配列番号1の配列、
配列番号1の配列に対して少なくとも80%の同一性を有する配列、
配列番号1の配列のうち連続する少なくとも500ヌクレオチドを含む配列、及び、
配列番号1の核酸配列又はその相補鎖と中度の厳格条件下でハイブリダイズできる配列、からなる群から選択される配列を含む。
本明細書で用いられる「核酸」又は「ポリヌクレオチド」は、リボ核酸又はデオキシリボ核酸といった任意の長さの高分子型ヌクレオチドを意味する。従って、この用語は、以下のものに限られないが、一本鎖、二本鎖若しくは多重鎖のDNA若しくはRNA、ゲノムDNA、cDNA、DNA−RNAハイブリッド、又はプリン及びピリミジン塩基、若しくは他の天然の、化学的若しくは生化学的修飾を受けた、非天然の、若しくは誘導体化されたヌクレオチド塩基を含むポリマーを含む。ポリヌクレオチドの骨格は、(典型的にRNA又はDNAで見られるように)糖及びリン酸基を含み、又は修飾若しくは置換された糖若しくはリン酸基を含み得る。代替的に、ポリヌクレオチドの骨格は、ホスホロアミダート等の合成サブユニットを含んでもよく、またオリゴデオキシヌクレオシドホスホロアミダート(P−NH2)又はホスホロアミダート−ホスホジエステルオリゴマー混合物であってもよい。本発明の核酸は、化学的合成、組換え及び突然変異誘発を含む当業者に周知のいずれの方法により調製されてもよい。好ましい実施形態に置いて、本発明の核酸は、好ましくは当業者に周知の組換え方法により合成されたDNA分子である。

(動物)
全ての試験を、国立衛生研究所における実験動物の管理と使用のガイドに従って行った。プロトコールは地方動物倫理委員会により承認され、欧州議会の2010/63/EU指令に従って行った。この試験で用いられた全てのマウスは、JanvierLaboratories (Le Genest Saint Isle, France)から入手されたC3H/HeN(rd1マウス)若しくはC57Bl6Jマウス(野生型)であり、又は外国起源のカニクイザル(macaca fasicularis)が用いられた。
組換型AAVをプラスミドコトランスフェクション法(Choi et al. Curr. Protoc. Hum. Genet. 2007;Chapter 12:Unit 12.9)により作製し、それに記載されているように、得られた溶解液をイオジキサノール勾配超遠心法により精製した。簡単に40%イオジキサノール分画を濃縮し、Amicon Ultra-15 Centrifugal Filter Unitsを用いてバッファー置換を行った。その後、ベクターストックを、標準に関連して(Aurnhammer C et al. Hum Gene Ther Methods. 2012 Feb;23(1):18-28)リアルタイムPCRによって、DNase耐性ベクターゲノムに作用した。
マウスをケタミン(50mg/kg)、キシラジン(10mg/kg Rompum)を用いて麻酔した。瞳孔を広げ、超微細な30ゲージの使い捨て針を赤道にあり角膜縁に隣接する強膜に通過させ、硝子体腔内に入れた。107から1011粒子のAAVを含む1μlのストックの注射を、硝子体腔の中心における針の直接観察により行った。霊長類では、10:1mg/kgのケタミン:キシラジンを用いて麻酔した。瞳孔を広げ、1×1011又は5×1011のウィルス粒子を含む100μLのウィルスベクターを、それぞれの眼の硝子体内に、30ゲージの針を用いて角膜縁の約4mm後ろの胸膜を通って注射した。眼科用ステロイド及び化膿止めを注射後の角膜に適用した。
形質導入されたマウスの網膜を、解剖し、4%ホルムアルデヒドにより30分間室温で固定しPBSで洗浄した。その後、網膜を、1%TritonX−100、0.5%Tween 20及び5%ウシ血清アルブミンブロッキングバッファーを含むPBSにより室温で1時間処理した。マウスの網膜を抗GFPポリクローナル抗体(Life Technologies; 1:2000)及び抗Brn3aモノクローナル抗体(MilliporeChemicon; 1:100)を含む半希釈ブロッキングバッファーにより4℃で一晩インキュベートした。その後、網膜をそれぞれAlexa TM594、Alexa TM488及びAlexa TM647とコンジュゲートされた抗ウサギIgG、抗マウスIgG二次抗体を含む半希釈ブロッキングバッファーにより1時間室温でインキュベートした。霊長類の網膜を、類似の条件で、抗Brn3a、抗GFP及び抗チャンネルロドプシン抗体を用いてラベルした。その後、細胞核を、検体と4’,6−ジアミジノ−2−フェニルインドール(Sigma-Aldrich; 10μg/mL)とを共にインキュベートすることにより染色した。網膜をリンスし、共焦点観察を開始する前に、2つのカバーガラスの間にマウンティング媒体を用いてフラットマウンティングした。先にラベルされたフラットマウントの組織切片を、マウントの解除、凍結保存及び凍結切片(15μm)の前にOCTで包埋することにより得て、共焦点顕微鏡により観察した。
共焦点顕微鏡観察は、オリンパスFV1000レーザースキャニング共焦点顕微鏡を用いて行った。画像は、励起及び放射のクロストークを低減するために連続的に、一行ごとに得て、ステップサイズをナイキスト・シャノン標本化定理に従って規定した。最終画像における過飽和な画素を最小化する露光設定を用いた。その後、FIJIを用いて12ビットイメージを処理し、Z断面を、Z−投影機能下での最大強度を用いて単一平面に投影し、最終的に8ビットのRGBカラーモードに変換した。
マウスを麻酔し、迅速な頚椎脱臼によって屠殺し、霊長類には致死量のペントバルビタールを与えた。眼球を取り出し、Amesメディウム(Sigma Aldrich A1420)において、室温で95%O2及び5%CO2で泡立てた。単離された網膜をセルロース膜上に置き、RGCに対する電極を有するMEA(MEA256 100/30 iR-ITO, Multichannel systems,ドイツ)に対して穏やかに押し付けた。網膜を、試験中1〜2ml/minで34℃の泡立つAmesメディウムを連続的に浸み込ませた。代謝型グルタミン酸受容体アゴニストであるL−(+)−2−アミノ−4−ホスホノ酪酸(LAP-4, Tocris Bioscience, cat No. 0103)及びグリシン受容体アンタゴニストであるストリキニン塩酸塩(Sigma Aldrich S8753)を、それぞれ50μM及び10μMの濃度に新しく希釈し、記録前に灌流システムに10分間適用した。全視野の光刺激を、STG2008刺激発生器(MCS)により作動されるPolychrome V monochromator(オリンパス)を用いて行った。アウトプットライトの強度を、1.1014photons/cm2/sから1.1017photons/cm2/sの範囲に調整した。刺激は10秒間隔で2秒間行った。反応の波長感度は、400nmから650nmまでを10nm刻みで10回刺激することにより決定した。試験された波長の順序は、網膜の適応を防ぐためにランダムにした。
25倍の水浸対物レンズ(XLPLN25xWMP/NA1.05,オリンパス)、フェムト秒パルスレーザ(InSightDeepSee - Newport Corporation)を備えた特注の二光子顕微鏡を、CatCh−GFP陽性網膜神経節細胞をイメージングするために用いた。rd1マウスからのAAV処理網膜を、酸素を入れた(95% O2, 5% CO2)Amesメディウム(Sigma-Aldrich)において単離した。ライブ二光子イメージングのために、網膜切片(300μm)をrazor blade tissue chopper (Stoelting)を切断して顕微鏡の記録チャンバに置き、Zスタックを930nmの波長の励起レーザを用いて得た。画像をImageJを用いてオフラインで処理した。イメージングの際に、網膜に対して、酸素を入れたAmesメディウムで表面を灌流した。
マウスを低容量のケタミン−キシラジン注射(ケタミン:100mg/kg及びキシラジン:10mg/kg)により鎮静させ、その後ウレタン(1.0 g/kg, 生理食塩水中10% w/v)を用いて麻酔した。動物を定位ホルダに置いた。温度は37℃に維持し、ビタミンA(Allergan)で覆われたカバーガラスを、角膜脱水を防止するために両目の上に置いた。処理された眼の反対側における半球におけるV1の上の開頭(1mm2)は、ラムダ点から3mm横側で且つ0.5mm吻側を中心とした。硬膜を除き、電極を皮質表面に30°の角度で3軸マイクロマニピュレーター(Sutter Instruments)を用いて挿入した。それを800μm進めて、露出表面をアガロース(1.2%、cortexバッファー中)で覆った。
GFPの蛍光画像(眼底自発蛍光モード:励起波長488nm及びバリアフィルタ500nm)及び眼底の赤外線写真及びOCT画像は、瞳孔拡張後にSpectralis HRA+OCTシステム(HeidelbergEngineering, Heidelberg, Germany)を用いて得られた。細隙灯生体顕微鏡(PortableSlit Lamp model SL-14, Kowa)及び倒像検眼鏡(Indirect BinocularOphthalmoscope model HK 150-1 uno, Heine)からなる眼科的試験を、投薬前2週間、その後毎月、全てのマカクで行った。
高用量が注射されたNHPからの眼を注射後3ヶ月で摘出し、針を挿入し、眼球が膨張するまで0.15〜0.3mlの固定液を注射した。眼を固定液に浸漬して一晩固定し、全体構造を横切る水平方向断面を作製するように処理した。所望の眼構造のすべてが存在する網膜の断面をNanozoomer (Hamamatsu, Japan)で画像化した。
<hSNCGのプロモーター配列>
SNCG遺伝子は第10染色体(10q23.3)に位置する(図1)。それは+1転写サイトから5番目のエクソンの端部まで3.5kbにわたって延びる5つのエクソンを含む。マルチメリン2遺伝子(MMRN2)の第1のエクソンは、+1SNCG転写サイトの863bp下流に位置する。SNCGプロモーターのヒト配列を抽出するために、−785から+163の領域(図1において矢印で示す)をHEK293T細胞から増幅させた。
フォワードプライマー:5’−CACAAGCCAGTTCCTGTCC−3’ (配列番号2)、及び、
リバースプライマー:5’−GGGTGTGCAGGGTTGTG−3’ (配列番号3)。
(構築)
SNCGプロモーター(配列番号1)及びPGKプロモーター配列を、Gateway法を用いた相同組換えにより、5’ITRから3’ITRまで:(Gatewayシステムを用いた相同組換えを許容する)Rfaインサート、GFPをコードするcDNA及びウシ成長ホルモンのポリアデニル化シグナル(bGHポリA)を含むAAV産物のためのシャトルプラスミドに導入した。
発現カセット、rep2及びcap2−Y444F遺伝子を含む組換型AAV2ゲノムを運ぶプラスミドと、ヘルパープラスミドとのHEK293T細胞へのコトランスフェクション(カルシウムリン酸共沈法を使用)によりAAV粒子を生成した。その後、不連続勾配イオジキサノール超遠心法により、トランスフェクションされた細胞の細胞質抽出液からAAV粒子を精製及び濃縮した。ウィルス力価を、AAV粒子のゲノムDNAにおけるITR配列の定量的PCR増幅により測定した。
C57BL6/Jの成体マウス(6週)に、PBSで1011vg(ベクターゲノム)の量に希釈された配列番号1のプロモーター及びGFPタンパク質をコードする核酸を含むAAV−2/Y444Fベクター(実施例1及び図1を参照)を硝子体内注射した。注射から1ヵ月後に、眼底及び網膜は、ベクター又は手術による毒性が無いことを示し、正常外観を有していた(図2A)
網膜におけるhSNCGプロモーターの発現パターンを特徴付けるために、本発明者らは、hSNCGプロモーター(配列番号1)を、網膜において高いレベルで発現することが知られているユビキタスプロモーターであるmPGKプロモーターと比較した。
我々は、Brn3aとの共標識による発現の位置を調べることにより、RGCにおける遺伝子発現の強さ及び特異性を評価した。Brn3aは、RGCに特異的に発現し、この転写因子に対する抗体は、マウスRGCを同定する信頼できるマーカーとして考えられる(Quina et al. J. Neurosci. 2005;25(50):11595-11604)。
AAV2−CMV−GFP又はAAV2−SNCG−GFPベクターのそれぞれの5×1011ウィルス粒子を、100μlの容量で、正常のマカク(macaca fascicularis)の眼に硝子体内注射した。我々は、眼底の蛍光イメージングにより、カニクイザルの網膜におけるGFP発現を評価した。GFPの蛍光画像(眼底自然蛍光モード:励起波長488nm、バリアフィルタ500nm)は、瞳孔拡張後に、Spectralis HRA+OCTシステム(Spectralis HRA+OCT;Heidelberg Engineering, Heidelberg, Germany)を用いて得られた。蛍光画像は、CMVプロモーターを含むAAV−GFPベクターの場合(図4B)と比較して、形質導入領域(中心窩の周縁の輪)やSNCGプロモーターを含むAAV−GFPベクターのバックグラウンドできわめて強いGFP発現を示した。この試験は、高い導入遺伝子発現がSNCGプロモーター(配列番号1)を用いた非ヒト霊長類で得られることを証明する。
(配列番号1のプロモーター配列を用いたマウス網膜における強いRGC特異的発現)
ヒトγシヌクレイン(SNCG)の制御領域由来の実施例1で得られたプロモーター配列を、GFPの上流にクローニングした。我々は、マウスの網膜におけるAAVを介した発現パターンを特徴付け、RGCにおける高レベルのGFP発現を観察した。また、我々は、AAV骨格におけるヒト化CatCh(L132C変異を有するヒトコドン最適化チャンネルロドプシン)の上流にこの配列をクローニングした。AAV2ベクターを、GFPと融合したhCatchの発現を作動するCMV又はSNCGプロモーターを用いて作製した。5つのrd1マウスの眼にhCatCh−GFPの発現を作動するCMV又はSNCGを注射した。
hCatCh−GFPの選択的RGC標的化が、盲目のrd1の網膜(12週齢超過)において視覚機能を回復できることを証明するために、我々は、マルチ電極アレイ(MEA)を用いて網膜神経節細胞からのスパイク活動を記録した。CMV又はSNCG(配列番号1)下でhCatCh−GFPをコードするAAV2を3つの用量で誕生から4〜8週のマウスに注射し、注射後8〜12週にMEA記録を行った(図6A〜C)。光遺伝学的反応の光感受性は、1014から1017photons/cm2/sの範囲の光条件において252の電極アレイで読み出された情報として測定された。光励起スパイク活動は、2秒の全視野フラッシュを用いて処置されたrd1網膜を刺激した際に観察され(図6A〜E)、一方で、コントロールのrd1網膜では光に応答したスパイク活動の増大は示されなかった(図示せず)。光に反応する細胞の割合は、ウィルス量に依存した(図6A〜B)。1014photons/cm2/sの刺激により、高用量のAAVのSNCGプロモーターで注射されたrd1動物において多くの細胞が反応した(図6A)。この改善された感受性は、SNCGプロモーター(配列番号1)がRGCで高レベルのCatCh発現を作動し、高い割合の細胞が同一のウィルス力価で光感受性になることを裏付ける。発火率頻度は強度依存的であり、5×109vg/眼のウィルス用量で、SNCGプロモーター下で発現するCatChにおける1014photons/cm2/sでの強い光応答が起こる(図6A)。正規化された発火率は、光強度の上昇に従って増大する(図6C〜D)。なお、CMVプロモーターを有する最も高いウィルス用量でのみ、1014photons/cm2/sでの光応答が生じた(図6A)。さらに、わずかの細胞がこれらの条件で光に反応した。最も高い光強度でさえ、中程度のウィルス用量(5×108vg/眼)において反応する細胞はほとんど無かった(図6B)。この試験は、RGCにおける機能性タンパク質の発現の作動のための、SNCGプロモーター(配列番号1)の高い有効性を証明した。
マウスの試験において、我々は、5×107から5×109vg/眼の範囲の用量を用いた。我々は、ヒトにおける光毒性を最小にするために低光強度を用いることが必要であることを観察し(1014〜1015photons/cm2/sの範囲)、MEAにおける光応答を得るのに必要な特定の数字は、5×108から5×109vg/眼の範囲である。マカクの硝子体の容積はマウスの硝子体容積の約100倍大きいので、我々は、我々の非ヒト霊長類試験におけるこの用量範囲の医薬的等量を使用することを決めた。5匹の非ヒト霊長類は、それらの血清におけるAAV2に対する中和抗体力価が無いことに基づいて選択された(Kotterman et al. Gene Ther. 2015 Feb;22(2):116-26)。これらの成体のマカクの4匹に、SNCGプロモーター下でhCatChをコードする1×1011(n=4眼)又は5×1011粒子(n=4眼)のAAV2を硝子体内注射した(表1)。
注射後3ヶ月の高用量群においてわれわれが観察した1つの眼でのみ組織学的病変が見られた。眼全体を固定し、切断し、断面を40倍の分解能を有するnanozoomer技術を用いて観察した(図8)。炎症を示す構造的変化(繊維柱帯網及び虹彩角膜角におけるリンパ球若しくはプラズマ細胞、硝子体における炎症性細胞、又は網膜における血管周囲リンパ球の存在)は無かった(図8B〜F)。重要なことに、5×1011vg用量群のこの動物(NHP3)は、注射後1〜2ヶ月における倒像検眼鏡による評価において、硝子体混濁及び細胞の可変レベルを示した(図7)。3ヶ月において炎症細胞及び網膜のダメージがないことは、進行性の前眼房フレア又は硝子体混濁が網膜構造において永続的変化を引き起こさないこと、並びに網膜全体及び眼の前領域がダメージ又は炎症のサインを回避したことを示す。従って、SNCGプロモーター(配列番号1)の制御下での光遺伝子発現は、炎症反応を誘導しないことが明らかである。

Science 2010;3(June):413-7)、抗Brn3a抗体及びGFP抗体(存在する場合)で染色した。NHP1からの組織を、緑色の抗GFP抗体及び赤色の抗Brn3a抗体で標識した(図9B)。Brn3a及び抗チャンネルロドプシン抗体が同一種で生産されているため、CatChの局在を示す抗体と共に我々がRGC特異的マーカーであるBrn3aを用いることができたのは、網膜のみである。中心窩の中心から1mm未満におよぶ四角形の範囲のこの組織において、我々は中心窩の周囲の600μmの半径の半円内におけるBrn3a(+)及びCatch−GFP(+)細胞をカウントした。この領域において、約37%のBrn3a(+)細胞はGFP陽性であった(1413個のBrn3a陽性細胞中、523個のGFP陽性細胞)。NHP2から得られた組織において、我々は、一つの眼の半分において1351個のCatCh陽性細胞をカウントし、反対側の眼の中心窩の中心から600μmにわたる範囲において455個カウントした。高用量群の網膜の1つはMEA記録後に損傷し、免疫蛍光標識を用いることができなかった。これらの結果は、中心窩の周縁におけるRGC細胞の少なくとも4分の1が、5×1011vgで注射した後にCatChで標識されたことを示す。
AAV2-SNCG-hCatCh-GFPが注射されたNHP1を注射の3ヵ月後に屠殺した。網膜を解剖し、GFP由来緑色蛍光が中心窩領域で最大であるため(図9A)、中心窩領域をMEA及びパッチクランプ記録のために注意深く二等分した。この発現パターンは以前のNHP研究と一致していた(Dalkara et al. Sci Transl Med. 2013 Jun 12;5(189):189ra76; Yin etal. Invest Ophthalmol Vis Sci. 2011 Apr 25;52(5):2775-83)。我々の試験の全てにおいて、NHPの網膜における内在性光反応を、浴用液中の50μMの代謝型グルタミン酸受容体アゴニストであるL−(+)−2−アミノ−4−ホスホノ酪酸(L−AP4)を用いて防止した。我々は、以前に、マウス(Nagel et al. Proc. Natl. Acad. Sci. U. S. A. 2003;100(24):13940-5)及びヒト(Tomita et al. Mol Ther. 2014 Aug;22(8):1434-40)の網膜における野生型網膜の全てのON反応に対するL−AP4防止を検証している。
我々は、光強度の関数として、反応性細胞の数(図11A〜C)及び最大発火頻度(図11D)又は光電流(図11E、高用量のみ)に関して4つの群を比較した。我々は、両ウィルス用量において、AAV2-SNCG-hCatChを注射された動物由来の記録された中心窩周辺細胞が、AAV2-CMV-hCatChを注射された動物由来の記録された細胞よりも光応答する可能性が高いことを発見した。また、我々の結果は、SNCG応答性細胞群(両用量)がCMVプロモーターを含む群よりも光感受性が高い(発火活動が速い)ことを示した。
これらの光電流が個体群レベルでRGC活性をどのように制御しているか定義するために、網膜フラットマウントをマルチ電極アレイ(MEA)技術で記録した。図12Bは、NHP1の左眼の単一ユニット記録からのラスタープロットを示す(高用量)。光反応は、網膜フラットマウントで見られる中心窩周辺のGFP発現と相関した(図12A〜D)。5倍低い用量を注射した反対側の眼では反応は得られなかった。次に、我々は、GFPタグが付いていない高用量のAAV2-hCatChを注射された他の動物を屠殺した。高用量群から得られた4つの網膜の全ては、90%以下の光に反応する記録されたRGCを含むL−AP4での阻害下で同様の光反応を示した(図12A)。同様に、光応答性細胞の分布は、AAV2-SNCG-hCatChベクターを含む場合でさえ中心窩領域が常に中心であった。これらの結果は、GFPタグがRGCにおける機能的CatCh発現を得るのに必要でないことを示す。発火頻度のスペクトル同調を算出し、480nmの光に対する最も高い周波数応答を示し、それはChR2の励起ピークに対応する。応答性細胞の発火率頻度は、1017photons/cm2/sで適用された最大光強度で最大に達する強度依存性であった(図12D)。

このベクターは、SNCGプロモーター(配列番号1)の制御下でChrimsonR-tdTomato(Klapoetke et al., NatMethods, 2014 Mar;11(3):338-46)のコード配列をカプシドで包み、網膜への導入を最適化されたAAVカプシド変異体であるAAV2−7m8(Dalkara et al., Sci Trans Med, 2013 Jun 12;5(189):189ra76)からなる。
ChrimsonR-tdTomatoの選択的RGC標的化が霊長類における視覚機能を修復できることを証明するために、我々は、マルチ電極アレイ(MEA)を用いて網膜神経節細胞からのスパイク活動を記録した。MEA記録を注射の6ヶ月後に行った(図15)。
Claims (38)
- 目的のポリペプチド又は核酸をコードする核酸に作動可能に連結された網膜神経節細胞においてプロモーター活性を有する核酸を含む発現カセットであって、
前記プロモーター活性を有する核酸は、1.5kb未満の長さを有し、且つ、配列番号1の配列又は配列番号1の配列に対して少なくとも80%の同一性を有する配列番号1の配列の機能性変異体を含む又はからなる発現カセット。 - 前記プロモーター活性を有する核酸は、網膜神経節細胞に特異的なプロモーター活性を有する請求項1に記載の発現カセット。
- 前記プロモーター活性を有する核酸は、配列番号1の配列に対して少なくとも80%の同一性を有する配列を含む又はからなる請求項1又は2に記載の発現カセット。
- 前記プロモーター活性を有する核酸は、配列番号1の配列に対して少なくとも90%の同一性を有する配列を含む又はからなる請求項1〜3のいずれか1項に記載の発現カセット。
- 前記プロモーター活性を有する核酸は、配列番号1の配列からなる請求項1〜4のいずれか1項に記載の発現カセット。
- 前記プロモーター活性を有する核酸は、1.2kb未満の長さを有する請求項1〜5のいずれか1項に記載の発現カセット。
- 前記プロモーター活性を有する核酸は、1kb未満の長さを有する請求項1〜6のいずれか1項に記載の発現カセット。
- 前記プロモーター活性を有する核酸は、目的のポリペプチドをコードする核酸に作動可能に連結されている請求項1〜7のいずれか1項に記載の発現カセット。
- 前記目的のポリペプチドは、治療用タンパク質、光遺伝学的アクチュエータ又はレポータータンパク質である請求項8に記載の発現カセット。
- 前記目的のポリペプチドは、治療用タンパク質である請求項9に記載の発現カセット。
- 前記治療用タンパク質は、MT−ND4、MT−ND1、MT−ND6、MT−CYB、MT−CO3、MT−ND5、MT−ND2、MT−COI、MT−ATP6、MT−ND4L、OPA1、OPA3、OPA7及びACO2からなる群から選択される請求項10に記載される発現カセット。
- 前記治療用タンパク質は、GDNF、VEGF、CNTF、FGF2、BDNF及びEPOからなる群から好ましくは選択される神経栄養因子、BCL2及びBCL2L1からなる群から好ましくは選択される抗アポトーシスタンパク質、エンドスタチン、アンジオスタチン及びsFltからなる群から好ましくは選択される抗血管新生因子、IL10、IL1R1、TGFBI及びIL4からなる群から好ましくは選択される抗炎症因子、又は杆体由来の錐体生存因子(RdCVF)である請求項10に記載の発現カセット。
- 前記目的のポリペプチドは、光遺伝学的アクチュエータである請求項9に記載の発現カセット。
- 前記光遺伝学的アクチュエータは、ロドプシン、フォトプシン、メラノプシン、ピノプシン、パラピノプシン、VAオプシン、ペロプシン、ニューロプシン、エンセファロプシン、レチノクロム、RGRオプシン、赤色にシフトした分光特性を有するReaChR、Chrimson若しくはChrimsonR等の微生物性オプシン、Gi/Oシグナルをリクルートできる短波長脊椎動物オプシン若しくは長波長脊椎動物オプシン等の脊椎動物オプシン、Chlamydomonas属の微細藻類由来のチャンネルロドプシン−1及びチャンネルロドプシン−2(Chlamydomonas reinhardtii由来)等のチャンネルロドプシン、並びにそれらの変異体からなる群から好ましくは選択される光遺伝学的活性化因子である請求項13に記載の発現カセット。
- 前記光遺伝学的アクチュエータは、ハロロドプシン(NpHR)、増強型ハロロドプシン(eNpHR2.0及びeNpHR3.0)及び赤色にシフトしたハロロドプシン、Halo57、古細菌ロドプシン−3(AR−3)、古細菌ロドプシン(Arch)、増強型バクテリオロドプシン(eBR)等のバクテリオロドプシン、プロテオロドプシン、キサントロドプシン、Leptosphaeria maculans真菌オプシン(Mac)、クラックスハロロドプシンJaws、並びにそれらの変異体からなる群から好ましくは選択される光遺伝学的阻害因子である請求項13に記載の発現カセット。
- 前記目的のポリペプチドは、蛍光タンパク質、カルシウムインジケーター、アルカリホスファターゼ、βガラクトシダーゼ、βラクタマーゼ、ホースラディッシュペルオキシダーゼ及びそれらの変異体からなる群から好ましくは選択されるレポータータンパク質である請求項9に記載の発現カセット。
- 前記プロモーター活性を有する核酸は、siRNA、shRNA、RNAi、miRNA、アンチセンスRNA、リボザイム及びDNAザイムからなる群から好ましくは選択される目的の核酸をコードする核酸に作動可能に連結されている請求項1〜7のいずれか1項に記載の発現カセット。
- 前記目的のポリペプチドは、SNCGタンパク質ではない請求項1〜17のいずれか1項に記載の発現カセット。
- 前記目的のポリペプチドは、ルシフェラーゼではない請求項1〜18のいずれか1項に記載の発現カセット。
- 請求項1〜19のいずれか1項に記載の発現カセットを含むベクター。
- ウィルスベクターである請求項20に記載のベクター。
- 前記ウィルスベクターは、モロニーマウス白血病ウィルスベクター(MoMLV)、MSCV、SFFV、MPSV若しくはSNV、ヒト免疫不全ウィルス(HIV)、サル免疫不全ウィルス(SIV)、ネコ免疫不全ウィルス(FIV)、ウシ免疫不全ウィルス(BIV)若しくウマ伝染性貧血ウィルスに好ましくは由来するレンチウィルスベクター、アデノウィルス(Ad)ベクター、アデノ随伴ウィルス(AAV)ベクター、シミアンウィルス40(SV−40)ベクター、ウシパピローマウィルスベクター、エプスタイン・バールウィルス、ヘルペスウィルスベクター、ワクチニアウィルスベクター、ハーベイマウス肉腫ウィルスベクター、マウス乳がんウィルスベクター及びラウス肉腫ウィルスベクターからなる群から選択される請求項21に記載のベクター。
- 前記ウィルスベクターは、レトロウィルスベクターであり、好ましくはレンチウィルスベクター又は非病原性パルボウィルスである請求項21又は22に記載のベクター。
- アデノ随伴ウィルス(AAV)ベクターである請求項20〜23のいずれか1項に記載のベクター。
- 請求項20〜24のいずれか1項に記載のベクターを含むウィルス粒子。
- 請求項24に記載のベクター及びAAV由来カプシドを含むAAV粒子。
- 前記AAV由来カプシドは、AAV−2、AAV−5、AAV2−7m8、AAV−9及びAAV−8血清型カプシドから選択される請求項26に記載のAAV粒子。
- 前記AAV由来カプシドは、AAV−2由来カプシドであり、好ましくはAAV−2及びAAV2−7m8カプシドから選択される請求項27に記載のAAV粒子。
- 請求項1〜19のいずれか1項に記載の発現カセット、請求項20〜24のいずれか1項に記載のベクター又は請求項25〜28のいずれか1項に記載のウィルス粒子により形質転換された細胞。
- 網膜神経節細胞である請求項29に記載の細胞。
- 請求項1〜19のいずれか1項に記載の発現カセット、請求項20〜24のいずれか1項に記載のベクター、請求項25〜28のいずれか1項に記載のウィルス粒子又は請求項29若しくは30に記載の細胞と、医薬的に許容可能な賦形剤とを含む医薬組成物。
- 眼疾患の治療のための、請求項1〜19のいずれか1項に記載の発現カセット、請求項20〜24のいずれか1項に記載のベクター、請求項25〜28のいずれか1項に記載のウィルス粒子、請求項29若しくは30に記載の細胞、又は請求項31に記載の医薬組成物の使用。
- 前記眼疾患は、網膜神経節細胞変性又は光受容体細胞変性に関連する疾患である請求項32に記載の使用。
- 前記眼疾患は、網膜神経節細胞変性に関連する疾患であり、遺伝性視神経症、圧迫性視神経症、自己免疫性視神経症、糖尿病性網膜症、緑内障を含む緑内障性視神経疾患、動脈炎性虚血性視神経症、非動脈炎性虚血性視神経症、浸潤性視神経症、感染性視神経症、脱髄疾患による視神経炎、放射線治療後視神経症及び腸性肢端皮膚炎からなる群から選択される請求項33に記載の使用。
- 前記眼疾患は、レーバー遺伝性視神経萎縮症又は優性視神経萎縮症から選択される遺伝性視神経症であり、好ましくはレーバー遺伝性視神経萎縮症、視神経萎縮1、視神経萎縮3及び視神経萎縮7から選択される請求項33に記載の使用。
- 前記眼疾患は、光受容体細胞変性に関連する疾患であり、加齢黄斑変性、レーバー遺伝性視神経萎縮症、錐体杆体変性、レーバー先天性黒内障、シュタルガルト病、糖尿病性網膜症、網膜剥離、ベスト病、網膜色素変性、先天性脈絡膜欠如又は色素上皮網膜変性から選択される請求項33に記載の使用。
- 網膜神経節細胞に目的のポリペプチド又は核酸をコードする核酸を発現するための、請求項1〜19のいずれか1項において定義された網膜神経節細胞においてプロモーター活性を有する核酸、請求項1〜19のいずれか1項に記載の発現カセット、請求項20〜24のいずれか1項に記載のベクター、又は請求項25〜28のいずれか1項に記載のウィルス粒子の使用。
- 網膜神経節細胞に目的のポリペプチド又は核酸を発現させる方法であって、
網膜神経節細胞に、請求項1〜19のいずれか1項に記載の発現カセット、請求項20〜24のいずれか1項に記載のベクター、又は請求項25〜28のいずれか1項に記載のウィルス粒子を導入することを含む方法。
