Abstract
Description
本明細書で用いられる場合、用語「ミニエマルジョン」とは、平均サイズが典型的には50〜500nmの範囲である安定な液滴を有する特定のタイプのエマルジョン(emulsion)のことを意味する。粒子サイズは複数の因子の影響を受ける。複数の因子には、存在する界面活性剤の量、系全体の粘度、液滴の生成に用いられる剪断速度が含まれる。ミニエマルジョンのための典型的な粒度分布曲線(たとえば、動的光散乱を用いて測定)は、ガウス状で、比較的幅が狭いものである。本発明のミニエマルジョンの多分散性指数(PDI)は、好ましくは0.3以下、さらに好ましくは0.2以下、たとえば約0.1である。
本発明のミニエマルジョン内の重合可能なモノマーは少なくとも1種類のアルキルシアノアクリレートモノマーからなる。これらは、ナノ粒子の調製における使用が広く報告されている生物分解性モノマーである。アルキルシアノアクリレートモノマーは、単官能アクリレートまたは2官能アクリレートである、すなわち単一のまたは複数のアクリレート官能基を含んでいる。いかなる線状のまたは分枝状のアルキルシアノアクリレートモノマーまたはその誘導体が用いられてもよいが、好ましいモノマーは、C1〜C10アルキルシアノアクリレート、さらに好ましいモノマーはC2〜C8アルキルシアノアクリレートである。単一の種類のモノマーが用いられてもよいしまたは異なる種類のアルキルシアノアクリレートの混合物が用いられてもよい。好ましいアルキルシアノアクリレート類としては、エチルシアノアクリレート、ブチル(n−ブチル)シアノアクリレート、イソヘキシルシアノアクリレート、オクチルシアノアクリレート、およびそれらの混合物が挙げられる。とくに、ブチルシアノアクリレート、イソヘキシルシアノアクリレートおよびオクチルシアノアクリレートが好ましい。
本発明のミニエマルジョンは、ポリエチレングリコール類(PEG)、ポリプロピレングリコール類(PPG)またはそれらの混合物からなる群から選択される少なくとも2種類のポリアルキレングリコールを含有しており、少なくとも2種類のポリアルキレングリコールのうちの少なくとも一方は、ターゲットモイエティと共有結合している。好ましくは、少なくとも2種類のポリアルキレングリコールのうちの少なくとも一方が陰イオン重合反応を開始するようになっている。
活性剤は、医療用途を有するいかなる物質、たとえば治療薬、造影剤であってもよい。活性剤は水溶性であってもよいし非水溶性であってもよいが、非水溶性であることが好ましい。活性剤が、水溶性である場合、油の中に微細パウダーとして加えられるのが一般的である。ある実施形態では、活性剤は、油相の好ましくは1〜75wt%、さらに好ましくは30〜60wt%を構成する。
本発明にかかるプロセスは、ポリ(アルキルシアノアクリレート)のホモポリマーまたはコポリマーから形成されるターゲットナノ粒子を調製することを含んでいる。かかる方法は、水中油型ミニエマルジョンの陰イオン重合を単一ステップで行うことを含んでいる。かかるミニエマルジョンは、(i)少なくとも1種類のアルキルシアノアクリレートモノマーを含む重合可能なモノマーと、(ii)ポリエチレングリコール類(PEG)、ポリプロピレングリコール類(PPG)またはそれらの混合物からなる群から選択される少なくとも2種類のポリアルキレングリコールであって、少なくとも2種類のポリアルキレングリコールのうちの少なくとも一方がターゲットモイエティと共有結合する、少なくとも2種類のポリアルキレングリコールと、(iii)任意選択的に、1種類以上の活性剤とを含んでいる。かかるミニエマルジョンは、上述の少なくとも2種類のポリアルキレングリコールを少なくとも1種類のアルキルシアノアクリレートモノマーと任意選択的な1種類以上の活性剤とを含んでいる水中油型ミニエマルジョンへ加えることにより調製されるようになっていてもよい。ある好ましい実施形態では、この加えるステップおよび陰イオン重合ステップは連続的に行なわれる、すなわち加えるステップの直後(0〜10分、たとえば0〜5分以内)に陰イオン重合ステップが実行される。
本発明にかかるプロセスにより製造されるナノ粒子は、エマルジョン重合により製造されるナノ粒子とは構造が異なっている。具体的にいえば、エマルジョン重合により製造されるナノ粒子は、活性剤がポリマーマトリクス内で物理的にかつ均一に分散しているポリマーマトリックスを有している。それに対して、ミニエマルジョン界面重合により製造されるナノ粒子はナノ粒子のコア内に活性剤が存在しポリマーのシェルにより周りを取り囲まれている小胞状のシステムである。
このようなナノ粒子およびその組成物は、医療、とくに薬物送達用途およびイメージング用途において用いることができる。したがって、本発明は医療分野において用いられる本発明にかかるナノ粒子に関するものである。さらなる実施形態では、本発明は、特定の障害および疾病の治療、防止および診断に用いられる本発明にかかるナノ粒子に関するものである。本発明に従って治療または防止されうる障害または疾病の例としては、癌、たとえば肺癌、乳癌、前立腺癌、頭頸部癌、卵巣癌、皮膚癌、睾丸癌、膵臓癌、結腸直腸癌、腎臓癌、子宮頸癌、胃腸の癌およびそれらを組み合わせたものが挙げられる。
実施例1:ミニエマルジョン法を用いて形成された約1%RGDターゲットモイエティを含むステルス粒子(PEG層内にRGDが埋め込まれた短いPEG)
溶液1(PEG/開始剤およびターゲット配位子):短い(約15PEG単位)mPEG−PPO−NH2(MW935、m=OMe保護基)を330mgと、ペプチド−PEG−PPO(H2N−PPO−PEG15...RGDfK)の70mg/mlの溶液を280μlと、蒸留水を3.2gとをガラスバイアル内で混合し、5MHClを用いてpHをpH6に調節し、N2を15分用いてガス抜きした。

溶液1(PEG/開始剤):短い(約15単位)mPEG−PPO−NH2(MW935、m=OMe保護基)を330mgと、蒸留水を3.5gとをガラスバイアル内で混合し、pHを5MHClを用いてpH6に調節し、N2を15分間用いてガス抜きした。
溶液1(PEG/開始剤/安定化物質およびターゲット配位子):Kolliphor HS 15(15PEG単位、MW960、Sugma社)を50mgと、Brij L23(23PEG単位、MW1225、Sugma社)を50mgと、c(RGDfK(PEG−PEG))(PEG=8−アミノ−3、6−ジオキサオクタン酸、MW894、Peptides International社、アメリカ)を0.85mgと、0.1MHClを8mlとをガラスバイアル内で混合した。
溶液1(PEG/開始剤/安定化物質およびターゲット配位子):Kolliphor HS 15(15PEG単位、MW960、Sugma社)を50mgと、Brij L23(23PEG単位、MW1225、Sugma社)を50mgと、c(RGDfK(PEG−PEG))(PEG=8−アミノ−3、6−ジオキサオクタン酸、MW894、Peptides International社、アメリカ)を42mgと、0.1MHClを8mlとをガラスバイアル内で混合した。

溶液1(PEG/開始剤/安定化物質):Kolliphor HS 15(15PEG単位、MW960、Sugma社)を150mgと、Brij L23(23PEG単位、MW1225、Sugma社)を150mgと、Jeffamine M−1000(MW 1000、Huntsman社)を3mgと、0.1MHClを25mlとをガラスバイアル内で混合した。
溶液1(PEG/開始剤):300mgのJeffamine M−2070(MW2000、Huntsman社)をガラスバイアル内で7mlの蒸留水に溶かし、pHを、5MHClを用いてpH6に調節した。
溶液1(PEG/開始剤):1gのJeffamine M−2070(MW2000、Huntsman社)を20mlの蒸留水にガラスバイアル内で溶かし、pHを5MHClを用いてpH6に調節した。
Claims (18)
- ポリ(アルキルシアノアクリレート)のホモポリマーまたはコポリマーから形成されるターゲットナノ粒子を調製するためのプロセスであって、
前記方法が、
水中油型ミニエマルジョンの陰イオン重合を単一ステップで行うことを含み、
前記ミニエマルジョンが、
(i)少なくとも1種類のアルキルシアノアクリレートモノマーと、
(ii)ポリエチレングリコール類(PEG)、ポリプロピレングリコール類(PPG)またはそれらの混合物からなる群から選択される少なくとも2種類のポリアルキレングリコールであって、該少なくとも2種類のポリアルキレングリコールのうちの少なくとも一方がターゲットモイエティと共有結合する、少なくとも2種類のポリアルキレングリコールと、
(iii)任意選択的に、1種類以上の活性剤とを含む、プロセス。 - ポリ(アルキルシアノアクリレート)のホモポリマーまたはコポリマーから形成されるターゲットナノ粒子を調製するためのプロセスであって、
前記方法が、
ポリエチレングリコール類(PEG)、ポリプロピレングリコール類(PPG)またはそれらの混合物からなる群から選択される少なくとも2種類のポリアルキレングリコールを水中油型ミニエマルジョンへ加えることと、
得られた混合物を陰イオン重合により重合することとを含み、
前記水中油型ミニエマルジョンが、
(i)少なくとも1種類のアルキルシアノアクリレートモノマーと、
(ii)任意選択的に、1種類以上の活性剤とを含む、プロセス。 - 前記水中油型ミニエマルジョンが、前記少なくとも1種類のアルキルシアノアクリレートモノマーと任意選択的に前記1種類以上の活性剤とを含む水中油型ミニエマルジョンに前記少なくとも2種類のポリアルキレングリコールを加えることにより調製される、請求項1に記載のプロセス。
- 前記少なくとも2種類のポリアルキレングリコールのうちの少なくとも一方が前記陰イオン重合反応を開始する、請求項1乃至3のうちのいずれか一項に記載のプロセス。
- 前記ナノ粒子がステルスナノ粒子である、請求項1乃至4のうちのいずれか一項に記載のプロセス。
- 前記水中油型ミニエマルジョンがラジカル重合可能な架橋剤、好ましくは無水物またはアクリレートをさらに含む、請求項1乃至5のうちのいずれか一項に記載のプロセス。
- 前記少なくとも1種類のシアノアクリレートモノマーが、エチルシアノアクリレート、ブチルシアノアクリレート、イソヘキシルシアノアクリレート、オクチルシアノアクリレートおよびそれらの誘導体および混合物からなる群から選択される、請求項1乃至6のうちのいずれか一項に記載のプロセス。
- 前記少なくとも2種類のポリアルキレングリコールのうちの少なくとも一方が、疎水性の化学成分、たとえばポリプロピレンオキシド成分をさらに含むポリエチレングリコールである、請求項1乃至7のうちのいずれか一項に記載のプロセス。
- 前記少なくとも2種類のポリエチレングリコールのうちの少なくとも一方がポリエチレングリコールとポリプロピレングリコールとのブロックコポリマーであり、該コポリマーが、好ましくは、ポリプロピレンオキシドブロックと結合するヒドロキシ末端基またはアミノ末端基を含む、請求項8に記載のプロセス。
- 前記ターゲットモイエティが、ペプチド、ミニタンパク質、抗体片、アフィボディ分子、ナノボディおよびアプタマーからなる群から選択される、請求項1乃至9のうちのいずれか一項に記載のプロセス。
- 前記ターゲットモイエティが、ペプチド、抗体片、アフィボディ分子、ナノボディおよびアプタマーからなる群から選択される、請求項1乃至10のうちのいずれか一項に記載のプロセス。
- 前記活性剤が、治療薬であり、好ましくは、ドセタキセル、カバジタキセル、パクリタキセル、5−フルオロウラシル、ソラフェニブ、AMG900、テムシロリムスまたはエベロリムスである、請求項1乃至11のうちのいずれか一項に記載のプロセス。
- 前記活性剤が、カルボプラチン、オキサリプラチン、ピコプラチン、テトラプラチン、サトラプラチン、シスプラチン、プラチナ−DACHまたはオルマプラチンである、請求項1乃至12のうちのいずれか一項に記載のプロセス。
- 前記活性剤が、造影剤であり、好ましくは、金属類、金属塩類、近赤外線染料類、蛍光染料類、PET/SPECTキレート試薬類、MRIまたはラマン分光分析法に適する物質類および放射性医薬品類からなる群から選択される、請求項1乃至13のうちのいずれか一項に記載のプロセス。
- 請求項1乃至14のうちのいずれか一項に記載のプロセスにより調製されるポリ(アルキルシアノアクリレート)のホモポリマーまたはコポリマーから形成されるターゲットナノ粒子。
- 請求項15に記載のターゲットナノ粒子と、1つ以上の薬学的に受け入れ可能な担体、希釈剤または賦形剤とを含有する医薬組成物。
- 医療に用いられる請求項15に記載のターゲットナノ粒子。
- 薬物送達または分子イメージングに用いられる請求項15に記載のターゲットナノ粒子。
