本発明は、新しいポリシストロニック(多シストロン性)発現系および本発現系を用いて真核細胞において抗体を作製するための方法に関する。より詳細には、本発明は、真核生物のポリシストロニック発現系に関し、この系では、抗体の重鎖遺伝子および軽鎖遺伝子が、同じプロモーターから転写され、そして好ましくは、抗体の重鎖遺伝子および軽鎖遺伝子が、1つ以上の内部のリボソーム進入部位(IRES)によって分離される。
ポリシストロニック発現ベクターを利用する組換え宿主細胞における選択した遺伝子の発現の方法は、周知である。歴史的には、転写領域の5’末端側での所望される産生遺伝子配列および3’側選択マーカー遺伝子配列を組み込むポリシストロニック発現ベクターが、利用されてきた。このようなベクターは、ポリシストロニックmRNAの5’末端側での所望される遺伝子配列を優先的に翻訳する間に、3’側選択可能遺伝子の非効率的な翻訳を示す。所望される遺伝子産物の高いレベルを発現する組換え宿主細胞は、選択可能な培地中で最初のトランスフェクト体を培養する工程を包含する1つの工程の方法を介して得られる。このようなベクターの欠点は、予測不可能な影響であり、上流のリーディングフレームが、選択マーカー配列の翻訳に対して影響を及ぼし得る。Kaufman,Meth.Enzymol.,185:487(1990)を参照のこと。
本発明は、真核生物宿主細胞中でのポリシストロン性発現系を介した、適切なレベルの発現での、機能的(抗原結合)抗体の発現に関する。
好ましい実施形態の詳細な説明を提供する前に、以下の定義を提供する。
適度なレベルの抗体産生を生じるかどうかは一般に予測不可能である。この予測不可能性は、例えば、多シストロン性複合体の第2の所望の遺伝子が第1の遺伝子と比べてかなり低いレベルで発現され得るために生じる。従って、多シストロン性ベクターの好ましい実施形態は、抗体重鎖発現に対する抗体軽鎖発現の割合を約10:1〜約1:1の範囲内で提供すべきである。より好ましくは、重鎖遺伝子発現に対する軽鎖遺伝子発現の割合は、約3:1〜約1.5:1である。
ポリシストロンDNA発現ベクターを、本発明に従って構築した。これは、ヒト化抗TAG72抗体の重鎖および軽鎖を発現し、HuCC49と命名される。
図1に示される発現構築物は、ネオマイシンホスホトランスフェラーゼ遺伝子をコードする配列の発現を駆動するように構築物中に配置された、マウスβ−グロブリン主要プロモーター(β)を含む。ネオマイシンホスホトランスフェラーゼ遺伝子(これは、細菌起源である)は、2つのエキソン(N1およびN2)からなり、そして人工イントロンを含む。発現ベクター系における遺伝子産物発現を増強するように設計された選択マーカーの、イントロン挿入の方法は、例えば、米国特許第5,648,267号、米国特許第5,733,779号、米国特許第6,017,733号、および米国特許第6,159,730号(全て、Reffに対する)に開示されている。これらの特許の各々の全内容は、本明細書中に参考として援用される。選択される抗体は、固体腫瘍を処置するための適切性により当該分野において公知である。高レベルの組織浸入が、固体腫瘍細胞に影響を与えるために必要とされ、従って、この抗体は、Fabフラグメントを用いて見られるより比較的長い、血清半減期を有するべきである。この実施例において使用される、ドメインが欠失された構築物は、ヒトγ1重鎖定常ドメインを含み、ここで、ヒンジ領域に続くCH2の最初の9個のアミノ酸を除く、CH2ドメインの大部分が欠失されている。
SV40エンハンサーを利用して1つの重鎖を発現する発現ベクターを含むプラスミドを、一過性トランスフェクションプロトコルと、高い発現能力を有する安定なトランスフェクトされた細胞株を得るために設計されたプロトコルとの両方を使用して、CHO細胞にトランスフェクトした。これらのトランスフェクションを、独立した軽鎖および重鎖のカセット(CMV−Light−BGH)(CMV−Heavy−BGH)を含む従来の発現構築物と並行して実施した。実験結果は、起こった発現レベルに明らかな差異がないことを示し、このことは、非効率なIRES機能の結果としての重鎖翻訳の減少が検出されなかったことを意味する。従って、本発明のポリシストン構築物の実施形態は、第一のシストロンおよび引き続くシストロンに含まれる、他の目的の遺伝子配列を、同様に、満足な量で発現すると予測される。実験は、本明細書中以下に議論されるように、これらの構築物が、二シストロン構築物における下流重鎖遺伝子を、従来の非ポリシストン発現系を使用して得られるレベルと等価なレベルで発現することを示す。
(A.G418耐性細胞株の生成)
この実施例における、ポリシストロン発現構築物は、ヒトTAG 72タンパク質に対する特異性を有する、改変されたヒト化抗体をコードする。この抗体は、ヒト化マウス可変ドメイン(軽鎖と重鎖との両方)ヒトκ軽鎖定常ドメイン、およびヒトγ1重鎖定常領域から構成され、CH2ドメインが特異的に欠失している。この発現構築物は、「HuCC49 CH2 Linker in N5KIRESG1DelCH2」と称され、これのマップは、図3に示されており、そしてそれの配列は、図6に示されている。HuCC49 CH2 Linker in N5KIRESG1DelCH2を、CHO−DG44親細胞株にトランスフェクトした。トランスフェクトを、96ウェルプレートあたり4×106個のCHO細胞あたり0.5μg、1.0μg、2.0μg、または4.0μgの線状化DNAを使用して実施した。安定にトランスフェクトされる細胞の優勢な選択を、ネオマイシン耐性マーカーをコードする発現構築物およびG418を含有する培地(CHO−S−SFM II + HT)を介して達成した。
これらの細胞株の生成と並行して、同じ抗体のNEOSPLA発現構築物を構築し、そしてトランスフェクトした。HuCC49 CH2 Linker in N5KG1DelCH2(図3に示される)は、免疫グロブリンの軽鎖遺伝子および重鎖遺伝子を、独立した転写カセット上にコードし、各々の翻訳は、5’CAP翻訳開始複合体によって媒介される。この発現構築物のトランスフェクションによって単離した、特定のG418耐性細胞株を、14H9と命名した。14H9由来の培養上清およびポリシストンの単離された22B6の分析を実施して、分泌された軽鎖と重鎖との相対比を試験した。分析を、軽鎖または重鎖のいずれかに特異的な二次抗体で捕捉および検出することにより、ELISAによって実施した。二連サンプルの結果を、表1に示す。データは、免疫グロブリン標準と比較した、mg/Lとして表されている。比の欄において、測定された全軽鎖(κκ)を、測定された全構築抗体(γκ)と比較する。
NEOSPLAまたはポリシストロン発現系のいずれかによって産生された、種々の細胞株由来の培養上清の分析を、後に行って、分泌された軽鎖および重鎖の相対比を試験した。この分析を、軽鎖または重鎖のいずれかに特異的な二次抗体で捕捉および検出することにより、ELISAによって実施した。二連のサンプルの結果が、本明細書中に示され、そして免疫グロブリン標準と比較したmg/Lとして表される。

(A.ゲノム増幅を介する発現の増加)
NEOSPLA発現系およびポリシストロニック発現系の両方は、マウスDHFR遺伝子をコードする発現カセットを含む。発現に使用される親CHO−DG44細胞株は、DHFR酵素活性を完全に欠損しているので(二重欠失)、統合された標的遺伝子(マウスDHFR)の増幅は、メトトレキサート(MTX)を含む培地における増殖選択により可能である。この増幅の間、直接連結された免疫グロブリン遺伝子は、同時に増幅される。従って、上昇した量の免疫グロブリンを産生する細胞株を単離することが可能である。G418耐性細胞株の本発明者らの第1回目の選択を、5nMのMTX中で実施する。最も高いレベルの産生体(ELISAによって決定されるように)を同定し、次いで、漸増の濃度のMTX(50nM、次いで、500nM)での2つの引き続く回の増幅に供する。
(発現構築物の生成のための方法)
本発明に従うポリシストロニック発現構築物の生成についての一般的方法を、本明細書に記載する。この方法論において、最終発現構築物を、単一のクローニング工程における目的の遺伝子(すなわち、免疫部グロブリン軽鎖および重鎖)をコードするDNAフラグメントの挿入により生成する。DNAフラグメントは、従来の組換えDNA技術(オリゴ合成および連結、オーバーラップPCR、完全DNA合成、または以前に記載されたようなこれらの方法論の任意の組み合わせ)の使用によって生成され得る。
(ゲノム増幅を介する発現の増加)

Claims (35)
- 真核生物宿主細胞において機能的抗体を発現するためのポリシストロニックベクターであって、該ベクターは、5’から3’の方向で作動可能に連結された以下のエレメントを含むポリシストロニック転写系を含み:
(i)真核生物細胞において作動可能なプロモーター;
