Abstract
Description
The present invention relates to a laser microscope.
A scanning confocal microscope (a type of laser microscope) forms illumination light from a laser light source on a specimen through an objective lens, and scans spot-shaped light irradiated on the specimen two-dimensionally. Observation. When the sample is irradiated with laser light, reflection, absorption, fluorescence, scattering, and the like occur in the irradiated region due to the optical characteristics of the sample. Reflected light and fluorescence generated in the irradiation region are collected by the objective lens and then detected by the detector. Usually, a plurality of detectors are provided, and light of a desired wavelength is detected by the detector using a filter or the like (see Patent Document 1).
However, in the above prior art, the size of the focused spot is limited to the wavelength of the laser beam due to the limitation of the light diffraction limit. For example, when the thickness direction of the specimen is the Z direction, There was a problem that the resolution was limited.

According to the present invention, in the scanning confocal microscope, the resolution of the specimen in the XY plane can be improved.
Hereinafter, an embodiment according to a scanning confocal microscope of the present invention will be described with reference to the drawings. FIG. 1 is a diagram illustrating a configuration of a scanning confocal microscope according to the present embodiment. 1, 3, and 4, the XYZ coordinate system is used, the stage surface on which the
The
FIG. 2 is a diagram illustrating a schematic configuration of the
The
The parallel light from the
Here, the single polarization component defines a cross section perpendicular to the traveling direction of the beam, is a uniform polarization state (linearly polarized light, elliptically polarized light, circularly polarized light, etc.) at any coordinate in the plane, and The phase state of the polarized light in the plane can be regarded as almost constant. On the other hand, depending on the coordinate position in the cross section, the plurality of polarization states are at least one of having a non-uniform distribution of the polarization state or having a distribution of the in-plane polarization state according to the coordinates. It satisfies.
3 is a diagram schematically illustrating the configuration of the
The divided
The divided
By the way, when the wavelength of the laser light emitted from the
In FIG. 3A, for easy understanding, the optical axes expressed in the divided
Therefore, the optical axes of the divided areas facing each other with respect to the center point are orthogonal to each other. For example, the optical axis of the divided
In the present embodiment, a beam composed of a single polarization component composed of linearly polarized light whose polarization axis is along the Z direction is made incident on the
Next, the polarization state of the light emitted from the
Specifically, the polarization axes of linearly polarized light emitted from a pair of divided regions facing the center point are parallel. And a vibration direction is opposite in a pair of division area which opposes. Further, the polarization axis of the light emitted from the adjacent divided regions is shifted by 45 °. For example, the polarization axis of the light emitted from the divided
As described above, in the present embodiment, when linearly polarized light along a predetermined direction is incident on the
In addition, by increasing the number of divided regions of the
Specifically, for example, when the number of divisions is 16, the optical axis of the wave plate is shifted by 11.25 ° from the adjacent division region. As a result, the optical axes are orthogonal to each other in the divided areas facing each other. When a polarization axis having a certain angle is incident on the polarization control element, linearly polarized light is emitted as pseudo radial polarized light.
In the present embodiment, the pseudo-radially polarized light emitted from the
Here, the axially symmetric polarized donut beam in the present embodiment is attenuated in the intensity near the center in the beam cross-sectional intensity distribution than the axially symmetric polarized beam of normal radial polarization obtained by solving the Maxwell equation on cylindrical coordinates. Or it means a beam with a small area of weak intensity.
Therefore, the light that has passed through the
FIG. 5 is an enlarged view of the
The laser light emitted from the
FIG. 6B shows the irradiation state of the laser light L at an arbitrary time, and reflection, absorption, fluorescence, scattering, and the like occur in the irradiation region of the laser light L due to the optical characteristics of the
In the present embodiment, since the laser light L passes through the
Fluorescence emitted from the
The fluorescence is imaged on the
As shown in FIG. 2, the fluorescence transmitted to the
According to the present embodiment, the
The

In the above-described embodiment, the case where the
Moreover, although the said embodiment demonstrated the case where each division | segmentation area | region 151-158 of the
Moreover, even if it is the structure which changes each phase difference (optical rotation) by controlling each division area 151-158 independently, and converts the laser beam which passed the
The case where the
Examples of the optical device include a capturing device using an optical tweezers (LOT) technique that intentionally grabs and moves fine particles existing inside and outside the cell using the energy of irradiation light. In such an optical tweezer technology, it is preferable to increase the gripping force. For this purpose, it is necessary to increase the numerical aperture of the objective lens. However, in general, an objective lens having a large numerical aperture has a large magnification and cannot be used in a wide-field observation state. On the other hand, if the present invention is employed, the use of the
Further, by using the above-described optical device as illumination light, the present invention can be applied to a scanning multiphoton excitation laser microscope (for example, a scanning two-photon excitation laser microscope). Specifically, for example, even in a scanning two-photon excitation laser microscope in which a deflection control element is installed between a light source unit and an objective lens, the resolution (XY resolution) of a plane for scanning light can be improved. .
The fluorescence intensity distribution during one-photon observation is proportional to the first power of the excitation light PSF (Point Spread Function), whereas the fluorescence intensity distribution during two-photon observation is the square of the excitation light PSF. Proportional. Therefore, the side lobe intensity with respect to the peak intensity of the fluorescence intensity is lower in the two-photon observation than in the one-photon observation. Therefore, at the time of two-photon observation, even if the PSF has a strong side lobe, there is a merit that it is possible to prevent a decrease in signal noise ratio (SN ratio) and a decrease in XY resolution due to the side lobe. Therefore, it can be said that the two-photon observation is a preferable application example of the present invention.
When the above-described optical device is used as illumination light, the polarization state of the beam collapses and changes as the beam passes through an optical element such as an interference filter or a lens. Therefore, it is preferable to dispose the polarization control element immediately before joining with the imaging system, which is hardly affected by the optical element. According to this configuration, a beam close to an ideal state can be obtained.
In addition, the above-described optical device can be used as illumination light and applied to light stimulation. In the microscope image analysis system using, for example, a laser scanning microscope or a CCD camera, light stimulation uses light of the same or different wavelength as that of imaging (often laser light) from the same or different optical path as that of imaging. A part of the target cell (specimen) is intentionally irradiated with light, and various biochemical phenomena occurring in the cell are followed, for example, following a site that has been discolored by intentionally fading due to light irradiation. It refers to general so-called light manipulation such as investigation or follow-up investigation of a discolored site using photoconversion of fluorescent protein by light irradiation. Unlike the imaging, this light operation does not require the light used for the operation to be returned through the objective lens. That is, the
In addition, the microscope according to the above-described embodiment is preferably used for fluorescence correlation spectroscopy. Fluorescence Correlation Spectroscopy (FCS) is used to examine the molecular motion of a fluorescent substance and passes through the spatial illumination area of the constricted light produced by confocal microscope irradiation. The technology is based on photon counting of fluorescence emitted by molecules labeled with fluorescent dyes or expressed with fluorescent proteins. In order to improve the accuracy in fluorescence correlation spectroscopy, it is necessary to make the irradiation region smaller. On the other hand, when the present invention is employed, the use of the
