本出願は、2002年10月1日出願の米国仮特許出願第60/415,319号に基づきかつ優先権主張をしており、引用によりその全体を本明細書に包含させる。
本試験は、米国国立衛生研究所(NIH)から、助成金CA81512号により援助された。したがって、米国政府は本発明の主題にある権限を有する。
本発明の主題は、一般に、低酸素細胞中で、制限増殖コンピテント(conditionally replication competent)アデノウイルスベクターを増殖させる方法に関する。より特には、方法は、低酸素細胞、例えば腫瘍中の低酸素細胞を、低酸素細胞中で複製するアデノウイルスベクターのような制限増殖コンピテントアデノウイルスベクターで感染させ、細胞を殺すことに関する。
医学的研究および技術の著しい進歩にもかかわらず、癌は米国および世界中で死亡の主な原因の一つのままである。米国だけで100万人を超える癌の新症例が報告され、50万人以上がこの国で毎年癌により死亡している。
本発明の主題は、最小プロモーターおよび低酸素反応エレメント(HRE)を含む転写制御エレメント(TRE)の転写制御下にアデノウイルス遺伝子を含むアデノウイルスベクターである。一つの実施態様において、アデノウイルス遺伝子はE1A遺伝子、E1B遺伝子、E2A遺伝子、E2B遺伝子およびE4遺伝子からなる群から選択される。一つの実施態様において、アデノウイルスベクターは、転写制御エレメント(TRE)の転写制御下に第2のアデノウイルス遺伝子を含む。一つの実施態様において、最小プロモーターは、サイトメガロウイルス(CMV)最小プロモーター、ヒトβ−アクチン最小プロモーター、ヒトEF2最小プロモーターおよびアデノウイルスE1B最小プロモーターからなる群から選択される。他の実施態様において、CMV最小プロモーターは配列番号1を含む。一つの実施態様において、低酸素反応エレメント(HRE)はヒト血管内皮細胞増殖因子(VEGF)遺伝子に由来する。他の実施態様において、HREは配列番号2の5つのタンデムコピーを含む。一つの実施態様において、アデノウイルスベクターはさらにトランス遺伝子を含む。一つの例において、トランス遺伝子は第2のアデノウイルス遺伝子を含む。他の例において、トランス遺伝子は、免疫刺激分子をコードする核酸を含む。さらに別の例において、トランス遺伝子は自殺遺伝子を含む。
本発明の主題は、一般に制限増殖コンピテントアデノウイルスベクターを、転写因子低酸素誘引因子1(HIF−1)を発現する細胞内で増殖させる方法に関する。一つの実施態様において、方法は、低酸素細胞、例えば腫瘍中の低酸素細胞を、低酸素細胞中で複製するアデノウイルスベクターのような制限増殖コンピテントアデノウイルスベクターで感染させ、細胞を殺すことを含む。
正常組織酸素張力より低い状態である低酸素は、近年、癌を含むヒト疾患と関連している。それは、腫瘍増殖および発育に主に関連する。特に、低酸素は変異の促進および悪性腫瘍細胞の選択に重要な役割を担うことが判明した。それはまた腫瘍血管形成の促進に関与する。
以下の用語は当業者には十分理解されていると考えるが、以下の定義を本発明の主題の説明を容易にするために示す。
本発明の主題にしたがって用いる核酸分子は、配列番号1および2の任意の一つ;配列番号1および2の任意の一つの配列と実質的に同一な配列;その保存的変異体、その部分配列および伸長した配列、相補的DNA分子、および対応するRNA分子の単離核酸分子の任意の一つを含むが、これらに限定されない。本発明の主題は、また記載の核酸配列を含む遺伝子、cDNA、キメラ遺伝子およびベクターも包含する。
本発明の主題と関連して使用するポリペプチドは、下記に定義のような治療ポリペプチド;下記に定義の治療ポリペプチドと実質的に同一のポリペプチド;下記に定義の治療ポリペプチドのポリペプチドフラグメント(一つの実施態様において生物学的に機能的なフラグメント)、下記に定義の治療ポリペプチドを含む融合タンパク質、その生物学的に機能性のアナログならびに、抗体、特に下記に定義の治療ポリペプチドを認識する抗体と交差反応するポリペプチドを含むが、これらに限定されない。本発明の主題と関連して使用するポリペプチドは、単離ポリペプチド、ポリペプチドフラグメント、記載のアミノ酸配列を含む融合タンパク質、生物学的に機能性のアナログ、および、抗体、特に記載のポリペプチドを認識する抗体と交差反応性のポリペプチドを含むが、これらに限定されない。
ヌクレオチドまたはポリペプチド配列に関連する“同一”またはその割合“同一性”なる用語は、比較し、最大一致に対して整列させた場合、本明細書に記載の配列アルゴリズムの一つを使用してまたは目視により測定して、同じまたは同じであるアミノ酸残基またはヌクレオチドの特異的な割合を有する2個またはそれ以上の配列または部分配列を意味する。
一つの実施態様において、本発明の主題のアデノウイルスベクターは、制限増殖コンピテントである。すなわち、それらは誘導プロモーターの転写制御下に、複製に必要な1個またはそれ以上の機能遺伝子を含む。これはインビボで非制御の複製を阻止し、ウイルス感染の望ましくない副作用を減少させる。複製コンピテント自己制御式または自己破壊式ウイルスベクターも、複製欠損ウイルスベクターが使用され得るように、使用できる。
本発明の主題の方法は、細胞中での複製のためにアデノウイルスベクターを使用し、それにより細胞の融解をもたらす。アデノウイルスベクターを含む細胞をより効率的に殺すために、本発明の主題はまたトランス遺伝子を含むアデノウイルスベクターも提供する。現在提供している対象にしたがい、トランス遺伝子は、腫瘍サプレッサー遺伝子、アポトーシス誘導遺伝子、抗脈管形成遺伝子、自殺プロドラッグ変換酵素遺伝子、細菌毒素遺伝子、アンチセンス遺伝子、腫瘍サプレッサー遺伝子、免疫刺激性遺伝子またはこれらの組み合わせを含む、治療遺伝子を含むことができる。
本発明の主題の治療法は、腫瘍中の低酸素細胞とアデノウイルスベクターを接触させ、それによりベクターが細胞内に入り、腫瘍増殖を阻害することを含む。例えば、記載のアデノウイルスベクターは、***の原発性および転移性固形腫瘍および癌腫;結腸;直腸;肺;中咽頭;下咽頭;食道;胃;膵臓;肝臓;胆嚢;胆管;小腸;腎臓、膀胱および尿道内皮を含む尿管;子宮頸、子宮、卵巣、絨毛癌および妊娠性絨毛性疾患を含む、女性生殖管;前立腺、精嚢、精巣および生殖細胞腫瘍を含む、男性生殖管;甲状腺、副腎および下垂体を含む内分泌腺;血管腫、黒色腫、骨またな軟骨組織起源の肉腫およびカポジ肉腫を含む皮膚;星状細胞腫、神経膠腫、神経膠芽細胞腫、網膜芽腫、神経腫、神経芽腫、神経鞘腫および髄膜腫を含む脳、神経、眼および髄膜の腫瘍;白血病のような造血転移由来であり、クロローマ、形質細胞腫、プラークならびに、菌状息肉腫および皮膚T細胞リンパ腫/白血病由来の腫瘍を含む、固形腫瘍;ホジキン病および非ホジキン病リンパ腫を含むリンパ腫の治療に有用であり得る。
ほとんどの実施態様において本発明の主題で処置する対象は、ヒト対象が望まれるが、本発明の主題の原則は、本発明の主題が、非脊椎動物および哺乳動物を含むすべての脊椎動物に対して有効であり、これを用語“対象”に包含することを意図することは理解されるべきである。さらに、哺乳動物は、癌の処置または予防が望まれる任意の哺乳動物種、特に農業用または飼育用哺乳動物種を含むことは理解される。
本発明の主題のアデノウイルスベクターは、実施態様において、薬学的に許容される担体を含む医薬組成物を構成する。任意の適当な医薬製剤を、対象に投与するためにアデノウイルスベクターを調製するために使用できる。
本発明の主題のアデノウイルスベクターの適当な投与法は、静脈内または腫瘍内注射を含むが、これらに限定されない。あるいは、アデノウイルスベクターを任意の他の方法で、例えばアデノウイルスベクターを含む組成物を肺経路内噴霧することにより、処置が必要な部位に付着させることができる。本発明の主題を投与する特定の形態は、処置する細胞の分布および発生量、用いるベクター、ベクターの付加的組織または細胞ターゲティング特性および投与部位からベクターが代謝または除去する機構を含む、種々の因子に依存する。例えば、相対的に表面の腫瘍は腫瘍内注射できる。対照的に、内部の腫瘍は、静脈内注射により処置できる。
本発明の主題のアデノウイルスベクター組成物の有効な投与量を、それを必要とする対象に投与する。“治療的有効量”は、測定可能な反応(例えば、処置している対象における細胞融解反応)を産生するのに有効な治療組成物の量である。一つの実施態様において、腫瘍増殖を阻害する活性を測定する。本発明の主題の医薬組成物中の活性成分の実際の用量レベルは、特定の対象のための所望の治療的反応を達成するのに十分な活性化合物(複数もある)の量を投与するように、変化し得る。選択する投与レベルは、治療組成物の活性、投与経路、他の薬剤または処置との組み合わせ、処置する状態の重症度および処置する対象の状態および投薬歴に依存する。しかしながら、所望の治療効果を達成するのに必要なものより低いレベルで化合物の投与を開始し、所望の効果が達成するまで徐々に投与量を増加させることは、当業者の範囲内である。
以下の実施例は、本発明の主題の形態を説明するために包含させる。以下の実施例中のある態様は、現在の共発明者により、本発明の主題の実施に十分働くように発見されまたは意図された技術および方法の観点を記載している。これらの実施例は、共発明者の標準研究業務を説明する。本明細書の記載および当分野の一般的技術レベルに照らして、当業者は、以下の実施例が例示のみを意図し、多くの変化、修飾および置換を本発明の範囲から逸脱することなく用いることができることを認め得る。
低酸素に暴露した細胞におけるEGFPのインビトロ発現
VEGFプロモーター中のHIF−1結合エレメントに基づいたプロモーターを構築した。低酸素反応性プロモーター(HRP)は、サイトメガロウイルス(CMV)由来の最小プロモーターに連結した、ヒトVEGFプロモーターのHREの5つのタンデムコピーを含んだ。このプロモーターの活性を試験するために、HRPが緑色蛍光タンパク質(EGFP)遺伝子の発現を制御する、図1に記載のプラスミドを構築した。HRP−EGFP構築物を使用して、安定な二つの腫瘍細胞系:HCT116(ヒト結腸癌細胞系);および4T1(マウス***腺癌)を確立した。低酸素条件(0.5から1.5%の酸素張力)に暴露した、安定に形質導入されたサブラインからの細胞は、インキュベーション24時間後、EGFPを強く発現した。
皮下腫瘍におけるHRP−駆動EGFP発現
腫瘍を、105−106細胞をマウス皮下に注射することにより確立した。注射した細胞は、構築物(HRP−EGFP;図1参照)を安定に形質導入され、EGFP遺伝子の発現を制御する低酸素反応性プロモーターを含む、4T1細胞であった。腫瘍を直径約5−8mmまで増殖させた。腫瘍を切除し、マウスを殺す直前、マウスにピモニダゾールを腹腔内に注射した。ピモニダゾール染色は、腫瘍内の低酸素領域の同定のための標準法である(Raleigh et al., 1998)。腫瘍の凍結切片を次いで抗ピモニダゾール抗体で染色し、蛍光顕微鏡下で観察した。同じ切片のEGFP発現パターンも観察した。EGFP発現とピモニダゾール染色の一致したパターンが各切片で観察され、低酸素腫瘍領域に伝達するためのHRP−EGFPレポーターの適切性が確認された。
制限増殖コンピテントアデノウイルスベクターのインビトロ複製
アデノウイルスE1A遺伝子をHRPプロモーターの制御下に含むアデノウイルスベクターを構築した(AdHRP−E1A−dsRed2;図2参照)。赤色蛍光タンパク質(dsRed2)をコードするレポーター遺伝子をベクター内に操作して入れ、ウイルス感染と複製の追跡を容易にした。このベクターを次いでHCT116ヒト結腸癌細胞系で試験した。低酸素はこのウイルスベクターの活性化を導いた。蛍光顕微鏡測定は、低酸素に暴露した細胞において、有意に多くのウイルスが複製および感染したことを証明した。フローサイトメトリーで測定した場合、dsRed2発現の差は少なくとも100倍であり、これはプラーク形成アッセイで確認された。E1Aタンパク質のウエスタンブロット分析は、E1Aが低酸素条件に付された細胞でのみ著しいレベルで発現されることを示した。
制限増殖コンピテントベクターのインビボ複製
HRP−EGFP構築物(図1参照)を形質導入されたHCT116細胞を使用して、ヌードマウスに腫瘍を確立した。これらの腫瘍を担持するマウスを、次いで、赤色蛍光タンパク質を運搬するアデノウイルスベクター(AdHRP−E1A−dsRed2;図2参照)で感染させた。腫瘍細胞はEGFPタンパク質をHRPの制御下に発現し、一方赤色蛍光マーカーをコードするウイルスベクターは、ウイルス複製と腫瘍低酸素の相対的発現パターンの比較を可能にした。
インビボ腫瘍増殖阻害
HCT116(ヒト結腸癌)細胞を、ヌードマウスに3.0×106細胞/マウスで皮下注射した。腫瘍が直径5−10mmに到達したとき、ウイルスベクターを腫瘍内注射した。制御グループ(図6B、黒四角)はAdCMV−dsRed2(図5)を注射し、一方、処置グループ(図6B、黒三角)はAdHRP−E1A−TNF−α(図6A)を注射し、これは、HRPに稼働し得るように連結したE1A遺伝子を含み、さらに構造性に発現するTNF−α遺伝子を含む制限増殖コンピテントアデノウイルスベクターであった。腫瘍あたり適当なウイルスの2.0×109pfuを腫瘍内注射した。腫瘍容量を2−3日毎に測定した。相対的容量を、0日目(すなわち、ベクター注射の直前の時点)の各腫瘍の容量を1.0と設定することにより計算した。図6Bに示されるように、制限増殖コンピテントアデノウイルスベクターを注射された腫瘍の増殖は、コントロールと比較してかなり遅い。
制限増殖コンピテントアデノウイルスベクター存在下でのE1−欠損AdCMV−EGFPの複製
制限増殖コンピテントアデノウイルスベクターが、複製欠損アデノウイルスベクターの複製を支持できる能力を試験した。構造性に活性なEGFP遺伝子をコードする複製欠損アデノウイルスベクター、AdCMV−EGFP(図5参照)を構築した。このベクターにおいて、E1およびE3遺伝子が欠損し、EGFP遺伝子(構造性に活性なCMVプロモーターの制御下)がウイルスのE1領域に挿入されている。構造性に活性なdsRedタンパク質をコードし、上記された制限増殖コンピテントアデノウイルスベクターAdHRP−E1A−dsRed2(図2参照)を使用した。

Claims (36)

- さらに第2のアデノウイルス遺伝子をTREの転写制御下に含む、請求項1記載のアデノウイルスベクター。
- 最小プロモーターがサイトメガロウイルス(CMV)最小プロモーター、ヒトβ−アクチン最小プロモーター、ヒトEF2最小プロモーターおよびアデノウイルスE1B最小プロモーターからなる群から選択される、請求項1記載のアデノウイルスベクター。
- CMV最小プロモーターが配列番号1を含む、請求項4記載のアデノウイルスベクター。
- HREがヒト血管内皮細胞増殖因子(VEGF)プロモーターに由来する、請求項1記載のアデノウイルスベクター。

- さらにトランス遺伝子を含む、請求項1記載のアデノウイルスベクター。
- トランス遺伝子が第2のアデノウイルス遺伝子である、請求項9記載のアデノウイルスベクター。
- トランス遺伝子が免疫刺激分子をコードする、請求項9記載のアデノウイルスベクター。
- 免疫刺激分子がIL2およびIL12からなる群から選択される、請求項11記載のアデノウイルスベクター。
- トランス遺伝子が自殺遺伝子である、請求項9記載のアデノウイルスベクター。
- 自殺遺伝子がTNF−α遺伝子、Trail遺伝子、Bax遺伝子、HSV−tk遺伝子、シトシンデアミナーゼ遺伝子、p450遺伝子、およびジフテリア毒素遺伝子、s−Flt1遺伝子、およびex−Flk1遺伝子からなる群から選択される、請求項13記載のアデノウイルスベクター。
- 請求項1記載のアデノウイルスベクターを含む、組成物。
- さらに薬学的に許容される担体を含む、請求項15記載の組成物。
- 腫瘍増殖を抑制する方法であり、腫瘍中の低酸素細胞と請求項1記載のアデノウイルスベクターを接触させ、それによりベクターが細胞内に入り、腫瘍増殖を阻害することを含む、方法。
- 接触が、アデノウイルスベクターの腫瘍内注射を介した投与の結果である、請求項17記載の方法。
- 接触が、アデノウイルスベクターの静脈内注射による投与の結果である、請求項17記載の方法。
- さらに腫瘍を治療的有効量の第2の処置に暴露することを含み、該第2の処置が電離放射線照射、化学療法および光線力学療法からなる群から選択される、請求項17記載の方法。

- 標的細胞に選択的細胞毒性を与える方法であり、HREが機能できる細胞と請求項1記載のアデノウイルスを接触させ、それによりアデノウイルスベクターが細胞内に入ることを含む、方法。
- HIF−1を発現する標的組織中でアデノウイルスを選択的に増殖させる方法であり、標的組織と請求項1記載のアデノウイルスを接触させ、それによりアデノウイルスが少なくとも1.0×107pfu/mlの力価まで増殖することを含む、方法。
- 標的組織の増殖を阻害する方法であり:
(a)標的組織中の低酸素細胞と第1のアデノウイルスベクターを接触させ、それにより第1のアデノウイルスベクターが細胞内に入り;そして
(b)低酸素細胞と複製欠損アデノウイルスベクターを接触させ、それにより複製欠損アデノウイルスベクターが細胞内に入ることを含む、方法。 - 標的組織が腫瘍である、請求項24記載の方法。
- 第1のアデノウイルスベクターが、HREを含むTREの転写制御下にアデノウイルス遺伝子を含む、請求項24記載の方法。
- 複製欠損アデノウイルスベクターが第2の遺伝子を含む、請求項24記載の方法。
- 複製欠損アデノウイルスベクターが構造性プロモーターの転写制御下に第2の遺伝子を含む、請求項27記載の方法。
- 複製欠損アデノウイルスベクターがHREを含むTREの転写制御下に第2の遺伝子を含む、請求項27記載の方法。
- (a)第1のアデノウイルスベクターが、HREを含むTREの転写制御下に少なくとも二つの必須アデノウイルス遺伝子を含み;そして
(b)複製欠損アデノウイルスベクターが、第1のアデノウイルスベクター中のHREを含むTREの転写制御下に必須アデノウイルス遺伝子の少なくとも二つが欠損している、請求項24記載の方法。 - 二つの必須アデノウイルス遺伝子が、各々E1A遺伝子、E1B遺伝子、E2A遺伝子、E2B遺伝子およびE4遺伝子からなる群から選択される、請求項30記載の方法。
- 第2の遺伝子が自殺遺伝子である、請求項27記載の方法。
- 自殺遺伝子が、TNF−α遺伝子、Trail遺伝子、Bax遺伝子、HSV−tk遺伝子、シトシンデアミナーゼ遺伝子、p450遺伝子およびジフテリア毒素遺伝子、s−Flt1遺伝子およびex−Flk1遺伝子からなる群から選択される、請求項32記載の方法。
- 第2の遺伝子が免疫刺激分子をコードする、請求項27記載の方法。
- 免疫刺激分子がIL2およびIL12からなる群から選択される、請求項32記載の方法。
- さらに標的組織を治療的有効量の第2の処置に暴露することを含み、該第2の処置が電離放射線照射、化学療法および光線力学療法からなる群から選択される、請求項24記載の方法。
