タイプVII分泌系(T7SS)は、細菌の分泌系において最近発見され、結核菌(Mycobacterium tuberculosis)において最初に同定されたものである。対応する遺伝子群は、ESX(ESAT−6分泌系)領域と呼称された4〜6。結核菌(M.tuberculosis)H37Rvのゲノムは、遺伝子重複イベントを通して進化し、T7SS分泌機構の構成要素を含む5つの遺伝子群を含有する。これらの群は、ESAT−6分泌系(ESX)1〜5と呼ばれる。ESX系は、従来のシグナルペプチドを欠くタンパク質を分泌することが示されている。さらに、ESX1〜5によって分泌されるタンパク質の大部分は分泌について相互依存に従う7。
Rv3017c(esxR)を除き、ESAT−6ファミリータンパク質をコードする遺伝子は、結核菌(M.tuberculosis)H37Rv染色体上の11座で、縦列対で配列され、pe−ppe遺伝子対がそれに先行する場合が多い。それらは、アミノ酸約100個の鎖長でESX1〜5系によって分泌されるタンパク質をコードする。
・in vitro細胞応答は、毒性マイコバクテリア(mycobacteria)に現在感染している、又は過去に感染した動物又はヒトから取り出したリンパ球からの、IFN−γなどの関連するサイトカインの放出によって、又はこれらのT細胞の増殖の検出によって測定される。ポリペプチド又は免疫原性部分を、ウェル当たり1×105個の細胞〜3×105個の細胞を含む懸濁液に添加することによって実施される誘導。血液、脾臓、肝臓又は肺から単離される細胞、及び懸濁液1ml当たり20μg以下の濃度になるポリペプチド又は免疫原性部分の添加、並びに2日〜5日実施される刺激。細胞増殖のモニタリングには、放射性標識したチミジンで細胞をパルス標識し、インキュベーションの16〜22時間後、液体シンチレーション計数による増殖の検出。バックグラウンドプラス2標準偏差より高い応答である陽性応答。IFN−γの放出は、当業者に周知のELISA法によって測定することができる。バックグラウンドプラス2標準偏差より高い応答である陽性応答。IFN−γ以外のサイトカイン、例えば、IL−12、TNF−α、IL−4、IL−5、IL−10、IL−6、TGF−βは、ポリペプチドに対する免疫学的応答をモニターする際に関連し得る。サイトカイン(例えばIFN−γ)の存在を測定する別のさらに高感度な方法はELISPOT法であり、血液、脾臓、肝臓又は肺から単離された細胞を1〜4×106個の細胞/mlの好ましい濃度に希釈し、1ml当たり20μg以下の濃度になるポリペプチド又は免疫原性部分の存在下で18〜22時間インキュベートする。その後、細胞懸濁液を1〜2×106/mlに希釈し、抗IFN−γでコートしたMaxisorpプレートに移し、好ましくは4〜16時間インキュベートする。IFN−γを産生する細胞を、標識した抗IFN−γ二次抗体及びスポットを生じさせる関連の基質の使用によって判定し、解剖顕微鏡を使用してこれを計数することができる。関連するサイトカインをコードするmRNAの存在を、PCR技術の使用によって判定することも実現可能である。通常、例えばPCR、ELISPOT又はELISAを利用して、1種又は複数種のサイトカインを測定することになろう。当業者には理解されるであろうが、特定のポリペプチドによって誘導されるこれらのサイトカインの何れかの量における有意な増加又は減少は、ポリペプチドの免疫学的な活性の評価に使用することができる。
本発明の好ましい実施形態において、ポリペプチドは、ポリペプチドの免疫原性部分、例えば、B細胞又はT細胞に対するエピトープを含む。ポリペプチドの免疫原性部分はポリペプチドの一部であり、動物又はヒトにおいて、及び/又は本明細書に記載する生物学的アッセイの何れかによって測定される生体試料において、免疫応答を引き出す。ポリペプチドの免疫原性部分は、T細胞エピトープ又はB細胞エピトープであり得る。免疫原性部分は、ポリペプチドの1個又は数個の比較的小さい部分に関連している可能性があり、ポリペプチド配列全体にわたって分散していたり、ポリペプチドの特定の部分に位置していたりする可能性がある。数種のポリペプチドについては、エピトープが、ポリペプチドの全配列にわたる全域に分散していることが示されている(Ravn et al 1999)。
「融合タンパク質」という用語は、任意の長さ及び配列のアミノ酸リンカー/スペーサーを伴って、又は伴わずに融合した、結核菌(M.tuberculosis)由来の順不同の2つ以上の免疫原性のポリペプチド又はそのアナログのことと理解される。下流生成でのタンパク質凝集を回避するために、融合タンパク質中の全てのシステインが任意のアミノ酸で置換され得るが、セリンは、その構造がシステインと類似性が高いため、好ましい置換基である。
リンカー又はスペーサーは、融合タンパク質中のポリペプチドパートナー同士の間に生じる短鎖ペプチド配列である。リンカーは、グリシン及びセリンのような可動性残基から構成されることが多いため、隣接するタンパク質ドメイン同士は互いに対して、分泌/産生中の独立した固有のフォールディングのために自由に動く。2つの隣接するドメインが、互いを立体的に干渉しないように確実にする必要がある場合、長めのリンカーが使用される。
「パラログ」という用語は、祖先が共通で、その後1つ又は複数の重複事象が生じたために、ある程度の相同性をもつタンパク質又は遺伝子のことであると理解される。パラログはゲノム内の重複が関連する遺伝子であり、一方、共通祖先遺伝子から種分化によって進化した異なる種における相同遺伝子であるオルソログ。ホモログという用語は、種分化の事象によって分離した遺伝子間の関係に適用される(オルソログ)か、又は遺伝子重複の事象によって分離した遺伝子間の関係に適用される(パラログ)。
配列アナログという用語は、互いが構造的且つ免疫原性的に類似しているが、アミノ酸組成において異なるポリペプチドを意味する。
本発明の別の部分は、本発明に係る融合タンパク質を含むワクチン組成物に関する。融合タンパク質本発明が動物に認識される有効なワクチンは、動物モデルにおいて、毒性マイコバクテリウム(Mycobacterium)の感染に曝露させた後、非ワクチン接種の動物と比較して、標的器官において細菌負荷を減少させ、生存期間を長期化させ、且つ/又は体重減少幅を減少させることが可能であろう。
本発明は、ワクチンとして投与されると、実験動物において結核菌(M.tuberculosis)感染症の重症度を減少させ、又は以前の感染の再活性化を予防するその能力に基づいて、治療ワクチンとして使用するための本発明の融合タンパク質の使用にも関する。治療ワクチン用に使用される組成物は、ワクチンについて上に記載したとおりに調製することができる。
マイコバクテリア(Mycobacteria)の分泌系は、細胞外環境へ、又は直接的に宿主細胞中へ、毒性因子を排出することを担っており、このため、細菌の毒性及び生存にきわめて重要な役割をする。ESX−1分泌系の一部は、弱毒化ウシ型結核菌(M.bovis)BCG及び病原性マイコバクテリア種の比較ゲノム解析中に同定された8。主なゲノムの差異の1つは、ウシ型結核菌(M.bovis)ゲノムにおける、分泌抗原CFP10及びESAT−6をコードする領域を含んだ大幅な欠失であった。この領域は、毒性に特に寄与することが観察され、この領域を修復すると、ESAT−6の分泌が可能になっただけでなく、ウシ型結核菌(M.bovis)BCGにおいて毒性が増大した4。
ESAT−6(Rv3875)は、CFP10と一緒に、ヘテロ二量体としてESX−1によって分泌される14。
EspR(Rv3849)は、それ自身の発現の転写活性化因子であり、espA(Rv3616c)、C及びDを含むオペロンである。EspRタンパク質は、ESX−1によって分泌される15。
EspC(Rv3615c)の遺伝子発現は、EspRによって制御される。EspCは、ESX−1によって分泌される11。
EspD(Rv3614c)の遺伝子発現は、EspRによって制御される。EspDは、espC及びespAと共転写される。EspDの分泌ではなく発現は、EsxAの分泌に必要である。EspDは、EspA〜EspCの複合体を安定化させる。EspDの分泌は、ESX−1系を必要とするだけというわけではない16。
EspA(Rv3616)の遺伝子発現は、EspRによって制御される。EspAは、ESX−1によって分泌される7。
EspB(Rv3881)は、ESX−1によって分泌される(Xu,2007 #912;McLaughlin,2007 #591)。
ESAT−6(Rv3875)は、CFP10と一緒に、ヘテロ二量体としてESX−1によって分泌される14。

EspC(Rv3615c)タンパク質配列のアミノ酸1−56。espCの遺伝子発現は、EspRによって制御される。EspCタンパク質は、ESX−1によって分泌される11。
EspR(Rv3849)は、それ自身の発現の転写活性化因子であり、espA(Rv3616c)、C及びDを含むオペロンである。EspRタンパク質は、ESX−1によって分泌される15。
PE35(Rv3872)は分泌PEタンパク質である。遺伝子pe35(Rv3872)を不活性化するとCFP−10及びESAT−6の発現が損なわれ、これは調節における役割を示唆するものである。
実施例1:H74融合タンパク質−予防TBワクチン接種モデルにおける免疫応答及び防御。
CB6F1マウスの各群に、0.01、0.1、1、5、又は25μgの融合タンパク質H74又は5μgの融合タンパク質H56の何れかを3回ワクチン接種した。両タンパク質は、注射前にリポソーム系アジュバントCAF01中に配合した。コントロール群には、等しい体積の塩水(200μL)を3回注射、又は200μLのBCG(5×107CFU/mL)を1回ワクチン接種した。ワクチン接種の間隔は2週間とし、3回目のワクチン接種から3週間後に、PBMC及び脾細胞(splenocytes)を単離し、ワクチンが誘導したT細胞応答を測定した。単離した細胞(5×106個/ウェル)を、2つの融合タンパク質中に存在する個々のタンパク質2μg又は融合タンパク質2μgで3日間インキュベートし、分泌されたIFN−gを培地中でELISAによって測定した(図1A−F)。H56をワクチン接種したコントロール動物では、H56タンパク質及びAg85Bの認識が強く、ESAT−6の認識は中程度である。H74をワクチン接種した動物は、Rv3881cに特異的な応答が強く、Rv3849c及びRv3616cに対する応答は中程度である。この近交系マウスでは、Rv3615c又はRv3614cに対する応答はない。生理食塩水を注射した動物に応答はなく、これらの応答はワクチンに特異的であることが確認された。H74応答の強さは、1又は5μgの何れかのH74タンパク質を接種された動物において組織に依存して最も強かった(図1A及びB)。しかし、0.1μgの低用量のH74でも、H56タンパク質5μgでのワクチン接種よりも高いIFN-γ放出が得られた。脾臓T細胞によって産生された更なるサイトカインを見てみると、応答しているT細胞の頻度は、1μgのH74群で最大であったが、この場合もやはり、0.01μgのH74が、5μgのH56よりも高い応答であった(図2A)。ワクチン用量とT細胞の多機能性に関して、IL−2産生T細胞の相対比率とH74ワクチン用量との間には逆相関関係があった(図2B)。
ワクチン接種用の最適なH74用量を決定するために、BCGワクチン接種動物群に、BCGワクチンに加えて、リポソーム系アジュバントCAF01中に配合した融合タンパク質H74を、様々な用量で3回ワクチン接種した。最初のH74ワクチン接種は、BCGワクチン接種の3カ月後に行った。コントロール動物には、一定体積の塩水(BCG無し、H74無し)を4回注射又はBCGを1回ワクチン接種した。H74ワクチン接種間の間隔は2週間であった。3回目のワクチン接種から6週間後に、動物に、毒性結核菌(M.tuberculosis)エルドマン(Erdman)をエアロゾルで曝露した。12週間後に全てのマウスを安楽死させ、個々の動物の肺における細菌数を、肺ホモジネートの希釈物をプレーティングし、コロニーの数を計数することによって求めた(図3A及びB)。
この実験には、4つのワクチン接種群及び1つの生理食塩水コントロール群がある。1のワクチン接種群には、BCGのみを接種し、もう1群には、5μgのH74/CAF01を3回接種した。残りの2群には、初回のワクチン接種として、一の注射器でBCGをワクチン接種し、別の注射器でH74/CAF01又はCAF01の何れかをワクチン接種した(並行ワクチン接種:side−by−side vaccination)。2回目及び3回目のワクチン接種では、H74/CAF01又はCAF01をそれぞれ接種した。3回目のワクチン接種から6週間後に、全てのマウスに結核菌(Mtb)株エルドマン(Erdman)を曝露し、6週間後に上記と同じように肺におけるCFU数を測定した(図5)。全てのワクチンで有意な防御が誘導され(p<0.001)、BCG、BCG:CAF01、及びH74についてのレベルは同様であった。しかし、BCG:H74及び2回のH74の追加免疫(boost)を接種した群の動物は、他のワクチン接種群、すなわちBCG、BCG:CAF01、及びH74よりも、肺における細菌が有意に少なかった(p<0.001)。
H64融合タンパク質は、結核菌(Mycobacterium tuberculosis)に由来する6つのタンパク質を含む。そのうちの2つ、Rv3865及びRv3615は、TB患者内で高い頻度及び特異性で認識され、従って、来たるTB診断キットにおけるタンパク質抗原として重要な価値を有する。抗原が重複するため、H64ワクチンの世界規模での使用は、この診断キットに支障をきたす。この問題を避けるために、H64からRv3865の全長及びRv3615タンパク質の半分を除去して、H78と命名した僅かに短い融合タンパク質を作成した。H64とH78の免疫応答を誘導する能力を比較するために、動物群に、CAF01アジュバントに配合した2μgのH64又はH78の何れかを3回ワクチン接種又は生理食塩水を3回注射した。3回目のワクチン接種から3週間後に、各群の動物3匹を安楽死させ、フローサイトメトリにより、脾臓細胞における最も重要な抗原に対するT細胞応答を測定した(図6)。H64及びH78の両方は、予期された抗原パターンを誘導している。H64は、調べた4つの抗原(ESAT−6、Rv3615c、Rv3614c、及びRv3865)全てに対するT細胞応答を誘導した。予期された通り、H78でワクチン接種された動物においてRv3865に対する応答はなかったが、極めて重要なことに、H78がRv3615cタンパク質の半分を欠損しているにも関わらず、H64でワクチン接種された動物において見られたものと同様のT細胞頻度を持つRv3615cに対する特異的応答がまだあった。

c)配列番号2(ESAT6)、配列番号3(Rv3614c)、配列番号9(Rv3615の一部)、配列番号6(Rv3849)及び配列番号8(Rv3872)、若しくは

- 請求項1又は2に記載の融合タンパク質において、硫黄架橋形成及びタンパク質凝集を回避するために、システインが別のアミノ酸で置換されていることを特徴とする融合タンパク質。
- 請求項3に記載の融合タンパク質において、前記システインがセリンで置換されていることを特徴とする融合タンパク質。
- 請求項1乃至4の何れか1項に記載の融合タンパク質において、融合パートナー同士がリンカー分子で結合していることを特徴とする融合タンパク質。
- 請求項5に記載の融合タンパク質において、配列番号7(H74)、配列番号9(H164)、配列番号10(H174)、配列番号11(H264)、配列番号12(H274)、配列番号13(H374)又は配列番号14(H78)のアミノ酸配列を伴うことを特徴とする融合タンパク質。
- 毒性マイコバクテリア(mycobacteria)によって、例えば結核菌(Mycobacterium tuberculosis)、アフリカ型結核菌(Mycobacterium africanum)又はウシ型結核菌(Mycobacterium bovis)によって引き起こされる感染に対するワクチン接種用の医薬組成物を調製するためであることを特徴とする、請求項1乃至7の何れか1項に記載の融合タンパク質又は抗原カクテルの使用。
- 請求項1乃至6の何れか1項に記載の融合タンパク質又は抗原カクテルを含むことを特徴とするワクチン。
- 請求項8に記載のワクチンにおいて、アジュバントをさらに含むことを特徴とするワクチン。
- 請求項9に記載のワクチンにおいて、前記アジュバントが、カチオン性リポソーム(例えばジメチルジオクタデシルアンモニウムブロミド(DDA))、Quil A、poly l:C、水酸化アルミニウム、フロイント不完全アジュバント、IFN−γ、IL−2、IL−12、モノホスホリルリピドA(MPL)、トレハロースジミコラート(TDM)、トレハロースジベヘナート(TDB)、ムラミルジペプチド(MDP)及びモノミコリルグリセロール(MMG)又はこれらの組合せからなる群から選択されることを特徴とするワクチン。
- 毒性マイコバクテリア(mycobacteria)によって、例えば、結核菌(Mycobacterium tuberculosis)、アフリカ型結核菌(Mycobacterium africanum)又はウシ型結核菌(Mycobacterium bovis)によって引き起こされる結核に対してヒトを含む動物を免疫する方法において、請求項8乃至10の何れか1項に記載のワクチンを前記動物に投与するステップを含むことを特徴とする方法。
