Abstract
Description
癌細胞は、同一の患者(自己)または異なる患者(同種異系)から供給することができる。
ワクチンに自己癌細胞を使用することは個別療法であり、実用上の困難を伴う。自己細胞は全ての患者において入手できない場合がある。入手できる場合でも、所望の質および/または量ではない場合がある。このアプローチはまた多大な時間を要する。このアプローチには規制のハードルもある。
これは、ワクチン中に多数の異種の同種異系癌細胞を使用するか、癌組織中に存在する抗原を同定し、それに対して特異的なワクチンを使用することによって克服できる。
癌/腫瘍の異種の問題を克服するため、抗原として2以上の細胞を用いることが示されている。エメンス(Emens)らは、2以上の同種異系細胞株を用いて、異種の腫瘍/癌の抗原レパートリーをカバーすることを実証した(エメンスLAら;J Clin Oncol. 2009年12月10日;27(35)巻:5911-8頁)。一方、ラヘル(Laheru) Dらは、GM CSFを使用して、癌治療のための同種異系癌細胞ワクチンの免疫原性を向上させることを実証した(Clin Cancer Res. 2008年5月1日;14(5)巻:1455-63頁)。
組織/器官に特異的な異種癌細胞は、同一の組織/器官に存在する/由来する癌細胞であるが、同一の組織/器官に存在する/由来する癌細胞によって生じる免疫反応を引き起こさず、および/または、反応しない。

本発明の目的は、癌細胞が組織/器官に特異的な異種癌抗原に対して免疫原性を有するような方法で、癌細胞の免疫原性プロファイルを変更することである。
本発明の目的は、組織/器官に特異的な異種癌抗原に対する免疫原性を有する、悪性腫瘍の治療に使用するための、治療用ワクチンを提供することである。
本発明のさらに別の目的は、組織/器官に特異的な異種癌抗原に対する免疫原性を有する、悪性腫瘍の治療に使用するための、治療用ワクチンの製造方法を提供することである。
本発明のもう一つの目的は、発癌性を誘導しない同種癌ワクチンを提供することである。
さらに別の目的は、組織/器官に特異的な同種および異種癌細胞に特異的な細胞性免疫反応を刺激する、悪性腫瘍のための治療用ワクチンを提供することである。
さらに別の目的は、組織/器官に特異的な同種および異種癌細胞に特異的な体液性免疫反応を刺激する、悪性腫瘍のための治療用ワクチンを提供することである。
従って、本発明によれば、ある器官が起源の癌細胞の免疫原性プロファイルは、マイコバクテリウムw(Mw)の存在下で変化する。変化した免疫原性プロファイルにより、癌細胞は、同一の器官/組織に存在するかそこから発生する同種細胞および異種細胞に対して、免疫反応を起こす。異種細胞に対する免疫反応の発生は、典型的には癌細胞では見られない。
本発明の癌細胞は、加熱または煮沸によって死滅させるか、スチームで処理される。
本発明の癌細胞は、アルデヒド、ケトン、酸、アルカリ、塩、エーテル、エステル等の化学物質/薬物を用いた処理によって死滅させる。
本発明では、癌細胞のマイコバクテリウムwに対する割合は、好ましくは1:10〜1:1000の範囲内である。
本発明では、癌細胞のマイコバクテリウムwに対する最も好ましい割合は、1:10〜1:100の範囲内である。
癌細胞の免疫原性プロファイルを変化させるために必要なマイコバクテリウムwの存在時間は、1分以上である。この時間は、癌細胞を体内に投与するまででもよい。
癌細胞の免疫プロファイルの変化が起こる温度は、1〜60℃の範囲である。
癌細胞の免疫プロファイルの変化に必要な培地環境は、生理食塩水、緩衝液、栄養培地またはこれらの組み合わせから選択される。栄養倍地は、癌細胞が繁殖および/または生存し続ける培地である。
本発明では、マイコバクテリウムwによって引き起こされる死滅は、全細胞の10%より多く、好ましくは30%より多く、最も好ましくは全癌細胞の60〜80%である。
本発明の目的のための癌細胞は、生きている癌細胞であるか、死滅した癌細胞でもよい。
本発明では、免疫学的プロファイルを獲得した癌細胞は、死滅しても、同一のプロファイルを保持する。本発明によって調製した癌細胞は、免疫反応を引き起こす。
自己癌細胞は、同一の生物/哺乳類/ヒト/患者から、単離、精製、誘導および/または修飾される。
治療用癌ワクチンの効果は、特異的抗原に対する免疫反応を誘導する能力、また、抗原に対して反応する能力によって測定する。本発明では、抗原は、癌細胞をマイコバクテリウムWと共にインキュベートすることによって免疫学的特性を変化させた癌細胞である。
同様に、異種免疫反応であるが、組織/器官に特異的な反応を誘導する能力も、非同種組織/器官に特異的な癌細胞株(異種)の刺激に反応した、特異的免疫反応によって測定した。異種反応もまた、インターフェロンγELISPOTを用いた細胞性反応およびウェスタンブロットを用いた液性反応の両方について評価した。
A) 同種Mia-paca-2癌細胞を採取し、ダルベッコのリン酸緩衝生理食塩水(DPBS)で洗浄して、微量の血清を除去する。生存細胞を計数し、「マイコバクテリウムW」を、細胞1個:「マイコバクテリウムW」比が1:100で、細胞に添加する。この細胞懸濁液を37℃で6時間インキュベートする。細胞懸濁液を350gで10分間遠心分離し、「マイコバクテリウムW」を分離除去する。細胞内p38値を測定する。p38値が上昇した細胞は、ワクチンとして使用するか、さらに調製してもよい。要すれば、アジュバントをそれに添加してもよい。
A. インターフェロンγELISPOTによって示される通り、治療用ワクチンは、同種癌細胞に対する細胞性免疫反応を引き起こす。
マウスモデルにおける免疫原性試験によって示される通り、実施例1に記載された方法によって調製された治療用癌ワクチンは免疫原性を有し、Th1型の免疫反応を引き起こす。簡単に説明すると、マウスを、賦形剤の対照薬か、2x106個の細胞を含むワクチン製剤で、0日目および21日目に皮内免疫した。28日目に、動物をCO2の過剰暴露によって安楽死させ、脾細胞のうちのIFN-g分泌細胞数による試験で、免疫状態を測定した。表1に示す通り、賦形剤対照群と比較して、ワクチン製剤で免疫した群においては、IFN-g分泌細胞の数の有意な増加(8.4倍)が見られた。
マウスをランダムに2群に分けた。マウスの第一群は、実施例1〜6で調製した治療用癌ワクチンで0および21日目に皮内に免疫し、第二群、即ち対照群はPBSで免疫した。試験28日目に全てのマウスから血清サンプルを分離し、ワクチンに対する抗体産生を検出した。
図1に示すウェスタンブロット分析から、治療用癌ワクチンでの免疫化によって、同種細胞溶解物に対する抗体が産生されることが分かる。
A. インターフェロンγ産生細胞におけるELISPOT上昇によって示される通り、治療用ワクチンは、組織/器官に特異的な異種癌細胞に対する細胞性免疫反応を引き起こす。
マウスを、対照薬か、実施例1の通りに調製した癌ワクチン製剤による2x106個のMia-Paca-2細胞で、0日目および21日目に皮内免疫した。28日目に、マウスをCO2の過剰暴露によって安楽死させた。各マウスから脾臓を採取し、脾細胞を単離した。各マウスから、5x105個の脾細胞を、IFN-gの捕捉抗体でコートした、R&DシステムズのELISPOTプレートに播種した。細胞をインビトロで、Mia-PaCa-2、Panc-1およびAsPC-1の溶解物10 ug/mlで刺激し、37℃および6% CO2で〜36時間インキュベートした。インキュベーション期間の最後に、プレートを製造元の使用説明書に従って展開した。簡単に説明すると、細胞を洗い流し、検出抗体を加えた。プレートを室温で2時間インキュベートした。次に、ストレプトアビジン-ALP結合酵素を加え、その後、沈殿基質BCIP-NBTを加えた。自動免疫スポットリーダーを用いて、スポット数を計測した。膵臓起源の同種癌細胞(Mia-paca-2細胞)および異種癌細胞(Panc-1およびAsPC-1)に対する癌ワクチン製剤の免疫反応性を、図2に示した通り、IFN-g分泌細胞数によって測定した。この知見より、mia-paca-2細胞を用いて本発明で調製された癌ワクチンは、Mia-paca-2細胞(同種)のみならず、Panc-1およびAsPC-1細胞(異種)に対する免疫反応を引き起こす能力があることが示唆される。種々の細胞型に対して引き起こされる免疫反応に有意差はない。
Balb/cマウスを、癌細胞(Mia-Paca-2)か、実施例1〜6に従って調製した治療用癌ワクチンで、0日目および21日目に免疫した。マウスを屠殺し、単離した脾細胞をMia-paca2およびPanc-1細胞株に対するエフェクター細胞として使用した。図3に示した結果から、治療用癌ワクチンは、癌細胞における交差免疫をもたらす能力があることが示される。治療用癌ワクチンは、両細胞株、即ち、膵臓起源の同種癌細胞(Mia-paca-2細胞)および異種癌細胞(Panc-1およびAsPC-1)に対してエフェクター機能を示す。
マウスの第一群は、MiaPaCa-2細胞で0日目および21日目に皮内免疫し、第二群は、実施例1〜6で調製した通りの治療用癌ワクチンで免疫した。試験28日目に、全てのマウスから脾細胞懸濁液(細胞107個/mL)を調製し、ELISPOTによりIFN-g分泌細胞を評価した。
対照群および試験群の両者の脾細胞を、ELISPOTプレートの別個のウェルに細胞1x106個/ウェルの密度で加えた。プレートを37℃および6% CO2で36〜48時間インキュベートした。インキュベーション期間の完了後、プレートから細胞をデカントし、DPBSで洗浄した。各ウェルに、1:100に希釈した検出抗体100μlを加えた。プレートを4℃で一晩インキュベートし、その後、洗浄し、軽く叩いて水分を切った。ストレプトアビジン-ALP結合体をPBS - 5% FBSで1:1000に希釈した。1:1000に希釈したストレプトアビジン-ALP結合体100μlを各ウェルに加えた後、暗所で、室温で1時間インキュベートした。プレートを洗浄し、軽く叩いて水分を切った。各ウェルにBCIP-NBT基質100μlを加えた。再度、プレートを、はっきりしたスポットが現れるまで、暗所で室温でインキュベートした。プレートを水洗することによって反応を停止した。プレートを37℃で一晩保持して乾燥させた。図4に示される結果から、本発明によって調製される治療用癌ワクチンは、異種癌細胞に対するマウスの免疫反応を高めることが明らかであった。
マウスを治療用癌ワクチンで、0日目および21日目に皮内免疫した。試験の28日目にマウスから血清サンプルを単離して、ワクチンに対する抗体の産生、およびマウス癌ワクチン抗体の、(異種の)膵臓起源の他の細胞、即ちSW1990およびAsPCとの異種反応性を検出した。
MiaPaCa-2、AsPC-1、SW-1990の溶解物、および、HEK-293(腎臓)、PC-3(前立腺)、MCF-7(***)、A549(肺)、PA-1(卵巣)等の種々の起源の癌によるウェスタンブロットを、治療用癌ワクチンに対してマウスにおいて産生された一次抗体を用いて実施した。溶解物抗原と結合した一次抗体を検出するため、HRP標識ヤギ抗マウスIgG抗体を、着色/検出剤としてのDAB(ジアミノベンジジン)と共に使用した。
ウェスタンブロット分析(図5)により、マウス抗癌ワクチン抗体が、膵臓起源(異種)の他の癌細胞の溶解物に対する異種反応性を有し、異なる組織/器官の癌細胞溶解物には反応性がないことが示される。
Balb/cマウスに、「マイコバクテリウムW」と1:100の割合で混合した、熱殺菌癌細胞(Mia-Paca-2膵臓癌)細胞、熱殺菌癌細胞(Mia-Paca-2膵臓癌)細胞を0日目および21日目に投与して免疫した。28日目にマウスを屠殺し、脾細胞を単離し、同種の癌細胞株に対するエフェクター細胞として使用した。
図6に示した結果から、「マイコバクテリウムW」の添加により、悪性腫瘍の治療に使用するための熱殺菌細胞を用いた治療用ワクチンの効果は向上し、これは、エフェクター機能である標的癌細胞の死滅によって示される通り、組織/器官に特異的な異種癌細胞に対する免疫反応を引き起こすことが示される。
Balb/cマウスを、ホルムアルデヒド処理した癌細胞(Mia-Paca-2膵臓癌)細胞、「マイコバクテリウムW」と種々の割合で混合したホルムアルデヒド処理した癌細胞(Mia-Paca-2膵臓癌)細胞で、0日目および21日目に免疫した。マウスを屠殺し、単離した脾細胞を、同種の癌細胞株に対するエフェクター細胞として使用した。
図7に示した結果から、「マイコバクテリウムW」の添加により、悪性腫瘍の治療に使用するための熱殺菌細胞を用いた治療用ワクチンの効果は向上し、これは、エフェクター機能である標的癌細胞の死滅によって示される通り、組織/器官に特異的な異種癌細胞に対する免疫反応を引き起こすことが示される。
Balb/cマウスを、種々のアジュバント、即ち「マイコバクテリウムW」、BB2、G1、Cadi OFF 10、およびこれらの組み合わせと混合した死滅癌細胞(Mia-Paca-2膵臓癌)で0日目および21日目に免疫した。マウスを屠殺し、単離した脾細胞を、IFN-γELISPOT同種癌細胞溶解物に使用した(図8)。
図8に示した結果から、アジュバントの添加は、悪性腫瘍の治療に使用するための死滅細胞を用いた治療用癌ワクチンの効果を向上させることが示される。
A. インビボでの腫瘍退縮:本発明の範囲を限定しない、本発明による哺乳類における癌の治療
雄のBalb/Cマウス(6〜8週)30匹を試験に用いた。動物は体重に基づいてランダムに分けた。マウスの後肢に1x105個のB16-F1細胞を皮下注射することによって、腫瘍誘導を行った。マウスは平均腫瘍サイズを100〜150 mm3まで増殖させ、腫瘍サイズに基づいて各15匹ずつ2群にランダムに分けた。マウスの第一群は、ランダム分類後0および10日目にメラノーマワクチン(実施例1により調製)で皮内免疫し、一方、第二群、即ち対照マウスは免疫しなかった(非処置)。腫瘍サイズが動物の体重の10%に達するまで、腫瘍サイズを週に2回記録した。
処置群では、非処置の動物と比較して、増殖の遅延および腫瘍量の退縮が示された。
雄のC57マウス(6〜8週)20匹を試験に用いた。動物は体重に基づいてランダムに分けた。マウスの後肢に1x105個のPan 02細胞を皮下注射することによって、腫瘍誘導を行った。マウスは平均腫瘍サイズを200 mm3まで増殖させ、腫瘍サイズに基づいて各10匹ずつ2群にランダムに分けた。マウスの第一群は、ランダム分類後0および10日目に膵臓癌ワクチン(実施例1により調製)で皮内免疫し、一方、第二群、即ち対照マウスは免疫しなかった(非処置)。腫瘍サイズが動物の体重の10%に達するまで、腫瘍サイズを週に2回記録した。
処置群では、非処置の動物と比較して、増殖の遅延および腫瘍量の退縮が示された。
マウスおよびエクスビボの免疫分析を用いる他の実験から、癌細胞ワクチンはエフェクター機能を引き起こす能力があることが示され、即ち、免疫系およびその一部は、組織/器官に特異的な異種性の標的癌細胞を効果的に死滅させる能力があることを意味する。また、このワクチンは、癌の治療において、インビボでの有効性を示す。
従って、癌ワクチンは、癌を遅延させ、高め、または治療する観点で癌の管理に使用した。また、このワクチンは、既存の腫瘍および癌細胞の負担を退縮させるために使用できる。
Claims (11)
- 抗原を発現する癌細胞、賦形剤、所望によりアジュバントを含む癌ワクチン組成物であって、前記抗原は、前記癌細胞がp38インデューサと接触する際に発現され、前記の発現された抗原が、前記癌細胞が属するものと同種の器官/組織に特異的である、上記組成物。
- 抗原を発現する癌細胞が104〜109である、請求項1に記載の組成物。
- 哺乳類に投与された場合に、癌細胞が属する組織/器官に特異的な細胞性免疫反応を引き起こす、請求項1に記載の組成物。
- 哺乳類に投与された場合、癌細胞が属する組織/器官に特異的な体液性免疫反応を引き起こす、請求項1に記載の組成物。
- アジュバントがマイコバクテリウムwである、請求項1に記載の組成物。
- 癌細胞が患者および/または細胞リポジトリより得られる、請求項1に記載の組成物。
- 請求項1に記載された組成物の治療的有効量を哺乳類に投与することを含む、哺乳類において癌を治療する方法。
- 癌が同種または異種細胞で構成される、請求項7、8及び9に記載の方法。
- 治療用ワクチン組成物が同一または他の哺乳類から得た癌細胞を含む、請求項7に記載の方法。
- 投与経路が非経口経路から選択される、請求項7に記載の方法。
- 好ましい投与経路が皮内である、請求項7に記載の方法。
