動物
Ner nu/nu雌マウス(Taconic Farms,German Town,NY)をDLD−1およびNCI−H460腫瘍モデルを含むすべてのインビボ研究に使用した。雌CB−17SCIDマウス(Taconic Farms,German Town,NY)をMia−PaCa−2腫瘍モデルを含む研究のために使用した。マウスは、バイエルIACUC,およびヒト処置および研究室動物のケアのための州および連邦ガイドラインに従ってComparative Medicine Department,at Bayer Corporation,West Haven,CT内に収容され、維持された。マウスは随意に食物および水を与えられた。
すべての研究に化合物A(ロット9910071)を用いた。化合物Aは白色ないしアイボリーもしくは淡黄の範囲の色を有する乾燥粉末である。
CamptsarTM(ロット番号09FDYおよび27FMR)はファルマシア−アップジョンによって製造され、20mg/ml溶液として供給された。それは包装添付文書に指示されたように室温で貯蔵された。

DOX(ドキソルビシンHCl)はBedford Laboratories(ロット110033)によって製造され、凍結乾燥した赤/オレンジ粉末として供給された。それは4℃において貯蔵され、そして光から保護された。
ゲフィニチブ(ZD1839)、化学名(4−(3−クロロ−4−フロロアニリノ)−7−メトキシー6−(3−モルホリノプロポキシ)キナゾリン)はAlbany Medical Research(Syracuse,NY)によって合成された。2D1839は使用まで室温で暗所に貯蔵された。
Cremophr EL/エタノール(50:50)(Sigma Cremophor EL カタログNo.C−5135;500g,95%エチルアルコール)がストック溶液として調製され、アルミ箔で包まれ、そして室温で貯蔵された。化合物AはこのCremophr EL/エタノール(50:50)溶液中に最大投与量の4倍(4×)において処方された。この4×ストック溶液は3日毎に新しく調製された。最終投与溶液は、使用日にエンドトキシンスクリーニングした蒸留水(GIBCO,カタログNo.15230−147)で1×へ希釈し、そして投与直前渦混合された。低い投与量は1×溶液をCremophor EL/エタノール/H2 O(12.5:12.5:75)で希釈することによって調製された。CamptosarTMのためのビヒクルは0.9%食塩水であり、NavelbineTMのためのビヒクルはD5Wであった。すべてのビヒクルおよび化合物は室温で貯蔵され、そして箔で包まれた。
DLD−1ヒト結腸カルチノーマおよびMiaPa−2ヒト膵臓カルチノーマはAmerican Type Tissue Culture Repositoryから得た。MX−1ヒト乳腫瘍はNCI tumor Repositoryから得た。腫瘍は12ゲージ套管針を用いてわき腹に移植したS.C.断片(3×3mm)の段階インビボ通過として継持された。
NCI−H460およびA549ヒト非小細胞肺カルチノーマラインはAmerican Type Tissue Culture Repositoryから得た。NCI−H460細胞は、1%熱不活性化ウシ胎児血清(JRH Bioscience カタログNo.1195−065;500ml)、L−グルタミン2mM(GIBCOカタログNo.25030−8),10mM HEPESドッファー(GIBCOカタログNo.15630−080)およびペニシリン−ストレプトマイシン(GIBCOカタログNo.15140ー122;5ml/50ml DMEM)を補給したDMEM(GIBCOカタログNo.11995−65;500ml)を用いてインビトロで維持され、通過させられた。A549細胞は、10%熱不活性ウシ胎児血清(JRH Bioscience カタログNo.12106−500M)を補給したRPMI 1640媒地(GIBCOカタログNo.11875;1000ml)を用いてインビトロで維持され、通過させられた。すべての細胞は37℃で、そしてFisher Scientific 610 CO2 インキュベーター中5%CO2 において維持された。
雌マウスにインビボ通過からのMX−1又はMia−PaCa−2腫瘍断片を移植した。NCI−460およびA549についての研究は、トリプシン−EDTA(GIBCOカタログNo.25200−056)を2分間加え、次に細胞をペレットに遠心し、そしてHBSS(GIBCOカタログNo.14025−092)中最終細胞カウント3〜5×107 生存細胞/mlへ再懸濁することによってインビトロ培養物から細胞を収穫することによって開始された。細胞懸濁液の0.1mlを各マウスの右脇腹にS.C.注射した。すべての処置は実験のすべてのマウスが100ないし150mgの範囲のサイズの腫瘍を確立した時に開始された。マウスの一般的健康状態をモニターし、死亡率を毎日記録した。腫瘍寸法および体重を処置1日目からスタートして週2回記録した。動物はバイエルIACUCガイドラインに従って安楽死させた。20%以上の死亡率および/または20%正味体重ロスが有毒と考えられた。
腫瘍重量は式(1×W2 )/2を用いて計算された。ここで1およびW各測定において集められた大きいおよび小さい寸法を意味する。各実験において、評価の終点は対照グループにおける腫瘍のための中間時間が処置の期間中よりも少し大きいサイズに達するように選定された。抗腫瘍有効度は、腫瘍が長さおよび幅において測定限界(3mm)以下に縮小したと定義される完全退行(CR)、腫瘍がそれらの初期サイズの50%以上であるがしかし100%未満縮小したと定義される部分的退行(PR)、および腫瘍成長抑制%(%TGS)として測定された。TGSは式〔(T−C)/C〕×100によって計算される。ここでTおよびCは、その評価のための評価サイズに達する処置(T)および無処置(C)グループのそれぞれの中間腫瘍のための時間を表わす。
がんの処置のための化合物Aの臨床開発において予想された最も強力な組合わせ化学療法は、各自が現行の臨床実験を構成する例えばCamptosar,Gemzar,NavelbineまたはDOXのような細胞毒剤/細胞増殖抑制剤の間歇投与を含んだ全期間を通じて投与される化合物の毎日投与を含むであろう。これらの剤の潜在的相互作用を探求するため、同じ日に出発する各実験における両方の治療について、我々の前臨床モデルにおいてこの予想された臨床スケジュールに個々の剤のスケジュールを(化合物Aについて1日4回×9およびCamptosar,Gemzar,NavelbineまたはDOXについて4日4回×3)を重ねることによりモデルとした。組合わせ処置の代替スケジュールは、Iressaのような細胞増殖抑制剤の連続投与を含んだ全期間を通じ化合物Aの毎日投与であろう。これらの剤の潜在的相互作用を探求するため、前臨床モデルは個々の剤のスケジュール(化合物AおよびIressaの両方について1日4回×9または10)を重ねることによって確立された。これらのスケジュールは“併用療法”と名付けられた。各研究は10〜20動物の未処置対照グループと、1群10マウスの処理グループよりなっていた。

Camptosarは単独剤として最小体重ロスと死亡なしでよく耐えた。40mg/kg投与レベルは完全または部分的腫瘍退行なしで71%のTGSを生成した。
化合物Aは単独剤として80mg/kg/投与において有意な体重ロスおよび死亡なしを生成した。化合物Aは100%のTGSを示した。
Camptosarと化合物Aの組合わせに関連した体重ロスおよび死亡の増加はなかった。併用療法の抗腫療有効度は少なくともTGS229%を生ずる付加であった。これはPR3例に関連していた。
第2の研究は4日4回×3スケジュールにおいて120mg/kg/投与におけるi.p.投与したGemzarと、1日4回×9スケジュールにおいて40mg/kg/投与におけるp.o.投与された化合物を評価した。すべての処置は、すべての動物が小さいがしかし108mgサイズ平均のMiaPaCaヒト膵臓腫瘍キセノグラフトを確立する時の移植後7日目に開始された。対照腫瘍は平均倍増時間4.1日ですべての動物において次第に成長した。成長遅延パラメータを計算するために使用された評価終点は塊倍増2例までの時間であった。このサイズへ達する未処置対照グループのための中間時間は5.8日であった。
Gemzarは単独剤として最小体重ロスと死亡なしでよく耐えた。この投与レベルは完全または部分的腫瘍退行なしで154%のTGSを生じた。化合物Aも80mg/kg投与レベルにおいて有意な体重ロスおよび死亡なしで良く耐えた。化合物Aは112%のTGSを生じた。Gemzarと化合物Aの組合わせに関連した体重ロスおよび死亡の増加はなかった。Gemzar120mg/kgと化合物A40mg/kgの併用療法は少なくとも222%のTGSを生ずる付加であった。これはPR2例に関連していた。
第3の例は40mg/kg/投与において1日4回×9のスケジュールでのp.o.投与された化合物Aと、6.7mg/kg/投与において4日×3スケジュールでi.v.投与されたNavelbineの組合わせの効果を証明する。すべての処置は、すべての動物が小さいがしかし100mgサイズ平均のNCI−H460ヒト非小細胞肺腫瘍キセノグラフトを確立する時の移植後6日目に開始された。対照腫瘍は平均倍増時間3.1日ですべての動物において次第に成長した。成長遅延パラメータを計算するために使用された評価終点は塊倍増3例までの時間であった。このサイズへ達する未処置対照グループのための中間時間は7.4日であった。
Navelbineの6.7mg/kg投与レベルは単独剤として処置期間の間の平均19%の体重ロスを生ずる大よその最大耐薬投与であった。これは32%のTGSと関連していた。化合物Aは有意な体重ロスおよび死亡例なしで良く耐え、そして104%のTGSを生じた。これらの処置の組合わせは死亡例なしでそして14%の平均体重ロス(Navelbine単独によって生じたよりも小さい)で良く耐えた。この組合わせの抗腫瘍有効度は133%のTGSの大よその付加であった。
第4の例は40mg/kg/投与の1日4回×9スケジュールにおいてp.o.投与された化合物Aと、4mg/kg/投与の4日4回×3スケジュールにおいてi.v.投与されたDOXの効果を証明する。すべての処置は、すべての動物が小さいがしかし66mgサイズの平均腫瘍に達した時の移植後7日目から開始された。対照腫瘍は平均倍増時間3.7日ですべての動物において次第に成長した。成長遅延パラメータを計算するために使用された評価終点は塊倍増4例までの時間であった。このサイズへ達する未処置対照グループのための中間時間は14.5日であった。DOXの4mg/kg投与レベルは良く耐え、単独剤として処置期間の間の平均5%の体重ロスを生じた。これは43%のTGSに関連していた。化合物Aは有意な体重ロスなしで良く耐え、そして46%のTGSを生じた。これら処置の組合わせは死亡なしで12%の平均体重ロスをもって良く耐えた。この組合わせの抗腫瘍有効度はやはり133%のTGSの大よその付加であった。
第5の例は80mg/kg/投与の1日4回×9スケジュールにおいてp.o.投与された化合物Aと、80mg/kg/投与の1日4回×9スケジュールにおいてp.o.投与されたゲフィニチブ(Iressa)の効果を証明する。すべての処置は、すべての動物が小さいがしかし110mgサイズの平均腫瘍を持った時の移植後15日目に開始された。対照腫瘍は平均倍増時間10.5日ですべての動物において次第に成長した。成長遅延パラメータを計算するために用いた評価終点は塊倍増1例への時間であった。Iressaの150mg/kg投与レベルは単独剤として処置期間の間体重ロスおよび死亡なしで良く耐えた。この処置は101%TSとPR例に関連していた。化合物Aも単独剤として体重ロスまたは死亡なしで良く耐え、CR1例およびPR2例をもって218%のTGSを生じた。これらの剤の組合わせはマウス10匹中1例の非特異的死と、平均10%の体重ロスをもって良く耐えた。この組合わせの抗腫瘍有効度は314%のTGSの大よその付加であった。この処置はCR6例およびPR3例を生じた。

- 1以上の薬学的に許容し得る担体分子と組合わせた請求項1の組成物。
- 前記細胞毒剤又は細胞増殖抑制剤は、DNAトポイソメラーゼI、DNAトポイソメラーゼII、DNAインターカレーター、アルキル化剤、微小管ディスラプター、ホルモン因子受容体アンタゴニスト/アゴニスト、又は成長因子受容体アンタゴニスト/アゴニストである請求項1の組成物。
- 前記細胞毒剤又は細胞増殖抑制剤は、イリノテカン、ビノレルビン、ゲムシタビン、ゲフィニチブ、パクリタクセル、タクソテレ、ドキソルビシン、シスプラチン、カルボプラチン、BCNN、CCNU、DTIC、メルファラン、シクロフォスファミド、araA、araC、エトポシド、ビンクリスチン、ビンブラスチン、アクチノマイシンD、5−フロロウラシル、メトトレキセート、ヘルセプチン、及びマイトマイシンCである請求項1の組成物。
- 前記細胞毒剤はイリノテカンである請求項1の組成物。
- 前記細胞毒剤はパクリタクセルである請求項1の組成物。
- 前記細胞毒剤はビノレルビンである請求項1の組成物。
- 前記細胞毒剤はゲムシタビンである請求項1の組成物。
- 前記細胞毒剤はドキソルビシンである請求項1の組成物。
- 前記細胞毒剤はゲフィニチブである請求項1の組成物。
- 前記組成物は、約0.1ないし約300mg/kg全体重の範囲の経口、筋肉内、静脈内、皮下、又は非経口投与量においてそれを必要とする患者へ投与される請求項1の組成物。
- 前記アリール尿素化合物は、N−(4−クロロ−3−(トリフロロメチル)フェニル−N’−(4−(2−メチルカルバモイル)−4−ピリジルオキシ)フェニル)尿素のトシレート塩である請求項1の組成物。
- アリール尿素化合物の治療的に有効量および(a)細胞毒剤又は(b)細胞増殖抑制剤又は(a)もしくは(b)の薬学的に許容し得る塩をそれを必要とする患者へ投与することよりなるがんの治療方法。
- 前記がんはrafキナーゼによって仲介される請求項16の方法。
- 前記アリール尿素化合物は、置換橋かけアリール尿素化合物、又は遠方リング上に置換基を有する少なくとも1つのアリール尿素構造を有する置換橋かけアリール尿素化合物、又は式Iの化合物
A−D−B (I)
であり:
式Iにおいて、Dは−NH−C(O)−NH−であり;
Aは式:−L−(M−L1 )q の40炭素原子までの置換分であり、ここでLはDへ直結した5員もしくは6員の環構造であり、L1 は少なくとも5員を有する置換環状基であり、Mは少なくとも1原子を有する橋かけ基であり、qは1ないし3の整数であり、そしてL及びL1 の各環構造は窒素、酸素及びイオウよりなる群から選ばれた0〜4員を含み;そして
Bは窒素、酸素及びイオウよりなる群から選ばれた、0〜4員を有するDへ直結した少なくとも1の6員環構造を有する置換もしくは未置換の30炭素原子までの3環までのアリール又はヘテロアリール部分であり;
ここでL1 は−SO2 Rx ,−C(O)Rx 及び−C(NRy )Rz よりなる群から選ばれた少なくとも1の置換基によって置換されており;
Ry は水素、又はN,S及びOよりなる群から選ばれたヘテロ原子を任意に含み、そしてペルハロまで任意にハロ置換された24炭素原子までの炭素系置換分であり;
Rz は水素、又はN,S及びOよりなる群から選ばれたヘテロ原子を任意に含み、そしてハロゲン、ヒドロキシ、及びN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲンによって任意に置換された24炭素原子までの炭素系置換基によって任意に置換された30炭素原子までの炭素系置換分であり;
Rx はRz 又はNRa Rb であり、ここでRa 及びRb は
a)独立して水素,
N,S,Oよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲン、ヒドロキシ、及びN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲンによって任意に置換された24炭素原子までの炭素系置換基によって任意に置換された30炭素原子までの炭素系置換分か、又は
−OSi(Rf )3 であり、ここでRf はN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲン、ヒドロキシ及びN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲンによって任意に置換された24炭素原子までの炭素性置換基によって任意に置換された30炭素原子までの炭素系置換分であり;又は
b)Ra 及びRb は、合体してN,S及びOよりなる群から選ばれた1−3ヘテロ原子の5−7員複素環構造か、又はハロゲン、ヒドロキシ、又はN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲンによって任意に置換された24炭素原子までの炭素系置換基によって置換された、N,S及びOよりなる群から選ばれた1−3ヘテロ原子の置換5−7員複素環構造を形成し;又は
c)Ra 及びRb の1つは、置換分Lへ少なくとも5員の環状構造を形成するように結合したC1 −C5 2価アルキレン基かもしくは置換C1 −C5 2価アルキレン基であり、ここで置換C1 −C5 アルキレン基の置換基はハロゲン、ヒドロキシ、及びN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲンによって任意に置換された24炭素原子までの炭素系置換基よりなる群から選ばれ;
Bが置換されている場合、Lが置換されているか又はL’が追加的に置換されており、この場合の置換基はペルハロまでのハロゲン、及びWn(nは0−3)よりなる群から選ばれ;
ここで各Wは−CN,−CO2 R7 ,−C(O)NR7 R7 ,−C(O)−R7 ,−NO2 ,−OR7 ,−SR7 ,−NR7 R7 ,−NR7 C(O)OR7 ,−NR7 C(O)R7 ,−Q−Ar,及びN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそして−CN,−CO2 R7 ,−C(O)R7 ,−C(O)NR7 R7 ,−OR7 ,−SR7 ,−NR7 R7 ,−NO2 ,−NR7 C(O)R7 ,−NR7 C(O)OR7 及びペルハロまでのハロゲンよりなる群から独立に選ばれた1以上の置換基によって任意に置換された24炭素原子までの炭素系置換分よりなる群から独立に選ばれ、ここでR7 は水素、又はN,S及びOよりなる群から選ばれたヘテロ原子を任意に含みそしてハロゲンによって任意に置換された24炭素原子までの炭素系置換分から独立に選ばれ;
ここでQは−O−,−S−,−N(R7 )−,−(CH2 )m −,−C(O)−,−CH(OH)−,−(CH2 )m O−.−(CH2 )m S−,−(CH2 )m N(R7 )−,−O(CH2 )m −CHXa −,−CXa 2 −,−S(CH2 )m −,及び−N(R7 )(CH2 )m −であり、ここでm=1−3及びXa はハロゲンであり;そして

- 前記がんは、結腸、胃、肺、膵臓、卵巣、前立腺、白血病、メラノーマ、肝細胞、腎臓、神経膠腫、又は頭部及び頸部がんである請求項16の方法。
- 前記細胞毒剤又は細胞増殖抑制剤は、DNAトポイソメラーゼI、DNAトポイソメラーゼII、DNAインターカレーター、アルキル化剤、微小管ディスラプター、ホルモン因子受容体アンタゴニスト/アゴニスト、又は成長因子受容体アンタゴニスト/アゴニストである請求項16の方法。
- 前記細胞毒剤又は細胞増殖抑制剤は、イリノテカン、ビノレルビン、ゲムシタビン、ゲフィニチブ、パクリタクセル、タクソテレ、ドキソルビシン、シスプラチン、カルボプラチン、BCNN、CCNU、DTIC、メルファラン、シクロフォスファミド、araA、araC、エトポシド、ビンクリスチン、ビンブラスチン、アクチノマイシンD、5−フロロウラシル、メトトレキセート、ヘルセプチン、及びマイトマイシンCである請求項16の方法。
- 前記細胞毒剤はイリノテカンである請求項16の方法。
- 前記細胞毒剤はパクリタクセルである請求項16の方法。
- 前記細胞毒剤はビノレルビンである請求項16の方法。
- 前記細胞毒剤はゲムシタビンである請求項16の方法。
- 前記細胞毒剤はドキソルビシンである請求項16の方法。
- 前記細胞毒剤はゲフィニチブである請求項16の方法。
- 前記組成物は、経口放出により又は静脈内注射もしくは注入によりそれを必要とする患者へ治療上有効な量において投与される請求項16の方法。
- 前記組成物は、錠剤、液剤、局所ゲル、吸入剤の形、又は持続性組成物の形でそれを必要とする患者へ治療上有効な量において投与される請求項16の方法。
- 前記組成物は、約0.1ないし約300mg/kg全体重の範囲の経口、筋肉内、静脈内、皮下又は非経口投与量においてそれを必要とする患者へ投与される請求項16の方法。
- 前記アリール尿素化合物は、N−(4−クロロ−3−(トリフロロメチル)フエフニル−N’−(4−(2−メチルカルバモイル)−4−ピリジルオキシ)フェニル尿素のトシレート塩である請求項16の方法。
- 患者内のがん細胞の増殖を阻止する方法であって、前記がん細胞を請求項1の組成物からなる製剤と接触させることよりなる方法。
