Abstract
Description
本出願は、米国特許法第119条(e)の下で2017年4月6日に出願の米国特許出願第62/482,644号の優先権を主張し、その全体を参照により本明細書に組み込む。
(a)細胞を初期温度から約−80℃の最終温度にドライアイス(solid carbon dioxide)を使用して冷却すること、または(b)細胞を初期温度から約−196℃の最終温度に液体窒素を使用して冷却することを含む。ある特定の実施形態では、ステップ(iii)が、本明細書に開示された急速凍結方法を使用して達成され得ることが認識される。
本発明の記載および添付の特許請求の範囲に使用される、単数形「1つの(a)」、「1つの(an)」および「この(the)」は、文脈が他に明らかに示さない限り、複数形を同様に含むことが意図される。
以下の略語は、以下に開示される実施例に使用される。
一般に、凍結保存の技術は、等張性緩衝剤の使用に頼っている。このような緩衝剤は、浸透圧効果に起因する水のシフトがないことから、氷晶形成を生成する性向が最も少ないことが公知である。
I.材料および方法
ヒト組織の供給。
インビトロ実験に関して、総肝管、胆管、胆嚢管、胆嚢、および胆膵管膨大部を含むヒト肝臓外胆嚢系を、General Surgery and Organ Transplantation、Sapienza University of Rome、Rome、Italyの「Paride Stefanini」部門の臓器提供者から得た。研究目的のために組織を使用するとのインフォームドコンセントを、発明者らの移植プログラムから得た。全てのサンプルは、19から73歳の間の成人に由来した。インビボ実験に関して、胎児肝臓から単離されたhBTSCsが、利用された。ヒト胎児(16〜22週の在胎月齢)を、婦人科学部門(Sapienza、University of Rome、Italy)の選択的な妊娠終結によって得た。インフォームドコンセントを、中絶前に母親から得た。研究は、Sapienza University Hospitalの地域倫理委員会によって承認された。プロトコールは本発明者らの治験審査委員会の承認を受け、処理手続きは現行のGood Manufacturing Practice(cGMP)に準拠した。研究プロトコールは、Ethic Committee of Umberto I University Hospital、Romeの倫理委員会によって審査および承認された。
組織検体を、以前に記載されているように処理した1、5、6、28〜30。簡潔に述べると、組織を、0.1% Octalbin 20%(Octapharma #5400454)、1nMセレン、抗生物質、300U/mlコラゲナーゼNB1 GMP(Serva #17452.01)、100U/ml Pulmozyme(Roche #18450.02)を添加されたGMP Serum−free Dendritic Cell Medium(CellGro #20801〜0500)で、37℃で30〜45分間頻繁に撹拌して消化した。懸濁液を、800ミクロンの金属性メッシュフィルター(IDEALE ACLRI9 inoxステンレススチール)を通して濾過し、再懸濁前に270gで10分間回転させた。その後、細胞懸濁液を、100および30ミクロン(μ)メッシュフィルターを通して続けて通過させた;次いで、細胞計数をFast−Read 102 (BiosigmaSrl、Venice、Italy)によって行い、細胞生存率をトリパンブルーアッセイ測定による細胞生存率を測定した(全細胞に対する生存細胞の%として表される)。細胞生存率(トリパンブルー排除)は、一貫して95%超であった。
細胞を、製造者(MiltenyiBiotec Inc.、Germany)によって示された通り磁気ビーズを使用することによって上皮細胞接着分子(EpCAM)の発現に関してソートした。簡単に説明すると、EpCAM+細胞を、EpCAMマイクロビーズ(MiltenyiBiotec Inc.、カタログ#130−061−101)で磁気的に標識した。次いで、細胞懸濁液を、MACS Separatorの磁場に配置したMACS LSカラム(Miltenyi Biotec Inc.、カタログ#130−042−401)にロードした。EpCAM+細胞を、1mL当たり300,000個細胞の濃度で基礎培地に懸濁し、最終的な細胞懸濁液として使用した。
将来的な臨床応用に関してcGMP条件でhBTSCsを産生するために、胆嚢を「欧州連合の医薬品管理規則」およびヒト使用に関する医薬品のためのgood manufacturing practicesの欧州ガイドライン(EudraLex − 4巻Good manufacturing practiceガイドライン)に従って処理した。
全ての培地を、滅菌濾過(0.22μmフィルター)し、使用前に暗所で4℃に保持した。RPMI−1640(全ての細胞培養に使用される基礎培地)およびウシ胎児血清(FBS)は、GIBCO/Invitrogen (Carlsbad、CA)から得た。全ての試薬は、他に指定されない限り、Sigma(St. Louis、MO)から得た。増殖因子(注記されるもの以外)は、R&D Systems (Minneapolis、MN)から購入した。
・肝細胞分化(HM)に関するHDMは、MKMに7μg/Lグルカゴン、2g/Lガラクトース、1nMトリヨードサイロニン3(T3)、10ng/mLオンコスタチンM(OSM);10ng/mL上皮細胞増殖因子(EGF)、20ng/mL肝細胞増殖因子(HGF)、および1μMデキサメタゾンを補充することによって調製された4、6。
・胆管細胞分化(CM)に関するHDM:MKMに20ng/mL血管内皮細胞増殖因子(VEGF)165および10ng/mL HGFを補充4、6。
・膵島細胞分化(PM)に関するHDM:MKMにヒドロコルチゾンなしで2% B27、0.1mMアスコルビン酸、0.25μMシクロパミン、1μMレチノイン酸を補充;bFGFを、最初の4日間添加しておき、次いで、50ng/mLエキセンジン−4および20ng/mLのHGFで置き換えた4、5。
細胞を、様々なプラスチック基層から解離させて採取し、凍結保存した。解離させた細胞培養を270gで10分間遠心分離し、凍結保存の溶液の1mLを細胞ペレットに添加した。最終的に細胞を含有する緩衝液を、Nuncバイアル(Unimed #6302598)に移した。これらを、Nalgene Cryo 1℃ Freezing Container(Nalgene、CAT No. 5100−0001)に配置した。使用した凍結保存の方法は、温度を毎分1℃で−80℃に低下させることによってであり;24時間後、細胞を、−196℃で液体窒素に配置した。

HAを、30mLのKM中に懸濁した200mgのヒアルロン酸ナトリウムを使用して調製した。
Nunc(Unimed #6302598)中で凍結した細胞を解凍し、20%ヒト血清由来アルブミンを有する1mLの培養培地を徐々(一滴ずつ)に添加した。次いで、内容物を15mLファルコンチューブに移し;容量をKMで徐々に5mLにし、次いで、270gで10分間の遠心分離に供した2。遠心分離後、上清を除去し、凍結保存に使用されたDMSOを排除した。細胞ペレットを、10%血清を補充したKMでのプレーティングに必要な容量まで再懸濁した1。分析研究には、異なる凍結保存溶液で凍結されていた解凍した細胞の細胞生存率、ならびに多能性幹細胞のマーカーである接着分子(ITGB1、ITGB4、CD44、CDH1)および内胚葉幹細胞のマーカー(PDX1、OCT4、SOX17、SOX2、Nanog)の両方のRT−qPCRの使用による遺伝子発現を評価するものが含まれる。
胆嚢組織検体から得られた、未ソートおよびソートされたEpCAM+細胞(およそ3X105)を、3cm直径プラスチック培養皿に播種し、10%FBSを有するKM中に一晩(約12時間)保持した。その後、細胞培養を無血清KM中に維持し、少なくとも2カ月観察した。hBTSCsのクローン増殖の試験に関して、単一細胞の懸濁液を得て、細胞を、無期限に約36〜40時間毎に自己複製する条件(特に低(2%)酸素条件での場合)で、KM中でクローン接種密度(500/cm2)33で培養プラスチックにプレーティングした。肝芽細胞は、これらの条件下で約5〜7日間のみ継続する(それらは、より長期の生存および増殖のために追加の因子を必要とする)。肝臓、胆嚢系および膵臓の成熟上皮細胞は、無血清KM中で一週を超えて生存しない。
細胞生存度を、トリパンブルー排除アッセイ(Sigma #302 643−25G)により決定した。青色に染色された細胞が死細胞であった;生存細胞は染色されなかった。この色素は、細胞溶液と1:1 v/vで使用された。細胞計数を、FAST−READ 102(Biosigma#BSV100)の使用により実施した。細胞生存率を、細胞の解凍後に直ちに計算した。
解凍した細胞の老化を、X−Gal試験(Sigma #CS0030)34によって決定した。本発明者らは2.6X104細胞/cm2の細胞密度を使用し、細胞を試験前に三日間成長させた。解凍において最高の生存率であることが実証されたものである、Sol1およびSol3中に凍結保存させた細胞を、X−Gal試験でさらに分析した。結果を、凍結保存されていなかった細胞である対照と比較した。対照は、新たに単離された細胞を生成するプロセスを模倣するアッセイのために、培養され、解離させられ、および、次いで、プレーティングさせられた細胞から構成された。
増殖率を、1×104細胞/cm2の密度で6マルチウェルプレートに播種し、7日間培養した、同じhBTSC集団で分析した。細胞計数は、以下の培養条件下で実施された:
・Sol1中で凍結保存されたhBTSCs
・Sol3中で凍結保存されたhBTSCs
・新たに単離されたhBTSCs(凍結保存されなかった)

Nは採取された細胞数であり、Nsは初期のプレーティングした細胞数である。
hBTSCコロニーは、プレーティング後の1から2週の間に出現し始め、光学顕微鏡で10Xで検査することによって容易に同定した。任意のサイズのコロニーを、大きいもので>3,000細胞または小さいもので<200細胞として計数した。8ウェルチャンバースライドの各ウェルを、コロニーについて10X拡大率を使用して評価し、培養の2〜3週間後に計数した。コロニー数、サイズ、および形態学の観察を注記した。解凍における最高の生存率が、緩衝剤Sol1およびSol3中で凍結保存された細胞によって与えられることを考慮して、これらの細胞をさらなるアッセイに供して凍結に対するそれらの応答を評価した19。
RNA抽出を、マウス肝臓またはhBTSC培養物からの組織で実施した。肝内および肝臓外胆嚢系由来細胞培養からの全RNAを、ChomczynskiおよびSacchiI36の手順によって抽出した。本発明者らは、それぞれインビトロおよびインビボデータの参照遺伝子としてGAPDHおよびβアクチンを使用した。
hBTSCsは、培養プラスチックにプレーティング後に無血清クボタ培地(KM)で自己複製期間を経験した。細胞を、KM中に3.8×105細胞/cm2の密度で播種した。培地を、3日毎に交換した。KMで1週間培養した後、培養物を、KM(対照)または肝細胞に適合させたHDMのいずれかに供した。アルブミン分泌実験を、培養のさらに2週間後に実施した。アッセイの全期間、細胞は継代されなかった。
hBTSCsは、培養プラスチックにプレーティング後に無血清KMで自己複製期間を経験した。細胞を、KM中に5.2×105細胞/cm2の密度で播種した。培地を、3日毎に交換した。KMで1週間培養した後、培養物を、KM(対照)または膵島に対する幹細胞の分化に適合させたHDMのいずれかに供した。グルコースチャレンジ実験を、培養のさらに2週間後に実施した。アッセイの全期間について、細胞は継代されなかった。
全ての動物実験は、動物実験に関するEU指令2010/63/EUおよびSapienza施設ガイドラインに基づき実施された。動物実験プロトコールは、Sapienza University of Rome and Umberto I University Hospital of Rome (Prot. 541)の倫理委員会によって審査および承認された。SCIDマウス(T/SOPF NOD.CB17 PRKDC/J)(N=4)は、雄性4週齡動物であり、ヒト細胞の移植に関する宿主として使用された。動物を、麻酔薬(2,2,2−トリブロモエタノール)で鎮静化した。その後、新たに単離されたまたは凍結保存および解凍された2×106個のhBTSCsを、100μl食塩水で懸濁し、脾臓を介して肝臓に注射した。シャム対照は、100μl食塩水のみを注入された。全ての動物を、回復するまで厳重にモニターし、食物および水を自由に与えた。全ての動物プロトコールは、本発明者らの施設ガイドラインを順守した。死亡は発生しなかった。細胞移植の30日後、マウスを屠殺し、さらなる分析のために肝臓を除去した。肝臓サンプルを、遺伝子分析のためにTrizol試薬または病理および免疫組織化学分析のために4%ホルマリンに配置した。血液サンプルを、心臓から採取し、遠心分離し、血清サンプルをELISA(ABCAM #ad108788)によるヒトアルブミンの定量のために−20℃で保存した。
検体を、10%緩衝ホルマリンに2〜4時間固定し、低温度融解パラフィン(55〜57℃)に包埋し、3〜4μm切片を、標準的なプロトコールに従って、ヘマトキシリンエオシンおよびSiriusレッド/Fastグリーンで染色した。IHCに関して、内因性ペルオキシダーゼ活性を、メタノール性過酸化水素(2.5%)中で30分間インキュベーションすることによってブロックした。販売会社によって示された通り、抗原を、プロテイナーゼK(Dako、コードSol3020)で10分間室温で適用することによって回復させた。切片を、次いで、一晩4℃、一次抗体でインキュベートした。
データを、平均±SDとして表した。統計学的分析を、SPSS統計ソフトウェア(SPSS Inc. Chicago IL、USA)によって実施した。非正規分布パラメータに関する群間の差を、Mann−Whitney U検定によって検定した。統計学的な有意性を、p値<0.05に設定した。
凍結保存されたhBTSCsによる生存率、老化およびコロニー形成。
hBTSCsを、基礎対照溶液(KM中の10% DMSO、1.5%ヒトアルブミン)で4〜12週間凍結保存し、次いで、解凍し、プラスチックに10,000細胞/mLの密度で播種した。図1および7は、基礎対照溶液での凍結保存の4週間後のhBTSC培養物の細胞生存率および形態を示す。解凍後、細胞を、クボタ培地(KM)で30日の期間成長させた。hBTSCsは、新たに単離された細胞によって生成されたものに形態学的に類似する細胞コロニーを形成できる(図7)。本発明者らは、様々な凍結保存緩衝剤を試験した。それらの全ては、10%ジメチルスルホキシド(DMSO)を添加された無血清KMから構成され、異なる濃度のヒトアルブミンおよびHA含有することで区別された。生存細胞のパーセントを、凍結保存の4週間後および解凍直後に評価した(N=9)。溶液1(Sol1:0.1% HA+15%組換えヒトアルブミンをさらに補充した)中の細胞は、72.78±5.65%の平均生存率を有していた。溶液3(Sol3:15%組換えヒトアルブミンをさらに補充した)中の細胞は、78.89±6.51%の平均生存率を有していた。Sol1およびSol3は、他の緩衝剤中の細胞よりも有意に高い(p<0.001)解凍後の生存率をもたらした。溶液2A(Sol2A:0.1% HAを補充した)の平均生存率は、53.33±13.23%であり;溶液2B(Sol2B:0.05% HAを補充した)の細胞は、50.56±5.27%であり、および対照溶液(CTRL:1.5%組換えヒトアルブミンを補充した)の細胞は、50.00±6.61%である。Sol1とSol3の間に細胞生存率の有意差は認められなかった(図1A)。
凍結保存が幹細胞表現型に影響するかどうかを評価するために、内胚葉幹細胞によって一般的に発現される中心的な遺伝子の発現を評価した。これらには、多能性遺伝子(OCT4、NANOG、SOX2)および内胚葉転写因子(SOX17、PDX1)が含まれる。これらを、凍結保存の前後1カ月で評価した。興味深いことに、幹細胞遺伝子は、新たに単離された細胞よりもSol1およびSol3で凍結保存されたhBTSCsで、より高度に発現した[SOX2(p<0.05)、PDX1(p<0.05)、NANOG(p<0.01)、SOX17(p<0.05)、およびOCT4(p<0.01);N=5](図2)。
多分化能遺伝子は、自己複製条件下でhBTSCsで発現され、次いで、成熟細胞に分化した際に消失する。出願人らは、Sol1(図3A、4A)、Sol3(データ示さず)対新たに単離された細胞(図3B、4B)での凍結保存後のhBTSCsの培養を試験した。分化条件に関して、出願人らは、成熟肝細胞(HM)、胆管細胞(CM)または膵島(PM)へのhBTSCsの分化を誘導するため特異的に適合させた、異なるホルモン規定培地(HDM)を使用した。ヒドロコルチゾンなしのKMを対照として使用した、というのも、この培地が細胞増殖を許容し、肝臓および膵臓の両方に向かう分化に関して中立的であるからである(糖質コルチコイドは、膵臓分化については回避しなければならない)。凍結保存されたhBTSCs(図3A)ならびに新たに単離された細胞(図3B)は、肝細胞(HM)、膵臓(PM)または胆嚢(CM)運命への幹細胞の分化に適合させたHDMでの培養の二週間後に、多能性遺伝子(NANOG、OCT4、およびSOX2)および内胚葉幹細胞遺伝子(EpCAM、PDX1、およびSOX17)の発現の減少を示した(p<0.05)。hBTSCs(Sol1および新たに単離された)がKMからHMに2週間移された場合に、成熟肝細胞特異的遺伝子の発現の有意な増加が観察された(例えば、アルブミン(Alb);N=5;p<0.01対KM;トランスフェリン(Transf);N=5;p<0.05対KMおよびチトクロムP450 3A4(CYP3A4);N=5;p<0.01対KMを含む)(図4)。同様に、hBTSCs(Sol1および新たに単離された)がPMまたはCMに2週間移された場合に、膵島特異的遺伝子発現(インスリン(Ins)、N=5、p<0.05;グルカゴンN=5、p<0.01 PM対KM)、および大胆管細胞特異的遺伝子発現(セクレチン受容体(SR)、N=5、p<0.01;嚢胞性線維症膜コンダクタンス制御因子(CFTR),N=5、p<0.01;アピカルナトリウム依存性胆汁酸トランスポーター(ASBT)、N=5、p<0.05 CM対KM)(図4)の有意な増加が観察された。HDM中のhBTSCsは、形態および表現型特性の特徴的な変化を発生させた。具体的には、HMで15日後、アルブミン(肝細胞マーカー)を発現する立方状細胞を観察し(図5A)(N=5);CMで15日後、サイトケラチン19(CK19)を発現する細胞のクラスターが出現した(図5A)(N=5);他方で、14日後、PMでのhBTSCsは、コロニーの端部から出芽し、インスリンを発現する細胞を含有する、凝集性細胞の高密度球を産生した(図5A)(N=5)。Sol1、Sol3および新たに単離された細胞の間に有意差は観察されなかった(N=5)。
凍結保存されたhBTSCsが免疫無防備状態マウス(immune−compromised mice)の肝臓に効果的に移植され、増殖することができるかどうかを決定するために、細胞をSCIDマウスの脾臓に移植した。次いで、肝臓を、以前5に記載されている通り細胞移植の30日後、ヒトミトコンドリアに対する抗体での免疫組織化学によって分析した。図6Aに示される通り、凍結保存された(Sol1)hBTSCsは、新たに単離された細胞と同じ効率で肝実質に移植された(N=3;p>0.05)。実際、SCIDマウスの肝実質でのヒトミトコンドリアの発現は、2.626±1.530%および3.722±0.639の宿主の実質細胞塊が、それぞれ、新たなまたは凍結保存されたhBTSCsに由来するヒト細胞を含んでいたことを示した(図6A)。移植された凍結保存hBTSCsのマウス肝臓への効果的な移植および分化を確認するために、本発明者らは、肝臓中のヒトアルブミンmRNAおよび血清中のヒトアルブミン(タンパク質)を測定した。肝臓でのヒトアルブミンmRNAの発現は、新たに単離されたhBTSCs(1.90*10−10±1.09*10−10)で移植されたマウスよりも、凍結保存されたhBTSCs(5.19*10−7±3.06*10−7)で移植されたマウスで有意に高かった(N=3;p<0.01)(図6B)。したがって、同じ動物での、ヒト血清アルブミンレベルは、新たに単離されたhBTSCs(24.13±1.44ng/mL)で移植されたマウスよりも、凍結保存されたhBTSCs(76.39±17.04ng/mL)で移植されたマウスで、有意に高かった(N=3;p<0.0001)(図6C)。
出願人らは、DMSO(10%)、HA(0.1%)および高濃度の組換えヒトアルブミン(15%)を補充した無血清クボタ培地(KM)から構成される、hBTSCsのための、成功を収めた凍結保存プロトコールを確立した。この結論が導かれる鍵となる所見は、以下である:1)hBTSCsは、この凍結保存緩衝剤に供した場合、120日間の凍結保存後の解凍時に、生存し、高い生存率(約80%)を有し得る;2)インビトロでの増殖率(集団倍加時間)およびコロニー形成能力は、HA(0.1%)を有する凍結保存緩衝剤の補充によって改善された;3)高アルブミン濃度±HAを含有する緩衝剤で凍結保存されたhBTSCsは、インビトロで成熟運命(肝細胞、胆管細胞、または機能的な膵臓β細胞)に効率的に分化した;4)高アルブミン濃度+HAを含有する緩衝剤で凍結保存されたhBTSCsは、SCIDマウスに効果的に移植され、移植後にインビボで分化した。

Claims (24)
- ヒト胆嚢系幹/前駆細胞の凍結保存のための方法であって、
(i)ヒト胆嚢系幹/前駆細胞を採取すること;
(ii)(a)脂質を含む基礎培地、(b)約0.05%から0.15%の間の濃度のヒアルロナン(HA)、(c)凍結保護物質、(d)抗酸化剤、および(e)約1%から5%の間の濃度のアルブミンを含む凍結保存溶液を前記細胞に添加すること;ならびに
(iii)前記細胞を初期温度から前記細胞が凍結される最終温度に冷却することを含む方法。 - 前記ヒアルロナンが、約0.1%の濃度である、請求項1に記載の方法。
- 前記凍結保護物質が、場合により約1%から20%の間の濃度の、糖、グリセロール、およびジメチルスルホキシド(DMSO)から選択される、請求項1に記載の方法。
- 前記凍結保護物質が、約10%の濃度のDMSOである、請求項4に記載の方法。
- 前記抗酸化剤は、セレン、ビタミンE、ビタミンC、および還元グルタチオンから選択される、請求項1に記載の方法。

(b)細胞を前記初期温度から約−196℃の前記最終温度に液体窒素を使用して冷却することを含む、請求項1に記載の方法。 - 凍結保存されたヒト胆嚢系幹/前駆細胞を解凍する方法であって:
(i)請求項1に記載の方法に従って凍結保存された細胞を解凍すること;
(ii)血清または血清代用培地を含む第一の緩衝液を添加すること;
(iii)前記細胞を凍結保存培地および前記第一の緩衝液から分離すること;および
(iv)前記細胞を、血清または血清代用培地を含む第二の緩衝液に再懸濁することを含む方法。 - 前記血清がウシ胎児血清である、または前記血清代用培地がGIBCO’s Knockout Serum Replacement Medium、もしくはアルブミン、場合によりヒト血清由来アルブミン、を添加されたクボタ培地である、請求項12に記載の方法。
- 前記血清が、約2%から20%の間、場合により、約10%から20%の間、約10%、または約20%の濃度である、請求項12に記載の方法。
- 前記血清代用培地は、約1%から5%の間の濃度でアルブミン、場合によりヒト血清由来アルブミン、を含む、請求項12に記載の方法。
- 培養培地および/または緩衝液が、解凍緩衝剤を含む、請求項12に記載の方法。
- ステップ(ii)が:
(a)前記細胞を遠心分離すること;

(iv)前記緩衝液を、ヒト胆嚢系幹/前駆細胞の成長および/または分化のために設計された培養培地で置き換えることを含む方法。 - ステップ(ii)が、約6から7時間の間で行われる、請求項18に記載の方法。
- ヒト胆嚢系幹/前駆細胞の成長および/または分化のために設計された前記培養培地は、クボタ培地および/または細胞の分化のためのホルモン規定培地(HDM)(例えば、肝細胞への系統に限定するためのもの、この場合HDM−H)を含む、請求項18に記載の方法。
