Abstract
Description
The present invention relates to a composite material used in the medical field and the like, and particularly to a titanium-based member having antibacterial properties.
Various composite materials including titanium-based members have been tried for biological implants such as artificial bones and artificial joints. A major problem when using such a material for a biological implant is infection after surgery, which is often difficult to treat. The most promising means for preventing this post-operative infection is to impart antibacterial properties to the materials used, but no such technology has yet been developed.
For example, in Japanese Patent No. 2775523 (Patent Document 1), a substrate made of titanium or a titanium alloy is immersed in an alkaline solution, and then heated to a temperature lower than the transition temperature of titanium, whereby a titanium oxide phase is formed on the surface of the substrate. Although a bone repair material is described in which an amorphous phase coating of alkali titanate is formed, this material is exclusively for promoting bone formation and has no antibacterial properties . Japanese Patent No. 3608167 (Patent Document 2) discloses a technique in which titanate nanotubes are coated by a method of applying and baking crystalline titania and further replacing metal ions such as calcium. The purpose is to promote bone formation, and no antibacterial viewpoint is considered.

An object of the present invention is to provide a new type of composite material having antibacterial properties suitable for use in biological implants and the like.
As a result of repeated studies, the present inventors have developed a technique for coating a titanium-based substrate with a crystalline titanate layer containing silver ions, and have derived the present invention.
That is, the present invention includes a titanium-based substrate, a crystalline alkali titanate nanosheet layer or nanotube layer formed on the substrate, and a silver titanate layer in which the alkali component of the alkali titanate is replaced with silver ions. An antibacterial titanate-coated titanium-based member is provided.
Furthermore, according to the present invention, there is provided a method for producing the above antibacterial titanate-coated titanium-based member, wherein (i) a step of subjecting the titanium-based substrate to hydrothermal treatment at a temperature of 110 to 180 ° C. in an alkaline aqueous solution. (Ii) a step of heat-treating the product after the hydrothermal treatment at 200 to 700 ° C., and (iii) a step of washing and drying the product after the heat-treatment and then immersing in a silver ion-containing aqueous solution. A method characterized in that is provided.
The present invention realizes a biological implant coated with a titanate layer containing (substituted) silver ions, which is the most effective antibacterial agent for preventing postoperative infection.
According to the present invention, unstable and difficult to coat titanate can be stably coated as a nanosheet or nanotube layer of crystalline alkali titanate. Moreover, the elution rate of silver ions can be controlled by controlling the alkaline hydrothermal reaction conditions or by post-treatment.
One of the features of the antibacterial titanate-coated titanium-based member of the present invention is that a “crystalline” alkali titanate nanosheet or nanotube layer is formed on a titanium-based substrate. Therefore, according to the present invention, the titanium-based substrate is subjected to hydrothermal treatment in an alkaline aqueous solution at a high temperature, that is, at a temperature of 110 to 180 ° C., preferably at a temperature of 150 to 170 ° C. The reaction time is generally 2 to 30 hours, and the film thickness (the thickness of the nanosheet layer or nanotube layer of crystalline alkali titanate) can be controlled by controlling the reaction time.
The aqueous alkali solution used in the hydrothermal treatment according to the present invention is most commonly an aqueous sodium hydroxide solution, but is not limited thereto, and for example, an aqueous potassium hydroxide solution can also be used. As described above, a nanosheet layer or a nanotube layer of crystalline alkali titanate can be obtained by performing hydrothermal treatment at a high temperature and controlling the concentration of the aqueous alkali solution. For example, when hydrothermal treatment is performed using a sodium hydroxide aqueous solution at a temperature of 150 to 170 ° C., a nanosheet layer can be obtained if the concentration of the sodium hydroxide aqueous solution is about 1 mol / l, and if the concentration is about 10 mol / l. A nanotube layer is obtained.
The type of crystalline alkali titanate constituting the titanium-based member of the present invention depends on the alkali used in the hydrothermal treatment step as described above, and when sodium hydroxide is used, it becomes sodium titanate, and when potassium hydroxide is used Is potassium titanate.
Further, as the titanium-based substrate used in the present invention, titanium alloy can be used in addition to titanium metal. Examples of titanium alloys suitable for use in the present invention include Ti-6Al-4V alloy, Ti-6Al -2Nb-1Ta alloy, Ti-15Zr-4Nb-4Ta alloy, Ti-6Al-7Nb alloy, Ti-15Mo-5Zr-3Al alloy, Ti-13Nb-13Zr alloy, Ti-12Mo-6Zr-2Fe alloy .
When the substrate has a fine or complex shape or surface structure, such as a wire-like substrate having a small outer diameter, a titanium source material is used in addition to the titanium-based substrate, and the titanium-based substrate is replaced with titanium. The alkaline hydrothermal treatment is preferably performed in the state of being close to the source material. Here, the state in which the titanium-based substrate is brought close to the titanium source material means that there is a void in which a liquid phase exists between the titanium-based substrate and the titanium source material, specifically, a void of 10 μm to 1000 μm. The state where the base material and the titanium source material are approaching each other. In order to carry out such an alkaline hydrothermal treatment, for example, when the titanium base material is in the form of a wire, the base material is wound around the titanium source material, or the gap is formed between the base material and the titanium source material. Alkaline in a state where a corresponding spacer is provided, or a wire having an outer diameter corresponding to the gap is wound around one of the base material and the titanium source material, and the base material and the titanium source material are stacked and fixed with an appropriate member. What is necessary is just to perform a hydrothermal treatment under the above conditions by immersing in an aqueous solution. As another method, the substrate may be subjected to hydrothermal treatment in a state where the substrate is fitted in the mold made of the titanium source material so that the voids exist.
According to the present invention, as described above, by changing the structure of the titanate layer or controlling the film thickness, the silver substitution amount described later is adjusted to control the elution (release) rate of silver ions. An antibacterial member can be obtained.
The hydrothermally treated product obtained as described above is then subjected to a heat treatment at 200 to 700 ° C., thereby forming (coated) crystalline titanate on the titanium-based substrate. The layer (nanosheet layer or nanotube layer) will be stabilized.
The alkaline water heat treatment method has been known as a good method for forming titanate nanotubes, but has a drawback that the coating layer is easily peeled off when the substrate is coated. Therefore, the present inventor has advanced earnestly research and succeeded in preventing the coating layer from being peeled off by performing heat treatment at 200 to 700 ° C. as it is without washing with water after alkali heat treatment. This is presumably because the heat treatment causes the polycondensation of hydroxyl groups at the interface between the titanium base and the dense sodium titanate layer below the titanate nanotube layer (or nanosheet layer), thereby improving the adhesion of the coating layer. That is, for the polycondensation reaction, a temperature of 200 ° C. or higher is necessary, and on the other hand, when the temperature is higher than 700 ° C., the adhesion itself is impaired.
Thus, according to a preferred embodiment of the present invention, the product after hydrothermal treatment is heat-treated at 200 to 700 ° C., preferably 300 to 600 ° C. without washing. This embodiment is particularly effective when a titanate layer having a large film thickness is formed (coated) on a titanium-based substrate.
In order to produce the antibacterial titanate-coated titanium-based member of the present invention, the product after the above heat treatment is washed and dried, and then immersed in an aqueous solution containing silver ions. Thereby, the antimicrobial member by which the sodium ion and potassium ion of crystalline alkali titanate were substituted by the silver ion is obtained. As a silver ion containing aqueous solution used for this immersion process, silver acetate aqueous solution is preferable. Of course, other silver ion-containing aqueous solutions such as an aqueous silver nitrate solution can also be used. This silver ion substitution step can be performed at room temperature or near room temperature under light shielding.
Furthermore, according to the preferable aspect of this invention, a part of silver ion substituted and produced | generated by the said process immersed in a silver ion containing aqueous solution is converted into silver oxide or metallic silver. For example, a part of the silver ions is converted into silver oxide or metallic silver by heat treatment or UV irradiation (ultraviolet light irradiation) of the product obtained by the immersion step. Moreover, a part of silver ion can also be coat | covered with metallic silver by giving silver sputtering to the product after the said immersion process.
Thus, according to the present invention, an antibacterial titanium member containing silver oxide (Ag 2 O) and metallic silver (Ag) in addition to silver ions (Ag + ) is obtained. The elution rate as silver ions is in the order of silver ions> silver oxide> metal silver. Therefore, the antibacterial member of the present invention can also control the elution rate of silver ions to release silver ions gradually, which is an important function for preventing infection after surgery particularly in a biological implant, This is one of the features of the present invention.
The shape of the titanium-based substrate used in the antibacterial titanate-coated titanium-based member of the present invention is not particularly limited, and is based on various shapes such as a plate shape (flat plate shape), a wire shape, a mesh shape, and a tube shape. Material is applicable.
Hereinafter, examples will be described in order to more specifically show the characteristics of the present invention, but the present invention is not limited to the following examples.
Titanate nanotube coating (1)
A 20mm x 20mm x 2mm pure titanium test piece (provided by Nippon Medical Materials Co., Ltd.) was degreased and washed with ethyl alcohol, then placed in a hydrothermal container with 20ml of 10mol / l sodium hydroxide aqueous solution, and placed in a 160 ° C oil bath For a predetermined time (3 hours or 20 hours). After the reaction was completed, the test piece was taken out, dried at 10 ° C. or lower overnight without being washed with water, and then heat-treated at 300 ° C. for 1 hour in an electric furnace. Thereafter, the test piece was repeatedly washed with distilled water until the pH became near neutral. After washing, it was dried overnight at 10 ° C. or lower.
When the obtained product was observed by TEM, it was found that most of the film was composed of nanotubes with an outer diameter of about 7 nm (Fig. 1b), and the result of EDX (energy dispersive X-ray) analysis (Fig. 2). ), The nanotube was found to be Na 2 Ti 4 O 9 H 2 O. SEM observation of the cross section of the thin film revealed that the TNT (titanate nanotube) phase was from the thin film surface to about 19 μm, and the dense sodium titanate phase was from the back surface (the surface in contact with the Ti plate) to about 1 μm. (Figure 1a). Therefore, first of all, the elution of Ti species from the Ti (titanium) plate by the reaction between the Ti plate and OH − and the reprecipitation reaction of the Ti species as sodium titanate occur simultaneously. It was found that the TNT phase grew after the dense phase of sodium was formed on the Ti plate.
In this example, in order to prevent peeling of the TNT thin film, it was dried without being washed with water after hydrothermal treatment as described above. Although NaOH crystals were observed on the plate, the thin film remained fixed on the plate. When this plate was washed with water after heat treatment at 300 ° C. for 1 hour in the air as described above, the NaOH crystals eluted, but the TNT thin film remained firmly fixed on the plate. Peeling was not recognized (the upper left of FIG. 3). In addition, no change in the nanotube morphology was observed during the heat treatment at 300 ° C. This sticking is considered to be because the dehydration condensation of the hydroxyl group progressed near the interface between the Ti plate and the sodium titanate phase by heat treatment at 300 ° C., and the sodium titanate was more strongly immobilized on the Ti plate. When XRD measurement (CuKα) of this plate was performed, only diffraction lines peculiar to TNT and metal Ti were observed (see FIG. 4). In addition, the formation of a TNT thin film was observed even after 3 hours of reaction, and the thickness of the thin film was 13 μm. Thus, it was confirmed that the film thickness could be controlled by controlling the reaction time.
Titanate nanotube coating (2)
In the same manner as in Example 1, synthesis of TNT was attempted using Ti wires having various outer diameters and lengths as a Ti source. As a result, it was found that a Ti source having a long and large outer diameter (for example, a Ti wire having an outer diameter of 203.7 μm and a length of 24 cm), that is, a Ti source having a larger surface area, tends to generate a TNT phase on the surface. On the other hand, a Ti wire with a short outer diameter (for example, a Ti wire with an outer diameter of 53.4 μm and a length of 5 cm), that is, a Ti source with a small surface area, generates a dissolved Ti wire or an amorphous sodium titanate phase. I found it easy.
Furthermore, even when a similar experiment was performed using Ti mesh or Ti sphere as the Ti source, TNT phase was generated on the surface while maintaining the shape of the mesh or sphere (upper right and lower left in FIG. 3), With increasing reaction time, the thickness of TNT phase also increased. The same experiment was performed on the Ti tube, and the central part of the obtained tube was cut and the inner and outer surfaces of the tube were observed (lower right in FIG. 3). The inner and outer surfaces of the tube were TNT. It was confirmed that it was covered.
Titanate nanotube coating (3)
As a base material, a Ti wire having a diameter of 53.4 μm was wound around a Ti plate that could serve as a Ti source and reacted in the same manner as in Example 1. As a result, the wire shape was maintained in both the reaction time of 3 hours and 20 hours. It was observed by SEM that a fibrous substance was densely formed on the surface of the wire, and it was confirmed by TEM that this fibrous substance was TNT having an outer diameter of about 7 nm.

When XRD measurement of the product was performed, only diffraction lines attributed to titanium metal and sodium titanate were observed (FIG. 5). FIG. 6 is an SEM photograph of the surface of the obtained product, and it can be confirmed that nanoscale sheet titanate is produced.
Silver substitution and silver ion elution test The test piece obtained in Example 1 was placed in a centrifuge tube protected from light with 15 ml of 0.005 mol / l silver acetate solution and reacted in a water bath at 40 ° C for 3 hours to replace silver. Went. After the reaction, it was repeatedly washed with distilled water to completely remove silver ions remaining on the test piece. Then, it was dried overnight at 10 ° C. or lower. FIG. 7 shows the result of EDX analysis of the titanate nanotube test piece obtained in Example 1 after silver substitution. Although the titanate structure is maintained, the peak attributed to Na (sodium) disappears, and it is understood that Na ions are replaced with silver ions.
Next, the silver ion-dissolved sample was immersed in fetal calf serum and subjected to a silver ion elution test. Experimental conditions and results are shown in Table 1. It was confirmed that the silver ion was surely eluted in fetal bovine serum, and that the titanium-based member according to the present invention has a function of releasing silver ion in the biological fluid.
