Description
The present invention relates to a detection method, a detection apparatus, a biochip screening method, and a screening apparatus.

In view of the circumstances as described above, an object of the present invention is to provide a detection method, a detection device, a biochip screening method, and a screening device capable of accurately recognizing the positional relationship between individual imaging results. また、例えば、センサ28は、PMT(photomultiplier tube)などの光検出器、や撮像素子を含む。 センサ28は、物体の像情報を取得可能であり、例えばCCD(charge coupled device)を含む。

The
The

FIG. 2 is a schematic diagram illustrating an example of the
The
The
FIG. 3B is a diagram showing the relationship between the wavelength of incident light and the transmittance as the optical characteristics of the

The
Further, the
That is, the
As shown in FIG. 1, the imaging
The
The
The driving
The

A part of the light (generated light L2) incident on the
As shown in FIG. 1, the

Next, an example of the
FIG. 4A is a diagram showing the shape of the
The
A row of biomolecule support regions is formed on the
In the present embodiment, a region (arrangement region) Sa including one or more biomolecule support regions S is formed on the
Each biomolecule support region S can take various shapes such as a rectangle, a circle, and a polygon in plan view. In this embodiment, as shown to Fig.4 (a), it forms in the rectangle, for example. In each biomolecule support region S, various measurement targets B that can specifically react with a labeled target contained in a specimen (eg, whole blood or serum) are arranged in a predetermined arrangement (FIG. 4). (See (b)). For example, when a target labeled with a fluorescent substance such as a fluorescent dye or a phosphorescent substance is used, a predetermined generated light (L2) such as fluorescence or phosphorescence is emitted by irradiating the biomolecule supporting region S with excitation light after the bioassay. : See FIG. 1). The generated light (L2) is emitted from the surface of the biochip 50 (substrate body 60) for each biomolecule support region S.
For example, in the measurement target B, a biomolecule is arranged as a probe. Further, the fluorescence measurement is performed after the specimen containing the fluorescently labeled target is dispensed into the biomolecule support region S of the
The
FIG. 5 is a diagram illustrating the configuration of the
As shown in FIG. 5, one
For example, in the present embodiment, the detection apparatus MS can capture an image in a range included in the
The shape of the detection range D100 can be set to, for example, the shape of the biomolecule support region S or the shape corresponding to the arrangement direction. The detection range D100 in the present embodiment is set to a rectangle (eg, a square), and is set to a region including a plurality of rows and a plurality of columns of biomolecule support regions S.
The size of the detection range D100 is smaller than the arrangement region (area of the
In such a case, the detection apparatus MS performs alignment so that the detection range D100 coincides with each of the detection areas (including D101 to D104). This alignment is performed by moving the
For example, the detection apparatus MS performs the measurement process of the arrangement area Sa by combining (stitching) a plurality of detection results (measurement results and imaging results) in the obtained detection areas D101 to D104. Also in this measurement process, the affinity between the biomolecule formed in the biomolecule support region S and the target contained in the specimen can be detected. Therefore, the processing result of the measurement process can be an affinity detection result. In the stitching, the detection range D100 to be measured is imaged a plurality of times while relatively moving sequentially, and the results are combined. The detection range D100 includes a detection target region such as a light receiving region or an imaging region in the
In the example shown in FIG. 5, the detection area D101 and the detection area D102 are adjacent to each other in the X direction. The detection area D103 and the detection area D104 are adjacent to each other in the X direction. The detection area D101 and the detection area D103 are adjacent to each other in the Y direction. The detection area D102 and the detection area D104 are adjacent to each other in the Y direction.
Further, the + X side end of the detection region D101 and the −X side end of the detection region D102 overlap each other (overlapping region D12). In the overlapping region D12, a plurality of biomolecule support regions S are arranged along the Y direction. Similarly, the + X side end portion of the detection region D103 and the −X side end portion of the detection region D104 overlap each other (overlap region D34), and the biomolecule support region S extends along the Y direction in the overlap region D34. Several are arranged. The number of rows of the biomolecule support regions S arranged in the overlap region D12 and the overlap region D34 in FIG. 5 is, for example, one row, but is not limited to this, and a plurality of rows of biomolecule support regions S are arranged. It may be a configuration.
Further, the −Y side end of the detection region D101 and the + Y side end of the detection region D103 overlap each other (overlapping region D13), and the overlapping region D13 includes a plurality of biomolecule support regions S along the X direction. Has been placed. Further, the −Y side end of the detection region D102 and the + Y side end of the detection region D104 overlap each other (overlap region D24), and the overlap region D24 includes a plurality of biomolecule support regions S along the X direction.
Next, using the detection device MS having the above-described configuration and an assay unit (dispensing device) AS, which will be described later, etc., light is emitted from each biomolecule support region S of the bioassay method and the
First, as shown in FIG. 6, a biomolecule formed in the biomolecule support region S by dispensing a specimen containing a fluorescently labeled target to each biomolecule support region S of the
After the biomolecule and the target are reacted in the dispensing step, the
Next, a step of drying the washed
After the cleaning and drying steps of the
Next, the

Next, the
From the biomolecule support region S irradiated with the excitation light L1, fluorescence is emitted from the biomolecule support region S in which the target and the fluorescent molecule labeled in the specimen are bound by a specific reaction (having affinity). The generated light (second light) L2 is generated. Thereby, for example, as shown in FIG. 7, a light emission pattern PL1 of the generated light L2 is formed on the
The generated light L2 generated in the biomolecule support region S passes through the
Next, the
After the first image Im1 is acquired, the
After detecting the affinity for the first image Im1, the
After acquiring the image of the
Moreover, the
After the second image Im2 is acquired, the
Thereafter, the
After acquiring the image of the
In addition, the
After the third image Im3 is acquired, the
After detecting the affinity for the third image Im3, the
After acquiring the image of the
Moreover, the
After the fourth image Im4 is acquired, the
Here, the + X side end portion of the detection range D100 arranged in the detection region D101 and the −X side end portion of the detection range D100 arranged in the detection region D102 become an overlapping region D12 (see FIG. 5). Yes. For this reason, as shown in FIGS. 11A and 11B, a portion corresponding to the overlapping region D12 (first image Im1: in FIG. 11A) in the first image Im1 and the second image Im2. The right end portion, the second image Im2: the left end portion in FIG. 11B) includes the same light emission pattern (pattern) P1. As a result, the
Similarly, the + X side end portion of the detection range D100 arranged in the detection region D103 and the −X side end portion of the detection range D100 arranged in the detection region D104 become an overlapping region D34 (see FIG. 5). Yes. For this reason, as shown in FIGS. 11C and 11D, the portion corresponding to the overlapping region D34 in the third image Im3 and the fourth image Im4 (third image Im3: in FIG. 11C). The right end portion, the fourth image Im4: the left end portion in FIG. 11D) includes the same light emission pattern (pattern) P2. Thereby, the
Further, the −Y side end portion of the detection range D100 arranged in the detection region D101 and the + Y side end portion of the detection range D100 arranged in the detection region D103 are an overlapping region D13 (see FIG. 5). . For this reason, as shown in FIGS. 11A and 11C, the portion corresponding to the overlapping region D13 (first image Im1: in FIG. 11A) in the first image Im1 and the third image Im3. The lower end, the third image Im3: the upper end in FIG. 11C includes the same light emission pattern (pattern) P3. As a result, the
Further, the −Y side end portion of the detection range D100 arranged in the detection region D102 and the + Y side end portion of the detection range D100 arranged in the detection region D104 are an overlapping region D24 (see FIG. 5). . Therefore, as shown in FIG. 11B and FIG. 11D, the portion corresponding to the overlap region D24 (second image Im2: in FIG. 11B) in the second image Im2 and the fourth image Im4. The lower end portion, the fourth image Im4: the upper end portion in FIG. 11D) includes the same light emission pattern (pattern) P4. As a result, the
FIG. 12A is a graph showing the output value (luminance) of the light emission pattern P1 among the luminance distributions of the light emission patterns PL1 to PL4 detected by the affinity detection operation. FIG. 12B is a diagram illustrating an output value (luminance) for the light emission pattern P2 in the luminance distribution of the light emission patterns PL1 to PL4 detected by the affinity detection operation.
As an example, the horizontal axes of FIGS. 12A and 12B indicate the Y coordinates of nine biomolecule support regions S arranged in the Y direction of the light emission patterns P1 and P2 (light emission pattern P1: Y01). To Y09, light emission pattern P2: Y09 to Y17). The vertical axis in FIG. 12 represents the output value (luminance: relative value) of the
As shown in FIG. 12, in the light emission patterns P1 and P2, the luminance of the generated light L2 changes while increasing or decreasing at random from Y01 to Y09 and Y09 to Y17, respectively. The transition of the luminance changes according to the affinity between the target and the biomolecule contained in the specimen (for example, reactivity or binding based on the presence / absence of fluorescence or the intensity of fluorescence). For example, in the present embodiment, when the target and the biomolecule have high affinity, the luminance of the generated light L2 increases. The affinity with the target varies depending on the biomolecule (eg, sequence).
When two different regions including a plurality of biomolecule support regions S are selected in the detection result, the possibility that the luminance distribution of one region matches the luminance distribution of the other region is substantially low. I can say that. For example, as shown in FIGS. 12A and 12B, for example, the light emission pattern P1 and the light emission pattern P2 have completely different distributions. However, at the coordinates Y09 corresponding to the same biomolecule support region S, the luminances of the light emission patterns P1 and P2 are the same. The same applies to the light emission patterns P3 and P4 having different arrangement directions from the light emission patterns P1 and P2. Note that the greater the number of biomolecule support regions S included in one region, the lower the possibility that the patterns (luminance distributions) match each other.
Thus, the light emission pattern of an arbitrary region selected to include a plurality of biomolecule support regions S is very likely to be a unique light emission pattern in the arrangement region Sa. Therefore, in the present embodiment, the combining
13 to 16 are diagrams illustrating how images (detection results) are combined in the combining step. In FIGS. 13 to 16, for the convenience of illustration, the biomolecule support regions S other than the biomolecule support region S included in the light emission patterns P1 to P4 are shown in white.
First, as illustrated in FIG. 13A, the
Similarly, as illustrated in FIG. 14A, the

For example, as illustrated in FIG. 16A, the
Further, as shown in FIG. 16C, the
Thereafter, the
Moreover, although illustration is abbreviate | omitted, the
As described above, according to this embodiment, the excitation light L1 is irradiated to the plurality of biomolecule support regions S arranged on the
In addition, a plurality of biomolecule support regions S of the
The technical scope of the present invention is not limited to the above-described embodiment, and appropriate modifications can be made without departing from the spirit of the present invention.
In the said embodiment, although the aspect which synthesize | combined an imaging result was mentioned as an example and demonstrated using the light emission patterns P1-P4, it is not restricted to this. For example, as will be described below, the
FIG. 17 is a flowchart showing an assay method and a detection method according to a modification.
As shown in FIG. 17, the
After the irradiation step S3, the
Thereafter, the
FIG. 18A is a plan view showing the positional relationship between the biomolecule support region S and the
As shown in FIG. 18A, in the present embodiment, the
The region Xa is a region arranged at one of the four corners of the
Next, the
In the region Xa and the region Xb, the luminance of the generated light L2 generated from Y0 to Y9 changes while increasing or decreasing randomly as described in the above embodiment. Therefore, for example, the luminance distribution (light emission pattern) in the region Xa and the luminance distribution (light emission pattern) in the region Xb are completely different distributions. As described above, when two different regions including a plurality of biomolecule support regions S having the same arrangement (the same detection region) are selected, the luminance distribution of one region and the luminance distribution of the other region can match. It can be said that the sex is substantially low.
Therefore, in this embodiment, the
With the above operation, the detection range D100 can be adjusted to the upper left corner (−X side end and + Y side end) of the
After aligning the
In the detection step, in the present embodiment, the
When relatively moving the measurement target region and the
Then, after the imaging of the first imaging area is completed, the
When moving the
Then, after the imaging of the second imaging area is completed, the
After performing such an operation on the entire measurement target region and obtaining an imaging result for the entire measurement target region, the
As an example, the detection device MS in the above embodiment uses a luminance distribution as a light emission pattern (light pattern received by a sensor) generated from the biomolecule support region S included in the regions Xa, Xb, and Ya. Although explained, it is not limited to this. For example, a wavelength spectrum (intensity distribution for each wavelength), an illuminance pattern (illuminance distribution in the sensor 28), an imaging pattern obtained by image recognition, or the like may be used as the light emission pattern.
Further, when the detection device MS performs the detection process a plurality of times on the
Further, the detection device MS may use a light emission pattern obtained continuously before and after moving the

The detection apparatus MS in the above embodiment has been described by taking as an example the case where the light emission pattern of the biomolecule support region S included in one region is used when aligning each imaging region, but is not limited thereto. There is nothing. For example, as shown in FIG. 20, the detection apparatus MS sets a plurality of regions Xc and Xd in one
In addition to the configuration of the above embodiment, as shown in FIG. 20, a configuration in which a
