プリオンは核酸を欠いた伝染性の粒子である。最も注目されるプリオン疾患は、牛海綿状脳症(BSE)、ヒツジスクレイピー、シカ科動物(シカ、エルク、ムース)における慢性消耗病(CWD)及びヒトにおけるクロイツフェルト・ヤコブ病(CJD)である。プリオンは、PrPScと称されるプリオン蛋白質(PrP)の修飾されたイソ型のみから構成されていると考えられている。正常な細胞PrP(PrPCと称される)は翻訳後プロセシングにより感染性のPrPScに変換される。このプロセシングにおいて、PrPCの構造は改変され、かつPrPの生理化学的性質の変化を伴う。プリオンは天然の細胞蛋白質(PrPC)の病原型へのリフォールディングを誘導する、配座変更された蛋白質の(PrPSc)の能力により疾患を引き起こすと考えられている。それは、感染した個体の脳に形成される特徴的な海綿状プラークの形成を最終的には生ずるこの蛋白質変換反応の増殖である。
プリオン疾患への感受性は遺伝学的に決定されるものであることがこれまでに検証されてきた。これは、ヒツジのスクレイピー及びエルクのCWDの場合には最も明らかに示されており、プリオン遺伝子のコード領域における特徴的なアミノ酸はCWD感染に対する感受性を調整する。エルクに関しては、プリオン遺伝子の132位にあるメチオニン残基の存在が感受性に対する劣性の決定因子である。シカにおける状況はそれほど明らかではないが、少なくとも4つの別個の遺伝子座が関係していると考えられている。CWDに対して遺伝的に感受性のある動物を最初に特定した。特定後、それらの動物を、FDC培養物を生成するためのドナーとして使用した。10匹のエルクと10匹のオジロジカからの血液サンプルを、プリオン遺伝子の遺伝シークエンシングの育種者から入手した。結果を表1に示す。
シカ及びエルクFDCの初代培養物は、遺伝学的に感受性の高い動物のリンパ節から単離した。実施例1に従って選択された動物は地方の農場より入手し、ケタミン/キシラジンを使用して麻酔をかけ、サウスダコタ獣医学診断用実験室の標準の手順に従う感電死によって屠殺した。次に、全ての咽頭後リンパ節及び腸間膜リンパ節が新たに屠殺された動物から得られ、単細胞懸濁液を生成するための標準の手順に従って処理した。次に細胞を、FDCと特異的に反応するものとしてこれまでに確認されている抗体とともに15分間培養し、市販のヤギ抗マウス抗体結合磁気ビーズで二次染色した。次に、これらの細胞をAutoMACSを使用して選択し、陽性の細胞は10%のウシ胎仔血清を含む富栄養組織培養培地中にて培養した。それらの同定は、細胞表面マーカ、形態学及び増殖能力により確認した。図2は3ヶ月の仔ヒツジに由来する回腸パイヤー斑及び咽頭後リンパ節組織の免疫組織化学染色を示す。細胞は3乃至4日の間隔にて新たな培地に供給され、最初のウェルがコンフルエンシーに達すると分割した。3代継代培養された後に、細胞をトリプシン処理し、表面プリオン蛋白質(6H4、スイス国のプリオニクス(Prionics)AG)に対する抗体と反応させた。全てのクローンは、インビトロにてプリオンの伝播を促進するのに必要な、有意なレベルのプリオン蛋白質を発現した。図1を参照されたい。
実施例2にて得られた細胞の、インビトロにおけるプリオンの伝播を促進する利用可能性を定義した。これらの実験の時間集中的な性質は、プリオンの伝播を促進するこれらの細胞の効能の最終的な試験において有意な影響を有した。特に、FDCは極端に成長の遅い細胞であり、コンフルエントな培養に到達すると、プリオンにて更に感染させるには、最低限でも2乃至4週間は必要であった。
FDCのプリオン感染の詳細なプロトコルは以下の通りである。
[全体計画]:細胞は、感染前及び感染時には、血清飢餓状態であった。感染性は24時間未満の期間にわたってのみ実施されたが、その後細胞は、PrPScの伝播を促進するために数週間まで培養した。
a)対照;
b)FDC+100μl希釈脳ホモジネート;
c)FDC+正常ヒツジ血清と1:1で前培養させた100μlの脳ホモジネート;
d)FDC+100μl希釈脳ホモジネート+B細胞(107/ウェル);
e)FDC+正常ヒツジ血清と1:1で前培養させた100μlの脳ホモジネート+B細胞(107/ウェル);
f)FDC+末梢血単核細胞(107/ウェル)+100μl希釈脳ホモジネート;
g)FDCS+末梢血単核細胞+正常ヒツジ血清と1:1で前培養させた100μlの脳ホモジネート。
3)各ウェルに、10%FCSを含んだ1.7mlの1X HBSSを加えて、37℃で1時間インキュベートする。
5)おおよそ処理された(即ち、前培養されている、又はされていない)100μlの脳ホモジネートを加える。
7)PBSで細胞を2回洗浄し、バイオハザードとして処分し、廃棄する前に漂白剤で処理する。
9)更なる実験のために各々の複数のアリコートを更に4乃至6週間凍結させる(10%DMSO/90%FCS中における凍結)。
B細胞の単離のための詳細なプロトコルは以下の通りである。スクレイピーに感染されていないが感受性のある動物から得られた末梢血単核細胞を、パーコール濃度勾配を使用して調製した。細胞を計数して、PBS−1%FCS中に108個の細胞/ml(1−2×108個の細胞、全量)、となるように再懸濁させた。CD4(17D)、CD8(6−87)、CD61(1−44−19)及びγδ−TcR(18−106)に対する抗体1mlを加え、4℃にて10分間インキュベートした。細胞をPBS−FCSにて2回洗浄し、108個の細胞に対して200μlのGAM−IgG磁気ビーズを用いて、107個の細胞/mlの最終濃度にて、4℃にて10分間インキュベートした。細胞を2回洗浄し、次にAutoMACSを使用してB細胞に対して負の選択をした。採取した細胞を計数して、感染させるために10−7細胞/mlにて100ng/mlのE.Coliリポポリサッカライド(LPS)を含む培地中に再懸濁させた。
3)各ウェルに、107個の細胞を加える。
4)37℃にてインキュベートし、付着B細胞を損傷しないように慎重に、4日毎に新たな培地を加える。
末梢血B細胞を2匹のヒツジから単離し、一方はスクレイピー脳ホモジネートの脳内注射を用いて予め2ヶ月感染させた。この単離に対して正常なインキュベーションは14乃至17ヶ月であると仮定すると、限られた数のB細胞のみがPrPScで感染させるために利用できるものであると考えられる。それにも関わらず、末梢血由来のB細胞は培養したFDC上に播種され、10日間共に培養した。外因性のPrPScは培養物中には播種しなかった。インキュベーション後、病原性プリオン蛋白質(15B3、プリオニクス(Prionics)社から研究の目的にて入手した)に特異的な抗体を使用して、PrPScの存在を確認するために培養物を染色した。感染動物からの培養物は標準的な免疫蛍光測定を用いて強い陽性を示し、一方未感染の動物から得られた培養物は陰性であった。未感染のヒツジ(左側)及びスクレイピーに感染したヒツジ(右側)に由来するB細胞とともにインキュベートを開始後の10日におけるFDC培養物の免疫蛍光染色を示す図11A及び11Bを参照されたい。右側のパネルにおいて、PrPScに特異的なモノクロナール抗体15B3を用いて細胞が強力に染色されたことを明記したい(矢印)。分散した、非特異的な染色のみが未感染の動物からの培養物においては認められた。
急性プリオン疾患におけるB細胞サブセットを解析した。リンパ節において、PrPScは最初の感染部位からFDCに遊走白血球を介して輸送されていると思われる。そこで一度、PrPCは、感染を受けたFDCとの相互作用により濃度が増え、同FDCにおいて、イコソームズ(iccosomes)を介して部分増殖するB細胞及び可染体マクロファージへと移動する。これら研究の全体の意味は、PrPScは感染を受けたリンパ節におけるB細胞の成長を選択的に阻害すべきであるということである。PrPScでの感染に対するリンパ節の部分的応答を試験するために、本発明者らは両側の前大腿リンパ節を排出する輸出リンパ管にカニューレを挿入した。固有の組織層からこれら2つのリンパ節へリンパ液が排出されると、一方のリンパ節に試験材料(PrPSc)を選択的に接種する一方で、他方のリンパ節を対照として確保することが可能である。この方法を用いて、本発明者らはスクレイピー陽性動物の10%脳ホモジネートの200μlを右側の前大腿リンパ節の排出領域に注入し、正常動物から得られた等量の10%脳ホモジネートを左側の前大腿リンパ節に注入した。引き続く10日間にわたり、一定の間隔にて輸出リンパ液を採取し、局所リンパ節における進行する免疫応答に影響を与える特異的な細胞型の産生における変化を決定するために表現型化した。全体の細胞の産生及び両方のリンパ節からのCD4陽性T細胞及びCD8陽性T細胞において同等の変化があったが、スクレイピー注入側からのB細胞の産生において有意な減少があった。スクレイピーを接種したリンパ節からのB−細胞の産生における低減を示す図13を参照されたい。スクレイピー感染脳ホモジネートの注入に続き、局所リンパ節においてB細胞の産生が一時的ではあるが、有意な減少を示した。上側の青色の線は正常脳;下側の赤色の線はスクレイピー脳。
遊走性B細胞によるプリオンの輸送について調べた。血液がプリオン疾患を効果的に伝達することは幾らかの場合において知られているが、感染性粒子の性質については疑問が残るままである。最近のデータによると遊走性B細胞は感染プリオン蛋白質を輸送することが提案されているので、本発明者らは既に述べたように、スクレイピーで実際に感染したリンパ節を排出して、輸出リンパ細胞及び輸出リンパ血漿を採取した。輸出リンパ血漿のサンプルは、スクレイピー感染リンパ節及び対照リンパ節のいずれから得られたものも決まって陰性とされたが、PrPScに対して陽性である細胞は、免疫組織化学及びドット−ブロットのいずれによってもスクレイピー注入リンパ節から排出されたもののみから認められた。図14を参照されたい。興味深いことに、細胞に関連したPrPScの濃度は、スクレイピーを局所注入後の約5日目から始まり、実験が完了する注入後10日目まで増大し続けた。3つの別々の実験において示されたように、遊走性白血球は感染を受けたリンパ節からPrPScを輸送することが可能ではあるが、このデータを確認するために、そして、B細胞がこの輸送に必要とされる細胞型であることを確認するために、更なる実験が必要である。
Claims (28)
- インビトロにおいて感染プリオン(PrPSc)を伝播させる方法において、
濾胞樹状細胞(FDC)の培養物を提供する工程と、
前記FDC培養物に感染プリオンを加える工程と、
感染した細胞を培養する工程と、
からなる方法。 - 末梢B細胞をFDC培養物に加えて結合した細胞培養物を得る工程を更に含む、請求項1に記載の方法。
- 動物又はヒトにおいて感染プリオン(PrPSc)を検出する方法において、
感染プリオンに感染している疑いのある動物又はヒトから末梢血B細胞を採取する工程と、
前記B細胞を培養した濾胞樹状細胞とともに培養する工程と、
特異的結合アッセイにより感染プリオンを検出する工程と、
からなる方法。 - 前記特異的結合アッセイは免疫学的アッセイである、請求項3に記載の方法。
- 前記免疫学的アッセイは免疫組織化学を含む、請求項4に記載の方法。
- 前記免疫組織化学はウエスタンブロットを含む、請求項5に記載の方法。
- 前記動物はヒツジであり、前記免疫学的アッセイはスクレイピーに特異的な抗体を必要とする、請求項4に記載の方法。
- 前記動物はシカ科の動物であり、前記免疫学的アッセイは慢性消耗病(CWD)に特異的な抗体を必要とする、請求項4に記載の方法。
- ヒトにおける感染プリオンを検出するための、請求項4に記載の方法において、前記免疫学的アッセイはヒトプリオン蛋白質(PrP)に結合する抗体を必要とする、方法。
- ウシにおける感染プリオンを検出するための、請求項4に記載の方法において、前記免疫学的アッセイはウシプリオン蛋白質(PrP)に結合する抗体を必要とする、方法。
- 動物又はヒトにおいて感染プリオン(PrPSc)を検出する方法において、
感染プリオンで感染している疑いのある動物又はヒトから、流体、細胞又は組織のサンプルを取得する工程と、
前記サンプルを濾胞樹状細胞の培養物に加えて、前記細胞を培養する工程と、
特異的結合アッセイにより前記培養物中の感染プリオンを検出する工程と、
からなる方法。 - 前記濾胞樹状細胞の培養物はB細胞を含む、請求項11に記載の方法。
- 前記特異的結合アッセイは免疫学的アッセイである、請求項11に記載の方法。
- 前記免疫学的アッセイは免疫組織化学を含む、請求項13に記載の方法。
- 前記サンプルは、血液、脳、脾臓、脊髄液、リンパ節及び扁桃腺からなる群より選択される、請求項11に記載の方法。
- インビトロにおいて感染プリオン(PrPSc)を伝播させる方法において、
プリオン疾患に感受性である動物を選択する工程と、
前記動物からリンパ節細胞を取得する工程と、
FDCに特異的な抗体と結合するリンパ節細胞を選択して、得られた細胞を培養する工程と、
プリオン蛋白質に特異的な抗体と結合する培養物から細胞を選択する工程と、
前記選択した細胞を感染プリオンに感染させる工程と、
前記感染した細胞を培養する工程と、
からなる方法。 - 前記動物を選択する工程は、プリオン疾患に対して遺伝的に感受性である動物を選択する工程を必要とする、請求項16に記載の方法。
- 前記動物はヒツジであり、前記プリオン疾患はスクレイピーである、請求項17に記載の方法。
- 前記動物はシカ科の動物であり、前記プリオン疾患は慢性消耗病(CWD)である、請求項17に記載の方法。
- 前記動物はウシであり、前記プリオン疾患は牛海綿状脳症である、請求項16に記載の方法。
- 生物サンプル中のプリオンを検出し、かつ選択的に定量する方法において、前記方法は、
前記生物サンプルを、FDC及びB細胞の混合培養物と、同サンプルの感染を可能にする条件下にて接触させる工程と、
前記培養した細胞の感染又は非感染を検出する工程と、を含み、前記感染の存在は前記サンプル中のプリオンにて示される、方法。 - 前記サンプルは、血液、リンパ節及び脳からなる群より選択される、請求項21に記載の方法。
- 前記FDC及びB細胞の混合培養物はプリオン疾患に対して遺伝的に感受性である動物から単離される細胞を含む、請求項21に記載の方法。
- 前記検出工程は免疫学的アッセイを含む、請求項21に記載の方法。
- 生物サンプル中の感染プリオン(PrPSc)を検出するためのキットであって、前記キットは、
培養された濾胞樹状細胞(FDC)と、
