Abstract
Description
The present invention relates to a fatigue improving composition for menopause.
So-called climacteric disorder is a so-called indefinite complaint caused by autonomic and psychogenic manifestations in menopause. It causes unpleasant symptoms such as and affects daily life. Such indefinite complaints are thought to be due to decreased secretion of female hormones (estrogens). In addition, even among younger women, hormonal balance is disrupted due to excessive stress and diet accompanying social advancement, and juvenile climacteric disorders exhibiting symptoms similar to climacteric disorders are increasing. Furthermore, in men, estrogen receptors exist in the brain, cardiovascular, skeleton, skin, and other tissues, and estrogen converted from male hormone (testosterone) acts by the action of the metabolic enzyme aromatase. Thus, menopause can also occur in men.
Treatment methods for climacteric disorders include administration of hormonal agents and tranquilizers, but these treatments all have side effects, albeit with minor differences. In addition, although psychotherapy and exercise therapy are considered to be effective, there are currently no effective treatment methods to be mentioned.
As one of the methods to relieve symptoms in menopause, it is known to take a substance that has similar action to estrogen, a female hormone, that lowers the secretion of female hormones. The isoflavones contained in Isoflavone is said to be one of the plant estrogens, and its chemical structure is similar to that of the female hormone estrogen, and it is known that it binds to the estrogen receptor and exhibits various biological actions in an accelerated or competitive manner. Although isoflavones are mildly effective, they are said to have no side effects even if they are taken on a daily basis, but on the other hand, they may also increase the risk of breast cancer, uterine cancer, etc. The Ministry of Health, Labor and Welfare has urged attention to isoflavone intake through supplements.
In view of these circumstances, it is necessary to consider the risk of side effects that are frequently consumed and consumed more frequently than taking substances similar to female hormones as a means to relieve symptoms in menopause. Drugs and supplements for menopausal disorders that are extremely safe and without safety are anxious.
By the way, it is effective in prevention and improvement of swelling in
The longhorn (Pipe longum) described in
Fatigue associated with menopause is thought to be due to decreased secretion of female hormones, local muscle fatigue or general fatigue due to physical exercise, or perceived mental fatigue due to stress load, prolonged duration The kind of chronic fatigue that comes from exercise and activity is completely different. Therefore, the fatigue-improving composition for recovering fatigue accompanying menopause is considered to have a different mechanism of action from the normal fatigue-improving composition, and is assumed to have an effect on general fatigue. However, it is not known whether it functions as a fatigue improving composition for recovering fatigue accompanying menopause.
This invention is made | formed in view of such a situation, and it aims at providing the novel fatigue improvement composition which can reduce the fatigue symptom accompanying menopause.
It is difficult to predict whether there is no clue such as isoflavone that has a similar molecular structure to estrogen, such as isoflavone, which has an effect on fatigue recovery accompanying menopause. In addition, when evaluating a fatigue improving composition that relieves symptoms of fatigue associated with menopause, it is extremely important to use objective evaluation rather than subjective evaluation using a monitor. As a result of intensive studies by the present inventors, we found an evaluation method that can objectively evaluate whether it is useful for recovering fatigue associated with menopause, and Hihatsu is effective in recovering fatigue associated with menopause. As a result, the present invention was achieved.
That is, the fatigue improving composition for menopause of the present invention is characterized by containing hihatsu. The fatigue-improving composition of the present invention is preferably a menopausal fatigue-improving composition containing a fatigue-improving agent for menopause consisting of hihatsu (a menopausal fatigue-improving composition compounded with hihatsu as a menopausal fatigue-improving agent). .
The fatigue-improving composition for menopause of the present invention contains hihatsu and can recover the fatigue accompanying menopause.
Hereinafter, the fatigue improving composition for menopause of the present invention will be described in detail. Menopause is the so-called menopause (the transition period between the genital (sexual maturity) and non-genital (aged) period, the period until the ovarian function begins to decline and disappears), and young people due to decreased estrogen secretion It is a broad concept that includes gender menopause and male menopause due to decreased estrogen secretion.
The fatigue improving composition for menopause of the present invention is characterized by containing an extract of Piperlongum as an active ingredient. Hihatsu is an evergreen vine that belongs to the genus Pepperaceae and is distributed in Southeast Asia. The fruit spikes are thick and cylindrical, and are used as spices. The most preferred site for the use of baboons in the present invention is fruit spikes, but roots, leaves, stems, flowers, or mixed sites thereof can also be used. The cherries that can be used as a fatigue-improving composition for the menopause of the present invention can be obtained by an extraction method generally used for the extraction of plants using these extraction raw materials.
For example, it can be obtained by drying the jade ears and then immersing them in an extraction solvent for a certain period of time or bringing them into contact with an extraction solvent that has been heated to reflux, followed by filtration and concentration. As the extraction solvent, any solvent that is usually used for extraction can be used. For example, water, methanol, ethanol, propylene glycol, 1,3-butylene glycol, glycerol and other alcohols, chloroform, dichloroethane, four Organic solvents such as carbon chloride, acetone and ethyl acetate can be used alone or in combination. The extract obtained by extraction with the above solvent is used as it is, or concentrated extracts are used, or these extracts are removed by using an adsorption method, for example, ion exchange resin, or a porous polymer (for example, Amberlite XAD- After adsorbing on the column of 2), elution with methanol or ethanol and concentration can also be used. Further, a partitioning method, for example, an extract extracted with water / ethyl acetate can be used.
In addition, a hihatsu extract and hihatsu extract powder prepared from an extract of hihatsu are commercially available and may be used.
There is no particular limitation on the composition for improving menopause's fatigue, which comprises an agent for improving menopause consisting of hihatsu. For example, oral drugs such as drugs and quasi-drugs formulated with hihatsu as an active ingredient and excipients. In addition to health foods such as pharmaceuticals, foods for specified health use, functional nutritional foods, and health foods such as supplements, sweets such as chewing gum, chocolate, candy, jelly, biscuits, and crackers, mixed with foods and beverages; ice Frozen confectionery such as cream and ice confectionery; Beverages such as tea, soft drinks, energy drinks and beauty drinks; Noodles such as udon, Chinese noodles, spaghetti and instant noodles; Seasonings; fats and oils such as margarine, butter, salad oil; bread, ham, soup, retort food All common food products such as frozen food, and the like.
Although the intake disorder of hihatsu is not particularly a problem, when the fatigue improving composition containing the hihatsu of the present invention is ingested, the intake is preferably converted into the amount of hihatsu as an active ingredient per adult day, preferably It is preferable to ingest so that it may become 1-3000 mg / kg, More preferably, it will be 10-200 mg / kg.
As mentioned above, fatigue associated with menopause is due to decreased secretion of female hormones, which is completely different from general physical fatigue and mental fatigue, and can recover general fatigue. It is not known whether the composition considered to be able to recover from fatigue due to decreased secretion of female hormones. The present inventors have repeated experiments using an ovariectomized model (OVX) mouse that produces a decrease in female hormone secretion. As a result, the amount of behavior of the ovariectomized mouse is lower than that of a normal mouse. It was discovered that there was a rest period in which the amount of activity decreased greatly in the middle of the activity period. Then, by focusing on the rest period in this activity period, it was found that Hihatsu is extremely effective for fatigue associated with menopause.
Specifically, the amount of behavior in the active phase of the OVX mouse before administration of hihitsu is measured by measuring the amount of behavior in the active phase of the OVX mouse before administration of hihatsu and the control mouse (normal mouse) before administration of hihatsu, respectively. The amount of behavior of the control mice in the time period corresponding to the resting period was confirmed, and HIX was administered to each of the OVX mice and the control mice. By objectively assessing whether hihatsu helps relieve fatigue associated with menopause by comparing the amount of behavioral change before and after administration of hihatsu in control mice in the time period corresponding to the rest period This has led to the present invention.
Below, while showing that a hihatsu is effective in the fatigue accompanying a menopause, an example shows the prescription example of the fatigue improvement composition containing a hihatsu by an Example.
(Measurement of activity during active period before administration of hihatsu)
Ovariectomy was performed on 6 OVX mice preliminarily bred for about 2 weeks after performing 11-week-old female ddY mice and the same 11-week-old female ddY mice in order to offset the effects of ovariectomy surgery. After performing a sham operation without performing the procedure, five Sham mice preliminarily raised for about 2 weeks were prepared as control mice. About each prepared mouse | mouth, the change of the action amount of 12 hours of active phases was measured with time. The amount of activity was measured using an activity amount measuring device (Merquest) and output as the number of reactions (count) of the infrared sensor. The results are shown in FIG. The graph shown in FIG. 1 shows the amount of action by counting per hour. In addition, the amount of action of FIG. 1 is shown as an average value +/- standard deviation about 5 Sham mice (n = 5) and 6 OVX mice (n = 6) (* and ** are Sham by student t-test). Each mouse represents a significant difference with a risk rate of 5% and 1%).
From the graph shown in FIG. 1, it can be seen that the OVX mouse has a time zone in which the amount of activity is greatly reduced in the middle of the active period compared to the Sham mouse (in addition, when only the amount of action of the OVX mouse is seen, However, the behavioral amount of the Sham mice is also reduced at the 12th hour, and this behavior is excluded from the rest period in the active period). Since the difference between OVX mice and Sham mice is whether or not ovariectomy is performed, this decrease in behavioral amount is due to ovariectomy. In other words, the behavioral amount given by the presence or absence of estrogen secreted from the ovary It is shown. For this reason, it is inferred that this time period during which the amount of action decreases greatly contributes to fatigue during menopause. Therefore, in order to confirm whether hihatsu exhibits a recovery effect (anti-fatigue effect) against fatigue associated with menopause, the amount of such activity decreases, that is, the amount of activity during the rest period in the active period It can be seen that it is important to identify and evaluate.
FIG. 2 is a graph obtained by integrating the rest period in the active period of the OVX mouse and the amount of action in the corresponding time zone of the Sham mouse. Specifically, the time when the decrease in the amount of action is most observed for the Sham mouse, In FIG. 1, the cumulative action amount of the OVX mouse for 6 hours from the second hour to the seventh hour of the active period including the fifth hour of the active period and the cumulative action amount of the Sham mouse during the same time period are shown. From FIG. 2, the cumulative behavioral amount for 6 hours including the time when the behavioral amount of the OVX mouse is most decreased with respect to the behavioral amount of the Sham mouse is reduced by 30% or more compared to the cumulative behavioral amount of the Sham mouse. It is perceived that there is. This value of 30% is the difference necessary to exclude the time zone in which the amount of action decreases in both OVX mice and Sham mice (from the 8th to 12th hours including the 12th active phase as described above). is there. In addition, although the comparison is made with the integrated action amount of 6 hours including the time when the decrease in the action amount is most observed with respect to the Sham mouse, the action amount of the OVX mouse is the most reduced with respect to the action amount of the Sham mouse. If it is a time zone that includes the observed time and falls by 30% or more with respect to the cumulative action amount of the Sham mouse, it can be arbitrarily specified in the range of 2 to 6 hours.
(Measurement of activity during active period after administration of Hihatsu)
Eleven weeks old female ddY mice were subjected to ovariectomy and 16 OVX mice preliminarily raised for about 2 weeks were divided into two groups. One group received hihatsu (hihatsu extract powder MF: manufactured by Maruzen Pharmaceutical) for one day. One dose of 500 mg / kg was suspended in a vehicle (tragacanth gum solution) and forcibly administered orally for 5 weeks, and the other group was administered only a vehicle containing no rabbits. The behavioral amount of the HIX-administered OVX mice and non-hihatsu-administered OXX mice (control mice) was measured by the same method as described above. The results are shown in FIGS. FIG. 3 is a graph showing changes in the amount of behavior of HIX-administered OVX mice and control OVX mice during 12 hours of activity, and FIG. 4
(Measurement of behavior of normal mice after administration of Hihatsu)
After 16 8-week-old female ddY mice (normal mice) were preliminarily raised for about 1 week, they were divided into two groups, and one group received 500 mg of hihatsu (hihatsu extract powder MF: manufactured by Maruzen Pharmaceutical) once a day. / Kg was suspended in a vehicle (tragacanth gum solution) and administered by oral gavage for 4 weeks, and the other group was administered only a vehicle containing no rabbits. In order to make a fatigue model by daily work on the 4th week from the start of the test, a mild fatigue load test was conducted. In the fatigue load test, a treadmill (manufactured by Osaka Microsystem Co., Ltd.) was used, and the running load was determined by running for 30 minutes at a speed of 25 m / min and an inclination of 7 degrees. Immediately after the fatigue load test, the amount of behavior was measured by the same method as described above. The results are shown in FIGS. FIG. 5 is a graph showing changes in the amount of behavior of normal and control normal mice after 12 hours of active phase, and FIG. 6
