Abstract
Description
The present invention relates to a thermochemical inspection device that detects heat of reaction between a reaction reagent and a detection reagent with a thermal sensor, and relates to an analyzer that can arbitrarily replace a reacting portion.
We also get various tests when we get sick or in regular health checks. Many of them include qualitative and quantitative detection of substances in living bodies. For example, there are detection of a virus invading a living body, detection of an enzyme that is abnormally increased in the living body, or detection of carbohydrates, lipids, and the like that are increasing in the living body. Conventionally, since these examinations have been performed by large-scale devices provided in hospitals, it takes time to detect and evaluate the results, and therefore, a simpler and less time-consuming method has been demanded.
Thus, the immunochromatography method was considered (for example, see Patent Document 1). FIG. 9 shows the simple configuration. The structure is composed of a
When a sample sample containing the antigen to be measured is applied to the
In addition, although the unreacted sample moves downstream as it is, the colored
In this way, it has become possible to easily test in-vivo substances by using the conventional immunochromatography method. However, there are several problems with the conventional method. First, the color change of the sample sample combined with the chromogenic substance is judged, but since it is judged by the human eye, there are differences between individuals, differences in experience, oversight, etc., and clear judgment itself Is difficult. In particular, it is sufficient that a large amount of antigen is contained in the sample, but as the amount decreases, there is often no clear color change. Alternatively, since there is a coloring substance that moves without causing an antigen-antibody reaction, the noise component cannot be determined by color change.
These are problematic because, as can be seen from the mechanism that indicates that the sample has reached the end point reliably in the conventional method, it is often unclear whether it is caused by chemical bonding with the sample. ing. In order to clarify this judgment, there is an attempt to incorporate it into an optical inspection apparatus, but if this is done, the apparatus will still be complicated.
Furthermore, in the conventional method, even if the presence of the target substance can be determined, the amount thereof cannot be determined. In other words, for example, when a virus is infected, the initial stage is difficult to understand, and if it increases to some extent, it can be determined that it is present, but it is difficult to determine the progress of whether it is increasing further or decreasing toward ease. .
Accordingly, an object of the present invention is to provide an inspection apparatus capable of detecting a biological material by a simple operation, and a clear result that does not include human error in the detection judgment is obtained. Furthermore, it is providing the thermochemical test | inspection apparatus which can also quantify a biological substance.
In order to achieve the above object, the following means are employed in the structure of the thermochemical inspection apparatus and the structure of the reaction chip for the thermochemical inspection apparatus of the present invention.
Reaction using a thermal sensor using a thermo module, a temperature stabilizing member in contact with the lower surface of the thermal sensor, a protective cover containing the thermal sensor, a flow path through which the liquid passes, and a detection reagent in the middle of the flow path And a reaction chip that is in contact with the upper part of the thermal sensor in the vicinity of the reaction part.
Moreover, the reaction chip is good if the chemical solution supply part is exposed to the outside from the protective cover, and can be removed from the heat sensor, and more preferably exchangeable.
Further, it is effective that the temperature stabilizing member is a combination of a heat sink and a Peltier element, or has two heat sensors, and only one of the heat sensors is in contact with the reaction portion of the reaction chip.
In addition, the reaction chip for a thermochemical test apparatus has two thin plates that are overlapped while forming a slight gap serving as a flow path, a junction between the two thin plates, and one end and the other end of the flow path. Is provided with a chemical solution supply section, a chemical solution discharge section, and a reaction section in which a detection reagent is arranged in the middle of the flow path, or a flow path formed of a film-like water absorbing member and one of the flow paths And the other end is formed with a chemical supply part and a chemical discharge part, and has a reaction part containing a detection reagent in the middle of the chemical supply part and chemical discharge part of the flow path.
More preferably, a preliminary reaction part is provided between the reaction part and the chemical solution supply part of the reaction chip, or at least two chemical solution discharge parts are provided, or a protective member is provided on at least one surface of the water absorbing member. .
Furthermore, it is effective if the liquid moves in the flow path by utilizing capillary action.
Since the thermochemical inspection apparatus of the present invention can directly see the thermal reaction of the sample specimen in the reaction chip, the operation is very simple and rapid inspection is possible.
In addition, since it has a very simple structure consisting of a heat sensor, a heat retention device, and a test chip, it can be made very small compared to a conventional large-scale analyzer, such as a table in a hospital examination room. Can be used at the examination site.
Since the reaction chip also has a simple structure with only a flow path and a reactant, it can be easily made at low cost and can be used in a disposable manner. This is by using a water absorbing member as a flow path,
Further advantageous. In addition, since various chip configurations with different reactants are possible, various inspections can be performed with the same inspection apparatus.
Furthermore, since the reaction chip uses capillary action to move the liquid, it is not necessary to add a complicated liquid feeding system to the apparatus.
In addition, since the thermochemical test apparatus of the present invention finally determines the thermal reaction occurring in the reaction chip by converting it into an electrical signal, it is clear without being bound by personal subjectivity like a conventional immunochromatograph. Judgment is possible. The electrical numerical value can be measured not only qualitatively but also quantitatively.
Furthermore, by using two thermal sensors and making a thermochemical inspection apparatus capable of detecting the difference between the reaction heat and the environmental temperature change, it is possible to perform a further highly sensitive measurement.
From the above, the thermochemical test apparatus of the present invention will be used for rapid immunodiagnosis and virus diagnosis in hospitals and personal clinics in the future, or as a simple test apparatus in test institutions, and further for infectious diseases at home. Various uses such as diagnosis can be expected. Furthermore, it is expected to be applied outside hospitals, such as environmental analysis and in-situ inspection of environmental pollutants.
[First embodiment]
Hereinafter, an optimal embodiment of a thermochemical inspection apparatus of the present invention will be described with reference to the drawings. First, FIG. 1 shows a sectional view of a first example in the thermochemical inspection apparatus of the present invention.
As shown in FIG. 1, a
When a temperature difference occurs in the vertical direction of the
The Peltier
Further, in the thermochemical inspection apparatus of the present invention, a
A
Further, both ends of the
And as shown in FIG. 3, the
Although the description returns to FIG. 1, the
The
Now, an inspection method using a thermochemical inspection apparatus will be described. First, a
As described above, a slight amount of thermally conductive grease is applied in advance to the upper surface of the
Subsequently, the
A current is supplied to the
After the
The heat is transmitted to the
The above is a method for detecting a biological substance using an enzyme reaction, but detection using an antigen-antibody reaction, for example, is also possible. In this case, the sample sample may be blood, saliva, urine, etc., but what is immobilized on the
However, the antigen-antibody reaction generally has a smaller unit calorific value than the enzyme reaction. Therefore, a certain amount of amplification is required when the sample concentration is very small. The
The
The sample solution is allowed to permeate from the chemical
By using the
In the above description, the reaction heat of an enzyme reaction or an antigen-antibody reaction has been described as an example. However, other immune actions such as hapten-antibody interaction, and lectin, sugar and other active substances having biological affinity. And receptors are also available. However, since the thermal change is further reduced in the case of an affinity interaction, an amplification reaction is often required.
[Second Embodiment]
Next, the
Since the structure of the
A characteristic of the
The
An inspection method using the second thermochemical inspection apparatus will be briefly described. The process is the same as described above until the
However, since the
As described above, in the second thermochemical inspection apparatus, it is possible to further reduce the influence of the external temperature, and therefore it is possible to perform an inspection in which only a slight reaction heat is obtained.
In the
[Third embodiment]
Subsequently, a third example of the

Further, a
In the
As shown in FIG. 1, the
The
And the
As described above, the
