Abstract
Description
本発明は、国立衛生研究所により授与されたFA9550−15−1−0517EB016912および空軍科学研究局により授与されたFA9550−15−1−0517のもとに政府の支援を受けてなされた。政府は、本発明に所定の権利を有する。

細胞質遊離Ca2+の濃度([Ca2+]i)を、以前に報告されているように1,2、Fura−2を用いたレシオメトリック蛍光イメージングによりモニタリングした。簡単に述べると、色素を装填した細胞を、IX71顕微鏡(Olympus America、Center Valley、PA)に取り付けられたガラス底チャンバに入れた。チャンバに、以下を含む溶液(単位はmM)を連続的にかん流させた:140のNaCl、5.4のKCl、1.5のMgCl2、2のCaCl2、10のグルコース、および10のHEPES(pH7.3、290〜300mOsm/kg)。Ca2+を含まない条件に対しては、CaCl2を2mMのNa−EGTAで置き換えた。いくつかの実験では、10μMのシクロピアゾン酸(CPA)とともにプレインキュベートすることにより、Ca2+を小胞体(ER)から枯渇させた。高速波長切替器Lambda DG4(Sutter Instruments、Novato、CA)を使用して340nmおよび380nmで交互に、Fura−2を励起した。iXon Ultra897背面照射CCDカメラ(Andor Technology、Belfast、UK)を用いて、発光を510nmで測定した。Metafluor v.7.5(Molecular Devices、Sunnyvale、CA)を用いて、[Ca2+]iをFura−2発光比から計算した。
この実施例では、CANCAN概念の実現可能性をさらに試験した。独立した2対のnsEP送達電極を使用して、アガロースゲルに埋め込まれたCHO−K1細胞の透過化を測定した。2つのnsEPを同期および重畳させると、各刺激電極対から離れた領域に、単極性パルスのそれに等しい、透過化の増強が惹起されたことが判明した。したがって、独立した2つのnsEPを重畳させることにより生物学的に有効な単極性パルスを遠隔的に作り出し、CANCANが成功したことを表す概念実証が初めて示された。この技術の最適化は、非侵襲的な深部組織のエレクトロポレーションまたは電気刺激に対して多くの意味を有する。
細胞系および培地
チャイニーズハムスター卵巣(CHO−K1)細胞をAmerican Type Culture Collection(ATCC、Manassas、VA)から購入した。細胞を、10%ウシ胎仔血清(Atlanta Biologicals、Flowery Branch、GA)、100IU/mLペニシリン、および0.1μg/mLストレプトマイシン(Gibco Laboratories、Gaithersburg、MD)を補充したF−12K培地(Mediatech Cellgro、Herndon、VA)中で培養した。
実験日に、既述の方法(55)と同様に、細胞をアガロースゲル3次元(3D)培地に埋め込んだ。簡単に述べると、7mLのF−12K増殖培地中2%低ゲル化温度アガロース(Sigma−Aldrich、St Louis、MO)で60mm皿の底部をコーティングした。細胞を回収し、増殖培地中0.75%アガロース中に、5x106細胞/mLの濃度で再懸濁させ、この懸濁液4mLを60mm皿内の2%アガロースベース層上に滴下して堆積させた。皿を4℃で5分間インキュベートしてアガロースゼリー化を促進するとともに細胞沈降を防止し、次いで、nsEP暴露前に少なくとも30分間、インキュベータに移した。nsEP暴露の5分前に、YO−PRO−1ヨウ化物(YP;PBS中1μM;Thermo Fisher Scientific、Waltham、MA)を各皿に添加し、37℃でインキュベートして、色素をアガロースゲル全体にわたって平衡化させた。
アガロース3D培地に埋め込まれた細胞へのnsEP送達は、線形アレイに配列された2対のステンレス鋼ニードル電極を使用して達成された(図4;直径1.66mmで電極間隔は2mm)。各電極対を独立したnsEP送達チャネルに接続し、電極1、2はチャネル1に接続し、電極3、4はチャネル2に接続した(図4C)。線形アレイの中央に位置する電極2および3はアクティブ(a)とし、周囲の電極1および4は接地(g)とした。細胞を含むアガロースゲル中への電極の正確かつ安定した挿入を可能にするために、線形アレイを、マイクロマニピュレータ上に取り付けた。
実験条件に合致する3Dモデルを、市販の有限要素法ソルバCOMSOL Multiphysics(登録商標)、Release5.0(COMSOL Inc、Stockholm、Sweden)を使用して実施した。
ここで、Vは、電場E=−∇Vおよび電流J=σEを計算するために使用される電位であり、σは培地の導電率である。静電条件下では、培地の分散特性は無視した。
nsEP暴露後、皿を15分間カバーしたままにし、次いでPBSで5回洗浄して、すべてのYPを除去した。Hamamatsu C9100EM−CCDカメラ(浜松、静岡県、日本)および0.8倍、0.12NA対物レンズを備えたOlympus SZX16蛍光立体顕微鏡(Olympus America、Hamden、CT)を使用して、電気透過化細胞の画像を取得した。X−Cite Series 120Q蛍光光源(Excelitas Technologies Corporation、Waltham、MA)およびGFPフィルタ(ex.460〜490nm/em.510−)を使用して、YP発光を検出した。
双極性および単極性のnsEPの同期化は、エレクトロポレーションの増強を惹起する。
この研究で、本発明者らは、2つの生物学的に無効な双極性nsEPを重畳させて生物学的に有効な単極性パルスとすることによる遠隔エレクトロポレーションを初めて示す。相殺の相殺またはCANCANと称されたこの効果は、各双極性nsEPの同時発生位相の間に生成される電場が反対方向であり、互いに無効化するときに発生し、電極から離れた領域における単極性暴露のみを残す一方で、他所では双極性のままである。結果的に、CANCANは、標的化エレクトロポレーションのための双極性nsEPの固有の非効率性に依存する。独立した2つのnsEPの送達を同期化することで、非同期nsEP送達(すなわち、10ms離して送達される)よりも最高で3倍大きなエレクトロポレーションが遠隔的に惹起された。CANCANによるエレクトロポレーションのこの単離された増強は、さまざまなnsEPパラメータを用いた異なる実験セットにおいて再現可能に観察された。したがって、本発明者らは、標的化された遠隔エレクトロポレーションを惹起するCANCAN概念のための概念実証を提示する。

Claims (29)
- 生物学的に有効な単極性ナノ秒電気パルスを生成する方法であって、第1の電極対から生成された第1の生物学的に無効な双極性ナノ秒電気パルスと、第2の電極対から生成された第2の生物学的に無効な双極性ナノ秒電気パルスとを重畳させて、前記第1および第2の電極対から離れた位置に前記生物学的に有効な単極性ナノ秒電気パルスを作り出すことを含む、方法。
- 前記生物学的に有効な単極性ナノ秒電気パルスは、前記重畳ステップの不在下で生成される前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスと比較して、増強された刺激効率を有し、
前記重畳ステップの不在下で生成される前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスは、前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスの第2の位相が前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスの第1の位相の刺激効果を相殺または低減することによって惹起される相殺効果を誘発し、
前記生物学的に有効な単極性ナノ秒電気パルスの前記増強された刺激効率は、前記相殺効果を相殺または低減することによって惹起される、請求項1に記載の方法。 - 前記第1および第2の電極対は、各々、独立したナノ秒電気パルス送達チャネルに接続される、請求項1または2に記載の方法。
- 前記第1および第2の電極対は、各々、線形アレイ内に整列される、請求項1〜3のいずれかに記載の方法。
- 生物学的に有効な単極性ナノ秒電気パルスを生成する方法であって、第1の刺激から生成された第1の生物学的に無効な双極性ナノ秒電気パルスと、第2の刺激から生成された第2の生物学的に無効な双極性ナノ秒電気パルスとを重畳させて、前記第1および第2の刺激から離れた位置に前記生物学的に有効な単極性ナノ秒電気パルスを作り出すことを含む、方法。
- 前記第1の刺激は第1の電極対を含み、かつ/または前記第2の刺激は第2の電極対を含む、請求項5に記載の方法。
- 前記単極性ナノ秒電気パルスは、対象者に非侵襲的に送達される、請求項1〜6のいずれかに記載の方法。
- 前記単極性ナノ秒電気パルスは、前記対象者の局在細胞、組織または器官に非侵襲的に送達される、請求項7に記載の方法。
- 前記組織は、深部組織である、請求項8に記載の方法。
- 前記単極性ナノ秒電気パルスは、試料に送達される、請求項1〜9のいずれかに記載の方法。
- 前記試料は、細胞を含む、請求項10に記載の方法。
- 前記生物学的に有効な単極性ナノ秒電気パルスの前記増強された刺激効率は、前記相殺効果の相殺または低減の程度に方向的に比例する、請求項2〜4および16〜19のいずれかに記載の方法。
- 前記第1および/または第2の生物学的に無効な双極性ナノ秒電気パルスは、二相または三相である、請求項1〜12のいずれかに記載の方法。
- 前記第1および/または第2の生物学的に無効な双極性ナノ秒電気パルスは、少なくとも第1の位相および第2の位相を有し、前記第2の位相の振幅は前記第1の位相の25%〜100%である、請求項1〜13のいずれかに記載の方法。
- 前記重畳ステップは、局所的に重畳させることを含む、請求項1〜14のいずれかに記載の方法。
- 前記生物学的に有効な単極性ナノ秒電気パルスは、前記重畳ステップの不在下で生成される前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスと比較して、増強された刺激効率を有し、
前記重畳ステップの不在下で生成される前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスは、前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスの第2の位相が前記第1または第2の生物学的に無効な双極性ナノ秒電気パルスの第1の位相の刺激効果を相殺または低減することによって惹起される相殺効果を誘発し、
前記生物学的に有効な単極性ナノ秒電気パルスの前記増強された刺激効率は、前記相殺効果を相殺または低減することによって惹起される、請求項5または6に記載の方法。 - 前記相殺効果を相殺または低減することは、刺激の極性を反転させるステップを含む、請求項2または16に記載の方法。
- 前記増強された刺激効率は、刺激効率を回復させることを含む、請求項2または16に記載の方法。

- 単極性ナノ秒電気パルス(nsEP)を生成する方法であって、少なくとも第1のnsEP送達チャネルから生成された第1のnsEPと、少なくとも第2のnsEP送達チャネルから生成された第2のnsEPとを重畳させて、前記第1および第2のチャネルから離れた位置に前記単極性nsEPを作り出すことを含む、方法。
- 前記第1および第2のnsEPは各々が双極性である、請求項20に記載の方法。
- 前記第1のnsEPは双極性であり、前記第2のnsEPは単極性である、請求項20に記載の方法。
- 生物学的に有効な単極性ナノ秒電気パルスを作り出すための装置であって、

前記第1および第2の刺激は、前記第1および第2の生物学的に無効なナノ秒電気パルスが重畳されて、前記第1および第2の刺激から離れた位置に前記生物学的に有効な単極性ナノ秒電気パルスを作り出すように構成されている、装置。 - 前記第1の刺激は第1の電極対を含み、かつ/または前記第2の刺激は第2の電極対を含む、請求項23に記載の装置。
- 前記第1および/または第2の電極対は、1対のタングステンロッドを含む、請求項24に記載の装置。
- 前記第1および/または第2の電極対は、少なくとも0.1mmの直径を有する、請求項24または25に記載の装置。
