Description

〔00228〕 ビオチン化アプタマーを、PBDCプライマー(5’光切断ビオチン、D−スペーサー及びcy3)を用いるプライマー伸長法によって、酵素的に調製した。固定のため、1pmolのPBDCアプタマーを20μlのMyOneストレプトアビジンC1ビーズ(10mg ml−1)に添加し、15分間振とうし、非捕捉上清画分中cy3測定に基づいて約90%効率の固定を行った。細菌は、LBブロス培養または5%ヒツジ血液及び0.1mmol l−1ジピリジルを含むトリプシンソイ寒天にて16時間35℃で増殖させ、鉄分制限条件を作り出した。50μlのSB18T中106細菌まで含有する細胞懸濁液を捕捉ビーズに添加した。振とうしながら1時間37℃でインキュベーションした後、ビーズを洗浄し、50μlのSB18Tに再懸濁した。非捕捉上清中の細胞、洗浄画分、及びビーズ上細胞を、段階希釈物を定量的にLBアガーへ播種して計数した。定量培養による捕捉効率を混合集団及びある範囲の細胞密度(101〜107CFU ml−1)について決定した。

Claims (63)
- 試料中の微生物の有無の検出方法であって、
a)前記試料と第一アプタマーを接触させ混合物を形成し、ここで前記第一アプタマーは前記微生物の第一細胞表面タンパク質に結合して複合体を形成可能であり、かつ第一タグを含み、ここで前記第一タグは固体支持体に結合可能であり、
b)前記第一タグの前記固体支持体への結合を可能にする条件下で、前記混合物と前記固体支持体を接触させ、
c)前記固体支持体を洗浄して前記複合体に関して前記混合物を濃縮し、及び/または前記固体支持体を洗浄して未結合物質を実質的に除去し、
d)前記混合物と第二アプタマーを接触させ、ここで前記第二アプタマーは前記微生物の前記第一細胞表面タンパク質または第二細胞表面タンパク質に結合可能であり、及び
e)アッセイを行って前記第二アプタマーを検出し、ここで前記第二アプタマーの検出は前記微生物が前記試料中に存在することを示し、前記第二アプタマーの未検出は前記微生物が前記試料中に存在しないことを示す、ことを含む、検出方法。 - 試料中の微生物の有無の検出方法であって、
a)前記試料を固体支持体に接触させ、ここで第一アプタマーは第一タグを介して前記固体支持体に結合し、及びここで前記第一アプタマーは前記微生物の第一細胞表面タンパク質に結合して複合体を形成可能であり、
b)前記固体支持体を洗浄して前記複合体に関して前記混合物を濃縮し、及び/または前記固体支持体を洗浄して未結合材料を実質的に除去し、
c)前記混合物を第二アプタマーと接触させ、ここで前記第二アプタマーは前記微生物の前記第一細胞表面タンパク質または第二細胞表面タンパク質に結合可能であり、かつ第二タグを含み、及び
d)アッセイを行って前記第二アプタマーを検出し、ここで前記第二アプタマーの検出は前記微生物が前記試料中に存在することを示し、前記第二アプタマーの未検出は前記微生物が前記試料中に存在しないことを示すことを含む、検出方法。 - 請求項1及び2に記載の方法であって、前記第一アプタマー及び前記第二アプタマーの少なくとも1つはさらに少なくとも1つのC−5修飾ピリミジンを含む、前記方法。

- 前記細菌細胞がブドウ球菌細胞である、請求項48に記載の核酸分子。
- 前記細菌細胞が黄色ブドウ球菌細胞である、請求項48に記載の核酸分子。
- 前記細胞表面タンパク質が細菌細胞表面タンパク質である、請求項40に記載の核酸分子。
- 請求項40に記載の核酸分子であって、前記細胞表面タンパク質が、SPA、ClfA、ClfB、FnbA、FnbB、IsdA、IsdB、IsdC、IsdH及びSasDからなる群より選択される、前記核酸分子。
- 請求項40に記載の核酸分子であって、前記第一細胞表面タンパク質が、SPA及びClfAからなる群より選択される、前記核酸分子。
- 請求項40に記載の核酸分子であって、GGCWWCGGGWACCWAWWAWNGGWWWAGCC(N)nGWC(配列番号14)の配列を含み、ここでWがそれぞれの存在について独立してC−5修飾ピリミジン、Nが非修飾または修飾ヌクレオチドのいずれか、及びnが0、1、2、3、4または5である、前記核酸分子。
- 請求項55に記載の核酸分子であって、その長さが少なくとも約32、33、34、35、36、37、38、39、40、41、42、43、44、45、46、47、48、49または50個のヌクレオチドである、前記核酸分子。
- 請求項55に記載の核酸分子であって、その長さが、約32〜約100個のヌクレオチド(または32、33、34、35、36、37、38、39、40、41、42、43、44、45、46、47、48、49、50、51、52、53、54、55、56、57、58、59、60、61、62、63、64、65、66、67、68、69、70、71、72、73、74、75、76、77、78、79、80、81、82、83、84、85、86、87、88、89、90、91、92、93、94、95、96、97、98、99もしくは100個のヌクレオチド)である、前記核酸分子。
- 請求項40に記載の方法であって、前記核酸分子が、AWCWGGWWC(N)nAWCWGGWWWWWAAG(配列番号15)の配列を含み、ここでWがそれぞれの存在について独立してC−5修飾ピリミジン、Nが非修飾または修飾ヌクレオチドのいずれか、及びnが0、1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、26、27、28、29または30である、前記核酸分子。
