Abstract
Description
The present invention relates to a gradient liquid feeding method and apparatus. In particular, the present invention relates to an improvement that exerts an effect on solution mixing in a high-pressure gradient.
In liquid chromatography, a gradient elution method is used in which sample components are separated and eluted while changing the composition of the mobile phase in order to shorten analysis time, improve separation, and improve peak shape. Moreover, there are a low pressure mixing method (low pressure gradient) and a high pressure mixing method (high pressure gradient).
The low pressure gradient is a method in which the solution is mixed on the inlet side of the pump while changing the mixing ratio of the solution using an electromagnetic valve or the like and then sent to the column.
On the other hand, the high-pressure gradient is a method of using two or more pumps, adjusting the flow rate of each pump, mixing the solution on the outlet side of the pump, and feeding the solution.
As a conventional method, in high-pressure gradients, pipes are extended from the outlets of multiple liquid-feeding units that feed different types of solutions, and the pipes are joined and mixed together via connectors, etc. To the chromatography system. Here, it is assumed that the initial solution fed from the liquid feeding unit α is “A liquid” and the second solution fed from the liquid feeding unit β is “B liquid”.
Liquid A: When liquid B starts to be fed from 100%: 0%, the liquid is first fed to the chromatography system with the initial composition of A: B = 100%: 0%. It is necessary to wait for stability. (Equilibration process). In this process, the liquid A is fed from the liquid feeding part α by 100% of the set flow rate, and the liquid B is sent from the liquid feeding part β to 0% of the set flow rate, that is, the liquid is not fed.
At this time, in the solution mixing section where the solution merging occurs, ideally, as shown in FIG. 1A, the B liquid is stopped at the interface inside the connector, and the entire amount of the A liquid is transferred to the system. Should be delivered.
However, since the gradient liquid supply is actually in a state where only the A liquid is supplied at the start, the phenomenon that the A liquid replaces the B liquid and flows into the B liquid flow path occurs. Further, due to diffusion at the boundary of the liquid, mixing of the mutual liquid occurs in the flow path (FIG. 2A).
Due to this phenomenon, theoretically, even when a program (FIG. 1B) for linearly changing the composition of the liquid A and the liquid B is used over time, at the stage where the liquid B starts to be delivered, Since the liquid A that has entered the pipe of the liquid B is sent out, the liquid A is sent to the system with a composition of almost 100%. This indicates a delay in the gradient start time (rise). Further, if the liquid feeding is continued in this state, the B liquid is delivered after all the A liquid that has entered the B liquid piping is sent out. However, since the gradient program has progressed for the time taken to send the liquid A that has entered the liquid B side, when the liquid B starts to be sent to the system substantially, it is set in the middle of the program. Thus, the feeding of the B liquid is started.
In other words, the liquid solution should be transferred with a linear solution composition as shown in FIG. 1B. However, as shown in FIG. 2B, the B liquid is hardly sent until a certain time, and then the B liquid at the set flow rate is rapidly sent. Therefore, the ratio of B liquid will increase rapidly.
By this phenomenon, a peak that should have been separated and detected near the start of gradient setting would be detected without being separated due to a sudden rise in gradient.
In order to avoid such a phenomenon, it is most desirable to completely prevent the solution from entering the other side at the boundary surface of the mixing portion of the two liquids. In other words, it is not allowed to have mutual solution contact before the start of the gradient. However, in practice, it is difficult to completely prevent liquid from entering each other at the interface.
As a method for preventing (reducing) intrusion into the B liquid side flow path, Japanese Patent Application Laid-Open No. 2004-61119 (Patent Document 1) “opens and closes the flow path in the vicinity of the solvent mixing portion of the solvent flow path for supplying the solvent. There has been proposed a "gradient liquid feeding device characterized by including an openable / closable means". According to this, “By providing an opening / closing means in the vicinity of the solvent mixing section, the inflow of the initial solvent, which is the above-mentioned problem, into the second solvent flow path can be greatly suppressed. The initial solvent does not flow into the flow path portion that occupies a relatively large capacity between the metering pump and the valve, which can be suppressed to a small volume area up to this point. "
However, even in this proposal, there is a distance between the valve and the solution mixing section, and inflow of a small volume cannot be avoided. In recent HPLC using capillary columns, even nanometer and microscopic changes in the flow rate and solution composition that can be ignored in conventional chromatography can be ignored. For liquid chromatography that handles the flow rate, it becomes a very big disturbance. In the prior art, it is difficult to deal with techniques that require a fluid flow without a continuous flow rate change using a capillary column.
In Japanese Patent Application Laid-Open No. 2004-271409 (Patent Document 2), the first and second sampling loops are connected to the flow paths from the first and second pumps and the flow path to the sample introduction unit, and temporarily hold the solution. The flow path switching means is provided, and the second pump pumps the solution in the second sampling loop to the sample introduction side while sending the solution from the first pump to the first sampling loop. A device that alternately repeats the first state of pushing and the second state of pushing the solution in the first sampling loop to the sample introduction side by the second pump while feeding the solution from the first pump to the second sampling loop. is suggesting.
However, in this apparatus, the solution intrusion is determined only by the first pump and the solution extruding is determined only by the second pump, and the gradient liquid supply during this time must be extremely stepwise, The above problem cannot be solved.
The present invention basically provides a second solution of the liquid A of the initial solution as the mobile phase in the gradient liquid feeding for separating and eluting the sample components while changing the composition of the mobile phase in the liquid chromatography analysis. It aims at preventing the inflow to the solution B liquid side. For this reason, the room for the A solution of the initial solution to flow into the B solution side is eliminated as much as possible, that is, the space where the A solution of the initial solution can flow into the B solution side is almost eliminated, thereby the boundary between the two solutions. The aim is to minimize mutual infiltration of liquids on the surface.
Another object of the present invention is to obtain an apparatus that does not place a distance between the opening / closing means used for blocking the flow path and the solution mixing portion in order to maximize the effect of suppressing the inflow of the initial solution.
In addition, the objective is to minimize the delay in the start of the gradient, minimize the sudden rise of the gradient due to the rapid increase in the ratio of solution B, and bring the mixing ratio of the solution close to the set value. .
It is another object of the present invention to avoid losing a peak that should not be separated and detected near the start of gradient setting due to the rapid rise of the B liquid.
In order to solve the above-mentioned problems and achieve the object, the present invention firstly connects two or more liquid-feeding parts composed of a pump unit and a valve by one key valve, and sends liquids according to the mixing ratio from each liquid-feeding part. In the gradient liquid feeding system configured as described above, a solution mixing section is provided in communication with the two ports of the key valve, and the solution enters the solution mixing section from both ends by switching the key valve, and the two solutions are contact-mixed. A gradient liquid feeding method is proposed.
Secondly, in a gradient liquid feeding system in which two or more liquid feeding parts consisting of a pump unit and a valve are connected by a key valve, and liquid is fed according to the mixing ratio from each liquid feeding part, the key valve is switched. Proposes a gradient liquid feeding method characterized by continuously feeding the mixed solution.
Third, a gradient liquid feeding method is proposed in which the initial solution and the second solution are isolated by switching the key valve.
Fourthly, in a gradient liquid feeding system in which two or more liquid feeding parts composed of a pump unit and a valve are connected by one key valve, and the liquid is alternately fed from both liquid feeding parts. A gradient liquid feeding device is proposed in which a solution mixing unit is provided between the two ports and communicated with the port, and the solution is fed to the analysis system via the solution mixing unit.
Fifth, two or more liquid feeding sections are connected to the key valve, and a liquid feeding section α for feeding the initial solution from the key valve is communicated with the solution mixing section through the key valve port. The first state where the liquid feeding part β for feeding the liquid is stopped by the key valve, and the liquid feeding part α for feeding the initial solution are communicated with the solution mixing part via the port of the key valve, and the second liquid is fed. A gradient liquid feeding device is proposed in which the
Sixth, a gradient liquid feeding device is proposed in which a solution mixing port is connected to an initial solution outlet port and a second liquid outlet port of a key valve.
Seventh, a pressure sensor for measuring the pressure to the receiving system is provided in the solution supply flow path, and the pump is connected to the solution suction flow path through at least one pump unit and a valve of each liquid feeding section. Proposed a gradient liquid feeding device characterized by providing a pressure sensor for measuring the pressure in the unit and the suction flow path, and a pressure sensor for measuring the pressure in the flow path U communicating with the liquid feed section β and the key valve. To do.
Eighthly, there is proposed a gradient liquid feeding device characterized in that a sample introduction unit communicating with a key valve is provided in a supply channel between a pressure sensor provided in a supply channel of a receiving system and a key valve.
Ninth, a three-way joint is installed in the flow path between the key valve and each liquid feeding section, and the flow path is provided between the three-way joint and the switching valve provided in the suction flow path of each liquid feeding section. We propose a gradient liquid feeding device characterized in that the is formed in communication.
According to the present invention, in liquid chromatographic analysis, solution A, etc., and so-called gradient liquid delivery in which the composition of the mobile phase is changed, basically, the initial solution A as the mobile phase Inflow to the second solution B liquid side can be prevented. In particular, the mutual entry of the liquid at the boundary surface between the two liquids can be minimized by the configuration of the key valve and the solution mixing section that eliminates the space where the initial solution A liquid can enter the B liquid side.
In addition, this makes it possible to minimize the delay in the start of the gradient, minimize the steep rise of the gradient due to the rapid increase in the ratio of the B solution, and bring the mixing ratio of the solution close to the set value. . And it can prevent that the peak which should not be separated and should be detected near the start of the gradient setting due to the rapid rise of the B liquid is lost.
The present invention will be described below with reference to examples shown in the drawings.

First, the liquid feeding part α for feeding the liquid A (initial solution) is connected to the valve port I, and the liquid feeding part β for feeding the liquid B (second solution) is connected to the port V. The
Liquid A (initial solution): Liquid B (second solution) = 100%: The operation of the valve and the liquid feeding unit in the gradient analysis starting from 0% is performed as follows.
(1) In the state of the
1) The liquid A sent out from the liquid sending part α enters the
2) Although the key valve ports IV and V communicate with each other in the flow path from the liquid feeding section β, the flow path for the B liquid is blocked because the port IV is closed. In this state, B liquid is pressurized so as to be the same pressure as the system pressure as preparation for sending B liquid (preliminary pressurization of B liquid) (indicated by a striped arrow in the figure).
(2) At the same time as switching to

(3) Thereafter, both liquid A and liquid B are fed according to the gradient program.
The
This rotation is switched from
In the same relationship, even in the flow path between the ports VI and I at the valve position 1 (between the ports I and II at the valve position 2), only the liquid A flows even when switching occurs.
Thus, since only the same solution passes through the flow path of the valve, it is possible to prevent different solutions remaining in the flow path from contacting each other in the valve flow path.
In other words, by using this valve, the contact between the A liquid and the B liquid is made only when the valve is switched. Phenomena such as mixing by substitution and diffusion do not occur.
That is, the contact boundary surface between the liquid A and the liquid B is generated only when the valve is switched and the liquid B is also fed. At this stage, there is a possibility that mutual substitution of liquids may occur, but both liquid A and liquid B are being sent out, and by reducing the pipe capacity until reaching the solution mixing part Q which is the merging part Mixing and replacement can only occur for a short time.
As a result, a multi-stage gradient curve as shown in FIG. 2B cannot be obtained, and a linear gradient curve as shown in FIG. 1B can be realized. When this structure is used, the influence on the gradient curve is that the valve position is switched to 2, the liquid B starts to be fed, and the time it takes for the liquid to reach the solution mixing part Q This is a delay in the start time (rise) of the gent. However, since the delay is due to the pipe to the solution mixing portion Q of the solution mixing tool, the delay can be reduced by reducing the capacity of the pipe.
