Description
a)0.1Mリン酸バッファー中に4%パラホルムアルデヒド

c)PLP(パラホルムアルデヒド、リシン、パラホルムアルデヒド)固定液:例えば0.1Mリン酸バッファー中に4%パラホルムアルデヒド、0.2%過ヨウ素酸及び1.2%リシン
d)0.05%グルタルアルデヒドを伴う4%パラホルムアルデヒド
(1)狭スペクトル特性、
(2)高感度(量子収率)、
(3)生体適合性、
(4)分離した吸収及び励起スペクトル、及び
(5)光安定性。
(a)細胞のサブセット及び/又は結合ドメインが結合した前記細胞のサブセットを含んでなる前記組織サンプルを含んでなる手段(例えばスライド)を提供する工程;
(b)場合によっては前記組織サンプルを固定化する工程;
(c)場合によっては前記組織サンプルを脱水する工程;
(d)場合によっては組織サンプルをパラフィン処理させる工程;
(e)結合ドメイン、従って細胞のサブセットを直接的に又は間接的に検出する工程。
培養された細胞を無菌ガラスカバースリップ又はスライド上で一晩増殖させる
PBSで短く洗浄する
所望であれば固定化する。手順は以下を含みうる::
PBS中に10%のホルマリンで10分(湿らせたまま)
氷冷メタノールで5分、空気乾燥させる
氷冷アセトンで5分、空気乾燥させる
PBSにおいて洗浄する
液体窒素又は液体窒素において事前冷却されたイソペンタンにおいて瞬間凍結した新鮮な組織をO.C.T.化合物に包埋させる。凍結塊を−80℃で保存する。
PBSにおいて洗浄する
切片をキシレンにおいて、2x5分脱パラフィン化させる。
100%エタノールで、2x3分水和させる。
95%エタノールで、1分水和させる。
100%エタノールで、約2x約1分水和させる;
90%エタノールで、約1分水和させる;
80%エタノールで、約1分水和させる;
70%エタノールで、約1分水和させる;
水で約1分。
流水で約0,5分〜2分洗浄する
水で約10秒
0,1%エオシン、約1分
80%エタノールで、約10秒水和させる;
90%エタノールで、約10秒水和させる;
100%エタノールで、約10秒水和させる(切片を顕微鏡において検査);
キシレン、約1分。
(i)結合ドメインに有利に応答する傾向の被験体の同定(前記被験体は好ましくは腫瘍を患っている被験体である)、又は
(ii)治療、好ましくは結合ドメインに基づく抗-癌/腫瘍治療のモニタリング(すなわち、結合ドメインが治療的に有効)、又は
(iii)インビボで標的に結合可能な結合ドメインの同定、
(iv)標的によって特徴付けられている腫瘍に治療的に有効に作用する傾向の結合ドメインの選択;
(v)腫瘍の典型化。
以下の実施例は発明を説明する。これらの実施例は、この発明の範囲を限定すると解釈されるべきではない。実施例は説明のために含まれ、本発明は請求の範囲によってのみ制限される。
これらの新規なアプローチの検証のために、我々は50マイクログラムのCy5標識ハーセプチン、オムニターグ及びゾレアを、Calu3(Her2陽性腫瘍)を有するマウスに注入した。ハーセプチン及びオムニターグはHer2に対する治療抗体であり(ポジティブコントロール)、ゾレアはヒトIgEに結合する抗体である(ネガティブコントロール)。注入の48時間後、強い蛍光シグナルがハーセプチン-Cy5及びオムニターグ-Cy5処置マウスの腫瘍領域で検出可能であるが、ゾレア-Cy5処置グループは腫瘍にnow蛍光シグナルを示した(図1a−c)。インビボイメージング後、腫瘍を外植し、ホルマリンに固定し、その後脱水しパラフィンに包埋した。パラフィン包埋組織を2μmのスライスにカットし、顕微鏡スライドの載せ、39°Cにより一晩インキュベートした。外植腫瘍組織のその後の分析は、Her2特異的抗体は腫瘍細胞にのみ結合し、マウス組織には結合しないことを示した。対照的に、ヒトIgEに対するコントロール抗体ゾレアでは、蛍光シグナルは生成されない(図3a−3c)。
我々は第一の実施例からの結果を第二の実施例において確認した。これのために、幾つかの更なるCy5標識抗体(アービタックス、抗IGF-1R、抗Her3)以外は、同じ実験設定を使用した。ハーセプチン-Cy5及びオムニターグ-Cy5からの強いインビボシグナルが、組織スライドにおける強いエクスビボシグナリングと相関した(図4a、4b、6a、6b)。一方、アービタックス-Cy5、抗IGF-1R-Cy5及び抗Her3-Cy5は、有意なインビボ及びエクスビボ蛍光を示さなかった。インビボ及びエクスビボイメージングの結果は、実施例1の既に示したデータと完全に一致する。
続く研究は、標識抗体が腫瘍細胞に特異的に結合するか証明することを目的とした。図1aからのスライドを、蛍光インサイツハイブリダイゼーションのために使用した。このアッセイでは、Her2遺伝子がホルマリン固定組織スライドにおいて検出できる。図7bは、Cy5標識抗Her2抗体(ハーセプチン)が、Her2遺伝子を発現する細胞にのみ結合することを示す。
Claims (15)
- 細胞のサブセットを、前記細胞のサブセットを特徴付ける標的に特異的な結合ドメインを用いて検出する免疫組織化学(IHC)方法であって、該方法が前記細胞のサブセットをエクスビボで検出することを含み、前記結合ドメインが、前記細胞のサブセットの摘出より前に、前記細胞のサブセットを含むか又は含むと仮定される被験体に投与される方法。
- 前記細胞のサブセットが腫瘍細胞から成る請求項1に記載の方法。
- 前記結合ドメインが好ましくは蛍光標識で標識されている請求項1又は2に記載の方法。
- 前記標的がタンパク質、ペプチド、酵素、核酸、多糖、脂質、受容体、細胞性標的及び/又は腫瘍抗原である請求項1〜3の何れか一項に記載の方法。
- 前記結合ドメインが抗体である請求項1〜4の何れか一項に記載の方法。
- 前記細胞のサブセットが直接的免疫組織化学によって検出される請求項1〜5の何れか一項に記載の方法。
- 前記細胞のサブセットが間接的免疫組織化学によって検出される請求項1〜6の何れか一項に記載の方法。
- 直接的免疫組織化学による前記検出が、前記(蛍光性)標識結合ドメインの存在を検出することを含む請求項7に記載の方法。
- 前記被験体が、前記組織サンプルの摘出より前に、治療的に有効な用量において前記結合ドメインを受けている請求項1〜8の何れか一項に記載の方法。
- 請求項1〜9の何れか一項に定められている方法における請求項1〜9の何れか一項に定められている結合ドメインの使用。
- 請求項1〜10の何れか一項に定められている方法において使用される診断用組成物の調製のための、請求項1〜10の何れか一項に定められている結合ドメインの使用。
- 請求項1〜11の何れか一項に定められている方法において使用される、請求項1〜11の何れか一項に定められている通りの結合ドメイン。
- 請求項1〜12の何れか一項に定められている通りの結合ドメイン、及び前記結合ドメインを被験体に投与するための手段を含んでなり、請求項1〜12の何れか一項に定められている方法を用いて前記結合ドメインを検出するための手段を含んでいてもよいキット。
- 請求項1〜13の何れか一項に定められている方法であって、
(i)治療的に活性な結合ドメインに有利に応答する傾向の被験体の同定、又は
(ii)治療のモニタリング、又は
(iii)インビボで標的に結合可能な結合ドメインの同定、又は
(iv)標的により特徴づけられている腫瘍に治療的に活性に作用する傾向の結合ドメインの選択、又は
(v)腫瘍の典型化
のための方法。 - 請求項1〜14の何れか一項に記載の方法によって同定される通りの前記結合ドメインに有利に応答する傾向の患者の治療における使用のための、治療的に活性な結合ドメインであり、例えばアレムツズマブ、アポリズマブ、セツキシマブ、エプラツズマブ、ガリキシマブ、ゲムツズマブ、イピリムマブ、ラベツズマブ、パニツムマブ、リツキシマブ、トラスツズマブ、ニモツズマブ、マパツムマブ、マツズマブ、rhMab ICR62、rhMab B-Ly1及びペルツズマブ等であり、その組合せを含む結合ドメイン。
