Lac-SPGおよびGal-SPG、Glc-SPGとアンチセンスオリゴヌクレオチド(DNA)との複合体の調製 実施例2で合成されたLac-SPGならびに実施例3で合成されたGal-SPG、実施例4で合成されたGlc-SPGをそれぞれ極性溶媒であるジメチルスルホキシド(以下DMSOと表記)に溶解させ、濃度を19.8 mg/mlに調整し、この溶液1μl、純水3μl、10mMのトリス緩衝液(pH7.8)1μlと、1本鎖のアンチセンスオリゴヌクレオチド水溶液(1mg/ml)5μlを混合した。得られた溶液はすべて透明で、均一であった。アンチセンスオリゴヌクレオチドは、癌原遺伝子として文献に報告されているc-myb遺伝子のセンス配列に相補的なホスホロチオエート結合を持つアンチセンス配列 5’-GTG CCG GGG TCT TCG GGC-3’を適用した。実際に使用したアンチセンスオリゴヌクレオチド(固相合成品)は、3’末端に40のdAをつけたシークエンスのc-mybホスホロチオエートアンチセンスオリゴヌクレオチド(配列番号1、以下AS-c-mybと表記)とした。

また、Lac-SPGおよびGal-SPG、Glc-SPGのみをHepG2細胞に投与しても細胞生存率にはほとんど影響を与えていない。
