Abstract
本发明一种基于超声限域聚合技术制备高分子材料的方法。本发明使用高频超声引发单体的自由基聚合,该方法无引发剂、催化剂引入,适用于医用高分子材料(如纳米颗粒、微纳米管、水凝胶等)的制备。同时,模板的引入给聚合反应提供了限域的空间,可以方便地使用不同单体制备不同形貌、粒径的高分子材料。在此基础上,得到的凝胶可以通过扩散、共价修饰以及亲和作用进行进一步修饰,可以得到具有各种特性的形貌均一可控的高分子材料。

技术领域
本发明涉及高分子材料制备技术领域,具体涉及一种基于超声限域聚合制备高分子纳米颗粒、微纳米管和水凝胶等多尺度材料的方法。
背景技术
公开该背景技术部分的信息仅仅旨在增加对本发明的总体背景的理解,而不必然被视为承认或以任何形式暗示该信息构成已经成为本领域一般技术人员所公知的现有技术。
目前利用现有技术得到的高分子材料往往不能满足相关领域的要求。一方面,聚合过程中引入的引发剂等有毒物质会限制高分子材料在生物医学领域的应用,其他残留的物质也可能对其性能产生影响。另一方面,微纳米材料的单分散性差等问题也会限制其在分离科学等领域的实际应用。因此,减少聚合过程中对后续应用不利的分子的引入以及保证材料的单分散性和可控性对于构建具有优异性能的高分子材料具有重要的意义。
为了得到尺寸、形貌更加均一的高分子材料,人们已经开发了分散聚合、种子乳液聚合等方法。但是这些方法一方面仅适用于特定粒径的高分子合成,难以使用统一的方法得到从微观到宏观各个尺度的高分子材料;另一方面,这些方法普遍对条件改变非常敏感,如果需要更换单体或引入其他物质往往需要重新对条件进行优化,这些都不利于快速制备功能、尺寸多样的高分子材料。同时,现有的方法在制备具有特殊形貌的高分子材料方面也存在一些困难。
发明内容
为了解决上述技术问题,本发明提供一种基于超声限域聚合制备多尺度高分子材料(如纳米颗粒、微纳米管、水凝胶等)的方法。本发明的方法可高效可控地合成特殊形貌的高分子材料,无需引发剂,具有良好的普适性及可拓展性;该方法所制备得到的高分子材料适合在生物领域中应用。
为解决上述问题,本发明提供以下技术方案:
一种基于超声限域聚合制备多尺度高分子材料(如纳米颗粒、微纳米管、水凝胶等)的方法,包括步骤如下:
将单体、交联剂配置成水溶液;结合模板法,除氧后在高频超声下聚合;去除模板后,得到均匀的高分子材料;
所述模板法为以反相微乳液为模板、以均匀孔道为模板、以毛细管为模板或以厘米级模具为模板。
进一步的,所述单体为丙烯酰胺、丙烯酸、3-[[2-(甲基丙烯酰氧)乙基]二甲基铵]丙酸酯、[2-(甲基丙烯酰基氧基)乙基]二甲基-(3-磺酸丙基)氢氧化铵、甲氧基聚乙二醇甲基丙烯酸酯、甲基丙烯酸2-氨基乙基酯盐酸盐中的一种或多种;交联剂为N,N’-亚甲基双丙烯酰胺、八臂聚乙二醇丙烯酸酯。单体与交联剂的质量比为100:0.01-100:20。
进一步的,聚合反应除氧的方式为鼓氮气30min并在氮气球保护下聚合。
进一步的,聚合反应温度为40℃,超声功率40W,超声频率为412kHz,聚合反应时间为20min-2h。
进一步的,以反相微乳液为模板时,将表面活性剂溶解在油相正己烷中;水溶液和油相混合,超声乳化,除氧后在高频超声下聚合。
根据本发明优选的,所述表面活性剂为吐温80和司班80;司班80与吐温80的质量比为3:2。
根据本发明优选的,表面活性剂在正己烷溶液中,表面活性剂的质量分数为10%-20%。
根据本发明优选的,微乳液体系中,水的质量分数为1%-10%。
根据本发明优选的,所述去除模板包括:旋转蒸发除掉低沸点的溶剂、使用含司班80的正己烷洗涤除去吐温80、使用正己烷洗涤除去残留的司班80、冷冻干燥以及透析。
根据本发明优选的,以反相微乳液为模板时,单体为甲氧基聚乙二醇甲基丙烯酸酯(mPEG-MA),交联剂为八臂聚乙二醇丙烯酸酯(8-arm-PEG-ACLT),聚合反应得到聚乙二醇纳米颗粒。更优选的,单体还包括甲基丙烯酸2-氨基乙基酯盐酸盐。
根据本发明优选的,得到的不同尺度的高分子凝胶后,可在颗粒上修饰功能分子;所述功能分子的实现是通过氨基与活性酯的反应或anti-PEG抗体实现的。所述功能分子为和靶向抗体。
根据本发明优选的,负载吲哚菁绿及抗体的聚乙二醇纳米颗粒在制备靶向光热治疗方面材料的应用。
进一步的,以均匀孔道为模板时,抽真空使溶液进入到阳极氧化铝模板。根据本发明优选的,为使溶液扩散到阳极氧化铝孔内,抽真空的条件为使用隔膜泵抽真空1h。根据本发明优选的,去除模板的方式为氢氧化钠溶液浸泡。去除模板后可得棒状高分子凝胶。
进一步的,以毛细管为模板时,将单体、交联剂和毛细管混合;根据本发明优选的,去除模板的方式为使用注射器吹出聚乙二醇凝胶。去除模板后可得毫米级圆柱形高分子凝胶。
进一步的,以厘米级模具为模板时,在所述水溶液中加入厘米级模具;去除模板的方式为直接从厘米级模具中取出高分子凝胶。去除模板后可得块状凝胶。
根据本发明优选的,所述mPEG-MA为mPEG2k-MA,8-arm-PEG-ACLT为8-arm-PEG20K-ACLT。
根据上述制备方法,通过不同的模板,不同的高分子单体可得到不同尺寸、不同形态、不同物质的高分子材料。
本发明与现有技术相比,具有以下有益效果:
1.本发明利用超声限域聚合制备多个尺度的高分子凝胶。利用超声聚合的方法,该方法无需加入引发剂、催化剂,适用于制备各种医用高分子材料。同时超声聚合还具有聚合时间短、单体转化率高等优点。
2.本发明利用了微乳液、AAO、毛细管等为模板,与分散聚合等反应体系相比,这些模板更加稳定,因此可以方便的根据需要更换单体、交联剂等。
3.本发明通过改变模板的形貌、尺寸、单体种类等,可以制备其他具有特殊结构的、均一的高分子凝胶。

为更好地理解本发明,下面结合具体实施例和附图来进一步说明。
实施例1
以玻璃瓶或硅胶为模板制备厘米级高分子凝胶。
将单体、交联剂溶解在一定量的水中,加入剪裁合适的方形硅胶模具,鼓氮气30min,向体系鼓氮气30min去除体系中的氧气,随后在频率412kHz、功率40W、温度40℃、氮气保护的条件下超声2h,聚合完成后敲碎玻璃瓶取出模具及高分子。得到的高分子外观如图1e、2a所示。
应用实验例1
1、超声限域聚合对单体的兼容性
分别在Schlenk瓶中加入200mg AM、AA、CBMA、SBMA,加入1mL水溶解,单体溶液中加入MBA作为交联剂,单体与交联剂质量比为100:1,mPEG-MA单体溶液中加入8-arm-PEG-ACLT作为交联剂,质量比为10:1。向体系鼓氮气30min去除体系中的氧气,随后在频率412kHz、功率40W、温度40℃、氮气保护的条件下超声1h,得到不同高分子凝胶。
根据图2a可以看出,这几种单体在同样的引发条件下都可以得到保留了玻璃瓶形貌的高分子凝胶。通过对其进行流变学性能进行表征,可以看到各个高分子的储能模量均大于损耗模量,具有类固体凝胶的性质。这证明了该方法不仅可以使用多种类型的小分子单体得到高分子材料,对于大分子单体也同样使用,具有良好的单体兼容性。
2、超声限域聚合对凝胶力学性能的调控
选取AM作为研究对象,超声聚合后,表征其流变性质,分别研究不同单体浓度、超声时间及单体与交联剂的质量比对凝胶流变性质的影响。单体浓度为20%,单体与交联剂的质量比分别为100:1,500:1,1000:1和5000:1,超声聚合时间为30min,研究单体与交联剂的质量比对凝胶流变性质的影响;单体浓度分别为15%,20%,25%和30%,单体与交联剂的质量比为100:1,超声时间为30min,研究单体浓度对凝胶流变性质的影响;单体浓度为20%,单体与交联剂的质量比为100:1,超声时间分别为10min,30min,1h和2h,研究超声时间对凝胶流变性质的影响。在剪切频率为1Hz下,对以上凝胶进行应力扫描,测试其弹性模量的变化。
如图3所示,在这里我们设想通过调控单体浓度,交联剂和单体的比例以及超声的时间来实现对凝胶材料机械性能的调控。从凝胶材料的流变学数据可以看出随着单体浓度,交联剂比例的增加以及超声时间的增长,高分子凝胶的弹性模量逐渐增大(图2-3),即机械强度逐渐增强。随着单体浓度增加,高频超声产生的羟基自由基引发的单体自由基浓度也增大,高分子浓度增大,因此其机械强度增大;单体与交联剂质量比降低,交联剂增多,凝胶的交联密度增大,机械强度随之增大;随着超声时间增长,单体转化率增大,凝胶机械强度增大。其中,超声时间对高分子凝胶弹性模量的影响最大,聚合2h后,弹性模量G’可达到1×103Pa以上。这证明了我们的策略可以通过调节单体浓度、交联剂与单体比例以及超声时间等实现对材料力学性能的调控。
3、高分子凝胶水溶性物质的负载
为了进一步扩展制备的水凝胶的应用,需要将其他分子负载于水凝胶上。在这里我们选择了水溶性的亚甲基蓝作为模拟物。根据图1e所示,经过12h的浸泡,亚甲基蓝已经深入到了凝胶块中,这证明了通过超声限域聚合的方法制备的水凝胶可以方便的负载水溶性的物质,有利于其后续的应用。
实施例2
毛细管模板中mPEG-MA的超声聚合
在Schlenk瓶中加入200mg mPEG-MA单体、20mg 8-arm-PEG-ACLT交联剂溶解于1mL水中,向体系鼓氮气15min,放入毛细管,溶液由于毛细现象进入毛细管中,继续鼓氮气15min后,在前述条件下超声2h,用注射器将其吹出,得到条状凝胶。
应用实验例2:条状水凝胶的染色以及毫米级图案的构筑
将得到的水凝胶浸泡在罗丹明溶液中染色。根据图1d可以看出,该凝胶很好的保留了毛细管的形貌,长度约为1mm,并且在浸泡染色后可以均匀的负载所选择的染料。后续,我们通过进一步的裁剪,将得到的条状凝胶重新组合,得到了毫米级的PEG图案。这证明了通过毛细管中超声限域聚合制备得到的毫米级柱状水凝胶可以负载水溶性染料并且可以进一步裁剪排列,用于构筑毫米级的图案。
实施例3
阳极氧化铝模板中SBMA单体的超声聚合及其表征
在Schlenk瓶中加入200mg SBMA单体、2mg MBA交联剂溶解于1mL水中,鼓氮气15min后,放入AAO模板,孔径为380nm,孔间距为450nm,孔深为20μm。抽真空1h使溶液进入AAO模板的孔洞中,前述条件下超声2h。聚合结束后,去掉AAO模板外部聚合成的凝胶,将AAO模板表面用水冲洗干净,浸泡在10%氢氧化钠溶液中3h以去除模板。将溶液在10000rpm转速下离心5min,用水清洗三次,得到棒状PSBMA纳米颗粒。
应用实验例3:棒状水凝胶的形貌表征
应用光学显微镜对制备得到的棒状凝胶颗粒进行表征。根据图1c可以看到凝胶颗粒很好的保留了AAO孔道的形貌和尺寸,这证明了该方法可以用于微米级具有特定形貌的粒子。
实施例4
微乳液模板中mPEG-MA的超声聚合及其表征
将90mg mPEG-MA单体、10mg 8-arm-PEG-ACLT溶解在0.9165mL水中配成水溶液。在另一离心管中将0.99g Span 80和0.66g Tween 80溶解在13ml的正己烷中,超声使表面活性剂均匀分散在油相中。将油相转移至Schlenk反应瓶中,向反应瓶中加入之前配好的水溶液,超声分散。向体系鼓氮气30min,前述条件下超声2h。将反应液在80℃条件下旋蒸除去溶液中的水和正己烷,随后用正己烷沉淀旋蒸后的反应液,离心得到粗产品。用含Span 80的正己烷溶液洗涤沉淀,去除少量的Tween 80,重复三次。再用正己烷洗涤沉淀三次,去除少量的Span 80,得到白色固体。40℃真空干燥12h后分散在2mL水中,透析2d,去除未反应的单体,得到纯的聚乙二醇纳米颗粒分散液。为了后续进一步修饰功能分子,我们同时制备了含AEMA单体的聚乙二醇纳米颗粒。在单体投料时加入单体量5%的AEMA分子,其他步骤相同。
应用实验例4

取10μL聚乙二醇纳米粒子原液稀释至1mL,然后取10μL稀释后的样品滴在新制的云母片上,-60℃冷冻干燥12h,利用原子力显微镜在气相轻敲模式下对样品的高度及形貌进行表征。分辨率:512*512,扫描速度:1kHz,针型号:60kHz,3N/m。图1b为反相微乳液模板(30nm)制备而得的聚乙二醇纳米颗粒的AFM照片,从图中可以看出纳米颗粒的尺寸约为50nm,高度为2-3nm,纳米颗粒发生的明显的坍塌现象,这主要是由于干燥过程中微凝胶失水导致的。这证明了我们的方法可以利用均匀的微乳液作为模板,制备粒径均一的高分子纳米颗粒。
2、聚乙二醇纳米粒子负载小分子
由于在聚合过程中引入了AEMA,因此,该颗粒后续可以利用氨基与活性酯的反应负载其他的功能性分子实现后续的应用。在这里我们选择了具有光热效果的ICG进行了实验。在PBS缓冲溶液中(pH 7.4,10mM),预先修饰了活性酯的小分子可以很容易的通过酰胺键修饰到颗粒上。根据图4的紫外可见吸收光谱,可以看到通过超声限域聚合制备得到的表面氨基化的聚乙二醇纳米粒子可以通过修饰活性酯的小分子进行进一步功能化。
3、负载ICG的聚乙二醇纳米颗粒的光热效果
为了考察负载后ICG的稳定性,我们分别对保存一段时间后的冻干粉末以及水分散体系进行紫外可见吸收光谱的表征,如图4所示,可以看出利用该策略修饰在聚乙二醇纳米颗粒上的小分子具有良好的稳定性。同时,负载后的小分子依然具有良好的功能,如图5所示,在808nm的激光器照射下,体系的温度上升了20℃左右。这证明了我们的方法可以保持小分子在颗粒表面修饰后的稳定性和相关的性能。
4、负载ICG及抗体的聚乙二醇纳米颗粒的靶向光热治疗效果
聚乙二醇纳米颗粒的抗体功能化修饰及靶向性能选用anti-PEG和anti-HER2的双特异性抗体对聚乙二醇纳米颗粒进行修饰。后续选择HER2高表达的SKBR3细胞和低表达的MCF-6细胞考察修饰抗体后聚乙二醇纳米粒子的靶向能力。如图6所示,流式细胞和共聚焦成像的结果都证明了经过抗体修饰后的纳米颗粒可以特异性的结合HER2高表达的SKBR3细胞,而与HER2低表达的细胞基本没有相互作用。后面使用同时负载有ICG和抗体的聚乙二醇纳米颗粒验证了细胞层面的靶向光热治疗效果。如图7所示,由于该颗粒仅与HER2高表达的SKBR3细胞结合,在将游离的颗粒去除后,使用808nm激光照射,可以看到仅对SKBR3细胞具有明显的细胞毒性。这证明了借助活性酯以及PEG抗体可以对得到的颗粒进行后续功能化修饰,这些粒子具有良好的靶向性和光热治疗效果。

Claims (15)
1.一种基于超声限域聚合制备多尺度高分子材料的方法,其特征在于,包括如下步骤:
将单体、交联剂配置成水溶液;结合模板法,除氧后在高频超声下聚合;去除模板后,得到均匀的高分子材料;
所述模板法为以反相微乳液为模板、以均匀孔道为模板、以毛细管为模板或以厘米级模具为模板;
所述单体为丙烯酰胺、丙烯酸、3-[[2-(甲基丙烯酰氧)乙基]二甲基铵]丙酸酯、[2-(甲基丙烯酰基氧基)乙基]二甲基-(3-磺酸丙基)氢氧化铵、甲氧基聚乙二醇甲基丙烯酸酯、甲基丙烯酸 2-氨基乙基酯盐酸盐中的一种或多种;交联剂为N,N’-亚甲基双丙烯酰胺、八臂聚乙二醇丙烯酸酯中的一种或两种;单体与交联剂的质量比为100:0.01-100:20;
其中,以厘米级模具为模板时,在所述水溶液中加入厘米级模具;去除模板的方式为直接从厘米级模具中取出高分子材料;
以反相微乳液为模板时,将表面活性剂溶解在油相正己烷中;水溶液和油相混合,超声乳化后,除氧后在高频超声下聚合;
以均匀孔道为模板时,抽真空使溶液进入到阳极氧化铝模板;
以毛细管为模板时,将单体、交联剂和毛细管混合。
2. 根据权利要求1所述的方法,其特征在于,聚合反应除氧的方式为鼓氮气30 min并在氮气保护下聚合;聚合反应温度为40 ℃,超声功率40 W,超声频率为412kHz,聚合反应时间为20 min-2 h。
3.根据权利要求1所述的方法,其特征在于,所述表面活性剂为吐温80和司班80;司班80与吐温80的质量比为3:2。
4.根据权利要求1所述的方法,其特征在于,表面活性剂的正己烷溶液,表面活性剂的质量分数为10%-20%。
5.根据权利要求1所述的方法,其特征在于,微乳液体系中,水的质量分数为1%-10%。
6.根据权利要求1所述的方法,其特征在于,以反相微乳液为模板时,所述去除模板包括:旋转蒸发除掉低沸点的溶剂、使用含司班80的正己烷洗涤除去吐温80、使用正己烷洗涤除去残留的司班80、冷冻干燥以及透析。
7.根据权利要求1所述的方法,其特征在于,以反相微乳液为模板时,单体为甲氧基聚乙二醇甲基丙烯酸酯(mPEG-MA),交联剂为八臂聚乙二醇丙烯酸酯(8-arm-PEG-ACLT),聚合反应得到聚乙二醇纳米颗粒。
8.根据权利要求7所述的方法,其特征在于,所述单体还包括甲基丙烯酸2-氨基乙基酯盐酸盐。
9.根据权利要求7所述的方法,其特征在于,得到聚乙二醇纳米颗粒后,在颗粒表面修饰功能分子;所述功能分子的实现是通过氨基与活性酯的反应或负载anti-PEG抗体实现的。
10.根据权利要求9所述的方法,其特征在于,所述功能分子为吲哚菁绿和靶向抗体。
11.根据权利要求1所述的方法,其特征在于,以均匀孔道为模板时,为使溶液扩散到阳极氧化铝孔内,抽真空的条件为使用隔膜泵抽真空1 h。
12.根据权利要求11所述的方法,其特征在于,去除模板的方式为氢氧化钠溶液浸泡。
13.根据权利要求1所述的方法,其特征在于,以毛细管为模板时,去除模板的方式为使用注射器吹出高分子材料。
14.根据上述权利要求1-13任一项所述方法制备得到的高分子材料。
15.根据权利要求14所述的高分子材料在制备靶向光热治疗材料领域中的应用,其特征在于,所述高分子材料为负载吲哚菁绿及抗体的聚乙二醇纳米颗粒。
