定義
本発明をより容易に理解し得るように、特定の技術および学術用語を以下で明確に定義する。本書類の別の箇所で明確に定義されない限り、本明細書で使用される他のすべての技術および学術用語は、本発明が属する技術分野の当業者によって一般的に理解される意味を有する。
本発明は、イヌPD−1に結合する単離された抗体(特にマウス抗イヌPD−1抗体およびそのイヌ化抗体)またはその抗原結合フラグメントならびにそのような抗体またはそのフラグメントの使用を提供する。具体的な実施形態では、イヌPD−1に結合することおよびイヌPD−1のそのリガンドであるイヌPD−L1への結合をブロックすることの両方が示されている、マウス抗イヌPD−1抗体由来のマウス抗イヌPD−1 CDRが提供される。これらのCDRをイヌ抗体の修飾されたイヌフレームに挿入して、イヌ化マウス抗イヌPD−1抗体を作製することができる。

本発明はさらに、本明細書で開示されるマウス抗イヌPD−1抗体と同じイヌPD−1のエピトープのアミノ酸残基に結合する抗体またはその抗原結合フラグメントを提供する。特定の実施形態では、マウス抗イヌPD−1抗体またはその抗原結合フラグメントはまた、イヌPD−1のイヌPD−L1への結合を阻害する/ブロックすることもできる。
本発明のイヌ化マウス抗イヌPD−1抗体は、例えば抗体の特性を改善するために、親(すなわちイヌ)モノクローナル抗体の可変ドメイン内のイヌフレームワークおよび/またはイヌフレーム残基への修飾を含有するように操作することができる。
本発明のマウス抗イヌPD−1抗体および/もしくはイヌ化マウス抗イヌPD−1抗体またはその抗原結合フラグメントはまた、イヌPD−1タンパク質の診断アッセイ、例えば特定の腫瘍細胞、組織または血清中でのその発現を検出する診断アッセイにおいても有用であり得る。そのような診断方法は、様々な疾患診断、特にイヌにおける特定の癌の診断において有用であり得る。
(a)マウス抗イヌPD−1抗体またはその抗原結合フラグメントで基質(例えばマイクロタイタープレートのウェル、例えばプラスチックプレートの表面)を被覆する段階;
(b)イヌPD−1の存在に関して試験する試料を基質に適用する段階;

(i)結合イヌPD−1またはそのフラグメントの存在に関して試験する膜または固体基質を本発明のマウス抗イヌPD−1抗体またはその抗原結合フラグメントと接触させること。そのような膜は、非変性PAGE(ポリアクリルアミドゲル電気泳動)ゲルまたはSDS−PAGE(ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動)ゲル中のイヌPD−1の存在に関して試験するタンパク質が転写された(例えばゲル中での電気泳動分離後)ニトロセルロースまたはビニルベースの[例えばポリビニリデンフルオライド(PDVF)]膜の形態を取り得る。マウス抗イヌPD−1抗体またはその抗原結合フラグメントと膜の接触前に、膜上の非特異的タンパク質結合部位に結合させるために、例えば脱脂粉乳等で膜をブロックしてもよい。

イヌ化マウス抗イヌPD−1抗体またはその抗原結合フラグメントの医薬または滅菌組成物を調製するために、イヌ化マウス抗イヌPD−1抗体またはその抗原結合フラグメントを医薬的に許容される担体または賦形剤と混合することができる。[例えばRemington’s Pharmaceutical SciencesおよびU.S.Pharmacopeia:National Formulary,Mack Publishing Company,Easton,PA(1984)参照]。
先に述べたように、本発明のイヌ化マウス抗イヌPD−1抗体もしくはその抗原結合フラグメントおよび/または抗原ペプチドは、1またはそれ以上の他の治療薬(例えば化学療法剤)と同時投与し得る。抗体を他の治療薬に連結してもよく(免疫複合体として)または他の治療薬とは別に投与することができる。後者(別々の投与)の場合は、抗体を他の治療薬の前、後もしくは治療薬と同時に投与することができるかまたは他の公知の療法と同時投与することができる。
さらに、PD−1に特異的に結合する、本明細書で論じる抗体または抗原結合フラグメント(例えばイヌ化マウス抗イヌPD−1抗体またはその抗原結合フラグメント)を含むがこれらに限定されない1またはそれ以上の成分を、本明細書で論じる、医薬的に許容される担体および/または化学療法剤を含むがこれらに限定されない1またはそれ以上の付加的な成分と共に含有するキットが提供される。結合組成物および/または化学療法剤は、純粋な組成物としてまたは医薬的に許容される担体と組み合わせて医薬組成物に製剤することができる。
[実施例1]
イヌPD−1およびPD−L1
イヌPD−1の同定とクローニング:
完全長イヌPD−1(cPD−1)をコードする核酸を、NCBI遺伝子バンクデータベース(アクセッション番号XM_543338.4、配列番号:1)の検索を通して同定した。翻訳されたアミノ酸配列、配列番号:2(アクセッション番号XP−543338.3)は、遺伝子バンク(NCBI)タンパク質データベースを検索し、同定されたアミノ酸配列をマウス、ネコおよびヒトPD−1アミノ酸配列と整列することを通してさらに同定した、推定上のイヌPD−1タンパク質に対応する。CHO細胞についてコドン最適化した、完全長イヌPD−1遺伝子に対応するDNA配列を合成し、p96793と称するプラスミドにクローニングした。予測されるイヌPD−1のDNAおよびタンパク質配列と公知のPD−1 DNAおよびタンパク質配列との比較は、イヌPD−1の細胞外ドメイン(ECD)をコードするDNA配列(配列番号:3)およびイヌPD−1のECDのアミノ酸配列(配列番号:4)の同定をもたらした。
完全長イヌPD−L1をコードする核酸を、NCBI遺伝子バンクデータベース(アクセッション番号XM_541302.4;配列番号:7)の検索を通して同定した。推定上のイヌPD−1タンパク質に対応する翻訳されたアミノ酸配列(アクセッション番号XP−541302.4;配列番号:8)は、遺伝子バンク(NCBI)タンパク質データベースを検索し、同定された配列と公知のPD−L1マウスおよびヒト配列とのアラインメントによって同定した。
PD−1 ECD−HIS、PD−1 ECD−Fc、PDL−1 ECD−HISおよびPD−L1 ECD−Fcタンパク質をコードする発現プラスミドをHEK 293細胞にトランスフェクトし、Fc融合タンパク質についてはプロテインAまたはHISタグタンパク質についてはニッケル(Ni2+)カラムクロマトグラフィを使用してトランスフェクト細胞の上清からタンパク質を精製した。精製したタンパク質を以下で詳述するようにELISAまたは結合アッセイに使用した。発現したタンパク質をSDS−PAGEゲルによって分析した。

抗イヌPD−1抗体
抗イヌPD−1モノクローナル抗体の作製:
合計3匹のBalb/cマウスを17日間にわたって複数回免疫した(毎回10μgで)。免疫する抗原はイヌPD−1 ECD−Fc融合タンパク質であった。免疫後、各々のマウスから血清を採取し、イヌPD−1 ECD−HISタグタンパク質との反応性に関して試験した。最も高い血清抗PD−1 ECD−HIS力価を有するマウスの脾細胞を骨髄腫P3X63Ag8.653細胞株に融合した。融合の約2週間後に、推定上のハイブリドーマ細胞からの上清を、PD−1 ECD−HISタグタンパク質に対するそれらの反応性に関してELISAによって試験した。ELISAにおいて強い陽性シグナルを生じるハイブリドーマを限界希釈によってサブクローニングし、イヌPD−1 ECD−HISタグタンパク質に対する反応性に関して再び試験した。
ハイブリドーマによって分泌される抗体の、イヌPD−1のECDに対する反応性をELISAによって確認した。ハイブリドーマ細胞を、CELLineバイオリアクター(Integra−biosciences)を使用して10〜30日間培養した。細胞を最初に、4mM L−グルタミンおよびGibcoからの10%Ultra Low IgGウシ胎仔血清(FBS)を添加したDMEM中で維持した。ハイブリドーマ細胞を、FBS濃度を20%に増加した同じ培地15mL中約2×106細胞/mLの細胞密度でCELLineバイオリアクターの細胞チャンバーに接種した。外側チャンバーに栄養培地(4mM L−グルタミンおよび2%標準FBSを添加したDMEM)1Lを満たした。細胞チャンバー中のハイブリドーマ細胞を3〜7日間かけて約2.5×107細胞/mLに増殖させた。次に、細胞懸濁液10mLを細胞チャンバーから採取し、新鮮培地と交換して、細胞を再増殖させ、その後採取した。この手順を必要に応じて反復し、各ハイブリドーマクローンから適切な量のmAbを得た。採取した細胞懸濁液を遠心分離し、上清を0.2ミクロンフィルター膜でろ過した。抗体精製のために、各クローンの上清を、Protein G Sepharose 4 Fast flow 5mLカラム(GE Healthcare)を使用して重力流によって精製した。Tris−EDTA(TE)緩衝液pH8.0で洗浄した後、0.1Mグリシン緩衝液、pH2.7を使用して結合抗体を溶出し、次いで1M Tris、pH8.0を使用してpHを中和した。抗体を濃縮し、Centriprep YM−10、10kDa NMWL遠心分離フィルターユニット(Millipore)を使用して緩衝液をリン酸緩衝生理食塩水(PBS)に交換した。分光測光法を用いて抗体濃度を定量した。
完全長イヌPD−1遺伝子をプラスミドp96793にクローニングした。このプラスミドにおいてPD−1タンパク質の発現はhCMVプロモーターによって駆動される。CHO DXB11細胞(dhfr−)を、10%ウシ胎仔血清を添加したMEM−α(Gibco)中で維持した。プラスミドp96793によるCHO細胞のトランスフェクションを、約6×106細胞を含む75cm2フラスコ中でリポフェクタミン(Invitrogen)を使用したリポソーム媒介性遺伝子送達によって実施した。48時間後、10%FBSおよび400μg/mLハイグロマイシンBを添加した、ヌクレオシドを含有しないMEM−α培地(選択培地)に細胞を継代した。dhfr+のハイグロマイシン耐性細胞のプールに関して限界希釈クローニングを実施した。クローンを免疫蛍光アッセイによってイヌPD−1の発現に関して評価した。簡単に述べると、細胞単層を80%アセトンで96ウェルプレートに固定した。次に、固定して乾燥した細胞単層をポリクローナルヤギ抗ヒトPD−1抗体(R&D Systems)と共に1時間インキュベートした。プレートをPBSで洗浄し、次にフルオレセイン標識ウサギ抗ヤギIgG抗体(KPL)と共に1時間インキュベートした。プレートをPBSで洗浄した。蛍光を示すクローンを増殖させ、細胞株を樹立した。
CHO細胞上のイヌPD−1とマウス抗イヌPD−1 mAbの反応性を、PD−1を発現するCHO細胞を使用した細胞ベースのアッセイによって測定した。簡単に述べると、イヌPD−1を発現するCHO細胞を培地(DMEM/HAM F12、10%FBS)50μl中で80〜100%の集密度まで培養した。次に、様々な濃度の精製mAbを含有する培地50μlを37℃で1時間添加した。PBS−Tweenで3回洗浄した後、培地中で1:1000希釈したヤギ抗マウスホースラディッシュペルオキシダーゼ(HRP)100μlを37℃で1時間添加した。PBS−Tweenでさらに3回洗浄した後、結合mAbをペルオキシダーゼ(perioxidase)基質(TMB)で視覚化した。450nmでのペルオキシダーゼ(perioxidase)活性による吸光度の増加をマイクロプレートリーダーで測定した。
イヌPD−1抗原の約70共鳴単位(RU)をアミンカップリングによって直接固定化した。親和性測定を、結合時間300秒、解離時間1200秒ならびに50、100、200(×2)、400および800ナノモル(nM)の濃度で無標識表面プラズモン共鳴ベースの技術(例えばBiacore(登録商標)T200)によって実施した。1:1結合の適合モデルを使用した。抗原(イヌPD−1)を、アミンカップリングを介してセンサーチップ上に固定化し、以下の表5に示す4つの抗体を、抗原表面を流れる分析物として使用した。結果は、イヌPD−1抗原についての本発明の抗イヌPD−1抗体の結合親和性が強力であり、ナノモルおよびさらにはサブナノモルの解離定数(Kd)を有することを明らかにした。さらに、マウス抗イヌPD−1モノクローナル抗体および同じクローン由来の対応するイヌ化マウス抗イヌPD−1モノクローナル抗体は、極めて類似のKd値を生じた(以下の表5参照)。
イヌPD−1を発現するCHO細胞株に基づく細胞ベースのELISA(CELISA)アッセイを、イヌPD−1(cPD−1)と反応するマウスmAbに使用した。ビオチン化cPD−L1/Fcタンパク質と共にこのアッセイを使用してリガンド遮断を確認した。簡単に述べると、種cPD−1 CHO細胞を4×104細胞/ウェルで96ウェルプレートに入れ、細胞が95〜100%集密になるまで37℃で18〜24時間細胞をインキュベートした。細胞培養培地を吸引除去し、プレートをPBS+0.05%Tween20で3回およびCHO培地で1回洗浄した。30μg/mLから出発して、CHO培地中で抗cPD−1 mAbの3倍段階希釈物を作製し、各抗体希釈物50μL/ウェルをプレートに添加した。37℃、5%CO2で振とうしながら30分間インキュベーションを実施した。cPD−L1−Fc−ビオチン(CHO培地ストック中2μg/ml)50μL/ウェルを添加し、37℃、5%CO2で振とうしながら45分間インキュベーションを続けた。プレートをPBS+0.05%Tween20で6回洗浄した。CHO培地中のストレプトアビジン−HRP(1:2000)100ul/ウェルを添加し、次いで37℃/5%CO2で30〜60分間インキュベートした。プレートをPBS+0.05%Tween20で5回洗浄し、次にTMB発色基質100μl/ウェルを添加した。1Mリン酸50μl/ウェルを添加することによって発色を停止させた。ELISAプレートリーダーを使用してA450〜A620での光学密度(O.D.)を測定した。
PBMCを、Ficoll分離を使用して健常イヌおよび癌を有するイヌから得たEDTA血液試料より調製した。PBMCを、2.5×105細胞/ウェルの濃度で添加したFACS緩衝液(PBS、1%FBSおよび0.1%アジ化ナトリウム)に再懸濁し、様々な濃度の試験モノクローナル抗体(mAb)と共にインキュベートした。細胞を室温で30分間インキュベートし、その後2回洗浄した。次に細胞を再懸濁し、Alexa−488結合ロバ抗マウスIgG(H+L鎖)と共に室温で30分間インキュベートして、その後2回洗浄した。次に細胞をイヌCD4およびCD8に対するPBおよびPE結合抗体と共に30分間インキュベートし、その後洗浄した。次に細胞をFACS緩衝液に再懸濁し、PD−1 mAbの結合に関して陽性のCD4またはCD8 T細胞のパーセンテージを決定するためにフローサイトメトリによって分析した。対照には、二次抗体だけまたは無関係なアイソタイプがマッチするmAbと共にインキュベートした細胞が含まれた。
PBMCを、Ficoll分離を使用して健常イヌおよび癌を有するイヌから得たEDTA血液試料より調製した。細胞を3回洗浄し、96ウェルプレート中の三つ組みのウェルにおいて2.5×105細胞/ウェルの濃度で完全組織培養培地に再懸濁した。細胞をコンカナバリンA 1μg/mlで活性化した。試験抗体を様々な濃度で添加し、培養物を96時間インキュベートした。対照には、抗体なしでconAと共に、またはconAおよび無関係なアイソタイプがマッチする抗体と共にインキュベートした細胞が含まれた。96時間の培養後、上清を採取し、市販のイヌIFN−γ ELISAキット(R&D Systems)を使用してIFN−γ放出に関して検定した。
マウスVHおよびVL鎖のDNA配列ならびにそれらのCDRをコードするDNA配列を、標準的な分子生物学方法を用いて各ハイブリドーマからのmRNAの単離後に同定する。これらのハイブリドーマからのCDRの予測されるアミノ酸配列の配列番号を以下に列挙する:

[実施例3]
PD−1に特異的な突然変異体イヌIgG−B抗体
イヌIgGの4つの公知のIgG重鎖サブタイプが存在し、それらはIgG−A、IgG−B、IgG−CおよびIgG−Dと称される。2つの公知の軽鎖サブタイプはλおよびκと称される。しかし、イヌ免疫細胞の結合および活性化に加えて、PD−1に対するイヌまたはイヌ化抗体は2つの属性を有する:
1.抗体依存性細胞傷害(ADCC)および補体依存性細胞傷害(CDC)などのエフェクター機能の欠如、ならびに
2.プロテインAクロマトグラフィに基づくもののような業界標準技術を用いて大規模で容易に精製される。

抗イヌPD−1抗体のエピトープマッピング
緒言
抗体とそれらの同種タンパク質抗原との相互作用は、標的抗原の特定アミノ酸(エピトープ)と抗体の特定アミノ酸(パラトープ)との結合を通して媒介される。エピトープは、免疫グロブリンによる特異的反応を生じさせる抗原決定基である。エピトープは抗原の表面の一群のアミノ酸から成る。関心対象のタンパク質は、異なる抗体によって認識されるいくつかのエピトープを含有し得る。
抗PD−1 mAbについてのエピトープを形成するアミノ酸を同定するために、15アミノ酸長であり、10個のアミノ酸がオーバーラップしている合計28個のペプチドを化学的に合成した。オーバーラップするペプチドのこのライブラリは、完全長イヌPD−1タンパク質をカバーするように設計された。これらのペプチドの配列を以下の表6に列挙する。ペプチド−抗体結合の測定を、抗体試料をProArray Ultra(登録商標)ペプチドマイクロアレイに取り付け、次いで蛍光標識二次抗体と共にインキュベートすることによって実施した。すべてのペプチドを別々に合成し、その後ProImmune(登録商標)マウスIgG対照と共にProArray Ultra(登録商標)スライド表面に結合する。この最適化工程は、ペプチドが、対応するタンパク質領域の特性を密接に模倣し、遊離ペプチド自体の固有の物理化学的変動を回避して、適合するペプチドとタンパク質の組合せアレイプラットフォームを生成するように、ペプチドをアレイ上に提示することを確実にする。試験分析物(ペプチド)を別々のスポットでProArray Ultra(登録商標)スライドに分注し、適切なgalファイルは、生じたアレイ特徴を沈殿した分析物に正確に整列することを可能にする。mAbの非特異的結合を低減するために有効なブロッキング緩衝液を使用してProArray Ultra(登録商標)スライドをブロックした。次にそれらをmAb試料と共にインキュベートし、次いで特異的蛍光標識二次抗体と共にインキュベートした。数回の洗浄段階後、ProArray Ultra(登録商標)アレイを乾燥し、高分解能蛍光マイクロアレイスキャニングシステムを用いて走査した。蛍光標識ProArray Ultra(登録商標)スライドを走査した後、スキャナーが画像を記録し、これを、走査したマイクロアレイスライド上の各蛍光スポットに関連する蛍光強度のレベルの解釈および定量化を可能にする、画像解析ソフトウエアを用いて評価した。この実験の結果は、イヌPD−1ペプチドの一部が評価したmAbの一部によって認識されることを示した。mAbの同一性およびこれらのmAbによって認識されるアミノ酸配列を表7に列挙する。この試験は、mAb 2H9が、配列番号:84によって表されるアミノ酸配列から成るイヌPD−1の細胞外ドメインに位置するエピトープを認識することおよびmAb 1A1が、配列番号:84によって表されるアミノ酸配列および配列番号:83によって表されるアミノ酸配列によって表されるオーバーラップするアミノ酸配列を含有するエピトープを認識することを指し示す。
抗イヌPD−1によって認識される潜在的に不連続なエピトープを同定するために、化学的架橋および質量分析検出に基づく方法を使用した(CovalX(登録商標)Instrument Incorporated)。イヌPD−1のエピトープマッピングへのこの技術の適用は、表8に列挙されている指示mAbによって認識されるエピトープの少なくとも部分の同定をもたらした。表8からわかるように、mAb 3B6は、配列番号:99によって表されるアミノ酸配列内のイヌPD−1の細胞外ドメインに位置するエピトープの少なくとも一部分を認識し、およびmAb 2G9は、配列番号:100によって表されるアミノ酸配列内のエピトープの少なくとも一部分を認識する。他方で、mAb 1E4およびmAb 1B5は、それぞれ配列番号:101によって表されるアミノ酸配列および配列番号:102によって表されるアミノ酸配列内のエピトープの少なくとも一部分を認識する。

(a)CDRL1が、配列番号:13、配列番号:13の保存的に修飾された変異体、および3の正準構造クラスを構成する配列番号:13の変異体から成る群より選択されるアミノ酸配列を含有し;
(b)CDRL2が、配列番号:20、配列番号:20の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:20の変異体から成る群より選択されるアミノ酸配列を含有し;
(c)CDRL3が、配列番号:25、配列番号:25の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:25の変異体から成る群より選択されるアミノ酸配列を含有し;
(d)CDRH1が、配列番号:27、配列番号:27の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:27の変異体から成る群より選択されるアミノ酸配列を含有し;

(a)CDRL1が、配列番号:13、配列番号:13の保存的に修飾された変異体、および3の正準構造クラスを構成する配列番号:13の変異体から成る群より選択されるアミノ酸配列を含有し;
(b)CDRL2が、配列番号:16、配列番号:16の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:16の変異体から成る群より選択されるアミノ酸配列を含有し;
(c)CDRL3が、配列番号:22、配列番号:22の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:22の変異体から成る群より選択されるアミノ酸配列を含有し;
(d)CDRH1が、配列番号:30、配列番号:30の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:30の変異体から成る群より選択されるアミノ酸配列を含有し;
(e)CDRH2が、配列番号:31、配列番号:31の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:31の変異体から成る群より選択されるアミノ酸配列を含有し;

(a)CDRL1が、配列番号:15、配列番号:15の保存的に修飾された変異体、および2Aの正準構造クラスを構成する配列番号:15の変異体から成る群より選択されるアミノ酸配列を含有し;
(b)CDRL2が、配列番号:18、配列番号:18の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:18の変異体から成る群より選択されるアミノ酸配列を含有し;
(c)CDRL3が、配列番号:24、配列番号:24の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:24の変異体から成る群より選択されるアミノ酸配列を含有し;
(d)CDRH1が、配列番号:29、配列番号:29の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:29の変異体から成る群より選択されるアミノ酸配列を含有し;
(e)CDRH2が、配列番号:33、配列番号:33の保存的に修飾された変異体、および2Aの正準構造クラスを構成する配列番号:33の変異体から成る群より選択されるアミノ酸配列を含有し;

(a)CDRL1が、配列番号:15、配列番号:15の保存的に修飾された変異体、および2Aの正準構造クラスを構成する配列番号:15の変異体から成る群より選択されるアミノ酸配列を含有し;
(b)CDRL2が、配列番号:21、配列番号:21の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:21の変異体から成る群より選択されるアミノ酸配列を含有し;
(c)CDRL3が、配列番号:26、配列番号:26の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:26の変異体から成る群より選択されるアミノ酸配列を含有し;
(d)CDRH1が、配列番号:29、配列番号:29の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:29の変異体から成る群より選択されるアミノ酸配列を含有し;
(e)CDRH2が、配列番号:35、配列番号:35の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:35の変異体から成る群より選択されるアミノ酸配列を含有し;
(f)CDRH3が、配列番号:114、配列番号:114の保存的に修飾された変異体、および11の正準構造クラスを構成する配列番号:114の変異体から成る群より選択されるアミノ酸配列を含有し、ならびに
前記抗体およびその抗原結合フラグメントがイヌPD−1に結合し、およびイヌPD−1のイヌプログラム死リガンド1(PD−L1)への結合をブロックする、
単離された哺乳動物抗体またはその抗原結合フラグメント。 - 3つの軽鎖相補性決定領域(CDR):CDR軽鎖1(CDRL1)、CDR軽鎖2(CDRL2)およびCDR軽鎖3(CDRL3);ならびに3つの重鎖CDR:CDR重鎖1(CDRH1)、CDR重鎖2(CDRH2)およびCDR重鎖3(CDRH3)を含有する、イヌプログラム死受容体1(イヌPD−1)に特異的に結合する単離された哺乳動物抗体またはその抗原結合フラグメントであって、
(a)CDRL1が、配列番号:14、配列番号:14の保存的に修飾された変異体、および4の正準構造クラスを構成する配列番号:14の変異体から成る群より選択されるアミノ酸配列を含有し;
(b)CDRL2が、配列番号:17、配列番号:17の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:17の変異体から成る群より選択されるアミノ酸配列を含有し;
(c)CDRL3が、配列番号:23、配列番号:23の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:23の変異体から成る群より選択されるアミノ酸配列を含有し;
(d)CDRH1が、配列番号:28、配列番号:28の保存的に修飾された変異体、および1の正準構造クラスを構成する配列番号:28の変異体から成る群より選択されるアミノ酸配列を含有し;
(e)CDRH2が、配列番号:32、配列番号:32の保存的に修飾された変異体、および2Aの正準構造クラスを構成する配列番号:32の変異体から成る群より選択されるアミノ酸配列を含有し;
(f)CDRH3が、配列番号:37、配列番号:37の保存的に修飾された変異体、および13の正準構造クラスを構成する配列番号:37の変異体から成る群より選択されるアミノ酸配列を含有し、ならびに
前記抗体およびその抗原結合フラグメントがイヌPD−1に結合し、およびイヌPD−1のイヌプログラム死リガンド1(PD−L1)への結合をブロックする、
単離された哺乳動物抗体またはその抗原結合フラグメント。 - 前記哺乳動物抗体がマウス抗体である、請求項1、2、3、4、5、6、7または8に記載の単離された哺乳動物抗体またはその抗原結合フラグメント。
- イヌ化抗体である、請求項1、2、3、4、5、6、7、8または9に記載の単離された哺乳動物抗体またはその抗原結合フラグメント。
- 以下の特性:
(i)1×10−5Mから1×10−12Mの解離定数(Kd)でイヌPD−1に結合する;
(ii)1×102M−1秒−1から1×107M−1秒−1のオン速度(kon)でイヌPD−1に結合する;
(iii)1×10−3秒−1から1×10−8秒−1のオフ速度(koff)でイヌPD−1に結合する;
(iv)腫瘍または病原体に対する抗原特異的記憶応答を刺激する;
(v)インビボで抗体応答を刺激する;および
(vi)動物被験体における免疫応答を刺激する
のうちの1、2、3、4、5または全部を示す、請求項1、2、3、4、5、6、7、8、9または10に記載の哺乳動物抗体またはその抗原結合フラグメント。 - イヌプログラム死受容体1(イヌPD−1)に特異的に結合する単離された哺乳動物抗体またはその抗原結合フラグメントであって、イヌPD−1に結合する場合、前記抗体が配列番号:103内の少なくとも1個のアミノ酸残基に結合し、前記抗体およびその抗原結合フラグメントがイヌPD−1に結合し、およびイヌPD−1のイヌプログラム死リガンド1(PD−L1)への結合をブロックする、単離された哺乳動物抗体またはその抗原結合フラグメント。
- 請求項1、2、3、4、5、6、7、8、9、10または11に記載の単離された哺乳動物抗体またはその抗原結合フラグメントであって、イヌPD−1に結合する場合、前記抗体が配列番号:103内の少なくとも1個のアミノ酸残基に結合し、前記抗体およびその抗原結合フラグメントがイヌPD−1に結合し、およびイヌPD−1のイヌプログラム死リガンド1(PD−L1)への結合をブロックする、単離された哺乳動物抗体またはその抗原結合フラグメント。
- 請求項12または13に記載の単離された哺乳動物抗体またはその抗原結合フラグメントであって、イヌPD−1に結合する場合、前記抗体が、配列番号:83、配列番号:84、配列番号:99、配列番号:100、配列番号:101、配列番号:102および配列番号:104から成る群より選択される1またはそれ以上のアミノ酸配列内の少なくとも1個のアミノ酸残基に結合する、単離された哺乳動物抗体またはその抗原結合フラグメント。
- 請求項12、13または14に記載の単離された哺乳動物抗体またはその抗原結合フラグメントであって、イヌPD−1に結合する場合、前記抗体が配列番号:104内の少なくとも1個のアミノ酸残基に結合する、単離された哺乳動物抗体またはその抗原結合フラグメント。
- 請求項12、13、14または15に記載の単離された哺乳動物抗体またはその抗原結合フラグメントであって、イヌPD−1に結合する場合、前記抗体が、配列番号:2のR62、R69、R72、R75およびR90から成る群より選択される1個またはそれ以上のアミノ酸残基に結合する、単離された哺乳動物抗体またはその抗原結合フラグメント。
- 請求項1、2、3、4、5、6、7、8、9、10、11、12、13、14、15または16に記載の1またはそれ以上の哺乳動物抗体と、イヌPD−1との結合に関して交差競合するモノクローナル抗体またはその抗原結合フラグメントであって、前記抗体およびその抗原結合フラグメントが、イヌPD−1に結合し、およびイヌPD−1のPD−L1への結合をブロックする、モノクローナル抗体またはその抗原結合フラグメント。
- 請求項1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16または17に記載の抗体の軽鎖をコードする単離された核酸。
- 請求項1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16または17に記載の抗体の重鎖をコードする単離された核酸。
- 配列番号:13、14、15、16、17、18、19、20、21、22、23、24、25、26、27、28、29、30、31、32、33、34、35、36、37、38または114から成る群より選択される1またはそれ以上のアミノ酸配列をコードする、請求項18または19に記載の単離された核酸。
- 請求項18、19または20に記載の単離された核酸を含有する発現ベクター。
- 請求項21に記載の1またはそれ以上の発現ベクターを含有する宿主細胞。
- 配列番号:103のアミノ酸配列を含有する60個またはそれ未満のアミノ酸残基から成る単離された抗原ペプチド。
- 配列番号:103と95%同一であるアミノ酸配列を含有し、および請求項1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16または17に記載の単離された哺乳動物抗体またはその抗原結合フラグメントに結合する、60個またはそれ未満のアミノ酸残基から成る単離された抗原ペプチド。
- 前記アミノ酸配列が、配列番号:83、配列番号:84、配列番号:99、配列番号:100、配列番号:101、配列番号:102および配列番号:104から成る群より選択される、請求項23または24に記載の抗原ペプチド。
- 請求項23、24または25に記載の抗原ペプチドを含有する融合タンパク質。
- 非イヌ哺乳動物IgG抗体のFc領域も含有する、請求項26に記載の融合タンパク質。
- 前記非イヌ哺乳動物IgG抗体が、マウス、ヒト、ウマ、ブタおよびウシから成る群より選択される供給源に由来する、請求項27に記載の融合タンパク質。
- 前記非イヌ哺乳動物IgG抗体が、IgG1、IgG2a、IgG3およびIgG4から成る群より選択される、請求項27または28に記載の融合タンパク質。
- 請求項23、24もしくは25に記載の抗原ペプチド、または請求項26、27、28もしくは29に記載の融合タンパク質をコードする単離された核酸。
- 請求項30に記載の単離された核酸を含有する発現ベクター。
- 請求項1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16もしくは17に記載の抗体、または請求項23、24および25に記載の抗原ペプチド、または請求項26、27、28もしくは29に記載の融合タンパク質、または請求項18、19、20に記載の核酸、または請求項21もしくは31に記載の発現ベクター、またはそれらの任意の組合せ、および医薬的に許容される担体または希釈剤を含有する医薬組成物。
- 免疫細胞の活性を増大させる方法であって、それを必要とする被験体に、請求項32に記載の医薬組成物の治療有効量を投与することを含む方法。
- (i)癌の処置;
(ii)感染もしくは感染性疾患の処置;
(iii)ワクチンアジュバントとして;または
(iv)それらの任意の組合せ
のために使用される、請求項33に記載の方法。
