Description
本発明は、分子生物学の分野に関する。より詳細には、本発明は、DNA配列および対応するタンパク質に関する。

(a)ストリンジェントな条件下で、配列番号6またはそのフラグメントにハイブリダイズし得、そしてネイティブヒトDP−75mRNAにハイブリダイズし得る核酸配列を含有する、アンチセンスポリヌクレオチドを提供する、工程;および、
(b)上記アンチセンスポリヌクレオチドを、上記DP−75に接触させる、工程。
・項目41.以下の(a)、(b)、および(c)の工程を包含する、サンプル中の過剰増殖細胞を検出するための方法:
(a)ストリンジェントな条件下で、配列番号6またはそのフラグメントにハイブリダイズし得る配列を含有する、プローブポリヌクレオチドを提供する、工程;
(b)ポリヌクレオチドハイブリッドの形成を可能にする条件下で、上記プローブと、上記サンプル細胞のポリヌクレオチドとを接触させる、工程;および、
(c)上記ハイブリッドを検出する、工程。
・項目42.上記プローブが、10塩基と50塩基との間の長さである、項目41に記載の方法。
・項目43.以下の(a)、(b)、および(c)の工程を包含する、サンプル中のDP−75ポリペプチドを検出するための方法:
(a)DP−75ポリペプチドに特異的に結合する抗体を提供する工程であって、上記DP−75ポリペプチドが、配列番号6またはそのフラグメントにハイブリダイズし得る配列を含有するポリヌクレオチドによってコードされる核酸配列を含有する、工程;
(b)抗体/抗原複合体の形成を可能にする条件下で、上記抗体と上記サンプルとを接触させる、工程;および、
(c)上記複合体を検出する、工程。
・項目44.以下の(a)および(b)の工程を包含する、過剰増殖細胞の増殖を阻害するための方法:
(a)ネイティブDP−75mRNAの発現を阻害するために、被験体にリボザイムベクターまたはアンチセンスベクターを投与する、工程;および、
(b)上記リボザイムベクターまたはアンチセンスベクターと、上記過剰増殖細胞のポリヌクレオチドとを接触させる、工程。
・項目45.以下の(a)および(b)の工程を包含する、細胞の複製を阻害する方法:
(a)DP−75ポリペプチドに特異的に結合する抗体を提供する工程であって、上記DP−75ポリペプチドが、配列番号6またはそのフラグメントにハイブリダイズし得る配列を含有するポリヌクレオチドによってコードされるアミノ酸配列を含有する、工程;および、
(b)抗体/抗原複合体の形成を可能にする条件下で、上記抗体と上記サンプルとを接触させる、工程。

「相同性」は、xおよびy間の類似性の程度をいう。1つの型とその他の型との配列間の対応は、当該分野で公知の技術によって決定され得る。例えば、それらは、ポリヌクレオチドの配列情報の直接比較によって決定され得る。代表的には、ポリヌクレオチドまたはポリペプチドのいずれかの2つの配列は、その配列が、少なくとも45%の配列同一性、より代表的には50%配列同一性、より代表的には55%配列同一性、より代表的には60%配列同一性、さらに代表的には65%配列同一性、よりさらに代表的には70%配列同一を示すときに、相同である。一般的には、2つの配列は、その配列が、少なくとも75%配列同一性、より一般的には80%配列同一性、よりさらに一般的には85%配列同一性、よりさらに一般的的には90%配列同一性、そしてよりさらに一般的には95%配列同一を示すときに、相同である。

DP−75に対する部分核酸配列が、一本鎖分子として、配列番号1として下記に記載されている。
ドットブロットアッセイにおいて、DP−75を、ガンおよび正常の両組織からのRNAとハイブリダイズさせた。ガン組織の供給源は、腎臓、甲状腺、***、結腸、尿管、肺、鼻、胃、食道、肝臓、リンパ腫、子宮、膀胱、直腸、および脳を包含する。
配列番号6を、Stratagene,La Jolla,California,USA.から購入した、ファージベクターを使用する前頭皮質ライブラリーから単離した。そのライブラリーを、約1×106cpm/mlの最終放射性カウントで、ランダムプライム標識によって生成した配列番号1でプローブした。プローブを、Amersham,ArlingtonHeights,Illinois,USAから購入したRediPrime(登録商標)DNA標識キットを用い、製造者の指示に従って標識した。
