特許文献3に記載の方法に従って、AAV9由来の変異カプシドタンパク質、AAV3由来のITR配列などを含むrAAVベクターを調製した。詳細は以下の通りである。
(a)ベクターゲノムプラスミドの作成
マウスSPR遺伝子をインサートDNAとしてAAVベクターに組み込み、治療用AAV9−mSPR(配列番号10)を作成した。AAVベクターは、特許文献3に記載される、実験動物において経静脈投与で良好な脳への遺伝子導入が確認されている9型ベクター(AAV9由来の変異カプシドタンパク質、AAV3由来のITR配列などを含む)を用いた。本研究では、広い組織で遺伝子の発現を誘導できるCMVプロモーターの下流にSPR遺伝子を導入した。

(c)ウイルスベクターの生成
以下の手順に従って、塩化セシウムCsClの密度勾配による超遠心を行い、rAAVベクターを精製した。超遠心チューブ内に1.5Mおよび1.25MのCsClを重層し密度勾配を作製した。rAAVベクターを含む細胞破砕溶液を重層後、超遠心(30,000rpm、2.5時間)を行った。屈折率を計測してRI:1.365〜1.380のrAAVベクターを含む画分を回収した。この分画を再度CsCl溶液上に重層し、超遠心(36,000rpm、2.5時間)を行ってrAAVを含む分画を得た。

精製したrAAVの10-2〜10-6の希釈系列を作製した。GFP配列をスタンダードとしたプライマーセット(配列番号11及び12)を使用して、Applied Biosystems 7900HT Fast リアルタイムPCRシステム(Applied Biosystems社)で定量した。

実施例2
マウスへのrAAVベクターの腹腔内投与
生後4週齢の野生型BALB/cマウス(日本クレア社)、及び、BALB/cマウスとの交配を繰り返して遺伝的背景をBALB/cとしたSpr KOマウス(Lexicon Pharmaceuticals社)に対して、実施例1で調製されたrAAVベクター(2.2 x 1013 vg/ml, 5μl)及びPBS(15μl)の混合液、又はコントロールであるPBS(20μl)を腹腔内投与した(いずれも単回投与)。試料投与後のマウスの体重変化及び体長変化を測定した。結果を図2及び3に示す。Spr KOマウスにrAAVベクターを投与することで、投与開始2〜3日後から体重の上昇が確認された。投与開始2週間後には、rAAVベクターを投与したSpr KOマウスの体重及び体長は、野生型マウスと同程度となった。生体内のドーパミン量が増大すると体重増加することが知られている。したがって本実施例の結果は、本発明のrAAVベクターの投与によってマウス体内のSPR及びBH4の量が増大し、チロシンのL−ドーパへの変換が良好に進行して最終的にドーパミン量の増大につながったためであると考えられる。
Example 2

実施例3
マウスにおける総ビオプテリン量の変化
実施例2と同様にして、野生型マウス及びSpr KOマウスに対して実施例1で調製されたrAAVベクター(2.2 x 1013 vg/ml, 5μl)又はコントロールであるPBS(15μl)を腹腔内投与した(いずれも単回投与)。投与してから4週間後、マウスの脳及び肝臓を採取し、中脳、線条体及び肝臓における総プテリジン量を測定した。プテリジン量の測定は、0.1 mM EDTA, 1 mM アスコルビン酸を含む0.2 M過塩素酸溶液でタンパク質成分を除いた組織抽出液を、HPLC-蛍光検出器で分離定量することにより行った。プテリジン類の分離にはInertsil ODS-3カラムを用い、励起波長375nm、蛍光波長465nmにて検出した。総プテリジン量は、生体内におけるBH4の量を反映している。結果を図4に示す。rAAVベクターの投与によって、Spr KOマウスの肝臓における総ビオプテリン量が、野生型マウスの肝臓における総ビオプテリン量と同程度となった。Spr KOマウスの脳における総ビオプテリン量も、rAAVベクターの投与によって増加傾向が確認された。

実施例4
マウスにおけるモノアミン量の変化
実施例2と同様にして、野生型マウス及びSpr KOマウスに対して実施例1で調製されたrAAVベクター(2.2 x 1013 vg/ml, 5μl)又はコントロールであるPBS(15μl)を腹腔内投与した(いずれも単回投与)。投与してから4週間後、マウスの脳を採取し、中脳及び線条体におけるドーパミンの量、並びに中脳におけるセロトニンの量を測定した。モノアミン量の測定は、0.1 mM EDTAを含む0.2 M 過塩素酸溶液を加えてタンパク質成分を除いた組織抽出液を、HPLC-電気化学検出器で分離定量することにより行った。モノアミン類の分離にはEICOMPAK SC-5ODSカラムを用い、750 mVの印加電圧にて測定した。結果を図5に示す。rAAVベクターの投与によって、Spr KOマウスの脳内ドーパミン量及びセロトニン量は顕著に増大した。

実施例5

実施例2と同様にして、野生型マウス及びSpr KOマウスに対して実施例1で調製されたrAAVベクター(2.2 x 1013 vg/ml, 5μl)又はコントロールであるPBS(15μl)を腹腔内投与した(いずれも単回投与)。投与してから4週間後、マウスの脳及び肝臓を採取し、線条体及び肝臓におけるPhe/Tyr値(チロシンの量に対するフェニルアラニンの比率)を測定した。PheとTyrの定量は、過塩素酸でタンパク質成分を除いた組織抽出液を、HPLC-蛍光検出器で分析することにより行なった。PheとTyrの分離にはInertsil ODS-3カラムを用い、励起波長240 nm、蛍光波長287 nmにて検出した。結果を図6に示す。rAAVベクターの投与によって、Spr KOマウスのPhe/Tyr値は顕著に減少した。本発明のrAAVベクターの投与によってマウス体内のSPR及びBH4の量が増大し、フェニルアラニンからチロシンへの変換が良好に進行したためであると考えられる。

実施例6
マウスにおけるSPRの量の変化
実施例2と同様にして、野生型マウス及びSpr KOマウスに対して実施例1で調製されたrAAVベクター(2.2 x 1013 vg/ml, 5μl)又はコントロールであるPBS(15μl)を腹腔内投与した(いずれも単回投与)。投与してから4週間後、マウスの脳、肝臓及び副腎を採取し、肝臓及び副腎におけるSPRの発現を、抗SPR抗体(Ikemoto K. et al., Brain Res 954: 237-46 に記載の方法に従って調製)を使用するウェスタンブロッティングによって定量した。それぞれの組織から組織抽出液を作製しタンパク質量を測定して、一定量のタンパク質(100μg)をSDS−ポリアクリルアミドゲル電気泳動により分離した。タンパク質量は、Bradofordの試薬と組織抽出液の一部を混合し、595 nmの吸光度を測定することにより定量した。分離後にタンパク質をPVDF膜に転写し、目的タンパク質をそれぞれの特異的抗体を用いて検出した。結果を図7に示す。rAAVベクターの投与によってSPRタンパク質の増加傾向が確認された。

参考例
BH4の腹腔内投与
42日齢の野生型マウスおよびSpr−KOマウスに対して、BH4−2HCl(50mg/kg,サントリー)を含むリン酸緩衝液に溶解し、1日2回、11時間から13時間の間隔をあけて腹腔内に7日間連続投与した。対照群にはBH4を含まないPBSをマウスの体重1グラム当たり0.01 mL(1匹あたり約0.1〜0.2 mL)投与した。マウスの体重変化を図8に示す。BH4の投与によってSpr−KOマウスにおいて体重増加が認められたが、1週間後でも野生型の半分程度の体重までにしか回復しなかった。これは、AAV−Sprでは持続的にBH4が増加し続けるのに対して、BH4投与ではBH4が体内で速やかに分解排出されてしまうので、1日2回の投与でも十分な効果が得られないためであると考えられる。
