Abstract
Description
本発明は、発光強度の高い半導体ナノ粒子集積体に関する。 The present invention relates to a semiconductor nanoparticle assembly having high emission intensity.
近年、バイオイメージングにおける標識剤として蛍光発光する半導体ナノ粒子が用いられている。そして、半導体ナノ粒子の一粒子当たりの輝度が高いほど視認性の点で有利なことから、一粒子当たりの輝度のより高い粒子が望まれている。 In recent years, semiconductor nanoparticles that emit fluorescence are used as labeling agents in bioimaging. And since the brightness | luminance per particle | grain of semiconductor nanoparticles is so advantageous in terms of visibility, the particle | grains with higher brightness | luminance per particle | grain are desired.
As semiconductor nanoparticles emitting fluorescence, II-VI group and III-V group compound semiconductor nanoparticles are widely known. In general, the emission intensity is low only with the core semiconductor nanoparticles, but the emission intensity is improved several to several tens of times when the semiconductor nanoparticles have a core / shell structure in which a shell is added to the core particles. This is thought to be due to the fact that a quantum well structure is formed by using a semiconductor material having a wider band gap than the core particles as the shell, and the luminous efficiency is significantly improved by the quantum confinement effect.
また、バイオ標識体をより高輝度化させる1つの手法として、半導体ナノ粒子の集積化が考えられている。即ち、半導体ナノ粒子を集積化させることにより、1粒子あたりの発光強度を向上させる技術である。 In addition, integration of semiconductor nanoparticles is considered as one technique for increasing the brightness of biolabels. That is, it is a technique for improving the emission intensity per particle by integrating semiconductor nanoparticles.
However, since semiconductor nanoparticles are generally spherical, when they are integrated, voids and spaces that do not participate in light emission are formed between the semiconductor nanoparticles. This space is a region where excitation light cannot be received. When considering the luminous efficiency of one particle of the semiconductor nanoparticle assembly, the luminous efficiency per unit volume of the particle is lower than that of the semiconductor nanoparticle before the integration. Become. For example, in recent years, fluorescent glass particles have been proposed in which semiconductor nanoparticles are confined so as to be dispersed and fixed in a matrix such as glass. In this regard, Patent Document 1 discloses a phosphor by dispersing cadmium telluride nanoparticles and zinc selenide nanoparticles in a matrix containing silicon using a sol-gel method in the presence of an organoalkoxysilane. Is obtained. Patent Document 2 describes fluorescent glass particles obtained by using a sol-gel method in which cadmium telluride nanoparticles are present in the presence of an organic alkoxysilane.
In such fluorescent glass particles, the space between the semiconductor nanoparticles is filled with a glass material, but the glass material generally has a large band gap energy and cannot absorb the visible light normally used as excitation light for bioimaging, A decrease in luminous efficiency per unit volume of the aggregate occurs. In addition, there are often localized levels such as dangling bonds in the outermost layer part of the semiconductor nanoparticle assembly, and in this level, recombination of electron-hole pairs that do not contribute to light emission occurs. This also causes a problem that the luminous efficiency decreases.
本発明は、上記課題・状況に鑑みてなされたものであり、その解決課題は、高い発光強度を有する半導体ナノ粒子集積体を提供することである。 The present invention has been made in view of the above-described problems and circumstances, and a problem to be solved is to provide a semiconductor nanoparticle assembly having high emission intensity.

ここで本発明は、外部から与えられたエネルギーを内部に閉じ込めるために、半導体ナノ粒子集積体101をシェル材102が被覆した構造を有しているという点に最大の特徴がある。これについて、図1を用いて説明する。
Here, the present invention is characterized in that it has a structure in which the
In the conventional
ただ、従来の半導体ナノ粒子集積体200では、外部から与えられたエネルギーの一部が、集積体内部に伝達することなく外部に流出することから、外部から与えられたエネルギーから光エネルギーへの変換が充分に行われない。
However, in the conventional
On the other hand, in the shell material-coated
Note that in this specification, “fluorescence” refers to light emission in the process from the excited state to the ground state when excited by irradiation with external X-rays, ultraviolet rays, or visible light. Therefore, the “fluorescence” referred to in the present invention is not limited to the transition mode when returning from the excited state to the ground state, and may be narrowly defined fluorescence that is light emission accompanying deactivation from the excited singlet. In addition, phosphorescence accompanying deactivation from a triplet may be used.
-Semiconductor The semiconductor used in the present invention is a substance that exhibits intermediate electrical properties between a good conductor and an insulator, and is a process in which electrons are excited by receiving a certain amount of energy from the outside and then return to the ground state from the excited state. There is no particular limitation as long as it causes fluorescence emission.
However, in the present invention, the semiconductor constituting the

In the present specification, “semiconductor nanoparticle aggregate” refers to a particle containing an aggregate in which a plurality of semiconductor nanoparticles are in contact with each other.
半導体ナノ粒子
本発明において、半導体ナノ粒子集積体101を構成する半導体ナノ粒子110は、本発明のシェル材被覆半導体ナノ粒子集積体100において、外部から与えられたエネルギーを受けて励起し、励起状態から基底状態に戻る過程で蛍光を発する役割を有する。

本発明において、半導体ナノ粒子110は、上記「半導体」の項で上述した半導体を含んでおり、このような半導体として、特に、CdSe、InPがより好ましく用いられる。 In the present invention, the semiconductor nanoparticles 110 include the semiconductor described above in the section “Semiconductor”, and CdSe and InP are particularly preferably used as such a semiconductor.
The semiconductor constituting the semiconductor nanoparticles 110 may be a single type or a combination of two or more types. Here, when two or more kinds of semiconductors are used as the semiconductor nanoparticles 110, two or more kinds of semiconductor nanoparticles composed of one kind of semiconductor may be combined to be used as the semiconductor nanoparticles 110, and the semiconductor nanoparticles 110 are composed of a mixture of two or more kinds. One type of semiconductor nanoparticle may be used as the semiconductor nanoparticle 110, or a combination of two or more types of semiconductor nanoparticles made of a mixture of two or more types may be used as the semiconductor nanoparticle 110.
Further, the semiconductor nanoparticles 110 may have a uniform structure as a whole, may have two or more structures having different composition components, or the composition of the component components depends on the position. You may have the structure which changes continuously. In the present invention, it is particularly preferable that the semiconductor nanoparticles 110 have a core / shell structure from the viewpoint of high luminous efficiency. That is, in a preferred embodiment of the present invention, the semiconductor nanoparticles 110 are core-shell semiconductor nanoparticles comprising a
また、粒径によっても発光波長が変わるという半導体ナノ粒子の性質に着目すると、2種以上の発光波長を有するシェル材被覆半導体ナノ粒子集積体100を得るために、構成成分が同じで粒径のみが異なる2種以上の半導体ナノ粒子を、半導体ナノ粒子110として用いることもできる。
Further, focusing on the property of the semiconductor nanoparticles that the emission wavelength varies depending on the particle size, in order to obtain the shell-coated
ただ、いずれの場合にも、半導体ナノ粒子110を構成する半導体は、本発明のシェル材被覆半導体ナノ粒子集積体100を構成するそれ以外の部分を構成するいかなる半導体に対しても、同等またはそれより小さいバンドギャップエネルギーを有することが好ましく、少なくとも、シェル材102を構成する材質よりも小さいバンドギャップエネルギーを有することが好ましい。
However, in any case, the semiconductor constituting the semiconductor nanoparticles 110 is equivalent to or equal to any semiconductor constituting the other parts constituting the shell material-coated
The semiconductor nanoparticles 110 may also contain other components that do not impair the function of the constituent semiconductor as a phosphor, such as stabilizers, surfactants, and solvents that can be used in the synthesis process.

In the present invention, the

In the present invention, it does not prevent the use of a material other than the semiconductor as the material constituting the

Specific examples of the method include a liquid phase method and a gas phase method.
Liquid Phase Method In the present invention, the liquid phase method can be suitably used as a method for producing the semiconductor nanoparticles 110. In the liquid phase method, each semiconductor constituting the semiconductor nanoparticles 110 is obtained by chemically reacting a corresponding semiconductor precursor in an appropriate solvent. Examples of the production method based on the liquid phase method include a precipitation method, a coprecipitation method, a sol-gel method, a uniform precipitation method, and a reduction method. In addition, the reverse micelle method, the supercritical hydrothermal synthesis method, and the like are also excellent methods for producing nanoparticles (for example, JP 2002-322468, JP 2005-239775, JP 10-310770 A). No., JP 2000-104058 A, etc.).

Further, in the reaction by the liquid phase method, an embodiment having a step of performing the reaction of the semiconductor precursor in the presence of a surfactant is preferable. Also, in the presence of a stabilizer such as tri-n-octylphosphine oxide (TOPO) so that the formed semiconductor nanoparticles 110 (and
The reaction temperature at the time of introducing the semiconductor precursor to the required semiconductor is not particularly limited as long as it is not lower than the boiling point of the semiconductor precursor and not higher than the boiling point of the solvent, but is preferably in the range of 70 to 110 ° C.
<Reducing agent>
As the reducing agent for reducing the semiconductor precursor, various conventionally known reducing agents can be selected and used according to the reaction conditions. In the present invention, from the viewpoint of the strength of reducing power, lithium aluminum hydride (LiAlH 4 ), sodium borohydride (NaBH 4 ), sodium bis (2-methoxyethoxy) aluminum hydride, trihydride (sec- Reducing agents such as lithium (butyl) boron (LiBH (sec-C 4 H 9 ) 3 ), potassium tri (sec-butyl) borohydride, and lithium triethylborohydride are preferred. In particular, lithium aluminum hydride (LiAlH 4 ) is preferable because of its reducing power.
〈溶媒〉
半導体前駆体の分散用溶媒としては、従来周知の種々の溶媒を使用できるが、エチルアルコール、sec−ブチルアルコール、t−ブチルアルコール等のアルコール類、トルエン、デカン、ヘキサンなどの炭化水素類溶媒を使用することが好ましい。本発明においては、特に、トルエン等の疎水性の溶媒が分散用溶媒として好ましい。

Various known solvents can be used as the solvent for dispersing the semiconductor precursor. Alcohols such as ethyl alcohol, sec-butyl alcohol, and t-butyl alcohol, and hydrocarbon solvents such as toluene, decane, and hexane are used. It is preferable to use it. In the present invention, a hydrophobic solvent such as toluene is particularly preferable as the dispersion solvent.
<Surfactant>
As the surfactant, various conventionally known surfactants can be used, and anionic, nonionic, cationic, and amphoteric surfactants are included. Of these, tetrabutylammonium chloride, bromide or hexafluorophosphate, tetraoctylammonium bromide (TOAB), or tributylhexadecylphosphonium bromide, which are quaternary ammonium salt systems, are preferred. Tetraoctyl ammonium bromide is particularly preferable.
なお、液相法による反応は、液中の溶媒を含む化合物の状態により大きく変化する。単分散性の優れたナノサイズの粒子を製造する際には、特に注意を要する必要がある。例えば、逆ミセル反応法では、界面活性剤の濃度や種類により、反応場となる逆ミセルの大きさや状態が変わってくるため、ナノ粒子が形成される条件が限られてしまう。したがって、界面活性剤と溶媒とを適切に組み合わせることが必要となる。 The reaction by the liquid phase method greatly varies depending on the state of the compound containing the solvent in the liquid. When producing nano-sized particles with excellent monodispersity, special care must be taken. For example, in the reverse micelle reaction method, the size and state of the reverse micelle serving as a reaction field vary depending on the concentration and type of the surfactant, so that the conditions under which nanoparticles are formed are limited. Therefore, it is necessary to appropriately combine the surfactant and the solvent.

[各種測定方法]
・粒径の測定方法
本願において、コアシェル構造を持つ半導体ナノ粒子及び半導体ナノ粒子集積体の粒径(体積平均粒径)は、動的光散乱法による粒径測定装置(Malvern Instruments社製、Zetasizer Nano S)を用いて、半導体ナノ粒子又は集積体作製直後(凝集前)の粒径分布を測定することにより求めた。なお、平均粒径(体積平均粒径)は、粒径分布のピーク(中心)位置の粒径とした。
EXAMPLES Hereinafter, although an Example demonstrates this invention in detail, this invention is not limited to this.
[Various measurement methods]
-Particle diameter measurement method In the present application, the particle diameter (volume average particle diameter) of the semiconductor nanoparticles having a core-shell structure and the semiconductor nanoparticle aggregate is determined by a particle diameter measuring apparatus (Malvern Instruments, Zetasizer, by dynamic light scattering method). Using Nano S), it was determined by measuring the particle size distribution immediately after the production of semiconductor nanoparticles or aggregates (before aggregation). The average particle size (volume average particle size) was the particle size at the peak (center) position of the particle size distribution.
・各集積体への半導体ナノ粒子の内包数の算出方法
また、半導体ナノ粒子集積体、および、シェル材被覆半導体ナノ粒子集積体に内包される半導体ナノ粒子の計算は以下のようにして行った。
-Calculation method of the number of semiconductor nanoparticles included in each aggregate The calculation of the semiconductor nanoparticles included in the semiconductor nanoparticle aggregate and the shell material-coated semiconductor nanoparticle aggregate was performed as follows. .
First, the elemental ratio of the semiconductor nanoparticles is measured using a sequential high frequency plasma emission analyzer (ICPS-7500, manufactured by Shimadzu Corporation). Then, the concentration used for re-shelling is calculated by measuring the element ratio of the re-shelled semiconductor nanoparticle assembly using the high-frequency plasma emission spectrometer (ICP-AES). Since the density of the semiconductor nanoparticles and the semiconductor nanoparticle assembly is known, the inclusion number can be estimated together with the average particle diameter calculated by the dynamic light scattering method. The larger the number of inclusions, the better. For example, in a semiconductor nanoparticle assembly having a particle size of 100 nm, the number of inclusions is preferably 100 or more.

By removing, the CdSe semiconductor nanoparticle assembly precursor 22 was obtained. Next, 2 ml of octadecene was added to the CdSe semiconductor nanoparticle assembly precursor 22, and the solvent was re-substituted with octadecene. At that time, the CdSe semiconductor nanoparticle assembly precursor 22 did not re-disperse into the individual CdSe semiconductor nanoparticles 1, but maintained the form and particle size before the addition of octadecene. The octadecene mixed liquid of the CdSe semiconductor nanoparticle integrated precursor 22 is heated to 80 ° C. in a nitrogen atmosphere, and 100 mg of cadmium acetate, zinc acetate, and dodecanethiol are added to 1 mg of the raw material CdSe semiconductor nanoparticles 1. At that time, the CdSe / CdS core / shell semiconductor nanoparticle assembly precursor 55 did not re-disperse into the individual CdSe / ZnS core / shell semiconductor nanoparticles 4 and maintained the form and particle size before the addition of octadecene. It was. The octadecene mixed solution of the CdSe / CdS core-shell semiconductor nanoparticle integrated precursor 55 is heated to 80 ° C. in a nitrogen atmosphere, and cadmium acetate, zinc acetate, and dodecanethiol are added to 1 mg of the raw material CdSe / CdS core-shell semiconductor nanoparticles 4. Then, 100 mg each was added, and then the temperature was raised from 80 ° C. to 230 ° C. and reacted for 30 minutes to obtain a mixed liquid of CdSe / CdS core-shell semiconductor nanoparticle aggregates 5.

また、半導体ナノ粒子及び半導体ナノ粒子集積体の粒径(体積平均粒径)は、動的光散乱法による粒径測定装置(Malvern Instruments社製、Zetasizer Nano S)を用いて、半導体ナノ粒子又は集積体作製直後(凝集前)の粒径分布を測定することにより求めた。なお、平均粒径(体積平均粒径)は、粒径分布のピーク(中心)位置の粒径とした。 In addition, the particle diameter (volume average particle diameter) of the semiconductor nanoparticles and the semiconductor nanoparticle aggregate is determined by using a particle diameter measuring apparatus (Malvern Instruments, Zetasizer Nano S) by a dynamic light scattering method. It was determined by measuring the particle size distribution immediately after fabrication of the aggregate (before aggregation). The average particle size (volume average particle size) was the particle size at the peak (center) position of the particle size distribution.
Moreover, the calculation of the inclusion number of the semiconductor nanoparticles was performed as follows.
First, the element ratio of the semiconductor nanoparticles was measured using a sequential high-frequency plasma emission analyzer (ICPS-7500, manufactured by Shimadzu Corporation). Then, the concentration used for the re-shelling was calculated by measuring the element ratio of the re-shelled semiconductor nanoparticle assembly using the high-frequency plasma emission spectrometer (ICP-AES). Since the densities of the semiconductor nanoparticles and the re-shelling compound are known, the inclusion number was estimated together with the average particle diameter calculated by the dynamic light scattering method.
