手術3日後にElevated Body Swing 試験(The Journal of Neuroscience, July 1995, 15(7): 5372-5378)(改変)を行った(コントロール群 N=4;シリコン微粒子投与群 N=7)。Elevated Body Swing 試験は運動調節機能を反映する行動テストである。ラットの尻尾を掴んで逆さまに吊した状態にすると、健常ラットでは、前肢を地面に伸ばし姿勢を垂直に保持するが、脳機能障害を持つラットでは、体をひねる(スウィングする)。今回の試験では、スウィングの有無のみで評価した。コントロール群では4匹中4匹がスウィングした。シリコン微粒子投与群では7匹中3匹がスウィングした。全評価個体におけるスウィングした個体の割合を図7に示す。脳虚血もしくは再灌流による脳への障害が、シリコン微粒子投与により大きく軽減された。

C−1 サンプル調製
C57BL/6Jマウス(雄、7週齢)を日本SLCより入手した。シリコン微粒子投与群には、上記シリコン微粒子含有食を、コントロール群には、通常の飼料(通常食)(オリエンタル酵母工業株式会社製、型番AIN93M)を各群5匹ずつ1週間与えた。各々のマウスについて、深麻酔下で大腸を摘出し、盲腸、結腸及び直腸の3つに分けた。腸管内包物を取り出した各部位の一部(約2cm)をまとめて、重量を計測した。計測後、粉末状ドライアイスにて急速凍結し、マウス1匹の大腸サンプルとする。1群5匹の総計10匹の凍結大腸サンプルは、サルファーインデックス分析(株式会社ユーグレナ)に用いた。後日同様にサンプル調製を行い、1群5匹の総計10匹の凍結大腸サンプルを調製し、同様にサルファーインデックス分析(株式会社ユーグレナ)に用いた。

C−2 分析前処理
1回目のサンプル調製で得られた同群の凍結マウス大腸サンプル(5つ)を合わせ、内部標準化合物を含むメタノール抽出液を添加し(1 ml /g(臓器))、ペッスルですり潰した。その後、遠心分離を行い、上清100μlをサンプルとした。遠心後のサンプル上清100μlに対して硫黄化合物標識試薬等を添加し(計130μl)、懸濁した。遠心分離した上清(87μl)を遠心型エバポレーターで乾固した。水60μlに再懸濁後遠心分離した上清5μlをサルファーインデックス分析用サンプルとした。2回目のサンプル調製で得られたサンプルも同様に処理しサルファーインデックス分析用サンプルを得た。サルファーインデックス分析に用いたサンプルは、シリコン微粒子投与群は2サンプル(5匹からの混合サンプルが2つ)、コントロール群も2サンプル(5匹からの混合サンプルが2つ)である。

The samples used for the sulfur index analysis were 2 samples in the silicon fine particle administration group (2 mixed samples from 5 animals) and 2 samples in the control group (2 mixed samples from 5 animals).
C−3 サルファーインデックス分析
調製したサンプルに含まれる硫黄化合物は、サルファーインデックスメソッドを用いてLC MSMS 8040(島津製作所製)で分析を行った。具体的には、表1及び表2の測定対象化合物種うち、内部標準化合物(No. 53; Camphorsulfonate)およびチオール基修飾剤(No. 40; Monobromobimane)を除く、全61種の硫黄関連化合物種で相対定量を実施した。相対定量には、得られたマスクロマトグラムのピーク面積(内標化合物で標準化)を用いた。大腸サンプルにおいて計35種の化合物が検出された。検出された硫黄関連化合物データに基づく多変量解析に基づく、サンプル間の類似度のマッピング解析(Rソフトveganパッケージを活用)を行った。
