Abstract
Description
a)好適な基質(substrate)に、受入番号DSM23589のラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はその突然変異体若しくはバリアントを植え付け;
b)標準的な発酵条件などのビタミンK2生産及び/又は細菌の代謝に好ましい条件下で基質を発酵させ;
c)場合により、追加的な微生物及び/又は添加物を基質に加え;そして
d)場合により、発酵食品又は飼料をパッケージングすること
を含む、前記方法に関する。
a)細菌の代謝及び/又はビタミンK2生産に好ましい条件下で、ラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はその突然変異体若しくはバリアントを培養し;そして
b)生産されたビタミンK2を回収すること
を含む、前記方法に関する。
a)食用製品、その出発物質、又はその中間調製物に、本発明の第6の態様による乳酸発酵体、及び/又は本発明の第7の態様による方法によって生産されたビタミンK2、及び/又はラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はその突然変異体若しくはバリアントを加え;そして
b)ビタミンK2が豊富な食用製品を得ること、含む。
定義
本明細書の内容において、用語「乳(milk)」は、哺乳類又は植物の乳を含む。乳の例として、乳牛の乳(牛乳(bovine milk))、ラクダ乳、水牛の乳、山羊乳、羊乳及び豆乳が挙げられる。随意に乳を、例えば酸(クエン酸、酢酸又は乳酸)を添加することにより酸性化し、又は例えば水と混合する。乳は生のまま、又は例えば濾過、滅菌、低温殺菌、均質化することにより加工してもよく、あるいは再構成された粉乳であってもよい。本発明による「牛乳(bovine milk)」の重要な例は、低温殺菌された乳牛の乳である。乳は、細菌を植え付ける前、最中及び/又は後で酸性化、混合又は加工されてもよいことが理解される。
用語「細菌(bacteria)」は、本明細書の内容において、ここではたった1つの単独の細菌だけを含む組成物について話しても意味がないので、複数形である。
本発明の発明者らは、数種の乳酸菌、具体的には、ラクトコッカス・ラクティス種のスクリーニングにおいて、ラクトコッカス・ラクティス亜種クレモリス株CHCC12675が、ビタミンK2生産性に優れ、そして最大22μg/100mL、つまり220μg/LのビタミンK2を生産することを見出した(実施例1を参照)。よって、本発明は、乳酸菌株の生物学的に純粋な培養物を提供し、そしてそれは、多量のビタミンK2を生産できるかについて選択された。
a)好適な基質に、ラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はその突然変異体若しくはバリアントを植え付け;
b)ラクトコッカス・ラクティス亜種クレモリス株CHCC12675の代謝及び/又はビタミンK2生産に好ましい条件下で基質を発酵させ;

d)場合により、発酵食品又は飼料をパッケージングすること
を含む、前記方法に関する。
a)ビタミンK2生産に好ましい条件下でラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はその突然変異体若しくはバリアントを培養し;そして
b)それによって生産されたビタミンK2を回収すること
を含む方法によって生産され得る。
ビタミンK2は、ラクトコッカス・ラクティスやその他の微生物の細胞膜における呼吸鎖の必須成分であり、そこではビタミンK2が電子を往復させている。よって、細胞は、呼吸できるようにビタミンK2を必要としている。そのため、我々は、呼吸型生産条件下で高収量で増殖した細胞が、多量のビタミンK2を生産し得ると推測した。
培養条件は以下のとおりであった:30℃にて振盪しながら、100mLのM17(Difco(商標)M17)+1%のラクトース、5ppmのヘミン。100mLの体積で1.0のOD600をもたらす細胞の量を取り出し、そして4℃での遠心分離によって細胞を濃縮した。細胞を、5mLの脂肪分3.5%の

Claims (21)
- 受入番号DSM23589としてDeutsche Sammlung von Mikroorganismen und Zellkulturenに寄託されたラクトコッカス・ラクティス(Lactococcus lactis)亜種(subsp.)クレモリス(cremoris)株CHCC12675、又はそれから誘導された菌株である乳酸菌株。
- 発酵食品又は飼料を製造する方法であって:
a)好適な基質に、受入番号DSM23589としてDeutsche Sammlung von Mikroorganismen und Zellkulturenに寄託されたラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はそれから誘導された菌株を植え付け;
b)好ましい条件下で前記基質を発酵させ;
c)場合により、さらなる微生物及び/又は添加剤を前記基質に添加し;そして
d)場合により、発酵食品又は飼料をパッケージングすること
を含む、前記方法。 - 前記発酵食品又は飼料が、発酵乳製品であり、そして前記好適な基質が乳である、請求項2に記載の方法。
- 前記乳に、1×106〜1×1010CFU/mlのラクトコッカス・ラクティス亜種クレモリス株、又はそれから誘導された菌株が植え付けられる、請求項3に記載の方法。
- 受入番号DSM23589としてDeutsche Sammlung von Mikroorganismen und Zellkulturenに寄託されたラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はそれから誘導された菌株を含む発酵食品又は飼料。
- 対象の骨強度を増強する方法であって、請求項5に記載の発酵食品又は飼料を前記対象に投与するステップを含む、前記方法。
- 前記対象がヒトである、請求項6に記載の方法。
- 受入番号DSM23589としてDeutsche Sammlung von Mikroorganismen und Zellkulturenに寄託されたラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はそれから誘導された菌株を含む乳酸発酵体。
- 前記乳酸発酵体が、乾燥状態、凍結乾燥状態又は液体状態で存在する、請求項8に記載の乳酸発酵体。

a)好ましい条件下で、受入番号DSM23589としてDeutsche Sammlung von Mikroorganismen und Zellkulturenに寄託されたラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はそれから誘導された菌株を培養し;そして
b)生産されたビタミンK2を回収すること
を含む、前記方法。 - 食用製品を製造する方法であって:
a)食用製品、その出発物質、又はその中間体調製物に、請求項8又は9のいずれか1項に記載の乳酸発酵体、及び/又は、受入番号DSM23589としてDeutsche Sammlung von Mikroorganismen und Zellkulturenに寄託されたラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はそれから誘導された菌株を加え;そして
b)食用製品を得ること
を含む、前記方法。 - 前記哺乳動物がヒトである、請求項11に記載の方法。
- 受入番号DSM23589としてDeutsche Sammlung von Mikroorganismen und Zellkulturenに寄託されたラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はそれから誘導された菌株を含む食用製品。
- 前記食用製品が、乾燥された、単離された受入番号DSM23589のラクトコッカス・ラクティス亜種クレモリス株CHCC12675、又はそれから誘導された菌株を含む、請求項13に記載の食用製品。
- 薬剤として使用するための、請求項13又は14に記載の食用製品。
