本発明は、活性化B細胞において発現されるタンパク質TSPAN33に関する。
a)リンパ腫は、ホジキンリンパ腫、非ホジキンリンパ腫、前駆T細胞白血病/リンパ腫、濾胞性リンパ腫、びまん性大細胞型B細胞リンパ腫、マントル細胞リンパ腫、B細胞性慢性リンパ性白血病/リンパ腫、MALTリンパ腫、バーキットリンパ腫、バーキットリンパ腫、末梢性T細胞リンパ腫−特記のないもの、結節硬化型のホジキンリンパ腫、または混合細胞質サブタイプのホジキンリンパ腫であり得、
b)リンパ腫は、ホジキンリンパ腫または非ホジキンリンパ腫であり得、
c)投与は、患者においてTSPAN33+B細胞の低減された数をもたらし得、
d)抗TSPAN33抗体が、モノクローナル抗体、中和抗体、もしくはヒト化抗体、またはこれらの組み合わせであり得、または

いくつかの実施形態において、TSPAN33/BAAMに対する抗体標的療法が、TSPAN33/BAAM陽性リンパ腫に対する治療として用いられ得る。例えば、マントル細胞リンパ腫(NHL)、侵攻型非ホジキンリンパ腫、およびリード・シュテルンベルク細胞含有ホジキンリンパ腫を有する患者から生検体を採取した。その組織を切片にし、H&E染色に対する、HRP結合抗マウスIgGを用いてTSPAN33に対して染色した。ホジキンリンパ腫および侵攻型非ホジキンリンパ腫は活性化Bリンパ球に由来すると考えられており(25)、一方、マントル細胞リンパ腫は、ナイーブ前胚中心Bリンパ球(naive, pre-germinal center B lymphocyte)に由来すると考えられており(26)、そのため、非活性化Bリンパ腫の一形態を表す。ホジキンリンパ腫および侵攻型非ホジキンリンパ腫の切片のみが、TSPAN33陽性であった。このことから、TSPAN33/BAAMは、TSPAN33/BAAM陽性リンパ腫における治療的モノクローナル抗体の有効な標的であると考えられる。
いくつかの実施形態において、TSPAN33/BAAMは、診断検査としての、活性化Bリンパ球および病的Bリンパ球のバイオマーカーとして用いられる。これらの実施形態は、TSPAN33/BAAMが休止B細胞においては陰性であるが、12時間後の抗CD40+IL−4による活性化の後に転写は40倍超に増加しているという知見に基づいている。例えば、106細胞/mLの精製ヒトB細胞および2E2ヒトB細胞株を、0.1μg/mLの抗CD40(G28.5mAb)および4ng/mLのIL−4で刺激した。これらの細胞を溶解し、Qiagen社製RNeasyキットを用いてRNAを回収した。500μgを用い、Qiagen社製QuantiTect Rev. Transcription Kitを用い、ランダムヘキサマーでcDNAを作製した。ロシュ社製Lightcycler 480システムを用いて、細胞可溶化液に対してRT−qPCRを行った。lightcyclerプライマー設計プログラムを用い、フォワードプライマー5’−caacatgctcttctgggtga−3’(配列番号4)およびリバースプライマー5’−attagccgagcgtagacacc−3’(配列番号5)を用い、UPLプライマー#9を用いて、Tspann33プライマーを開発した。フォワードプライマー5’−aacaaaatctctactttgatggaactt−3’(配列番号6)およびリバースプライマー5’−gcaaggcctactgctgagtt−3’(配列番号7)を用い、UPLプライマー#60を用いて、CD20を増幅した。18SおよびGAPDH発現の平均値を用いて、発現を正規化した。このように、当業者によって作製されたTSPAN33/BAAMに結合する抗体またはタンパク質は、ELISA、フローサイトメトリー、またはELISPOTを含むがこれらに限定はされないアッセイを用いて、活性化B細胞または病的B細胞に対するスクリーニングツールとして用いられることが企図される。いくつかの実施形態は、活性化B細胞または病的B細胞の検出のためのスクリーニングツールとしての、TSPAN33を検出するためのRT−PCR、RT−qPCR、またはPCR等のPCRに基づく方法の使用にまで及ぶ。

活性化B細胞での発現を含む制限発現パターンを有する膜貫通タンパク質をコードする遺伝子Tspan33を特定した。TSPAN33はテトラスパニンファミリーに属する。TSPAN33は休止B細胞では発現しないが、活性化後のヒトのプレB細胞で強く誘導されている。バーキットリンパ腫由来B細胞の活性化及び分化のモデルであるヒト2E2細胞も活性化に伴いTSPAN33の発現量が増加している。TSPAN33はホジキンリンパ腫及びびまん性大細胞型B細胞リンパ腫を含む複数のリンパ腫で発現している。関節リウマチ患者、全身性エリテマトーデス(SLE)、及びMRL/Faslpr/lprマウス(SLEのモデルマウス)由来の脾臓B細胞を含む、B細胞が病態に関与している一部の自己免疫疾患においてもTSPAN33が発現している。TSPAN33を診断的バイオマーカー又は特定のB細胞リンパ腫若しくは自己免疫疾患の治療に用いる医療用抗体の標的して利用可能であると結論付ける。
序論
細胞系列特異的なマーカーの発見及び評価は複雑な免疫系の根底にある細胞サブセットの特定に役立っている。CD3e(全T細胞マーカー)、CD4(ヘルパーT細胞)、CD8(細胞障害性T細胞)及びB220/CD45R(B細胞)などの細胞表面のマーカーはリンパ球集団を分化させるために日常的に使用されている[1−2]。フローサイトメトリーの標識技術の進歩により、細胞系列特異的な転写因子の検出に基づいたCD4サブタイプ(Th1、Th2、Th17及びTreg細胞)の評価が行えるようになった[3]。CD1dhiCD5+であり、IL−10を分泌するB細胞の小サブセットの特定に基づき、制御性「B10細胞」が発見された[4−6]。さらに、細胞系列特異的表面マーカー(例えばB細胞マーカーであるCD20)は、その病原性B細胞除去能力により種々のリンパ腫のみならず関節リウマチ(RA1)等の自己免疫疾患に対して有効であることが示された医療用mAbの開発の標的として有望である[7−8]。
ヒトリンパ球の新規マーカーの特定を目指し、免疫系細胞を含む異なる105のヒト臓器におけるヒトの遺伝子発現に関する広範なデータベース(Body Index of Gene Expression(BIGE)データベースとして知られている)を解析した[9−10]。このデータベースは特定の臓器又は細胞に関係する新規遺伝子の特定に有用である[11]。B細胞とはこれまで無関係と思われていた膜貫通型タンパク質をコードする遺伝子(Tspan33)を特定した。テトラスパニンスーパーファミリーは高度に保存されたシステイン−システイン−グリシン(CCG)モチーフを含むシステインリッチな長い細胞外ループ(LEL)からなる保存領域の構造(Pfam00335)により定義される[12]。これらの特徴により、インテグリン又は他のテトラスポニン等その他のタンパク質とより大きな分子複合体を形成しやすくなり、増殖、接着、運動性及び分化を含む様々な機能を仲介する。いくつかのテトラスパニンは成人の組織に広く発現しているが、他(CD82、CD151及びCD37を含む)はより限定的な発現プロファイルを示し、免疫系の細胞系列特異的に高い発現が認められる[13]。
TSPAN33は当初、マウス骨髄中の赤血球前駆細胞のサブポピュレーション内で検出されたため造血に携わっていると考えられており、これまでPenumbra(前赤芽球のnu膜)として報告されている[14]。マウス骨髄中のTspan33の発現は骨髄細胞(赤芽球)のTER119+分画で検出され、好中球、T細胞、単球、NK細胞又は(休止)B細胞では検出されていない[14]。それは実際にマウスpre−CFU赤血球系細胞及びマウス骨髄中で発現している[15]。これらの結果は、骨髄総RNAに対するこれらTspan33+赤血球前駆細胞の寄与が少ないことを示唆している。興味深いことに、Heikensら[14]が作製したTspan33−/−マウスのうちいくつかのマウスで3カ月以内に赤血球形成異常が、1歳で脾腫大が認められた。しかし、ここに示した様に、ヒトの正常骨髄中でのTSPAN33の発現は非常に低く(図3)、その代わりに活性化Bリンパ球において特異的に強発現している。
TSPAN33の発現をマウスとヒト両方のB細胞で確認した。これらの結果をまとめると、TSPAN33が活性化B細胞の新規マーカーであることが示される。休止及び活性化B細胞のどちらにも存在している他のB細胞特異的な抗原(すなわちCD20、CD19)とは対照的に、TSPAN33は活性化B細胞でのみ発現している。次に、TSPAN33が、活性化悪性B細胞が関与するヒトの疾患でも発現しているかを確認することを試みた。そこで、ホジキンリンパ腫(HL)、様々な種類の非ホジキンリンパ腫(NHL)並びに全身性エリテマトーデス(SLE)及び関節リウマチ(RA)の2種類の自己免疫疾患においてTSPAN33の発現を測定した。
方法
マイクロアレイ解析
Body Index of Gene Expressionデータベース(BIGE)の作成については既に報告されている[9−10]。簡単に述べると、全RNAを死後3〜5時間経過した男性4人、女性4人のヒトのドナーから、又は市販のヒト組織のRNA(Clontech、Palo Alto、CA)を追加し得た。ゲノムワイドな遺伝子発現データはAffymetrix Human Genome U133 Plus 2.0遺伝子アレイ(Affymetrix、Santa Clara、CA)を用いて取得し、データの正規化及び要約はArray Assistソフトウェア(Iobion Labs、La Jolla、CA)を用いて行った。
製造元の説明書に従いQiagenRNeasy(登録商標)キット(Qiagen、CA)を用いて、ヒトの細胞株/細胞又は組織からRNAを単離した。QuantiTect(登録商標) Reverse Transcription(Qiagen、CA)を用いてRNAをcDNAに変換した。Roche LightCycler(登録商標) 480 Real−Time PCRシステムとTSPAN33、CD19、CD20、CD138及びGAPDH検出用のプローブ(Roche、Pleasanton、CA)を用いてqPCRを行った。配列番号4及び5記載の配列を有するTSPAN33用プライマーを用いた。
抗ヒトベータアクチン(Santa Cruz biotech、Santa Cruz、CA)、抗ベータチューブリン(MP Biomedicals、Santa Ana、CA)及び抗Tspan33/TSPAN33(Abeam、Cambridge、MA)ポリクローナルウサギ抗体をウエスタンブロッティングに用いた。
ヒトB細胞株2E2は既に報告されている[16]。ヒトT細胞株のジャーカットはATCC(American Type Culture Collection、Manassas、VA)より入手した。マウス細胞株A20−2Jは既に報告されている[17]。全てのDLBCL系列はDavid Fruman(UC Irvine Institute for Immunology)より提供して頂いた。ヒトのドナー由来のPBMCはフィコール密度勾配法により単離した。マウス脾臓B細胞はフィコール密度勾配分離法を用いて濃縮し、抗CD3 mAb(Biolegend、San Diego、CA)及び抗CD11c mAb(Biolegend)をコートしたプレート上でパンニングした。簡単には、抗CD3及び抗CD11cを37℃で2時間、10cm組織培養用プレートにコーティングした。フィコール密度勾配分離法により単離した脾細胞をコーティングしたプレートで2時間インキュベートし、回収した非接着細胞を2回目の濃縮過程に供した。
B細胞への刺激をLPS(Sigma Aldrich、St Louis、MO)+マウス又はヒトrIL−4(Sigma)、抗CD40mAbクローンG38.5(Invitrogen、Carlsbad、CA)+rIL−4又はCpG+ヤマゴボウ***促進因子(PWM)+パンソルビン(Sigma)の何れかを用いて行った。T細胞への刺激を抗CD3mAb+抗CD28mAb(Biolegend)又はホルボール12−ミリスチン酸13−酢酸(PMA)+イオノマイシン(Sigma)を用いて行った。
C57Bl/6j(保存番号000664)及びMRL/faslpr/lprマウス(保存番号000485)をJackson Laboratory(Bar Harbor、ME)から取得した。全ての動物実験プロトコールはカリフォルニア大学アーバイン校のInstitutional Animal Care and Use Committee(IACUC)の承認を受けた。
ヒトPBMCをループス患者又は健常人の末梢血から静脈穿刺により採取した。本プロトコールはINNCMSZのInstitutional Review Board(IRB)により承認されており、試料はインフォームドコンセントを行った上で取得した。1982年にアメリカリウマチ協会が作成したSLEの新基準を4つ以上満たす者をループス患者とした[18]。臨床的疾患活動性はSLE疾患活動性インデックス又はSLEDAIを用いてスコア化した[19]。不活性疾患(SLEDAI<3)を持つ者を対照とし、活性疾患を伴う患者のうち3つ以上のインデックススコアを持つ者を活性疾患患者とした。cDNAを製造元の説明書に従い、M−MLV逆転写酵素を用いて作製した(Invitrogen、Carlsbad、CA)。
免疫組織化学的検査に用いたヒト組織試料は検死より得られたもので、Anatomy and Pathology Service of the University Hospitalof the UANLの記録用試料である。健常ヒト腎臓又はHL患者6名、濾胞性リンパ腫患者6名、DLBCL患者6名及びマントル細胞リンパ腫患者2名を含むヒトのリンパ腫生検を用いて、既報に従ったプロテアーゼ及び/又は熱処理による抗原賦活化(デマスキング)後に組織アレイを行った[20]。そして切片を抗TSPAN33抗体に続き、抗ウサギIgGロバ2次抗体酵素複合体(Abcam)を用いて染色した。
統計的有意差検定はスチューデントのt検定により行った。p<0.05の値を統計的有意と判断した。エラーバーは標準偏差(SD)を表す。
B細胞におけるTSPAN33の高度な発現
BIGEデータベースの発現解析により、TSPAN33をB細胞活性化特異的マーカーとして特定した(図3)。その発現プロファイルは特異的且つ制限的な発現を示し、抗CD40及びIL−4で活性化後の末梢血液B細胞、次いで腎臓で最も高い発現が認められた(表IにTspan33発現が高い上位10箇所の部位を示す;全リストは補足情報(SI1)に示す)。ヒトRNAに対してqRT−PCRを行い(SI2A)、BIGEデータベースのTspan33発現パターンを確認したが、骨髄、胸腺及び脾臓を含む他のほとんどの組織で低い又は検出限界以下の発現量であった。
B細胞活性化マーカーは診断ツールとして重要である。なぜなら、これら分子の一部、例えばsCD23、sCD27、sCD30、sCD44、CXCL13、IL−6及びIL−10の血清中濃度の増加[24−25]が、癌(例えば、NHL)と関連していると報告されているからである。その他の既知のB細胞抗原(すなわちCD19及びCD20)もNHLで高く発現している[26]。よって、TSPAN33はヒトリンパ腫においても発現していると仮定した。その検証のため、Tspan33発現についてqRT−PCRを行い、DLBCL(OCI−LY1、OCI−LY7、OCI−LY8、RC−K8、SU−DHL−2、SU−DHL4、SU−DHL−5、SU−DHL−6、SU−DHL−7並びにSU−DHL−8及びVAL)として特徴づけられるNHL細胞株を含む11細胞株並びに非刺激又は刺激(抗CD40mAb+IL−4)2E2細胞についてms4al(CD20)と比較した(図5A)。DLBCLは侵攻性NHLの最も一般的な種類で、通常のリンパ球の核の2倍以上の大きさの核を持つびまん性大型B細胞の増殖を共通の特徴として有する種々のリンパ腫のグループである[27]。DLBCLは中心芽細胞、免疫芽細胞又は未分化変異体(HLの高度に活性化されたリード・シュテルンベルク細胞に類似)を含んでおり、増殖係数は>90%である[28]。これらのリンパ腫細胞株において、ms4al/CD20mRNAレベルも測定し、Tspan33の発現量と比較した。ms4al/CD20及びTspan33は全てのDLBCL細胞株で検出された。実際に、DLBCLにおいてTspan33の発現量はCD20と同等であった。
B細胞活性化マーカーはいくつかの自己免疫疾患とも関連している。例えば、臨床SLEにおいては、CD25、HLA−DR、CD38及びBLySは全て増加しており、自己抗体産生と関連している[34−35]。新たにRAと診断された患者において、血清免疫グロブリン量及びB細胞関連のサイトカインIL−6、IL−21及びBLySは全て有意に上昇している[36−38]。BLySを阻害するとMRKL/faslpr/lprマウス(可溶性TACI)において病徴が削減され[39]、ヒトにおいても治療的に有益な効果が得られている(anti−BLySmAb:Benlysta)[40]。自己免疫疾患におけるTspan33の役割を明らかにするため、SLE患者のPBMC、RAの滑膜病変又はSLEのモデルマウス中のTspan33のmRNA発現量を測定した。
図3及びSI2Aに示すように、マイクロアレイ及びqPCRの何れで解析を行ってもTSPAN33mRNAは腎臓においても検出される。腎臓の生理学的に重要な役割を考慮し、腎臓中でのTSPAN33の発現部位を特定することにした。そこで、TSPAN33を検出するため、リンパ球浸潤が起こっている腎組織切片(図8A)を含む健常者の腎臓切片を用いて免疫組織化学的検査を行った(図8A−8D)。近位曲尿細管、遠位曲尿細管及び集合曲尿細管においてTSPAN33染色が検出された(図8B−8C)が、浸潤性のリンパ球(これまでの実験結果と一致)又は糸球体では検出されなかった。より高い倍率の解析により、TSPAN33は頂端膜及び近位曲尿細管の上皮刷子縁細胞の顆粒で発現していることが分かった(図8D)。通常これらの部位に抗体は入り込めないので、これらの結果よりTSPAN33が治療用の抗体開発の標的して支持される。
概要
我々はテトラスパニンファミリーに属する分子(TSPAN33)が活性化B細胞で強く発現しており、複数のリンパ腫及び病原性B細胞が関わる自己免疫疾患(SLE及びRAを含む)でも発現していることから、本分子が活性化B細胞のマーカーであることを発見した。
現在、B細胞の同定及び追跡のために、CD72、CD20、CD19及びCD24を含む多くのマーカーが使用されている[53]。活性化胚中心B細胞はGL7[54]、CD10及びBCL6[55]を含む様々な遺伝子を発現していると報告されている。MUM1/IRF4及びFOXP1、ならびにCD23やCD69等の他のB細胞活性化マーカー並びに全身性B細胞活性化マーカーであるCXCL13、sCD23、sCD27、sCD30、sCD44がマーカーとしてNHL及びRAの診断並びにリスク評価に用いられている[25、32、56]。重要なことは、これらの活性化マーカーは全て末梢において別の免疫細胞種と関わりがあるため、活性化B細胞でのみ発現しているマーカーが存在しないことである。よって、TSPAN33はB細胞特異的な活性化マーカーであり、B細胞の活性化が関与する疾患の診断ツールとして活用できる可能性がある。リンパ腫及び自己免疫疾患双方の予後バイオマーカーの候補としてのTSPAN33発現量の使用可能性はさらに研究が必要となる[57−59]。
TSPAN33のB細胞活性化マーカーとしての利用に加えて、TSPAN33は、33番目のテトラスパニンファミリーの分子で(TSPAN33)であるため、膜貫通タンパク質である。よって、TSPAN33は治療目的の抗TSPAN33mAbを作製するのにふさわしい候補である。CD20は近縁のタンパク質であり現在はmembrane−spanning 4−domains superfamily(MS4A1)に分類されているが、NHL、慢性リンパ性白血病(CLL)及びRAを含む特定の自己免疫疾患等の悪性B細胞の治療に効果的であると証明されている医療用モノクローナル抗体生産の重要なターゲットの一例である[51−52、60]。しかし、CD20は休止及び活性化B細胞の両方で発現しているため、抗CD20mAb治療の結果、骨髄では70%のB細胞が、末梢血液中では全B細胞が消失する[25、36、61]。そこで、TSPAN33のような活性化B細胞に限定されたB細胞マーカーの特定はB細胞関連病態を治療するための「第二世代」mAb開発の代替戦略となりえる[32]。別のテトラスパニン(CD151)は有望な治療抗体の標的として研究されている[62]。我々のデータから抗TSPAN33医療用mAbは治療を受ける患者の大部分の休止B細胞を消失させずにすむという大きな優位性を持つことが示唆される。
TSPAN33は当初、骨髄中の赤血球前駆細胞の小集団内で発現している分子として特定されたので、これまでPenumbra(前赤芽球のnu膜)として報告されてきた[14]。この様な発現パターンから、造血に関与していると報告された。また、Tspan33-/-マウスの一部が3カ月齢で異常な赤血球を産生したと報告されている[14]。ここで認められた真性赤血球無形成は、赤血球を欠く病気及び原因によっては自己治癒するヒトにおける病態と類似している[63]。これらの結果、ヒトにおけるTSPAN33の一時的な阻害による副作用は限定的又は対処が容易である可能性が示唆された。
B細胞中でのTspan33の機能は現在不明であるが、B細胞活性化に伴いTspan33の発現が強く誘導されることから、B細胞のシグナル伝達/活性化(すなわち、CD9及びCD81)、成熟化/生存(すなわちCD37)又は抗原提示(すなわちCD63)に関与していることが強く示唆される。なぜなら、他のB細胞で発現しているテトラスパニンがこれらの過程に寄与していることが知られているからである[66−69]。
TSPAN33は活性化及び悪性B細胞の重要なバイオマーカー候補であると同時に、複数種のB細胞リンパ腫(DLBCL、BL、HL)と活性化B細胞の表現型を呈する病原性B細胞と関連する一部の自己免疫疾患(SLE及びRA)の治療に用いる医療用mAb開発の標的候補である。

Claims (20)
- TSPAN33が上方制御されるリンパ腫または白血病を治療する方法であって、
前記リンパ腫または白血病を治療するのに有効な量で抗TSPAN33抗体を、そのような治療を必要とする患者に投与することを含む、方法。 - 前記リンパ腫は、ホジキンリンパ腫、非ホジキンリンパ腫、前駆T細胞白血病/リンパ腫、濾胞性リンパ腫、びまん性大細胞型B細胞リンパ腫、マントル細胞リンパ腫、B細胞性慢性リンパ性白血病/リンパ腫、MALTリンパ腫、バーキットリンパ腫、バーキットリンパ腫、末梢性T細胞リンパ腫−特記のないもの、結節硬化型のホジキンリンパ腫、または混合細胞質サブタイプのホジキンリンパ腫である、請求項1の方法。
- 前記リンパ腫がホジキンリンパ腫または非ホジキンリンパ腫である、請求項2の方法。
- 前記投与が、前記患者において低減された数のTSPAN33+B細胞をもたらす、請求項1の方法。
- 前記抗TSPAN33抗体が、モノクローナル抗体、中和抗体、もしくはヒト化抗体、またはこれらの組み合わせである、請求項1の方法。
- TSPAN33が上方制御される免疫疾患を治療する方法であって、前記免疫疾患を治療するのに有効な量で抗TSPAN33抗体を、そのような治療を必要とする患者に投与することを含む、方法。
- 前記免疫疾患は、アレルギーまたは自己免疫疾患である、請求項6の方法。
- 前記疾病は、関節リウマチ、乾癬、アトピー性皮膚炎、シェーグレン症候群、自己免疫性肝炎、原発性胆汁性肝硬変症、潰瘍性大腸炎、クローン病、強皮症、過敏性肺臓炎、自己免疫性甲状腺炎、橋本甲状腺炎、グレーブス病、強直性脊椎炎、セリアック病、特発性血小板減少性紫斑病、混合性結合組織病、多発性硬化症、多発性骨髄腫、尋常性天疱瘡、側頭動脈炎、白斑、または全身性エリテマトーデスである、請求項6の方法。
- 前記疾病は、関節リウマチまたは全身性エリテマトーデスである、請求項8の方法。
- 前記投与が、前記患者において低減された数のTSPAN33+B細胞をもたらす、請求項6の方法。
- 前記抗TSPAN33抗体が、モノクローナル抗体、中和抗体、もしくはヒト化抗体、またはこれらの組み合わせである、請求項6の方法。
- 抗TSPAN33抗体をリンパ球含有細胞製剤と混合することと、前記抗体により結合されたリンパ球を分離することとを含む、活性化Bリンパ球を精製する方法。
- 前記抗TSPAN33抗体が、モノクローナル抗体、中和抗体、もしくはヒト化抗体、またはこれらの組み合わせである、請求項12の方法。
- 前記分離が、蛍光標識細胞分取によるものである、請求項12の方法。
- 前記リンパ球におけるTSPAN33の上方制御された発現を検出することを含む、活性化および/または病的Bリンパ球を識別する、方法。
- 前記検出が、
前記リンパ球のタンパク質を含む試料に抗TSPAN33抗体を添加することと、
TSPAN33が前記試料に存在する場合に、前記抗体とTSPAN33との間の免疫複合体を形成することと、
前記免疫複合体を検出することとを含む、請求項15の方法。 - 前記検出が、
前記リンパ球のRNAからcDNAを調製することと、
前記TSPAN33遺伝子におけるヌクレオチド配列に特異的なプライマーにより前記cDNAを増幅すること、または前記TSPAN33遺伝子のヌクレオチド配列に対する前記cDNAをハイブリダイズすることと、
前記増幅反応の増幅生成物を検出することまたは前記cDNAと前記TSPAN33ヌクレオチド配列との間のハイブリッドを検出することとを含む、請求項15の方法。 - 前記リンパ球が患者由来であり、前記方法は、TSPAN33の上方制御された発現が検出される場合に、前記患者に抗TSPAN33抗体を投与することをさらに含む、請求項15の方法。
