)を濃度漸増の硫酸亜鉛と併用した場合は、S1(S.アウレウス(S.aureu
s))の増殖に対して、明確な相乗的阻害効果があった。このように、組成物Cu#1単
独は、3mm2のAoIを示したが、50、100、および200マイクログラム/ミリ
リットルの濃度で硫酸亜鉛と併用した場合にはそれぞれ、AoIが13、20、および2
8mm2に増加した。これらの結果から、驚くべきことに、抗菌性の酸可溶性銅−アンモ
ニウム錯体は、単独ではS1(S.アウレウス(S.aureus))に対して抗菌活性
を示さない硫酸亜鉛と相乗的に作用することができることがわかる。これらの結果から、
酸可溶性銅−亜鉛−アンモニウム錯体が特定の生物に対して有利な抗菌活性を有し得るこ
とが示唆される。

20および33mm2のAoIで阻害した。これに対して、ゲル中で硫酸亜鉛と併用する
と、組成物Cu#9およびCu#10により、S1(S.アウレウス(S.aureus
))の増殖阻害が増大し、それぞれ38および57mm2のAoIを示した。これらの結
果は、図2に示す結果を確認および拡張し、酸可溶性銅−アンモニウム錯体と硫酸亜鉛の
間に、S1(S.アウレウス(S.aureus))の増殖に対する明確な相乗的阻害効
果あることが示される。

00マイクログラム/ミリリットル)の硫酸銅溶液も調製した。組成物Cu−Zn#1(
表2を参照のこと)の保存溶液も試験用に1:200に希釈した。すべての稀釈液は蒸留
水とした。

試験した5種のCu#組成物のZoIは同様であったが、組成物の配合は、表1に示すよ
うにかなり異なっていた。Cu#3、Cu#9、およびCu#13は同じZoIを示した
が、銅−アンモニウム錯体を可溶化するために使用した酸は異なっており(それぞれリン
酸、塩酸、および硫酸)、またCu#13は、硫酸銅ではなく塩化銅を含む。Cu#12
(硫酸銅および硫酸)は、P.アクネス(P.acnes)に対して最も大きなZoIを
示したが、Cu#13とは、組成物に使用した銅塩が異なるのみである(表1を参照のこ
と)。
これらの組成物の酸性度が、硫酸銅と比較した場合における、それらの大きな抗菌活性の
原因であることが示唆され得るであろう。しかしながら、表3に示すように、Cu#10
の保存溶液は、硫酸銅の等価溶液よりも高いpHを示すが、それにもかかわらず、P.ア
クネス(P.acnes)の増殖に対して大きな阻害効果を示す。さらに、表3に示す化
合物/組成物のZoIとpHの間のノンパラメトリックなスピアマン相関検定により、相
関係数=−0.056およびp=0.81が得られ(GraphPad Softwar
e in La Jolla,CaliforniaによるPrism 6統計パッケー
ジを使用する)、この実験で試験した組成物のpHと抗菌活性の間に有意な相関がないこ
とが示された。
しか有していないにもかかわらず、最も阻害的な組成物の1つであり、等モル濃度の亜鉛
元素を併せもつ(保存溶液中で1.5グラム/デシリットル)。Cu−Zn#1の印象的
な活性は、銅イオンと亜鉛イオンの間に相乗作用があるという、図2および3に提示する
結果による結論を支持し、この酸可溶性銅−亜鉛−アンモニウム錯体が、P.アクネス(
P.acnes)に対して予想を超える阻害効果を示すことを表す。これらの結果から、
P.アクネス(P.acnes)に強く関連した疾患である座瘡の処置において、組成物
Cu−Zn#1が治療価値を有し得ることも示唆される。さらに、P.アクネス(P.a
cnes)が、「正常な」皮膚のpH範囲であるpH4.5〜5であまり増殖しないこと
が知られており、また酸可溶性銅−亜鉛−アンモニウム錯体Cu−Zn#1がP.アクネ
ス(P.acnes)の増殖を強力に阻害し、防腐剤を含むゲル中のCu−Zn#1の1
%(容量/容量)製剤のpH値が約pH4.6であるので、Cu−Zn#1は、座瘡の処
置に治療価値がある可能性がある。
好気性菌株アシネトバクター・バウマンニ(Acinetobacter bauma
nnii)(A.バウマンニ(A.baumannii))、メチシリン耐性黄色ブドウ
球菌(Staphylococcus aureus)(MRSA)、およびクレブシエ
ラ肺炎桿菌(Klebsiella pneumonia)(K.ニューモニエ(K.p
neumoniae))を、病棟で採取したスワブから単離した。大腸菌(Escher
ichia coli)(NCTC 9001;E.コリ(E.coli))は、患者の
膀胱由来の臨床分離株であり;すべての株は、RPMI−1640中で培養した。マイク
ロプレートアッセイでの2倍希釈が1%容量/容量濃度から開始するように、試験する化
合物/組成物の保存溶液を希釈した。試験する組成物の2倍稀釈液を、96ウェル組織培
養プレートにおいてRPMI−1640中で調製した。細菌培養物を新鮮なRPMIで1
:40に希釈し、組成物に加えた。P.アクネス(P.acnes)を実施例1(寒天プ
レート試験法)に記載するように培養し、サンプルを新鮮なBHIで1:100に希釈し
た。96ウェル組織培養プレート(100マイクロリットル)において、試験する組成物
の2倍希釈液を蒸留水で上記のように調製し、希釈したP.アクネス(P.acnes)
培養物の100マイクロリットルを加えた。次いで、プレートを37℃で24時間(P.
アクネス(P.acnes)のために嫌気性条件下で)インキュベートした。組成物の最
小阻止濃度(MIC)は、細菌増殖が視覚的に観察されない最低濃度として定義された。
1.表4に、表1および2に記載の硫酸銅、硫酸亜鉛、および種々の組成物に対するM
ICの結果を示す。実験#1では、試験した2種の銅ベース組成物(Cu#3およびCu
#9)は、硫酸銅の2倍の活性であった。驚くべきことに、硫酸亜鉛は、銅ベース生成物
のいずれよりも低いP.アクネス(P.acnes)に対するMICを示し、酸可溶性銅
−亜鉛−アンモニウム製剤Cu−Zn#1は、試験した最も活性な組成物であった。Cu
−Zn#1と同量の銅元素を有するが亜鉛元素が1/6であるCu−Zn#3(表2を参
照のこと)の活性は、Cu−Zn#1の1/4であった。
Cは、第1の実験で観察の2倍であったが、Cu#3は依然として2倍の活性であった。
Cu#12およびCu#13は、Cu#3中のリン酸ではなく硫酸を含むにもかかわらず
、Cu#3と同じMICを示し;Cu#13は、Cu#3およびCu#12における硫酸
銅ではなく塩化銅を含んだ。これらの結果から、酸可溶性銅−アンモニウム錯体が硫酸銅
単独よりも概ね有効であることが示される。実験#1におけるように、Cu−Zn#1は
P.アクネス(P.acnes)の増殖に対して最も活性な組成物であり、再びCu−Z
n#3よりも4倍活性であった。

れらの実験結果から、Cu−Zn#1が座瘡に治療効果を有し得ることが確認される。
−Zn錯体を安定化するのに使用した塩酸の量を一定に保持しながら、銅元素と亜鉛元素
の比が様々であるCu−Zn組成物#1〜#5(表2を参照のこと)のMICの結果を示
す。この実験では、組成物Cu#3(実験間の参照として使用)は、硫酸亜鉛と同じよう
に、MIC=50マイクログラム/ミリリットルを示した。以前の実験のように、組成物
Cu−Zn#1は、Cu−Zn#3およびこの実験ではCu−Zn#2よりも、P.アク
ネス(P.acnes)の増殖に対する活性が4倍であった。Cu−Zn#4および#5
は両方とも同等に、活性がCu−Zn#2および#3の半分であり、Cu−Zn#1の1
/8であった。これらの結果から、等モルの銅元素および亜鉛元素を含む組成物Cu−Z
n#1が、試験したすべての中でP.アクネス(P.acnes)の増殖に対して最も活
性があることがわかる。Cu−Zn#2および#3に関する結果から、P.アクネス(P
.acnes)の増殖に対する銅の役割は、亜鉛よりもいくぶん大きいが、等モル等価(
Cu−Zn#1)の場合、一緒になった2種の金属の驚くべき相乗作用により、P.アク
ネス(P.acnes)の増殖に対して、断然最も有効な組成物が生成する。
A.バウマンニ(A.baumannii)、およびK.ニューモニエ(K.pneum
oniae)の増殖に対する硫酸銅ならびに種々のCu#およびCu−Zn#組成物のM
ICの結果を示す。MRSAおよびA.バウマンニ(A.baumannii)は両方と
も、E.コリ(E.coli)およびK.ニューモニエ(K.pneumonia)より
も、硫酸銅、Cu#組成物、およびCu−Zn#組成物の作用に対して明確に感受性があ
り、A.バウマンニ(A.baumannii)は、Cu−Zn#組成物の阻害作用に驚
くべき感受性がある。A.バウマンニ(A.baumannii)が、医療関連感染症(
HAI)の原因となる特に執拗な細菌であるので、組成物Cu−Zn#1による清浄およ
び/または噴霧は、この流行性で処置が困難な微生物の増加から病院および他のヘルスケ
ア施設を解放することに特に有用である可能性がある。
銅よりもCu#組成物の増殖阻害作用に感受性であり、一方A.バウマンニ(A.bau
mannii)およびK.ニューモニエ(K.pneumoniae)に対してはその反
対が成立する。しかしながら、E.コリ(E.coli)は、試験した組成物のすべてに
対して、概ね感受性がMRSAの約1/8であった。
クエン酸(Cu#11;表1に示さず、すなわち沈殿が24時間後に顕著であったので試
験していない)を使用して作製した特定の組成物が、調製の数日以内に沈殿し始めること
に気づいた。極めて類似した組成物を水酸化アンモニウムを使用して作製したところ、こ
れらの組成物(Cu#10aおよびCu#11a)は、驚くべきことに安定であり、調製
の数か月後でも沈殿の兆候を示さなかった。水酸化アンモニウムを使用して3種の組成物
を作製し(詳細については表1を参照のこと)、表6の下部に示すように、MRSA、A
.バウマンニ(A.baumannii)、およびE.コリ(E.coli)に対して試
験した。Cu#3は安定な組成物であり、Cu#3aは同等に安定であり、試験した3種
の菌株に対して活性であった。Cu10aはCu#10と同様の活性を示し、クエン酸を
含むCu#11aは他のCu#組成物と同様に活性であった。これら(および水酸化アン
モニウムで調製した他の組成物−下記を参照のこと)の組成物は安定性が増しているだけ
でなく、二酸化炭素が放出されないので作製するのが迅速かつ容易であり、また水酸化ア
ンモニウムで形成させたCu−/Cu−Zn−アンモニウム錯体は、可溶化に必要な酸が
少なく、そのため、組成物のpHをより高くすることができ、その製剤において利点があ
る。
るが、USA300 MRSA株も、特に皮膚のコミュニティー関連感染症において懸念
が高まっている。カルバペネム耐性K.ニューモニエ(K.pneumoniae)が過
去10年にわたって出現し、ほとんど完全に薬剤耐性の世界中に広がった院内病原体にな
っている。しかしながら、Cu#組成物およびCu−Zn#組成物に対する、試験した菌
株の感受性から(表6)、これらの組成物が、病院、家、および他の建物での清浄、噴霧
等により、これらの生物を除去するのに有用である可能性が示唆される。全体として、A
.バウマンニ(A.baumannii)もCu#組成物にかなり感受性があるので、C
u#3は、清浄および殺菌を行ってこれらの病原性微生物を建物から除去するのに特に有
効な組成物になり得る。
A−431(ATCC)を、37℃で空気雰囲気中5%CO2の加湿インキュベーター
中にて、10%ウシ胎児血清および2mM L−グルタミンを添加したRPMI−164
0培地(完全培地)で培養した。細胞傷害性実験のために、細胞をトリプシン処理し、洗
浄してカウントし、2×104細胞を、完全培地中にて平底96ウェルプレートのウェル
に播種し、3日間培養してコンフルエントな単層を形成させた。試験した生成物は次のと
おりであった:(i)図4に示す硫酸銅、Cu#9、Cu#10、およびCu−Zn#1
、ならびに(ii)図5に示す、防腐剤を含むアロエベラゲルで希釈した組成物Cu#9
およびCu−Zn#1、および以下の2種の座瘡クリーム剤:2%サリチル酸を含むCL
EARASIL(登録商標)(Reckitt Benckiser LLC of B
erkshire Englandの登録商標)超速効作用処置ゲルおよび10%過酸化
ベンゾイルを含むCLEARASIL(登録商標)(Reckitt Benckise
r LLC of Berkshire Englandの登録商標)毎日用透明座瘡処
置クリーム。試験生成物はすべて、完全培地で希釈した後、コンフルエントなA−431
細胞に三つ組で加え、次いでさらに24時間培養し、そこで下記のように、スルホロダミ
ンB(SRB)アッセイを使用して、細胞傷害性/細胞生存率を定量的に評価した。
1時間固定した。水道水で2回洗浄した後、細胞をSRB(1%酢酸中に0.4%w/v
のSRB)で室温にて30分間染色した。水道水で2回洗浄した後、残存する染色剤を1
0mMトリス塩基に溶解し、ウェルの吸光度をDynatech Multiplate
ELISAリーダー上で540nmにて測定した。試験生成物の吸光度を対照の吸光度
で除して100を乗じることによりパーセント細胞生存率を算出した。
図4から、硫酸銅ならびに組成物Cu#9および#10が、ヒト皮膚細胞についてほと
んど同一の細胞傷害性プロファイルを示し、50マイクログラム/ミリリットルの50%
細胞傷害性濃度(CC50)を有することがわかり、銅イオンが3種の生成物の細胞傷害
性効果の原因であることが示唆される。組成物Cu−Zn#1は、銅元素濃度が他の3種
の生成物のわずか37.5%であるにもかかわらず、おそらく銅イオンおよび亜鉛イオン
の両方が存在することにより、他の3種の生成物よりもわずかに細胞傷害性が強く(CC
50=30マイクログラム/ミリリットル)、亜鉛イオンがヒト皮膚細胞に対して銅イオ
ンよりもさらに一層細胞傷害性があること、および/または亜鉛イオンが銅イオンの細胞
傷害性を増強することが示唆される。
11a、#12、#13、ならびにCu−Zn#1および#3など、本明細書に記載の組
成物は、培養中のヒト皮膚細胞に細胞傷害性をもたらす濃度(約30マイクログラム/ミ
リリットル)より十分低い濃度(MIC 1.6〜6.3マイクログラム/ミリリットル
−表6を参照のこと)で、MRSAおよびA.バウマンニ(A.baumannii)な
どの細菌の増殖を阻害することがわかる。Cu−Zn#1はまた、ヒト皮膚細胞に対して
細胞傷害性でない濃度であるMIC=12.5マイクログラム/ミリリットル(表4)で
、P.アクネス(P.acnes)の増殖を阻害する。
9(400マイクログラム/ミリリットルの銅元素)およびCu−Zn#1(150マイ
クログラム/リットルの銅元素および亜鉛元素)を含有するゲル、ならびに2種の市販C
LEARASIL(登録商標)(Reckitt Benckiser LLC of
Berkshire Englandの登録商標)座瘡クリーム剤の効果を示す。一方は
活性成分として10%過酸化ベンゾイルを含有し、他方は2%サリチル酸を含有する、両
方のCLEARASIL(登録商標)(Reckitt Benckiser LLC
of Berkshire Englandの登録商標)製品は、それぞれ0.02%お
よび0.05%のCC50値を有し、約0.5%以上の濃度で100%細胞傷害性(0%
の細胞生存率)を示す同様の細胞傷害性プロファイルを生成した。座瘡の処置に好ましい
組成物と考えられる組成物Cu−Zn#1を1%含有するゲルは、4%のCC50を示す
が、組成物Cu#9を1%含有するゲルは、このアッセイでは10%容量/容量において
もCC50に達しなかった。これらのゲル製剤のCC50値は、組成物単独で得られたC
C50値と適合する(図4を参照のこと)。したがって、CLEARASIL(登録商標
)(Reckitt Benckiser LLC of Berkshire Eng
landの登録商標)製品は、ゲル製剤のいずれよりもヒト皮膚細胞に対して約200倍
を超える毒性があることが明らかである。
係は注意して解釈する必要があり、これらのインビトロの結果を臨床的な差に変換するこ
とはできないことに留意することは重要である。しかしながら、CLEARASIL(登
録商標)(Reckitt Benckiser LLC of Berkshire
Englandの登録商標)製品中の活性成分であり、座瘡処置製品中に幅広く使用され
ているサリチル酸および過酸化ベンゾイルは、座瘡に罹患する人々の皮膚上で多く曝露さ
れ得る皮膚細胞に特に有毒であるように見える。こうして、Cu−Zn#1製剤を含むゲ
ルは、座瘡罹患者の皮膚への損傷が少ない可能性がある一方、座瘡に関連した細菌である
P.アクネス(P.acnes)に対して極めて活性である(実施例1および2を参照の
こと)。
チャララ・フラキシネア(Chalara fraxinea)分離株196/28(
C.フラキシネア(C.fraxinea))、アスペルギルス・ニガー(Asperg
illus niger)(A.ニガー(A.niger))、およびウドンコ病から分
離した株(Pm;リンゴの葉から分離)を、ポテトデキストロース寒天(PDA)上で室
温(約22℃)にて培養した。真菌増殖に対する組成物の効果を評価するために、滅菌蒸
留水で希釈した試験組成物10マイクロリットルを、12ウェル組織培養プレートのウェ
ルに入れ、各ウェルに1ミリリットルのPDAをピペットで加えた。寒天の全体にわたっ
て均一に試験組成物が分散するようにプレートを撹拌し(蒸留水単独を対照として使用し
た)、次いで、寒天を凝固させた。確立した真菌培養物から真菌の菌糸を含有する寒天プ
ラグ(3mm2)を、メスを使用して注意深く切り取り、12ウェルプレートの各ウェル
の寒天の中心にある穴に挿入し、次いで室温(約22℃)で培養した。真菌の増殖速度は
株に応じて異なるので、実験は、Pmについては3日後、A.ニガー(A.niger)
については3〜4日の適切な間隔で、またC.フラキシネア(C.fraxinea)に
ついては約7日に評価した。真菌増殖に対する組成物の効果を評価するために、真菌の菌
糸の直径を、定規を使用して90°の角度で2回測定し、平均直径をミリメートル単位で
算出した。真菌増殖を視覚的に検出できない場合には、培養物を位相差顕微鏡(40倍)
で観察して、視覚試験で判断されるように増殖がない(NG)ことを確認する。
1.表7に、C.フラキシネア(C.fraxinea)の増殖に対する硫酸銅および
組成物Cu#16の効果を示す。おそらく抗真菌活性を有することが知られている亜リン
酸による、Cu#16中の銅−アンモニウム錯体の可溶化のために(表1を参照のこと)
、組成物Cu#16は、両方の時点で125マイクログラム/ミリリットルの濃度で真菌
増殖を強力に阻害し、硫酸銅よりも真菌の増殖に対して約30倍阻害的であった(3.7
5マイクログラム/ミリリットルのCu#16による結果は、125マイクログラム/ミ
リリットルの硫酸銅による結果に極めて類似していた)。
Zn#1およびCu−Zn#7の効果を示す。組成物Cu−Zn#1は、C.フラキシネ
ア(C.fraxinea)の増殖阻害において組成物Cu−Zn#7ほど有効ではなく
、2種の組成物の製剤は極めて類似しているが(表2)、組成物Cu−Zn#7は、銅−
亜鉛を可溶化する酸として亜リン酸を含有し、Cu−Zn#1は塩酸を含有する。亜リン
酸は、抗真菌活性を有することが知られており、組成物Cu−Zn#7中にそれを使用す
ると、アッセイの7日目および14日目にそれぞれ、Cu−Zn#1よりも3倍から10
倍の活性がある抗真菌組成物が得られる。
ため、Cu−Zn#7の効果を表7の硫酸銅およびCu#16の効果と比較すると、Cu
−Zn#7は、Cu#16よりもC.フラキシネア(C.fraxinea)増殖の阻害
剤としてはわずかに有効性が低いが、硫酸銅よりもかなり有効であることが示される。

真菌増殖を完全に阻害し、C.フラキシネア(C.fraxinea)で見られたように
(表7)、3時点すべてで硫酸銅よりも3〜10倍阻害的であった。
において硫酸銅よりも有効であったことを示す。組成物Cu#16は炭酸水素アンモニウ
ムを含んで作製され、一方Cu#27は水酸化アンモニウムを含有するが、前に示すよう
に(表6)、これは、組成物の抗真菌活性にあまり差を与えない。しかしながら、組成物
Cu#16については、室温で数日後に沈殿が認められるが、Cu#27に関してはこれ
は見られず、組成物を可溶化するために亜リン酸を使用する場合、水酸化アンモニウムは
銅−アンモニウム錯体の調製において驚くほど有利になり得ることが示唆される。Pm増
殖の同等に有効な阻害剤である組成物Cu−Zn#7およびCu−Zn#10は、それぞ
れの場合に亜リン酸で可溶化されるCu−Zn−アンモニウム錯体を生成させるために、
それぞれ炭酸アンモニウムおよび水酸化アンモニウムを使用する点で異なっていた(表2
)。しかしながら、亜リン酸で可溶化されるCu−Zn−アンモニウム錯体を調製するた
めに炭酸アンモニウムを使用する場合には、沈殿は認められなかった。驚くべきことに、
銅元素含量の点からすると、Cu−Zn#組成物は、Cu#組成物よりもPm増殖の有効
な阻害剤であった。したがって、1パーセント稀釈(150マイクログラム/ミリリット
ルの銅)では、Cu#組成物−ただし375マイクログラム/ミリリットルの銅元素−と
同様に、Cu−Zn#組成物は増殖を完全に阻害した。0.3パーセント稀釈(45マイ
クログラム/ミリリットルの銅)でも、Cu−Zn#組成物は依然としてPm増殖を強力
に阻害するが、Cu#組成物は、37.5マイクログラム/ミリリットルではPm増殖に
対してわずかに阻害的であるのみであった。計算によると、Pmの増殖を50%阻害する
のに必要とされる濃度は、Cu−Zn#組成物については約40マイクログラム/ミリリ
ットルであり、Cu#組成物については約170マイクログラム/ミリリットルであった
。したがって、これらの結果から、亜リン酸で可溶化したCu−Zn#組成物は、組成物
を形成させるために使用したアンモニウム塩にかかわらず、亜リン酸で可溶化したCu#
組成物よりもPmの増殖阻害において驚くほど有効であることがわかる。
リネコの木の枝枯れ病の原因である。A.ニガー(A.niger)は、ブドウ、タマネ
ギ、およびピーナッツなどの特定の果物および野菜のクロカビの原因であり、食物の通常
の汚染菌である。ブルメリア(Blumeria)属の真菌は、ブドウの木、ウリ科植物
(Curcubits)、タマネギ、リンゴおよびセイヨウナシの木を含む多くの植物の
葉のウドンコ病の原因となる広汎な植物病原菌である。上記の結果から、亜リン酸で可溶
化した銅−アンモニウムおよび銅−亜鉛−アンモニウムの錯体は、試験した種々の植物病
原真菌の増殖阻害において極めて有効であり、したがって、作物、植物、木、花、および
観賞用低木を保護する農業分野で実用的な用途を有し得ることがわかる。驚くべきことに
、Cu−Zn#7および#10は、Cu#16および#27よりもウドンコ病の増殖阻害
において顕著に有効であり、これらの組成物が特定の植物病原菌に対して有利な抗真菌活
性を有し得ることが示唆される。
よびクリプトコッカス・ネオフォルマンス(Cryptococcus neoform
ans)に関する抗真菌/酵母アッセイ。
カンジダ・アルビカンス(Candida albicans)(C.アルビカンス(
C.albicans))の3種の株を使用した:1−1、3−1、および4−1をそれ
ぞれ、ヒトの血液、膣、および中咽頭から単離した。クリプトコッカス・ネオフォルマン
ス(Cryptococcus neoformans)(C.ネオフォルマンス(C.
neoformans))株CCTP13を使用した。すべての株は、RPMI−164
0培地中で37℃にて培養した。硫酸銅およびCu#組成物は、200マイクログラム/
ミリリットルの高濃度で開始し、一方、Cu−Zn#組成物は、保存溶液(表2)の1%
となる高濃度で開始し、また硫酸亜鉛および亜リン酸は、Cu−Zn#7および#10中
のものと等価な高濃度で開始して(詳細については表11の説明文を参照のこと)、組成
物の2倍希釈液(100マイクロリットル)を96ウェル組織培養プレート中にてRPM
I−1640で調製した。C.アルビカンス(C.albicans)およびC.ネオフ
ォルマンス(C.neoformans)の一晩培養物をそれぞれ、40mM MOPS
緩衝剤を含有する新鮮なRPMIで希釈し、90マイクロリットルをプレート中の組成物
に加えた。レサズリンA(蒸留水中の0.0675%重量/容量溶液の10マイクロリッ
トル)を、増殖の指示薬として加え、培養物を37℃で24時間インキュベートした。化
合物/組成物の最小阻止濃度(MIC)は、レサズリンA指示薬が青色のままである最低
濃度として定義した(生細胞は青色色素をピンク色に変換する)。
表11に、試験した株がすべて、硫酸銅の形態であっても酸可溶性銅−アンモニウム組
成物(Cu#)の形態であっても、銅に驚くほど感受性であり、MRSAおよびA.バウ
マンニ(A.baumannii)で見られたものと同様のMIC(約6マイクログラム
/ミリリットル;表6)を有することを示す。
溶性Cu−Zn−アンモニウム錯体組成物#1および#7は、銅イオン濃度の点からする
と、硫酸銅およびCu#組成物よりもわずかに有効性に勝る、C.アルビカンス(C.a
lbicans)増殖の阻害剤であった(MIC=4.7マイクログラム/ミリリットル
のCu2+元素)。さらに、酵母C.ネオフォルマンス(C.neoformans)は
、銅に対してその形態にかかわらず極めて感受性であったが、C.アルビカンス(C.a
lbicans)株に比べるとCu−Zn#1に対する感受性は低かった。C.アルビカ

フォルマンス(C.neoformans)の増殖も硫酸亜鉛に影響を受けなかった。

極めて感受性であったが、Cu−Zn#1および硫酸亜鉛に対しては感受性がかなり低か
った。C.ネオフォルマンス(C.neoformans)は、HIV/AIDSの人々
には重篤な脳膜炎をもたらすため、銅含有錯体の脳への選択的な送達は、この致死的疾患
の有望な療法になる可能性がある。
組成物Cu#1、および1%容量/容量の組成物Cu#1を含む防腐剤含有アロエベラ
ゲルのサンプル(3ミリリットル)を、7ミリリットルの滅菌プラスチックチューブに入
れ、反復サンプルを以下の4つの条件下で保存した;加湿インキュベーター中で37℃、
戸棚中で室温(約22℃)、低温室中で4℃、および冷凍庫中で−20℃。保存5か月後
、Cu#1および1%Cu#1を含有するゲルの一連のサンプルを放置して室温にした。
ゲルサンプル(5マイクロリットル)およびCu#1のサンプル(蒸留水で1:1または
1:10容量/容量に希釈し、5マイクロリットルをペーパーディスク上に置いた)を、
寒天プレート試験で菌株S1(S.アウレウス(S.aureus))に対して試験し、
37℃で24時間の培養後、実施例1に記載のように、阻止帯の直径を測定した。
組成物Cu#1を1%含有するゲルのサンプルはすべて、S.アウレウス(S.aur
eus)に対して6ミリメートルの阻止帯を与え、広範囲の温度での保存がゲルの抗菌活
性に影響を与えないことが示された。

Claims (10)
- 銅塩と、
亜鉛塩と、
塩基性アンモニウム塩と、
硫酸、塩酸、リン酸、および亜リン酸からなる群から選択される無機酸である少なくとも1種の水溶性酸と、
を含む水溶液を含む抗菌性組成物。 - 前記銅塩が硫酸銅および塩化銅からなる群から選択される、請求項1に記載の抗菌性組成物。
- 前記塩基性アンモニウム塩が、水酸化アンモニウム、炭酸アンモニウム、および炭酸水素アンモニウムからなる群から選択される、請求項1に記載の抗菌性組成物。
- 前記水溶液の溶媒相が、蒸留水、脱イオン水、精製水、濾過水、医薬品グレード水、医療グレード水、および逆浸透水からなる群から選択される、請求項1に記載の抗菌性組成物。
- 前記銅塩が無水である、請求項1に記載の抗菌性組成物。
- 前記銅塩が水和されている、請求項1に記載の抗菌性組成物。
- 前記亜鉛塩が、無水硫酸亜鉛、無水塩化亜鉛、水和硫酸亜鉛、および水和塩化亜鉛からなる群から選択される、請求項1に記載の抗菌性組成物。
- 前記塩基性アンモニウム塩が水に溶解している、請求項1に記載の抗菌性組成物。
- ゲル、軟膏、油、ペースト、薬物、噴霧可能溶液、ドレッシング溶液、灌水溶液、クリーム、石鹸、界面活性剤、洗剤、および泡からなる群から選択される担体をさらに含む、請求項1に記載の抗菌性組成物。
- 前記抗菌性組成物が植物防疫製品である、請求項1に記載の抗菌性組成物。
