6〜8週齢のマウスをこの研究に使用した。全ての動物実験は、National Health Research InstitutesのInstitutional Animal Care and Use Committeeにより承認されたプロトコールに従って実施した。(プロトコール番号NHRI−IACUC−100003及びNHRI−IACUC−097077−A)
組み換えヒトGRO−α、GRO−β、GRO−γ、IL−4、GM−CSF及び組み換えマウスGM−CSFは、PeproTech Inc(Rocky Hill,NJ,USA)から購入した。
組み換えマウスGRO−α、GRO−β及びGRO−γは、R&D Systems(Minneapolis,MN,USA)から購入した。
N−(2−ヒドロキシ−4−ニトロフェニル)−N'−(2−ブロモフェニル)尿素(SB225002)は、Calbiochem(San Diego,CA,USA)から購入した。
リポ多糖(LPS、E.coli 055:B5)は、Sigma−Aldrich(St.Louis,MO,USA)から得た。
R−フィコエリトリン(PE)標識マウス抗ヒトCD11c、HLA−DR及びCD83抗体は、eBioscience(San Diego,CA,USA)から得た。
フルオレセインイソチオシアネート(FITC)抱合マウス抗ヒトCD86、DC−SIGN、CD40及びCD80抗体は、BioLegend(San Diego,CA,USA)から購入した。
臍帯間葉幹細胞(uMSC)を、臍帯血から精製し、以前に報告されたように(Lee et al.,2004)特徴決定した。uMSCは、10%ES−FBS(HyClone、Logan,UT,USA)を有する、α−MEMを含有する培養培地に維持した。uMSC(15cm2細胞培養皿中の3×105細胞/15mL、第7継代)を、プールAB型ヒト血清(Invitrogen,Carlsbad,CA,USA)を補充した培地において更に2継代拡大した。各継代の持続期間は、5日間であった。3×105個のuMSC(p9)を、15mLの完全培養培地(15cm2皿中の10%プールAB型ヒト血清及び1×ペニシリン−ストレプトマイシン−グルタミン(Invitrogen)を有するα−MEM)に再接種し、加湿CO2インキュベーターにより37℃で5日間培養した。uMSCの上澄みを300×g及び4℃で10分間遠心分離して、壊死細胞片を除去し、収集し、−80℃で保存し、次に調整培地(MSC−CM)として使用した。
10%ヒトプールAB型血清を有するα−MEMにおけるサイトカイン及び増殖因子の、uMSCの存在下又は不在下での分泌プロファイルを、製造会社の説明書に従ってHuman Cytokine Array C Kit(Transignal Human Cytokine antibody Arrays C series 1000.1,RayBio,Redwood City,CA,USA)を使用して確立した。化学発光シグナルは、Kodak BioMax Lightフィルム(Kodak,Rochester,NY,USA)によりECLシステム(Amersham Pharmacia Biotech,Aylesbury,UK)を使用して検出し、続いてデジタル化した。
シグナル強度は、AlphaImager 1220 Analysis and Documentation System(Alpha Innotech,Braintree,UK)を使用して、スポットデンシトメトリーにより定量化した。各スポットシグナルを近接するバックグラウンド強度に対して補正し、膜の陽性対照に正規化した。
ヒト単球由来樹状細胞(MDDC)は、健康な提供者から得た白血球除去輸血から生成した。末梢血単核細胞(PBMC)は、Fycoll−Hypaque Plus(GE Health−Pharmacia,Uppsala,Sweden)密度遠心分離により精製した。
CD14+単球は、製造会社の説明書に従ってヒトCD14+Cell Isolation Kit(MACS,Miltenyi Biotec,Inc.,Auburn,CA,USA)を使用して単離した。
続いて、精製CD14+細胞(1×106/mL)を、rhGM−CSF(80ng/mL)及びrhIL−4(80ng/mL)(Peprotech,NJ)を補充した、10%プールAB型ヒト血清を有する2mLのMSC−CM又はα−MEM完全培地において、GROケモカインの存在下又は不在下で培養して、DCへの分化を誘発した。
同じ濃度のrhGM−CSF及びrhIL−4を含有し、GROケモカインを有する又は有さない追加の1mLの培地を、3日目に各細胞群に加えた。培養培地の半分の量を、5日目に、同じ濃度のrhGM−CSF及びrhIL−4を含有する等量の新たな培地に代えた。MDDCの成熟化(成熟DC、mMDDC)を、1μg/mLのLPSを5日目にiMDDCの培養培地に加えることによって誘発し、細胞は更に48時間培養された。未熟DC(iMDDC)及び成熟DC(mMDDC)の表現型変化を、7日目にFACS分析によりモニターした。
研究プロトコールは、Academia SinicaのInstitutional Review Board of Human Subject Research Ethics Committee(AS−IRB01−10113)及びNational Health Research InstitutesのInstitutional Review Board of Research Ethics Committee(EC1001101)により承認された。
単球、iMDDC及びmMDDCの表現型プロファイルは、1×105個の細胞を、CD11c、HLA−DR、CD80、CD86、CD83、CD40及びDC−SIGNに対する蛍光色素標識抗体(Ab)で染色することによって得た。蛍光強度をフローサイトメトリーにより測定した。使用した対応するイソタイプ適合対照は、FITC−IgG1、FITC−IgG2a、PE−IgG2a及びPE−IgG2b(BD Biosciences,San Jose,CA,USA)であった。表面標識細胞は、FACS Caliburフローサイトメーター(BD Biosciences)を使用して分析した。細胞の精製では、選別は、FACS Aria細胞選別機(BO Biosciences)により実施した。個別の選別細胞個体群の純度は、95%を越えていた。
iMDDC又はmMDDCのエンドサイトーシス活性は、フローサイトメトリーにより定量化したFITC−デキストラン(40kD、FD40S、Sigma−Aldrich)の細胞取り込みを分析することによって測定した。細胞(5×104細胞/試料/96ウエルV底プレート)を、1mg/mLのFITC−デキストランを有するRPMI−1640培地において37℃で30分間インキュベートした。
インキュベートした後、細胞を染色緩衝剤で2回洗浄して、遊離試薬を除去し、次に1%パラホルムアルデヒドで固定した。細胞によりエンドサイトーシスされたFITC−デキストランからのシグナルを、FACSにより分析した。データ分析は、FlowJoバージョン5.7.2ソフトウエアを使用して実施した。FITC−デキストランが不在の培地でインキュベートした細胞から得たシグナルを、陰性対照として使用した。
異なる分化段階の単球由来細胞(3×104)に、X線生物照射器(X−ray biological irradiator)(X−ray R−2000、Rad Source Technologies,Inc,Alpharetta,GA,USA)を使用して30Gyで照射し、次に96ウエルU底プレートにおいて完全培養培地中で精製同種CD3+T細胞(1:10)と共に培養した。X線照射骨髄細胞及び同種Tリンパ球を、加湿CO2インキュベーターに37℃で入れた。96時間後、[3H]−チミジン(1μCi)を、T細胞と混合した、単球、iMDDC又はmMDDCのいずれかを含有する各ウエルに加え、16時間更にインキュベートした。
次に細胞を、Filtermate96ウエル採取機を使用して採取し、放射能(cpm)を、Pakardマイクロプレートシンチレーション及びルミネセンスカウンター(Perkin−Elmer−Packard;Waltham,MA,USA)により細胞増殖の指数として測定した。
全RNAをTrizol試薬により抽出し、製造会社の説明書に従ってReverTra Ace set(Toyobo Life Science,Osaka,Japan)を使用してcDNAに変換した。リアルタイムPCR分析を、ABI Prism 7900システム(Applied Biosystems,Foster City CA,USA)を使用して実施した。試料を以下のプログラムに従って処理した。5℃で2分間、94℃で10分間、そして、95℃で15秒間と60℃で1分間を40サイクル。分析は三重に実施した。それぞれの試料において、サイクル閾値(Ct)を決定した。
結果を、同じプレートのGAPDH遺伝子に正規化した。異なる細胞群のmRNA発現のレベルを、2ΔCt法の使用により計算した。それぞれの遺伝子に特異的なイントロンスパニングプライマー(intron−spanning primer)を設計した。

MDDC単独(5×104)、CD3+精製T細胞単独(ヒトCD3+Cell Isolation Kit、MACS,Miltenyi Biotec,Inc.により単離、5×105)又はCD3+精製T細胞(5×105)と同時培養したMDDC(5×104)の上澄みを採取し、上澄みにおけるIL−4、IL−10、IL−12、INF−γ、IL−6及びTNF−αの濃度を、製造会社のプロトコール(R&D systems)に従って市販のELISAキットを使用して三重に決定した。
MDDCを、GRO−γの存在下又は不在下で生成し、次にIDOタンパク質の細胞内発現について分析した。細胞(5×104)を、ポリ−L−リシン被覆スライドガラスで15分間平板培養し、0.5mLのPBSで素早く2回洗浄した。細胞を、0.5mLの1%パラホルムアルデヒド(Sigma−Aldrich)の添加により10分間固定し、次に続いてPBSで3回洗浄し、0.5mLの1%BSA/PBSにより室温で30分間インキュベートした。細胞を、0.1%(v/v)NP−40/PBSで3回洗浄し、1:50希釈マウス抗ヒトIDO抗体(Chemicon Inc,Temecula,CA,USA)を含有する0.5%BSA/PBSと共に、室温で1時間インキュベートした。細胞を0.1%(v/v)NP−40/PBSで更に洗浄して、非結合抗体を除去し、次にDyLight 488(Sigma−Aldrich)と抱合した1:50希釈抗マウスIgGと共に、室温で1時間インキュベートした。0.1%(v/v)NP−40/PBSで3回最終洗浄した後、スライドに10%グリセロール/PBSを載せ、マニキュア液で密閉した。蛍光画像は、Leica TCS SP5カメラ(Leica Camera AG,Solms,Germany)を使用し撮影した。
ネズミ骨髄由来樹状細胞(BM−DC)を採取し、前記のように樹状細胞に分化させた(Song et al.,2011)。簡潔には、C57BL/6マウスの骨髄細胞を、200U/mL(20ng/mL)の組み換えマウスGM−CSFを有する10mLの完全RPMI−1640培地を入れたペトリ皿にて、組み換えマウスGROケモカインの存在下又は不在下で、2×105細胞/mLの密度により培養した。3日目に、培養培地の半分の量を、GROと組み合わせた又は組み合わせない20ng/mLのrhGM−CSFを含有する完全RPMI培地に代えた。6日目に、異なる処理群のBMDCをそれぞれの皿から収集し、洗浄し、特徴決定した。
GRO−γ処理細胞のインビトロ免疫抑制活性を、OVA初回刺激DCにより刺激されたOVA特異的OT−1CD8+精製T細胞の増殖の阻害を測定することによって評価した。マウス(C57BL/6)BMDCを、GRO−γの存在下又は不在下で分化させ、6日目に収集し、次にLCM培地(5%FBS、50μg/mLのゲンタマイシン、20.25mMのHEPES、50μMの2−ME及び100U/mLのペニシリン、100μg/mLのストレプトマイシンを補充したRPMI1640)に再懸濁した。
OT−1CD8+T細胞(2×105細胞/ウエル)を、FACS Ariaフローサイトメーターを使用した細胞選別により収集し、次に96Uウエルマイクロプレート中の200μLのLCM培地において、OVA257-264CTLエピトープ(1μg/mL)若しくはヒトパピローマウイルスRAH対照ペプチド(1μg/mL)の存在下、単独で又はBMDC(1×105細胞/ウエル)若しくはBMDC/GRO−γ細胞(1×105細胞/ウエル)のいずれかと組み合わせて培養した。培養物を、5%CO2インキュベーターにより37℃で3日間インキュベートし、[3H]チミジン(1μCi)を各ウエルに加え、続いて更なるインキュベーションを16時間行った。次に細胞を、半自動試料採取機の使用により採取し、放射能(cpm)を、Packardマイクロプレートシンチレーション及びルミネセンスカウンターにより細胞増殖の指数として測定した。
個別のマウスから採取した脾細胞(2×105細胞/ウエル)を、96ウエルU底プレートにおいてOVA(1μg/mL)又はRAH(1μg/mL)ペプチドにより刺激した。培養物を、5%CO2により37℃で12、36及び60時間インキュベートした。HBSS群又はBMDC初回刺激マウスの異なる群のいずれかにおける脾細胞の増殖は、3H−チミジン取り込みにより決定した。アッセイは、二重に実施した。データは、3つの独立した実験における1群あたり3匹のマウスの代表値である。
統計分析は、GraphPad Prismバージョン5.02(GraphPad Software,Inc)を使用して実施した。データを、少なくとも3つの独立した実験の平均値ア平均値の標準誤差(SEM)として表す。群間の差の統計的有意性は、片側スチューデントt検定を使用して評価した。
(1)MSC調整培地は、MDDC分化及び機能に対して抑制効果を発揮する。
本発明者たちは、ヒトCD14+単球の、MDDCへの分化を誘発するインビトロ標準操作手順を確立し、分化経路の各段階における骨髄細胞の細胞表現型を特徴決定するアッセイを確立した。
本発明者たちは、MSC−CMにおいてどの可溶性因子が、MDDC分化に対する抑制活性の原因であるかを調査した。GRO濃度の有意な増加がMSC−CMにおいて検出された(図2A)。本発明者たちは、ヒトiMDDCの表現型に対するGROの異なるアイソフォームの生物学的効果を更に検査した。GRO−α及びGRO−γのみがCD40発現に対して有意な阻害効果を有することが見出された(図2B)。サイトカイン/ケモカインアレイにおいては、GRO−αの強度は、非常にわずかしか増加しかなかったので(図2A)、本発明者たちは、続く実験で、GRO−γの特性に研究を集中させた。
MDDC分化に対するMSC−CMに存在するGRO−γの効果を更に確認するため、本発明者たちは、iDC及びmDCの両方に対する調整培地の抑制効果が、中和抗GRO−γ抗体の添加により部分的に逆転されるが、そのアイソタイプ対照ではされないことを示した(図2C)。この抗体の存在下では、iDCにおけるCD80、DC−SIGN及びCD83の表面レベルでの発現は、アイソタイプ対照又はMSC−CMで処理された細胞と比較して有意に増加した(図2C、上側パネル)。同様に、抗GRO−γ抗体は、成熟DCにおけるCD80、DC−SIGN、CD40及びCD11cの表面発現に対するMSC−CMの抑制効果を実質的に逆転した(図2C、下側パネル)。
MDDC分化及び機能に対するGRO−γの抑制効果に加えて、GRO−γは、MDDC分化の際のサイトカイン発現のプロファイルに影響を及ぼす。リアルタイムPCRを実施して、7日培養のMDDCにおける選択された遺伝子の相対的mRNAレベルを検査した。結果は、炎症性サイトカイン遺伝子、すなわちTNF−α、IFN−γ及びIL−12のmRNAレベルが、GRO−γの存在下でのMDDC分化において有意に下方制御されたことを示した(図4A)。IL−10、IL−4、TGF−β、IL−1β及びIL−6遺伝子、並びにCOX2、プログラム死リガンド(PD−L1及びPD−L2)、マトリックスメタロペプチダーゼ9(MMP−9)、とりわけIDOをコードする遺伝子の発現レベルは、有意に上方制御された(図4A)。ヒトMDSCは、IL−10、IL−4、IL−1β及びIL−6を分泌する能力、並びにCOX2、PD−L1、MMP−9及びIDOを発現する能力によって特徴決定される。qPCR分析は、GRO−γによるMDDCの処理が、これらのMDSCマーカー遺伝子の発現を、未処理MDDC対照と比較して増加させることを明らかにした(図4A)。
GRO−γ処理MDDCの、T細胞応答への効果を更に調査するため、ヒトCD3+ Cell Isolation Kitを使用して精製されたCD3+T細胞を、GRO−γで処理された又は未処理の自己分化MDDCに曝露した。CD3+T細胞を同じ方法によりT/MDDC混合物から更に単離した。RNAを抽出し、選択されたサイトカイン遺伝子の発現を、リアルタイムPCRにより評価した。
本発明者たちは、GRO−γ初回刺激MDDCと同時培養されたCD3+T細胞が、未処理MDDC対照と同時培養されたT細胞と比較して、IL−4及びIL−10のmRNAをより高いレベルで発現する一方で、IFN−γ及びIL−12のmRNAをより低い量で発現することを見出した(図5A)。MDDC/T細胞同時培養培地のサイトカインプロファイルも、ELISAにより分析した。リアルタイムPCRデータと一致して、本発明者たちは、IL−4及びIL−10の分泌レベルが、非初回刺激MDDC対照T群と比較して、GRO−γ初回刺激MDDCと同時培養されたT細胞の上澄みにおいて有意に上昇しことを見出した(図5B)。同時培養培地中のIL−12及びIFN−γの濃度は、GRO−γ初回刺激MDDCの存在下で有意に低減した(図5B)。
GRO−γ初回刺激DCのインビボでの機能を更に調査するため、本発明者たちは、マウス系におけるGRO−γの存在下又は不在下でのDCの分化を研究した。分化したBMDCを収集し、CD11b及びGr−1蛍光抗体を使用して、マウスMDSCマーカーを染色した。二重陽性CD11b+Gr−1+細胞を、更なる分析のために更に選別した。本発明者たちは、BMDCの二重陽性CD11b+Gr−1+細胞の割合が、GRO−γの存在によって左右されないことを見出した(図6A、上側パネル)。
しかし本発明者たちは、ARG−1遺伝子及び誘発性NOシンターゼ(iNOS)遺伝子の発現を分析した。これらは、マウスにおいてT細胞の増殖及びアポトーシスを阻害することが知られているMDSCにおいて上方制御されることが知られている(Condamine and Gabrilovich,2011)。予測されたように、ARG−1及びiNOSのmRNAレベルは、GRO−γ処理BMDCにおいて上昇したことが見出され(図6A、下側パネル)、CXCR2インヒビターのSB225002の添加は、GRO−γ処理BMDCにおける両方の遺伝子の誘発を逆転した。本発明者たちのデータは、GRO−γの免疫抑制活性が、CD11b+Gr−1+二重陽性細胞の数の増加により仲介されるのではなく、むしろBMDCの特性における機能的変化の結果であることを示している。
次に、OVA257-264ペプチドパルスBMDCで免疫化した7日目後に動物を殺処分し、免疫化マウスの脾細胞を収集し、OVA257-264ペプチドによりインビトロで12、36及び60時間刺激した。
結果は、36及び60時間のOVA257-264ペプチド再刺激の後、GRO−γ初回刺激BMDCを注射したマウスの、OVA257-264ペプチド刺激脾細胞の増殖が、対照BMDCの投与を受けたマウスの細胞と比較して有意に低減したことを明らかにした(図6D)。異なる実験群のOVA257-264ペプチド刺激脾細胞の上澄みも、ELISAによりサイトカイン分泌について分析した。本発明者たちは、未処理BMDC群と比較して、GRO−γ初回刺激BMDC群におけるIFN−γ及びTNF−αレベルが低減したことを観察した(図6E)。
本発明者たちは、GRO−γ処理BMDC及びGRO−γ未処理BMDCにおけるCD11b+Gr1+細胞の数及び細胞特性を分析した。結果は、Arg−1遺伝子、iNOS遺伝子及びMDSC関連遺伝子の発現が、BMDC分化の際にGRO−γの存在下で上方制御されたばかりでなく、Arg−1及びiNOSのタンパク質発現及び酵素活性もGRO−γ処理BMDCにおいて増加したことを示した。一方、CD11b+Gr1+細胞の頻度は、対照とGRO−γ処理BMDCとの間で類似していた。
データは、無腫瘍マウスでは、総脾細胞又はCD11b-サブセットではなく、CD11b+サブセットのみが同種混合リンパ球反応(同種MLR)を実質的に抑制しうることを示した(図7B)。一方、腫瘍担持マウスから採取した総脾細胞、CD11b+細胞及びCD11b-サブセットは、全て、MLRを抑制する著しい能力を示した(図7B)。
MHC不適合野生型B6ドナーマウスの5×106個の骨髄細胞及び5×105個の脾細胞を、致死照射Balb/cマウス(レシピエント)に静脈内輸注することによって、Balb/cマウスにGvHDを誘発した。ドナーマウスの異なる細胞群がレシピエントに静脈内注射され、すなわち、5×106個の骨髄細胞単独、5×105個の脾細胞単独、骨髄細胞と脾細胞の組み合わせ及び1×107個のGRO−γ処理BMDCを補充した骨髄細胞と脾細胞の組み合わせが、移植の0日目、2及び7日後に注射された。対照マウスは、骨髄細胞のみの投与を受けた。レシピエントでは、生存、食餌摂取、体重減少及び臨床GvHDをモニターした。
対照マウスは、骨髄細胞及び脾細胞の両方の投与を受けたレシピエントと比較して、有意に高い生存率(100%)を有した(図9B)。GRO−γ処理BMDCの投与を受けたマウスの生存率は、骨髄細胞及び脾細胞のみの投与を受けたマウスより高かった(図9B)。

- 前記前駆体細胞はヒトCD14+単球であり、
前記培地は、組み換えヒトGRO−γを含有する請求項1に記載の免疫抑制細胞作成方法。 - 前記前駆体細胞はマウス骨髄細胞であり、
前記培地は、マウスGM−CSF(2〜100ng/mL)を含有する完全RPMI−1640培地である請求項2に記載の免疫抑制細胞作成方法。 - 前記GROケモカインは、マウスGRO−γ又はGRO−αである請求項6に記載の免疫抑制細胞作成方法。
- 前記免疫抑制表現型は、
(a)以下の遺伝子:IL−10、IL−4、TGF−β、IL−1β、IL−6、COX2、PD−L1、PD−L2、MMP−9、IDO、ARG−1及びiNOSの1つ以上の発現の増加、
(b)以下の遺伝子:TNF−α、IFN−γ及びIL−12の1つ以上の発現の減少、並びに、
(c)T細胞応答を抑制する能力を含む請求項1に記載の免疫抑制細胞作成方法。 - 請求項1に記載の方法により得られる免疫抑制細胞。
- 前記細胞は、CD11b+又はCD11b-である請求項9に記載の免疫抑制細胞。
- 前記細胞におけるCD40、CD83、CD80、CD11c、CD86及びDC−SIGNの発現が低減される請求項10に記載の免疫抑制細胞。
- 請求項9に記載の免疫抑制細胞を含む組成物。
- 使用対象における免疫応答の抑制に用いられる請求項12に記載の組成物の使用方法。
- 使用対象の状態の処置に用いられる請求項12に記載の組成物の使用方法であって、
該状態は、自己免疫性障害、炎症性障害、移植片対宿主病(GVHD)又は移植拒絶である組成物の使用方法。 - 前記状態は自己免疫性障害である請求項14に記載の組成物の使用方法。
- 前記状態は炎症性障害である請求項14に記載の組成物の使用方法。
- 前記状態は移植片対宿主病(GVHD)である請求項14に記載の組成物の使用方法。
