Abstract
【解決手段】分離、単離、またはペレット化した微生物サンプルは、その後、微生物のキャラクタリゼーションおよび/または同定に用いる測定値を提供する一以上の解析工程を受ける。本発明の一態様として、解析ステップは本願明細書に記載する分離装置または容器においてそのまま行うことができる。

(a) 広い内径を有する上部、
(b) 狭い内径を有する下部、および、
(c)容器の底面、上面および/または1以上の側面上の光学窓であって、近赤外、可視、および/または、紫外光スペクトルの少なくとも一部を透過する、光学窓。当該容器は、任意で、下部の狭い内径と上部の広い内径をつなげる中間の先細の部分を更に有してもよい。
(a)縦軸を持つ本体を有する円柱状容器であって、前記本体は、第1端部および第2端部を有する前記縦軸方向に細長い内部キャピラリ管を画定し、前記キャピラリ管の前記第1端部に連結された貯蔵器を更に画定する、円柱状容器。
(b)前記キャピラリ管の前記第2端部付近くの前記本体は、光学的に透明な物質からなる。
(c)前記貯蔵器への接近を可能にして、液体サンプルの供給を可能にする前記貯蔵器のカバー。
任意で、円柱状容器は、貯蔵器中に密度クッションを含んでもよい。加えて、容器は、貯蔵器およびキャピラリ管をつなぐ先細の部分を有してもよい。
PolymorphoPrep(商標))に対して例えば約75%〜約100%の濃度の、フルオロシリコンオイル、シリコーンゲル、メトリゾ酸-Ficoll(登録商標) (LymphoPrep(商標))、血液培養サンプルに対して例えば約25%〜約50%濃度のジアトリゾ酸-デキストラン、カルボキシメチルセルロース、ヒドロキシプロピルメチルセルロース、ポリエチレンオキシド(高分子量)、Pluronic(登録商標) F127、Pluronic(登録商標) F68、Pluronic(登録商標)化合物の混合物、ポリアクリル酸、クロス結合ポリビニルアルコール、クロス結合ポリビニルピロリジン、PEGジメチルエーテルメタクリル酸、ペクチン、アガロース、キサンタン、ゲラン、Phytagel(登録商標)、ソルビトール、Ficoll(登録商標)(血液培養サンプルに対して例えば約10%〜約15%濃度のFicoll(登録商標)400)、グリセリン、デキストラン(例えば、血液培養サンプルに対して約10%〜約15%濃度)、グリコーゲン、塩化セシウム(例えば、血液培養サンプルに対して約15%〜約25%濃度)、パーフルオロカーボン液体(例えばペルフルオロ-n-オクタン)、ヒドロフルオロカーボン液体(例えばVertrel XF)などの周知の物質を含む。一実施形態で、密度クッションは、一つ以上のコロイダルシリカ、イオジキサノール、イオヘキソール、塩化セシウム、メトリゾ酸-Ficoll(登録商標)、ジアトリゾ酸-デキストラン、ショ糖、Ficoll(登録商標) 400および/またはあらゆる組合せのデキストランから選択する。密度クッションはまた、物質の組合せ、例えばコロイダルシリカおよび油の組合せから作製することができる。上記の化合物の特定の組合せは、本発明の分離および読出ステップのために有益である。例えば、異なるUV消光性の化合物、例えば塩化セシウムおよびイオヘキソールの組合せである。
査ステップは、微生物のキャラクタリゼーションおよび/または同定に有効な計測を提供するために実施することができる。有効な解析手段は、従来技術で既知である。更なる解析手段は、先に記載した本発明の譲受人に譲渡された米国特許出願中に記述されている。
精製された微生物ペレットの生体内同定用装置および方法
分離装置中の微生物の速やかなin situ分離および同定の可能性を探求するため、本発明に従って、いくつかの装置をUV透過プラスチックから設計及び成形した。これらの装置は、閉鎖具、サンプル貯蔵器、下部および/または側面から沈殿した微生物のペレットを分光学的解析できるようにする先細の光学品質下部領域、および装置を分光蛍光計に連結することを容易にする構造等のいくつかの共通構造を有する。装置は、また、分離ステップの間、比較的大きい重力加速度に耐えることができなければならない。このチューブのいくつかの反復(iteration)は、微生物の回収および蛍光再現性を高め、および、迷散乱光の混入を減らすように設計された。当該チューブはまた、封止できるように設計された。
Claims (20)
- 微生物を単離および同定するための容器であって、
前記容器は、
(a)広い内径を有する上部、
(b)狭い内径を有する下部、および
(c)前記容器の底面、上部および/または1以上の側面上の光学窓であって、近赤外、可視、および/または、紫外光スペクトルの少なくとも一部を透過する光学窓、
を備える、微生物を単離および同定するための容器。 - 前記容器は、微生物のペレットを作製するのに十分なサンプルを保持するために十分な体積を有する請求項1に記載の容器。
- 前記上部および下部は、中間の先細部分によって連結される、請求項1に記載の容器。
- 前記下部は、容器全体の高さの半分未満である、請求項1に記載の容器。
- 前記容器は、当該容器を封止するように、閉鎖具を備え、又は閉鎖具を受けるようにねじ山を有する、請求項1に記載の容器。
- 前記光学窓は、クォーツ、石英ガラス、サファイヤ、アクリル、メタクリル酸エステル、環状オレフィン共重合体、シクロオレフィンポリマーまたはそれらのあらゆる組合せから成る、請求項1に記載の容器。
- 前記容器は、内部に密度溶液を含む、請求項1に記載の容器。
- 前記容器は、選択的な溶解溶液を更に含む、請求項7に記載の容器。
- 前記装置は、密度クッションおよび溶解溶液を分離する薄い膜を更に備える、請求項8に記載の分離装置。
- 前記容器は、ポリプロピレン・ボールを更に備える、請求項7に記載の容器。
- 使い捨ての分離装置であって、
(a)縦軸を持つ本体を有する円柱状容器であって、前記本体は、第1端部および第2端部を有する前記縦軸方向に細長い内部キャピラリ管を画定し、前記キャピラリ管の前記第1端部に連結された貯蔵器を更に画定する、円柱状容器を形成し、
(b)前記キャピラリ管の前記第2端部付近の前記本体は、光学的に透明な物質からなり、
(c)前記貯蔵器への接近を可能にして、液体サンプルの供給を可能にする前記貯蔵器のカバー
を備える、分離装置。 - 前記装置は、貯蔵器の中に含まれる密度クッションを更に備える、請求項11の分離装置。
- 前記装置は、前記密度クッション上部に重ねられる溶解溶液を更に含む、請求項12に記載の分離装置。
- 前記装置は、密度クッションおよび溶解溶液を分離する薄膜を更に備える、請求項13に記載の分離装置
- 前記貯蔵器は、0.5〜15mlの液体容量を有する、請求項11に記載の分離装置。
- 前記装置は、本体に固定される末端部であって、前記キャピラリ管の前記第2端部を閉鎖し、光学的に透明な物質からなる末端部を更に有する、請求項11に記載の分離装置。
- 前記装置は、前記貯蔵器および前記キャピラリ管の間にある先細部を更に備える、請求項11に記載の分離装置。
