Description
本発明は真菌感染の分野に属する。具体的には、本発明は、真菌感染および/または真菌敗血症、ならびに真菌起源の炎症性疾患を予防および/または治療するための可溶性CD5細胞外ドメインを含む医薬組成物に関する。
先天性免疫系による病原体の認識は、宿主によって共有されていない、その生存に必須な保存された微生物構造、いわゆる病原体関連分子パターン(PAMP)を同定するために進化した、限られた数の固定された生殖系列(germline)にコードされたレセプターに依存する(1、2)。PAMPの例には、グラム陰性菌由来のリポ多糖(LPS)、グラム陽性菌由来のリポテイコ酸(LTA)およびペプチドグリカン(PGN)、ミコバクテリア(抗酸菌)由来のリポアラビノマンナン、および真菌由来のβ-グルカンおよびマンナンがある。種々の宿主防御経路を誘導する種々の構造的および機能的に多様なクラスのパターン認識レセプター(PRR)が存在する。パターン認識に関与するタンパク質ドメインには、特に、樹状細胞(DC)レクチン由来のC型レクチンドメイン、Toll様レセプター(TLR)由来のロイシンリッチリピート(LRR)、およびスカベンジャーレセプターシステインリッチ(SRCR)が含まれる(2)。後者は、マウスタイプIクラスAマクロファージスカベンジャーレセプター(SR-AI)のクローニングに関して最初に記載された(3)。種々の他のタンパク質、例えばウニスペラクトレセプター、ヒトおよびマウスCD5、および補体因子Iとの配列比較から、SRCR-SFと名付けたタンパク質レセプターの新規スーパーファミリーの保存された100アミノ酸長モチーフの存在がわかった。このファミリーは、現在、下等無脊椎動物から哺乳動物までのほとんどの動物門に典型的な30以上の種々の細胞表面および/または分泌タンパク質からなる(4)。SRCR-SFのメンバーは、2グループに分けられ、グループAのメンバーは6個のシステインからなるSRCRドメインを含み、2つのエクソンによりコードされるが、グループBのメンバーは8個のシステインを含み、1つのエクソンによりコードされる。しかしながら、最近の構造データは、両グループAおよびBのSRCRドメインが同様の骨格(scaffold)(2つの逆平行βシートおよび1つのαらせんにより形成される中心コア)を共有し、主な違いは連結ループにみられる(5)。この状況は、進化が落ち着いた、多種多様な異なるタンパク質(例えば免疫グロブリンドメイン)を構築する免疫系の他の2、3の有効なタンパク質モジュールのそれを思い起こさせる。これらの保存されたドメインの多様性は、ドメイン構造を安定化させる重要な残基が進化を通して保存されるが、他は自由に進化し(特に外部ループのもの)大きな機能的多様性を生じる点にある(6)。したがって、高度の構造的および系統発生学的保存性にもかかわらず、SRCRドメインに対する統一的機能は報告されていない。それらのあるものはタンパク質-タンパク質相互作用に関与し、その最も良く研究された例は、CD6リンパ球レセプターとIgスーパーファミリーに属する貫膜接着分子であるCD166/ALCAMの相互作用(7、8)、およびCD163/M130マクロファージレセプターとヘモグロビン-ハプトグロビン複合体との相互作用(9)である。両グループA (すなわち、SR-AI/II、MARCO、およびSCARA5)およびB(すなわち、DMBT1、Spα、およびCD6)SRCR-SFの2、3のメンバーは、細菌表面、例えばLPS、LTA、およびPGN上に存在するPAMPと相互作用することも知られている。これら相互作用は、最初はSRCRドメインの外側にマップされていたが、最近の証拠は、それにSRCRドメインが直接関与することを示す(11-14)。したがって、病原体のスカベンジングがSRCR-SFのすべてのメンバーが共有するか、またはそのメンバーの選択したグループにのみ共有される一般的特性であるか否かはまだ分析されていない。
貫膜タイプIレセプターのCD5およびCD6は、SRCR-SFの2つのリンパ系グループBメンバーである。どちらも構造および機能的レベルで重要な類似性を共有し、共通の先祖遺伝子の複製に由来すると考えられる同じ染色体領域中の隣接遺伝子によりコードされる(15、16)。CD5およびCD6は、胸腺細胞にその発生の初期段階から発現し、成熟末梢T細胞、ならびにある種の自己免疫疾患およびB細胞慢性リンパ球性白血病において増大する多反応性天然抗体の産生に関与する成熟B細胞の小サブセットであるB1a細胞に発現する(17)。CD5およびCD6の細胞外領域は、もっぱら3連続グループB SRCRドメインのみからなり、広範囲に及ぶアミノ酸配列同一性を示す(5)。CD5とCD6の主な違いは、大きな細胞質領域にみられ、いずれも固有の触媒活性を欠くが、情報伝達における機能に適合する種々の構造的モチーフを含む(18、19)。その点で、CD5およびCD6は、T(TCR)およびB(BCR)細胞上に存在する抗原特異的複合体と物理的に結合し(20、21)、免疫学的シナプスの中心でそれと同時に局在する(21、22)。したがって、CD5およびCD6は、まだよくわかっていない抗原特異的レセプター(22〜26)および複合体情報伝達経路(23、27〜29)により生じる活性化および分化シグナルを正または負に調節するように良く位置している。これは、種々の細胞表面カウンターレセプターによるCD5およびCD6細胞外ドメインの関与を通して達成されよう。一方、CD6がCD166/ALCAMと結合することは十分証明されているが(30)、真正なCD5リガンドは依然検討中である(31-35)。興味深いことにCD5およびCD6は、CD166/ALCAMと結合するのに重要な残基が異なるようである(36)。
以前の研究では、血清中で循環する可溶性型として存在することも知られている(37、38)CD5およびCD6細胞外ドメインの細菌結合能が検討された。報告されたデータは、CD5ではなくCD6の可溶性型および膜型はいずれも特異的PAMP(すなわち、それぞれLPSおよびLTA)の認識を通してグラム陰性およびグラム陽性細菌の表面に結合することを示した(39)。
他の研究は、表面にCD5レセプターを発現するそれらの細胞、T細胞またはB細胞が多かれ少なかれC. neoformansおよびC. albicansの正常な発生を認識し、影響を及ぼす能力があることを示した(48、49)。
しかしながら、このレセプターが真菌細胞を認識し、または真菌細胞との親和性を有するメカニズムは記載も示唆もされていない。
ところで、本発明者らは、これらの研究を真菌構造に対するCD5およびCD6の認識および結合特性の分析に広げ、CD6に比べてCD5細胞外ドメインは真菌細胞表面上の保存された成分の認識によく適していることを示し、これはCD5レセプターから単離した該細胞外領域がC. neoformansおよびC. albicansなどの一般的真菌感染に対してそれ自身がin vivoでの予防をもたらすことができることを最初に示したものである。
本発明者らは、真菌細胞が可溶性型のCD5細胞外ドメインにより特異的に認識され、結合し、凝集することを示した。これは、可溶性CD5細胞外ドメインによる真菌細胞壁の保存された構造成分であるβ-グルカンの認識を通してなされる。
さらに、本発明者らは、驚くべきことに可溶性CD5細胞外ドメインがザイモサン誘発敗血性ショック様症候群のマウスモデルにおいて防御効果を有することをみいだした。
これらの結果は、真菌起源の敗血性ショック症候群または他の炎症過程を治療するための可溶性ヒトCD5細胞外ドメインの注入の治療的有用性を裏付ける。
(発明の目的)
本発明の目的は、CD5リンパ球レセプターの可溶性細胞外ドメインおよび少なくとも1の医薬的賦形剤を含む医薬組成物である。

本発明の別の目的は、真菌細胞および/またはβ-グルカンリッチ真菌細胞壁成分を凝集するための可溶性CD5細胞外ドメインの使用である。
最後に、本発明の目的は、真菌感染および/または真菌敗血症および/またはそれと関連するあらゆる炎症性疾患を予防および/または治療するための医薬を製造するための可溶性CD5細胞外ドメインの使用である。

本発明者らは、真菌細胞表面、すなわちβ-グルカン上の保存された成分の認識を介して真菌細胞と結合し、凝集することを示した。
したがって、第1の局面において、本発明は、可溶性形の全CD5細胞外ドメインまたはその部分と少なくとも1の医薬的賦形剤、例えばグリセロール、サッカロースなどを含む医薬組成物に関する。
細胞外ドメインは、細胞外スペース(細胞の外側のスペース)に広がる膜タンパク質の部分である。CD5の細胞外領域は、3種の連続グループB SRCRドメイン(CD5.D1、CD5.DII、およびCD5.DIII)からなる。CD5の3種全ての個々のSRCR細胞外ドメインは真菌細胞表面と相互作用する能力を保持した。したがって、本発明の文脈において、「可溶性CD5細胞外ドメイン」は、CD5.DI、CD5.DII、およびCD5.DIIIドメインまたはそのいずれかを含む組み合わせであると考えられる。
本発明の医薬組成物は、好ましくは静脈内または腹腔内に全身的に注入または注射することにより投与することができよう。好ましい態様において、可溶性CD5細胞外ドメインの医薬組成物は注射可能形である。
可溶性CD5細胞外ドメインの注入は、真菌感染および/または真菌起源の敗血症を予防および/または治療するのに有益である。さらに、可溶性CD5細胞外ドメインは、活発な臨床感染(または敗血症)が起きないにもかかわらず真菌成分によって引き起こされる炎症過程の予防および/または治療にも有用である。真菌壁成分は、臨床感染が継続するかしないかに関わらず、炎症反応を引き起こし、その場合には、可溶性CD5細胞外ドメインも炎症作用を予防するように働くので有効でありうる。
したがって、本発明の別の局面は、真菌感染および/または敗血症および/またはそれと関連するあらゆる炎症性疾患、例えばSIRSまたは全身性炎症反応症候群、無菌性漿膜炎などを予防および/または治療するための可溶性CD5細胞外ドメイン医薬組成物に関する。
該感染および/または敗血症および/または該炎症性疾患は腐生および非腐生真菌種により引き起こされる。具体的態様において、該真菌種はCandida albicansまたはCriptococcus neoformansである。
本発明の別の局面は、β-グルカンを結合および/または認識するための可溶性CD5細胞外ドメインの使用に関する。
該可溶性CD5細胞外ドメインの結合は、用量依存性で飽和可能であり、Ca2+により著しく促進される。
別の局面は、種々の真菌種(腐生性もしくは病原性)の真菌細胞および/またはβ-グルカンリッチ真菌細胞壁成分を凝集するための可溶性CD5細胞外ドメインの使用に関する。
CD5の可溶性細胞外ドメインが真菌細胞と結合するだけでなく凝集するという事実は、凝集は先天性免疫系のコンポーネントが用いる一般的戦略であるから、病原体の伝搬しにくさと食細胞による病原体のクリアランスの促進に関連する。
本発明の別の局面は、真菌感染および/または敗血症および/または真菌成分によって引き起こされるあらゆる炎症性疾患を予防および/または治療するための医薬を製造するための可溶性CD5細胞外ドメインの使用に関する。
該感染および/または敗血症および/または該炎症性疾患は、腐生および非腐生真菌細胞種によって引き起こされる。具体的態様において、これら真菌種は、Candida albicansまたはCriptococcus neoformansである。
組換え可溶性ヒトCD5 DIII細胞外ドメイン(rshCD5.DIII)の製造は以前に記載されている(5)。
現在、本発明者らは、組換え可溶性ヒトCD5 DIおよびDII細胞外ドメイン(rshCD5.DIおよびrshCD5.DII)のための発現構築物を製造するための新規プライマーを開発した。rshCD5 DI細胞外ドメインを増幅するためのオリゴヌクレオチドは配列番号1および配列番号2であり、rshCD5 DIIを増幅するためのオリゴヌクレオチドは配列番号3および配列番号4である。
したがって、本発明の別の局面は、以下を含む組換え可溶性ヒトCD5 DI細胞外ドメイン(rshCD5.DI)を含む方法に関する:
a)配列番号1および配列番号2の配列のプライマーを用いてDI細胞外ドメインをPCR増幅し、
b)増幅断片を発現ベクターにクローンし、
c)可溶性ヒト組換えCD5 DI細胞外ドメインを発現および精製する。

限定ではなく例示のために以下の実施例に本発明を示す。
ヒトCD5のSRCR細胞外ドメインは全真菌細胞浮遊物と結合する。
ヒトCD5およびCD6リンパ球レセプターの細胞外ドメインの微生物結合特性に関する研究をさらに拡大するために(39)、本発明者らは真菌に対する直接タンパク質結合アッセイを行った。これについて、一定量(15μg)の、ヒトCD5 (rshCD5)またはヒトCD6 (rshCD6)の3つのSRCR細胞外ドメインを含むアフィニティ精製組換え可溶性タンパク質のビオチン標識調製物を4℃で一夜、108個のS. pombe、C. albicans、またはC. neoformans細胞とインキュベーションした。これらrshCD5およびrshCD6タンパク質は、正常ヒト血清中に存在する等価な循環型と区別できない(見掛けの分子量、抗体反応性、および細胞結合特性)ことが以前に示されている(33、39)。徹底的に洗浄した後、タンパク質結合物をLaemmli試料緩衝液で可溶化してSDS/PAGEゲルに流し、さらにホースラディッシュパーオキシダーゼ標識ストレプトアビジン(HRP-SAv)に対してウエスタンブロッティングし、さらに化学ルミネセンスにより発光させた。結果は、rshCD5は試験した腐生(S. pombe)および病原性(C. albicans、C. neoformans)真菌細胞種と結合するが、rsCD6は腐生細菌細胞種とのみ結合することを示した(図1A)。種々の量(1〜20μg)のビオチン標識rshCD5のC. albicansに対する結合は、図1Aに示すように分析した。5mM EDTA存在下の20μgのビオチン標識rshCD5の結合も示す。rshCD5の結合は、用量依存性で飽和可能であることを示し、結合の減少がEDTAの添加後にみられたのでCa2+により著しく促進された(図1B)。同じタンパク質調製物について108個のグラム陰性(E. coli)またはグラム陽性(S. aureus)細菌との結合を試験すると、以前に報告されたデータに一致してrshCD5について結合はほとんどまたは全くみられなかった(図1C)(39)。これは、CD6とは反対に、CD5の細胞外領域は、細菌細胞壁構造ではなく真菌細胞壁構造の認識に十分適していることを示す。

CD5の細胞外領域の3つのSRCRドメイン(CD5.DI、CD5.DII、およびCD5.DIII)のどれが真菌結合、さらに全真菌細胞結合アッセイに関与するかを確認するため、rshCD5の個々の可溶性SRCRドメインを発現するHEK 293-EBNAトランスフェクタント(形質移入体)から得られる細胞培養上清を4℃で一夜108個のC. albicansまたはC. neoformansとインキュベーションした。非結合タンパク質(NB)を洗浄して除去し、10%トリクロロ酢酸(TCA)で沈殿させた。TCAで沈殿する細胞結合(B)タンパク質をSDS-PAGEゲル中で電気泳動し、ウサギポリクローナル抗CD5抗血清+HRP標識ヒツジ抗ウサギ抗血清を用いてウエスタンブロットで分析し、さらに化学ルミネセンスにより発光させた。図1Dに示すように、CD5の3つすべての個々のSRCR細胞外ドメインが真菌細胞表面と相互作用する能力を保持した。これは、CD5の3つすべてのSRCR細胞外ドメインが共有する保存された構造モチーフが真菌の除去(scavenging)に関与していることを示す。
可溶性CD5細胞外ドメインによる真菌細胞凝集の誘導。
ヒトCD5細胞外ドメイ上の複数の結合部位の存在が真菌の凝集をもたらすか否かを検討するため、FITC標識C. albicans細胞浮遊液を5または10μgの可溶性無標識タンパク質(BSA、rshCD5、およびrshCD6)と4℃で一夜インキュベーションし、次いで、落射蛍光顕微鏡で分析した。この条件下で、rshCD5は用量依存性に真菌細胞凝集をもたらしたが、rsCD6とBSAはいずれもそのような減少をもたらすことができなかった(図2、上段および中段パネル)。同じ結果がC. neoformans 真菌細胞をアッセイしたときにも得られた。興味深いことに、rshCD5誘導真菌細胞凝集は、アッセイを過剰量(20μg)のザイモサン(S. cerevisiae由来)またはβ-グルカン(オオムギ由来)存在下で行うと顕著に減少したがマンナン(S. cerevisiae由来)では減少はみられなかった(図2、下段パネル)。これは、rshCD5による真菌細胞の結合および凝集が特異的であり、真菌細胞壁、例えばβ-グルカンの特定の成分の認識によって仲介されるらしいことを示した。
ヒトCD5細胞外ドメイ上の複数の結合部位の存在が真菌の凝集をもたらすか否かを検討するため、FITC標識C. albicans細胞浮遊液を5または10μgの可溶性無標識タンパク質(BSA、rshCD5、およびrshCD6)と4℃で一夜インキュベーションし、次いで、落射蛍光顕微鏡で分析した。この条件下で、rshCD5は用量依存性に真菌細胞凝集をもたらしたが、rsCD6とBSAはいずれもそのような減少をもたらすことができなかった(図2、上段および中段パネル)。同じ結果がC. neoformans 真菌細胞をアッセイしたときにも得られた。興味深いことに、rshCD5誘導真菌細胞凝集は、アッセイを過剰量(20μg)のザイモサン(S. cerevisiae由来)またはβ-グルカン(オオムギ由来)存在下で行うと顕著に減少したがマンナン(S. cerevisiae由来)では減少はみられなかった(図2、下段パネル)。これは、rshCD5による真菌細胞の結合および凝集が特異的であり、真菌細胞壁、例えばβ-グルカンの特定の成分の認識によって仲介されるらしいことを示した。
可溶性CD5細胞外ドメインの細菌細胞壁ではなく真菌細胞壁の保存成分に対する直接結合。
可溶性ヒトCD5細胞外ドメインの真菌結合能をさらに確認し特徴づけるため、その精製真菌細胞壁調製物に対する直接結合を評価した。これについて、BSA、ザイモサン、LPS、PGN、またはLTAでコートした96ウェルELISAプレートを増加する量(0.01〜2μg)のビオチン標識rshCD5とインキュベーションした。結合タンパク質をHRP-SAvを加えて検出し、さらに3,3’,5,5’−テトラメチルベンジジン液体基質で発光させた。吸光度を450nmで測定した。図1に示す真菌および細菌細胞結合実験によれば、ビオチン標識rshCD5は、ザイモサンでコートした(コート)プレート(S. cerevisiae)と用量依存性に結合したが(図3A、上段)、LPS、LTA、またはPGNでコートしたプレートは結合しなかった(図3A、下段)。並行アッセイにおいて、ビオチン標識rshCD6は、予測通り(39)、LPS、LTA、またはPGNでコートしたプレートと用量依存性に結合した(図3B、下段)。同様な用量依存性がrshCD6についてザイモサンコートプレートに対してみられたが、吸光度値は常にrshCD5について得られるものより低かった(図3Bおよび3A、上段)。これは、細菌細胞壁成分ではなく真菌細胞壁成分の除去におけるヒトCD6細胞外ドメインと比較したヒトCD5細胞外ドメインの適合性に関する上記の記載を強める。

さらに競合ELISAアッセイを行って、ヒトCD5細胞外ドメインとの相互作用に関与する真菌細胞壁成分の特異性を検討した。これについて、一定量(2μg)のビオチン標識rshCD5およびrshCD6を、増加する量(0.01〜20μg)の無標識競合物(β-D-グルカン、ザイモサン、マンナン、またはBSA)の存在下または非存在下でザイモサンコートELISAプレートとインキュベーションした。結合タンパク質をHRP-SAvを加えて検出し、さらに3,3’,5,5’−テトラメチルベンジジン液体基質で発光させた。吸光度を450nmで測定した。図2左に示す真菌凝集成績によれば、マンナンではなくβ-D-グルカンおよびザイモサンは、ビオチン標識rshCD5のザイモサンに対する結合に用量依存性に競合することができた(図4A、左)。反対に、同じアッセイをビオチン標識rshCD6を用いて行うと、ザイモサンのみが結合に競合することができた(図4A、右)。
次に、ヒトCD5細胞外ドメインの真菌細胞壁構造物に対する結合と競合する、種々のβ-グルカン含有調製物の能力を分析した。これについて、一定量のビオチン標識rshCD5(15 μg)の全真菌細胞に対する結合は、増加する濃度のオオムギから精製したβ-グルカン、Euglena gracilisから精製したβ-1,3-グルカン、およびS. cerevisiae由来グルカン、ならびにそれぞれ陽性および陰性コントロールとして用いたザイモサンまたはマンナン(共にS. cerevisiae由来)と競合した。徹底的に洗浄した後、結合タンパク質を可溶化し、SDS-PAGEにかけた。ビオチン標識rshCD5の検出は、HRP-SAvを用いるウエスタンブロットにより行い、さらに化学ルミネセンスにより発光させた。図4Bに示すように、用いたすべてのグルカンは、ビオチン標識rshCD5のC .albicansおよびC. neoformans両方の細胞浮遊液に対する結合と用量依存性に競合した。これらの結果は、ヒトCD5細胞外ドメインと真菌との相互作用が真菌細胞壁の高度に保存された豊富な成分であるβ-1,3-グルカンの認識によって仲介されるようであることを示した。
ザイモサンは膜結合CD5と結合し、MAPKカスケードのCD5介在性活性化を誘導する。
次に、本発明者らは、膜結合形のヒトCD5レセプターも真菌細胞壁成分と相互作用することができるか否かを取り上げた。これについて、増加する量のFITC標識ザイモサンの非トランスフェクト(形質移入)2G5細胞またはCD5トランスフェクト2G5細胞(CD5およびCD6の両レセプターの発現欠損について選択したJurkat細胞誘導体)に対する結合が観察された(40)。増加する量(1〜30μg)のFITC標識ザイモサンを、非トランスフェクト2G5 Jurkat細胞、または野生型膜CD5レセプター(2G5-CD5.WT)を発現するトランスフェクト2G5 Jurkat細胞とインキュベーションした。染色細胞の蛍光強度はフローサイトメトリーで分析した。図5Aに示すように、野生型膜結合型のCD5(2G5-CD5.WT)を安定に発現する2G5細胞の蛍光強度は、非トランスフェクト親2G5細胞に比べて高かった。結果のさらなる確認は、一定量のFITC標識ザイモサン(15μg)が増加する量の(1〜30μg)の非標識ザイモサン(S. cerevisiae)、β-D-グルカン(オオムギ)およびマンナン(S. cerevisiae)と競合する2G5-CD5.WT安定トランスフェクタントを用いる競合結合実験から得られた。染色細胞の蛍光強度はフローサイトメトリーで分析した。図5Bに示すように、マンナンではなくβ-グルカンまたはザイモサンは、FITC標識ザイモサンの結合と用量依存性に競合することができた。これらの結果は、rshCD5について得られた結果を確認するものであり、CD5発現細胞が保存された真菌細胞壁成分の存在を感知するであろうことを示す。
次に、本発明者らは、膜結合形のヒトCD5レセプターも真菌細胞壁成分と相互作用することができるか否かを取り上げた。これについて、増加する量のFITC標識ザイモサンの非トランスフェクト(形質移入)2G5細胞またはCD5トランスフェクト2G5細胞(CD5およびCD6の両レセプターの発現欠損について選択したJurkat細胞誘導体)に対する結合が観察された(40)。増加する量(1〜30μg)のFITC標識ザイモサンを、非トランスフェクト2G5 Jurkat細胞、または野生型膜CD5レセプター(2G5-CD5.WT)を発現するトランスフェクト2G5 Jurkat細胞とインキュベーションした。染色細胞の蛍光強度はフローサイトメトリーで分析した。図5Aに示すように、野生型膜結合型のCD5(2G5-CD5.WT)を安定に発現する2G5細胞の蛍光強度は、非トランスフェクト親2G5細胞に比べて高かった。結果のさらなる確認は、一定量のFITC標識ザイモサン(15μg)が増加する量の(1〜30μg)の非標識ザイモサン(S. cerevisiae)、β-D-グルカン(オオムギ)およびマンナン(S. cerevisiae)と競合する2G5-CD5.WT安定トランスフェクタントを用いる競合結合実験から得られた。染色細胞の蛍光強度はフローサイトメトリーで分析した。図5Bに示すように、マンナンではなくβ-グルカンまたはザイモサンは、FITC標識ザイモサンの結合と用量依存性に競合することができた。これらの結果は、rshCD5について得られた結果を確認するものであり、CD5発現細胞が保存された真菌細胞壁成分の存在を感知するであろうことを示す。
ザイモサンの膜結合型CD5に対する結合に関するさらなる証拠は、野生型(2G5-CD5.WT)または細胞質側末端切断(2G5-CD5.K384stop)型のCD5を発現する安定な2G5トランスフェクタントにおけるMAPK情報伝達カスケードのメンバーの活性化から得られた(41)。これについて、2x106個の非トランスフェクトまたはトランスフェクト2G5細胞を37℃で40μg/mlのザイモサンにて種々の時間(0〜30分間)パルスした。次に、細胞溶解物試料をSDS-PAGE中で電気泳動し、ポリクローナルウサギ抗リン酸化ERK1/2(pERK1/2)、モノクローナルマウス抗リン酸化MEK(pMEK)、およびポリクローナルウサギ抗cdk4抗血清(後者はローディングコントロールとして用いる)を用いるウエスタンブロットにより分析した。徹底的に洗浄した後、膜を、それぞれHRP標識ヒツジ抗ウサギまたは抗マウスIg抗血清を用いる化学ルミネセンスにより発光させた。図6Aに示すように、ザイモサンに対する暴露は、非トランスフェクト親2G5細胞ではなく2G5-CD5.WT細胞中でMEKおよびERK1/2両方の時間依存性リン酸化をもたらした。興味深いことに、MEKおよびERK1/2両方のザイモサン誘発リン酸化は、ほとんどC末端88アミノ酸を欠く細胞質側末端切断CD5型を発現する2G5-CD5.K384stopトランスフェクタント(図6A、右)には観察されなかった(41)。これは、2G5細胞中のザイモサンによるMAPKカスケードの活性化がCD5の発現ならびにその細胞質ドメインの完全性に依存することを示す。
ザイモサンはCD5介在性サイトカイン放出を引き起こす。
真菌細胞壁成分の膜結合CD5との結合の生物学的結果をさらに検討するため、本発明者らはその後のサイトカイン放出現象を分析することにした。残念ながら、親2G5細胞および安定なトランスフェクト2G5細胞のいずれを刺激しても、種々の時点で顕著なサイトカイン放出は生じなかった。この不応答性は、高濃度のザイモサンへの暴露後、ならびに抗CD3および抗CD28mAbの組み合わせなどの強力なT細胞特異刺激への暴露後にみられ、2G5細胞のサイトカイン放出の遮断が存在するらしいことを示唆した。

これらの観察結果から、膜結合型のCD5は非リンパ系哺乳動物細胞系であるヒト胚腎(HEK)293細胞に発現した。親HEK 293細胞、ならびにザイモサンのよく知られたレセプターであるTLR2を安定に発現するHEK293細胞トランスフェクタントを、野生型(CD5.WT)および細胞質側末端切断(CD5.K384stop)のヒトCD5を発現させるために一時的にトランスフェクトした。次に、細胞を24時間ザイモサン(20μg/ml)に暴露し、細胞培養上清(100μl)中のIL-8濃度をELISAで測定した。図6Bに示すように、CD5.WT発現HEK 293細胞では非トランスフェクト細胞またはCD5.K384stop切断分子を発現する細胞に比べて有意なIL-8放出がみられた。興味深いことに、CD5.WT発現HEK 293細胞で検出されたIL-8レベルは、陽性コントロールとして用いたTLR2発現トランスフェクタントでみられたものと同様であった。さらに、CD5.WTおよびTLR2の同時発現は、ザイモサンに暴露後に相加効果や相乗効果をもたらさなかった。まとめると、これらの結果は、膜結合型のCD5は真菌細胞壁成分の存在を感知し、これがサイトカイン放出を生じる独立した情報伝達カスケードを開始することを示唆する。
可溶性CD5細胞外ドメインの注入は、マウスのザイモサン誘発敗血症性ショック様症候群から保護する。
CD5細胞外ドメインの真菌細胞壁成分との結合に対するさらなるin vivoでの検証は、ザイモサンにより誘発される敗血症性ショック様症候群のマウスモデルから得られた(42)。rshCD5が単高用量のザイモサン(500mg/kg)をi.p.投与後に誘発される全身性炎症および多臓器不全を予防できるか否かを評価した。これらの条件下で、ザイモサンは、18時間以内に急性腹膜炎および臓器損傷と、12日間にわたりマウスの死亡率の増加をもたらした。このために、CD1マウスを以下の群に割り当てた:BSA、BSA(25μg;i.p.)のみを注入されたマウス;BSA+ZYM、ザイモサン(500mg/kg;i.p.)注入前にBSA(25μg;i.p.)で前処置したマウス;rshCD5+ZYM、ザイモサン(500mg/kg;i.p.)注入前にrshCD5(25μg;i.p.)で前処置したマウス。図7A-Eに示すように、ザイモサン攻撃の1時間前にマウスに25μgの単回i.p.用量のrshCD5を投与すると、18時間における毒性スコア(図7A)、腹腔の総白血球数(図7B)、IL-6およびIL-1β血漿レベル(図7C)、およびミエロペルオキシダーゼ(MPO)活性により測定した肝臓の好中球浸潤(図7D)の有意な減少をもたらした。別の実験において、マウスの生存をモニターし、コントロールに比べて観察期間(12日間)の終了時の有意な増加(45%対15%)が、ザイモサン攻撃前にrshCD5の単回i.p.用量を投与された動物でみられた(図7E)。まとめると、これらの結果は、rshCD5でマウスを前処置するとザイモサンにより誘発される有害な全身性炎症を予防し、真菌性敗血症性ショックに対するrshCD5の治療的可能性が明らかであることを示す。

材料と方法
構築物。可溶性タンパク質rshCD5(43)、rshCD5.DIII(5)、およびrshCD6(22)に対する発現構築物の製造は他に記載している。rshCD5の細胞外ドメインDIおよびDIIは、以下のプライマーを用いてPCR増幅した:DIフォワード(配列番号1)およびリバース(配列番号2)。DIIフォワード(配列番号3)およびリバース(配列番号4)。該PCRをNheIおよびBamHIで制限し、適切に消化したpCEP-Puベクター中にクローンした。得られた構築物を以前に記載された様にHEK 293-EBNA細胞中にトランスフェクトした(44、45)。野生型(pHβ-CD5.WT)および細胞質側末端切断(pHβ-CD5.K384STOP)膜結合型のCD5をコードする発現構築物(41)をLipofectamine(登録商標)2000試薬(Invitrogen Life Technologies、Paisley、U.K.)を用い、使用説明書にしたがってHEK 293細胞中に一時的に導入した。
細胞。CD5-およびCD6-陰性2G5細胞をJurkat細胞のセルソーティング、さらにクローニングにより得(40)、次いで37℃、5%CO2で10% FCS(Invitrogen Life Technologies)、100U/mlペニシリンG(Laboratorios ERN、Barcelona、Spain)、および50μg/mlストレプトマイシン(Laboratorios Normon、Madrid、Spain)添加BioWhittaker RPMI 1640培地(Lonza、Verviers、Belgium)中でインキュベーションした。Epstein-Barrウイルスタンパク質EBNA-1を構成的に発現し、pCEP-Puベクターのエピソーム性複製を可能にするヒト胚腎HEK 293-EBNA細胞は、Dr. T. SasakiおよびDr. Timplの好意で分与された(Max Planck Institute for Biochemistry、Martinsried、Germany)。該細胞は、10% FCS、100U/mlペニシリンG、50μg/mlゲンタマイシン、および250μg/mlジェネテシン添加DMEM/F12(G418、Sigma、St Louis、MO)中で増殖させた。HEK 293細胞およびTLR2を安定に発現するHEK 293(HEK 293-TLR2)は、Dr. Golenbock(University of Massachusetts Medical School、Worcester、MA)の好意で分与された。プロマイシン(50μg/mL;Sigma)を、安定なHEK 293-EBNA/pCEP-PuおよびHEK 293-TLR2トランスフェクタントを選択するために培養液に加えた。
組換えタンパク質の発現、アフィニティ精製、およびビオチン標識。すべての組換え可溶性ヒトタンパク質rshCD5、rshCD6、rshCD5.DI、rshCD5.DII、およびrshCD5.DIIIを、エピソーマル発現系のpCEP-Pu/HEK 293-EBNAを用いて発現させた。rshCD5およびrshCD6タンパク質は、特異抗体を用いるアフィニティクロマトグラフィにより培養上清から精製した(22、43)。rshCD5の個々の細胞外ドメインは、成熟タンパク質由来のアミノ酸R1-L113(DI)、A135-F271(DII)、およびF271-D369(DIII)を含み、非分画無血清培養上清として用いた。タンパク質のビオチン化は、EZ-Link PEO-マレイミド活性化ビオチン(Pierce/Perbo Science、Cheshire、U.K.)を使用説明書にしたがって用いて行った。
細菌および真菌結合試験。この試験で用いた細菌株(S.aureusおよびE.coli)および真菌株(C. albicansおよびC. neoformans)は、Department of Microbiology of the Hospital Clinic of Barcelonaが標準的生化学的方法を用いて特徴づけた臨床分離株である。真菌株S. pombeは、Department of Cell Biology and Pathology of the University of Barcelonaの好意で分与された。細菌または真菌を、30〜37℃でLuria Bertoniブロス(LB)中で通気しながら一夜増殖させ、次いで3,500xgで10分間遠心して回収した。細菌または真菌ペレットを、TBS(20mM Tris-HCl、pH7.5、150mM NaCl)に最終密度細菌1010個/mlまたは真菌108個/mlとなるよう再浮遊させた。細菌/真菌希釈液を寒天に播いて定量した。組換え可溶性タンパク質(rshCD5、rshCD6)の結合を先に記載のごとく分析した(14)。
競合アッセイのために15μgのrshCD5を、種々の濃度の、S. cerevisiaeから精製したザイモサン(Sigma)、Euglena gracilis由来β-1-3-グルカン(Sigma)、S. cerevisiae由来グルカン(Sigma)、およびオオムギ由来β-D-グルカン(Sigma)、またはS. cerevisiae由来マンナン(Sigma)と4℃で1時間、前インキュベーションし、次いで細菌または真菌浮遊液とインキュベーションした。rshCD5細胞外ドメインDI、DII、およびDIIIを用いる真菌結合試験は、回転させながら4℃で一夜108個の真菌と1mlの各培養上清をインキュベーションして行った。非結合タンパク質は10%TCA沈殿により評価した。これら試料と結合タンパク質試料をLaemmli試料用緩衝液で可溶化し、SDS-PAGEで電気泳動し、自家製ウサギ抗CD5ポリクローナル抗血清+HRP標識ヒツジ抗ウサギIg抗血清(DAKO、Carpinteria、CA)を用いるウエスタンブロットにより分析した。
ELISAアッセイ。96ウェルマイクロタイタープレート(Nunc、Roskilde、Denmark)を、コーティング緩衝液(100mM NaHCO3、pH9.5)中の、20μgのLPS(E.coli O55:B5、Sigmaから精製)、リポテイコ酸(LTA;Sigma)、ペプチドグリカン(PGN;Sigma)、またはザイモサン(ZYM;Sigma)と4℃で一夜コートした。プレートを、3%BSA(Sigma)含有PBSで室温(RT)で1時間ブロックした。次に、種々の濃度のビオチン標識BSA、rshCD5、またはrshCD6をウェルに加え、室温で2時間インキュベーションした。結合タンパク質を、室温で1時間、1:2000希釈のHRP標識SAv(Roche Diagnostics GmbH、Mannheim、Germany)を加えて検出した。各インキュベーション工程の間に非結合タンパク質を、PBS 0,01% Tween-20で3回洗浄除去した。ELISAを3,3’,5,5’−テトラメチルベンジジン液体基質(TMB;Sigma)を加えて発光させ、吸光度を450nmで測定した。
競合ELISAアッセイは、ザイモサンコートプレートに加える前に、2μgのビオチン標識rshCD5またはrshCD6を種々の濃度のザイモサン、β-D-グルカン、またはマンナンと4℃で1時間プレインキュベーションする以外は上記のごとく行った。
IL-6およびIL-1βの血清レベルを決定するためのELISAは製造業者のプロトコール(R&D Systems、Minneapolis、MN)に従って行った。
真菌凝集アッセイ。種々の真菌株の蛍光標識は、100mM FITCで室温で1時間インキュベーションすることにより行った。細胞を3,000xgで5分間遠心して回収し、PBSで数回洗浄して非結合FITCを除去した後、真菌を300μl PBSに再浮遊させた。5または10μgのrshCD5またはrshCD6を加え、静かに軌道回転させながら4℃で一夜インキュベーションした。競合のために、10μgのrshCD5またはrshCD6を4℃で1時間、20μgのザイモサン、β-D-グルカン、またはマンナンとプレインキュベーションした。10μlの浮遊液をガラススライドに移し、蛍光顕微鏡(Leica Microsystems、Mannheim、Germany)で可視化した。画像をPhotoshop7.0(Adobe Systems、San Jose、CA)で分析した。
フローサイトメトリー分析。ザイモサンの2G5または2G5-CD5.WT細胞に対する結合は、2x105細胞をブロック用緩衝液(PBS、10%ヒトAB血清、2% FCS、および0.02%アジ化ナトリウム)中の種々の量のFITC標識S. cerevisiae由来ザイモサン(Sigma)とインキュベーションすることにより行った。4℃で1時間インキュベーションした後、細胞をPBS、2%FCS、および0.02%アジ化ナトリウムで洗浄し、次いでFACScan(Becton Dickinson、Mountain View、CA)で分析した。競合アッセイは、種々の量(1〜30μg)の非標識ザイモサン、β-D-グルカン、またはマンナン存在下、2G5-CD5.WT細胞を20μgのFITC標識ザイモサンとインキュベーションして行った。
IL-8サイトカイン放出アッセイ。HEK 293細胞およびTLR2を安定して発現するHEK 293をpHβ-CD5.WTまたはpHβ-CD5.H418STOPで、Lipofectamine(登録商標)2000試薬(Invitrogen)を使用説明書に従って用いてトランスフェクトした。トランスフェクション後24時間で培地を増殖培地(上記参照)に交換した。タンパク質の発現をSDS-PAGEおよびウサギポリクローナル抗CD5抗血清を用いるウエスタンブロットにより評価し、HRP標識ヒツジ抗ウサギ抗血清(DAKO)を用いる化学ルミネセンスにより発光させた。トランスフェクト細胞を20μg/mlのザイモサンで24時間パルスし、上清試料(100μl)を回収し、使用説明書に従ってELISA(BD OptEIA(登録商標)、ヒトIL-8 ELISAセット、BD Biosciencies、San Diego、CA)によりIL-8をアッセイした。
MAPキナーゼアッセイ。刺激のために、2x107 2G5、2G5-CD5.WT、または2G5-CD5.H418STOP細胞をFCS不含RPMI1640培地中で24時間飢餓させた。次に、細胞を37℃で10分間、300μlのRPMI1640培地に浮遊させ、次いで37℃で0、5、15、および30分間、40μg/mlのザイモサンで刺激した。細胞を、溶解用緩衝液(0.5μg/mlアプロチニン、10μg/mlロイペプチン、および1mM PMSF含有50mM Tris-HCl、pH7.6、50mM NaCl、1mM EDTA、および0.1%Triton X-100)中で崩壊させた。細胞抽出物中のタンパク質含有量をBradfordの方法(Bio-Rad Laboratories、Inc. Hercules、CA)により測定し、20〜30μgのタンパク質試料を、SDS-PAGEで分析し、ニトロセルロース膜(Millipore、Bedfore、MA)上に移した。シートを室温で1時間、5%脱脂乳粉末含有TBS-T(20mM Tris-HCl、pH7.5、150mM NaCl、0.05% Tween-20)とインキュベーションし、次いで振盪させながら、4℃で一夜、ウサギポリクローナル抗pERK1/2(sc-101760、1:1000希釈;Santa Cruz Biotechnology、Santa Cruz、CA)、マウスモノクローナル抗pMEK(sc-81503、1:1000希釈、Santa Cruz)、またはウサギポリクローナル抗cdk4(sc-260、1:200希釈;Santa Cruz)抗体でプローブした。TBS-Tで3回洗浄後、膜を室温で45分間、対応するHRP標識ヒツジポリクローナル抗マウスまたは抗ウサギIg抗血清(1:2000希釈;DAKO)とインキュベーションした。それをTBS-Tで3回、TBSで1回洗浄し、Super Signal West Dura Extended Duration基質(Pierce)を用いて化学ルミネセンスを増加させ、次いで、X-OMATフィルム(Kodak、Rochester、NY)に暴露して可視化した。
ザイモサン誘発敗血症性ショック用症候群。体重20〜22gの雄CD1マウス(Charles River、Milan、Italy)に250μlの無菌生理食塩水溶液中のザイモサン(500mg/kg)をi.p.注射した(46)。単回i.p.用量25μgのrshCD5またはBSAをザイモサン攻撃の1時間前に投与した。第3群のマウスには25μgのBSAを前投与した後、同容量の無菌生理食塩水溶液を投与した。ザイモサン攻撃後18時間に全身毒性の臨床的重症度を0〜3の範囲の主観的尺度でスコア付けした(ここで、0=なし、1=軽度、2=中等度、3=重度)。マウスで観察される各毒性兆候(昏睡、下痢、逆立った毛皮(ruffled fur)、および結膜炎)についてレンジングスケール(ranging scale)を用いた。各群の各毒性兆候値を加えて最終スコアを得た。
腹膜中の総白血球数を評価するため、5mlのPBSを白線を切開して腹腔に注射し、腹腔を10秒間マッサージした後に同量を回収した。自動細胞カウンター(Micros 60、ABX Diagnostics、Montpellier、France)を用いて測定した。肝臓試料を先に記載のごとく(47)ミエロペロキシダーゼ活性(MPO)の評価に用いるまで液体窒素中で凍結した。
各グループの死亡率を12日間にわたりモニターし、生存マウスのパーセンテージで表した。実験方法は、University of Barcelonaの倫理委員会により承認され、スペイン(RD 1201/2005)および欧州(86/609)の規制、およびNational Institutes of Health’s Guide for the Care and Use of Laboratory Animals(国立衛生研究所の実験動物の管理と使用に関する指針)に適合する実験動物管理ガイドラインに従って行った。
統計分析。結果は平均値±SEMで表す。対応のないt検定を統計的有意差を決定するために用いた。生存はlogrank検定により分析した。p値<0.05を統計的に有意とみなした。

- 可溶性CD5細胞外ドメインおよび少なくとも1の医薬的賦形剤を含む医薬組成物。
- 注射可能形の請求項1記載の組成物。
- 真菌感染および/または真菌敗血症および/または真菌成分によって引き起こされるあらゆる炎症性疾患を予防および/または治療するための請求項1または2に記載の組成物。
- 該感染および/または敗血症および/または該炎症性疾患がCandida albicansまたはCriptococcus neoformansによって引き起こされる請求項1記載の組成物。
- β-グルカンを結合および/または認識するための可溶性CD5細胞外ドメインの使用。
- 真菌細胞および/またはβ-グルカンリッチ真菌細胞壁成分を凝集させるための可溶性CD5細胞外ドメインの使用。
- 真菌感染および/または真菌敗血症および/または真菌成分によって引き起こされるあらゆる炎症性疾患を予防および/または治療するための医薬を製造するための可溶性CD5細胞外ドメインの使用。
- 該感染および/または敗血症および/または該炎症性疾患がCandida albicansまたはCriptococcus neoformansによって引き起こされる請求項7記載の使用。
- 以下を含む組換え可溶性ヒトCD5 DI細胞外ドメインを得る方法:
a) 配列番号1および配列番号2の配列のプライマーを用いてDI細胞外ドメインをPCR増幅、
b) 増幅した断片を発現ベクター内にクローニング、および
c) 可溶性ヒト組み換えCD5 DI細胞外ドメインを発現および精製。 - 発現ベクターがpCEP-Puである請求項9記載の方法。
- 可溶性ヒト組換えCD5 DI細胞外ドメインがHEK 293-EBNA細胞中で発現する請求項10記載の方法。
- 以下を含む組換え可溶性CD5 DII細胞外ドメインを得る方法:
a) 配列番号3および配列番号4の配列のプライマーを用いてDII細胞外ドメインをPCR増幅、
b) 増幅した断片を発現ベクター内にクローニング、および
