Cry1Abコア毒素を含むタンパク質およびCry1Cコア毒素を含むタンパク質の双方を発現するように形質転換された、微生物または植物細胞である宿主(該ポリヌクレオチド(群)は好ましくは、非バチルス・チューリンゲンシス(Bacillus thuringiensis)(に作動可能的に連結した/を含む)プロモーターの調節下の遺伝的構築物中に存在し、該ポリヌクレオチドは、植物中での高められた発現のためのコドン使用頻度を含むことができる);

植物(例えば、トウモロコシ植物、またはダイズもしくはワタもしくはシュガーケーンなど)およびこのような植物の種子であって、Cry1Caコア毒素を含むタンパク質をコードするDNAおよびCry1Abコア毒素を含むタンパク質をコードするDNAは、前記トウモロコシ植物中に遺伝子移入されている、種子
を含む鱗翅目有害生物を防除するための組成物を提供する。
構造化緩衝帯:
トウモロコシ地域の20%の非鱗翅目Btトウモロコシ緩衝帯
ワタ地域の50%の非鱗翅目Bt緩衝帯
ブロック:
1.内部(すなわち、Bt耕地内)
2.外部(すなわち、無作為接合を最大化するためのBt耕地の1/2マイル(可能なら1/4マイル)以内の分離された耕地)
圃場内帯状地
帯状地は、幼虫移動の効果を低減するために少なくとも4列の幅(好ましくは6列)でなければならない。
コーンボーラーのIRMの要件
・トウモロコシ耕地少なくとも20%に保護ハイブリッド種を植え付ける
・ワタを作る地域では、緩衝帯は50%でなければならない
・保護ハイブリッド種の1/2マイル以内に植え付けるべきである
・緩衝帯は、Bt圃場内に帯状地として植えることができ;緩衝帯の帯状地は少なくとも4列の幅でなければならない

キメラ毒素。別のCry毒素のプロトキシンセグメントに融合されたあるCry毒素のコア毒素ドメインを利用するキメラタンパク質は、例えば、米国特許第5593881号および米国特許第5932209号中に既に報告されている。

キメラタンパク質をコードする発現プラスミドの構築およびシュードモナス(Pseudomonas)中での発現
完全長DIG−152キメラタンパク質を産生するように遺伝子操作された、シュードモナス・フルオレセンス(Pseudomonas fluorescens)(Pf)発現構築物pMYC2547の構築では、標準的なクローニング法[例えば、Sambrook et al(1989)およびAusubelet al(1995)、ならびにこれらの更新版中に記載のような]を使用した。タンパク質の産生は、米国特許第5169760号中に開示のように、改変されたlacオペロンの挿入を有するシュードモナス・フルオレセンス(Pseudomonas fluorescens)株MB214(株MB101の誘導体;P.フルオレセンス(P. fluorescens)次亜種I)中で実施した。基本的なクローニング戦略は、DIG−152をコードするDNAフラグメントをプラスミドベクター中にサブクローニングすることを必要とし、それによって、該プラスミドベクターは、プラスミドpKK223−3(PL Pharmacia、Milwaukee、ウィスコンシン州)からのPtacプロモーターおよびrrnBTIT2ターミネーターの発現調節下に置かれる。1つのこのようなプラスミドは、pMYC2547と命名され、このプラスミドを抱えるMB214アイソレートはDpf108と命名される。
Cry1CaおよびCry1Abコア毒素タンパク質の調製、ならびに競合結合実験で使用するためのスポドプテラ・フルギペルダ(Spodoptera frugiperda)刷子縁膜小胞の単離
次の実施例は、昆虫消化管組織中の推定受容体に対するCry1コア毒素タンパク質の競合結合を評価する。125I標識化Cry1Caコア毒素タンパク質は、スポドプテラ・フルギペルダ(Spodoptera frugiperda)(フォールアーミーアーム)から調製された刷子縁膜小胞(BBMV)に対して高い親和性で結合すること、およびCry1Abコア毒素タンパク質はこの結合と競合しないことが示されている。代わりに、125I標識化Cry1Abコア毒素タンパク質は、S.フルギペルダ(S. frugiperda)から調製されたBBMVに対して高い親和性で結合し、Cry1Caコア毒素タンパク質は、この結合と競合しないことが示されている。
スポドプテラ・フルギペルダ(Spodoptera frugiperda)からのBBMVに対する125I標識化Cry1コア毒素タンパク質の結合
Cry1CaおよびCry1Abコア毒素タンパク質との結合アッセイで使用するためのBBMVタンパク質の最適量を決定するため、飽和曲線を作出した。0.5nMの125I放射能標識化Cry1コア毒素タンパク質を、結合緩衝液(8mM NaHPO4、2mM KH2PO4、150mM NaCl、0.1%BSA、pH7.4)中、0μg/mL〜500μg/mLの範囲のBBMVタンパク質の量で(全容積0.5mL)、28℃で1時間インキュベートした。BBMVタンパク質に結合された125I標識化Cry1コア毒素タンパク質を、非結合画分から、150μLの反応混合物を三つ組みで1.5mL分離遠心管中にサンプリングし、該サンプルを室温にて14,000×gで8分間遠心することによって分離した。上清液を、穏やかに除去し、ペレットを、氷冷結合緩衝液で3回洗浄した。ペレットを含む遠心管の底部を、切り離し、13×75mmのガラス製培養管中に配置し、サンプルを、ガンマーカウンター中でそれぞれ5分間計数した。得られたCMP(計数値/分)−バックグラウンドCMP(BBMVでないタンパク質との反応)を、BBMVタンパク質濃度に対してプロットした。結合アッセイで使用するためのBBMVタンパク質の最適濃度は、別の人(Luo et al, 1999)によって報告された結果と一致して、150μg/mLであると判定された。
S.フルギペルダ(S. frugiperda)からのBBMVに対するCry1AbおよびCry1Caのコア毒素タンパク質との競合結合アッセイ
同種および異種競合結合アッセイを、150μg/mLのS.フルギペルダ(S. frugiperda)BBMVタンパク質および0.5nMの125I放射能標識化Cry1Caコア毒素タンパク質を使用して実施した。真の結合競合を保障するために、反応混合物に添加される0.045nM〜300nMの範囲の濃度の競合非放射能標識化Cry1Abコア毒素タンパク質を、放射性Cry1Caコア毒素タンパク質と同時に添加した。28℃で1時間インキュベーションを実施し、BBMVに結合された(特異的結合)125I標識化Cry1Caコア毒素タンパク質の量を、前記のように測定した。非特異的結合は、1,000nMの非放射能標識化Cry1Caコア毒素タンパク質の存在下に得られた計数値によって表された。100パーセントの総結合を、全ての競合Cry1Abコア毒素タンパク質の不在下における結合量であるとみなした。

Claims (28)
- Cry1C殺虫性タンパク質をコードするDNAおよびCry1Ab殺虫性タンパク質をコードするDNAを含むトランスジェニック植物。
- 請求項1に記載の植物の種子。
- Cry1Ca殺虫性タンパク質をコードするDNAおよびCry1Ab殺虫性タンパク質をコードするDNAが遺伝子移入されている、請求項1に記載のトランスジェニック植物。
- 請求項3に記載の植物の種子。
- 非Bt緩衝帯植物、および請求項1に記載の複数のトランスジェニック植物を含む植物の圃場であって、前記緩衝帯植物が、前記圃場の全ての作物植物の40%未満を構成する、圃場。
- 前記緩衝帯植物が、前記圃場の全ての作物植物の30%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物が、前記圃場の全ての作物植物の20%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物が、前記圃場中の全ての作物植物の10%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物が、前記圃場の全ての作物植物の5%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物がブロックまたは帯状地にある、請求項5に記載の植物の圃場。
- 非Bt緩衝帯植物からの緩衝帯種子、および請求項7に記載の複数の種子を含む種子混合物であって、前記緩衝帯種子は混合物の全ての種子の40%未満を構成する、種子混合物。
