【背景技術】

最近の調査は、ヒトが近代社会においてアレルギー性副鼻腔炎、アレルギー性鼻炎、および喘息のようなアレルギー疾患にますます罹患していることを示している。例えば、アメリカだけでも、8700万人以上の人々が、何らかの形のアレルギー疾患に罹患していると推定される。治療の財政的負担は、全体で数十億ドルにも上るが、それはこれらの疾患が再発性であるためである。

‐単環式、二環式アリール、およびヘテロアリール化合物、
‐ならびにキナゾリン誘導体。 - 阻害剤が、4-(4-メチルピペラジン-1-イルメチル)-N-[4-メチル-3-(4-ピリジン-3-イル)ピリミジン-2-イルアミノ)フェニル]ベンズアミドである、請求項7記載の方法。
- c-kit阻害剤がIL-3の存在下で培養したIL-3依存的細胞の死を促進することができない、請求項3から8のいずれか一項に記載の方法。
- c-kit阻害剤が活性化c-kitの阻害剤である、請求項3から9のいずれか一項に記載の方法。
- 活性化c-kit阻害剤がSCF活性化c-kitを阻害することができる、請求項10記載の方法。
- 阻害剤が構成的に活性化された変異体c-kitを阻害することができる、請求項10記載の方法。
- 以下を含むスクリーニング法によって得られる選択的で強力かつ非毒性の活性化c-kit阻害剤である化合物を、そのような治療を必要とする哺乳類に投与する段階を含む、アレルギー疾患を治療する方法:
a)(i)活性化c-kit、および(ii)試験する少なくとも一つの化合物を、成分(i)と成分(ii)とが複合体を形成することができる条件下で接触させる段階、
b)活性化c-kitを阻害する化合物を選択する段階、
c)IL-3の存在下で培養したIL-3依存的細胞の死を促進することができない、段階b)において同定された化合物のサブセットを試験して選択する段階。 - スクリーニング法が、SCF活性化野生型c-kitを阻害することもできる活性化変異体c-kitの阻害剤である、段階b)において同定された化合物のサブセットを試験して選択することからなる段階をさらに含む、請求項13記載の方法。
- 活性化c-kitが、段階a)のSCF活性化野生型c-kitである、請求項13記載の方法。
- 推定の阻害剤が、段階a)において10 μMより高い濃度で試験される、請求項13から15のいずれか一項に記載の方法。
- IL-3が、好ましくはIL-3依存的細胞の培養培地中に0.5 ng/mlから10 ng/ml、好ましくは1 ng/mlから5 ng/mlの濃度で存在する、請求項13から16のいずれか一項に記載の方法。
- IL-3依存的細胞が、肥満細胞、トランスフェクトした肥満細胞、BaF3およびIC-2からなる群より選択される、請求項17記載の方法。
- 成分(ii)が活性化c-kitを阻害する程度がインビトロまたはインビボで測定される、請求項13から18のいずれか一項に記載の方法。
- 1μM未満の濃度で野生型c-kitを阻害することができる化合物を試験して選択することからなる段階をさらに含む、請求項13から18のいずれか一項に記載の方法。
- 試験がインビトロまたはインビボで行われる、請求項20記載の方法。
- 活性化変異体c-kitおよび/または野生型c-kitの阻害が、免疫沈降およびウェスタンブロットのような標準的な生化学的技術を用いて測定される、請求項13から21のいずれか一項に記載の方法。
- c-kitリン酸化の量が測定される、請求項13から21のいずれか一項に記載の方法。
- 同定および選択された化合物が、強力、選択的かつ非毒性の野生型c-kit阻害剤である、請求項13から23のいずれか一項に記載の方法。
- 以下を含むスクリーニング法によって得られるc-kit阻害剤を、そのような治療を必要とする哺乳類に投与する段階を含む、アレルギー疾患を治療する方法:
a)永続的に活性化されたc-kitである変異体c-kit(例えば、トランスホスホリラーゼドメインにおける変異)を発現する細胞を用いて、細胞死の程度を測定することによって、それぞれがIC50<10 μMを有する、活性化c-kitを標的とする候補化合物のサブセットを同定するために、複数の被験化合物について増殖アッセイを行う段階、

c)c-kitを発現する細胞を用いて、段階b)において同定された化合物のサブセットについて増殖アッセイを行い、細胞死の程度を測定することによって、それぞれがIC50<10 μMを有する、好ましくはIC50<1 μMを有する、野生型c-kitを標的とする候補化合物のサブセットを選択する段階。 - 細胞死の程度を3Hチミジンの取り込み、トリパンブルー排除法、またはヨウ化プロピジウムによるフローサイトメトリーによって測定する、請求項25記載の方法。
- ヒト、イヌ、およびネコにおけるアレルギー疾患を予防するか、発病を遅らせるか、または治療するための、請求項1から26のいずれか一項に記載の方法。
- 喘息、アレルギー性鼻炎、アレルギー性副鼻腔炎、アナフィラキシー症候群、蕁麻疹、血管浮腫、アトピー性皮膚炎、アレルギー性接触性皮膚炎、結節性紅斑、多形紅斑、皮膚壊死性細静脈炎、虫さされによる皮膚炎症および吸血寄生虫の外寄生からなる群より選択されるアレルギー疾患を予防するか、発病を遅らせるか、または治療するための、請求項1から27のいずれか一項に記載の方法。
- アレルギー疾患を治療する薬剤を製造するためのc-kit阻害剤の使用。
- チロシンキナーゼ阻害剤を含む、患者の呼吸器、鼻腔内、または局所投与の標的領域にエアロゾル化製剤を投与することを意図した組成物。
- チロシンキナーゼ阻害剤がc-kit阻害剤である、請求項30記載の組成物。
- エアロゾル投与に適した水溶液、エタノール溶液、水性/エタノール溶液、生理食塩液、コロイド懸濁液、および微結晶懸濁液から選択される溶液を含む、請求項30記載の組成物。
- 好ましくは定量弁を有する、請求項30から32のいずれか一項に記載の組成物を含むエアロゾル装置。
- 液体ガスシステム、懸濁液エアロゾル、または加圧ガスシステムを含む、請求項33記載のエアロゾル装置。
- チロシンキナーゼ阻害剤、より好ましくはc-kit阻害剤を含む、点鼻用点滴器または点鼻用スプレー装置。
- 蕁麻疹、アトピー性皮膚炎、アレルギー性接触性皮膚炎、結節性紅斑、多形紅斑、皮膚壊死性細静脈炎、虫さされによる皮膚炎症、ならびに特にイヌおよびネコにおける吸血寄生虫の外寄生のような皮膚アレルギー障害を治療するために、チロシンキナーゼ阻害剤、より詳細にはc-kit阻害剤を含む局所投与のために適した組成物。
- 蕁麻疹、アトピー性皮膚炎、アレルギー性接触性皮膚炎、結節性紅斑、多形紅斑、皮膚壊死性細静脈炎、虫さされによる皮膚炎症、ならびに特にイヌおよびネコにおける吸血寄生虫の外寄生のような皮膚アレルギー障害を治療するために、チロシンキナーゼ阻害剤、より詳細にはc-kit阻害剤を含む経口投与のために適した組成物。
