本発明は、DNAやRNAなどの核酸、自然界に存在する環境物質および新規化学物質の微量分析および物質創製に用いられるマイクロリアクタに関するものである。

従来のマイクロリアクタにおいては、流路内に残滴が残りにくくするために次のようなマイクロリアクタであった。その中では、超音波照射によって微小流路内での流体の混合攪拌手段が記載されている(例えば特許文献1参照)。

また、超音波の照射方向が互い違いに非対称になっていることで超音波の強度分布が流路内で非対称となり効果的に音響流が矢印10の方向に発生する。
上記のようなマイクロリアクタにおいて、流路内に何ら層流を乱す構成を持たせることなく滑らかな管壁から超音波を照射して攪拌することで、流路抵抗の上昇ないし試料溶液を攪拌通過させることが可能である。また、10MHz未満の振動数の超音波を用いることで、キャビテーションの発生を抑制することができるので、細胞などの生物試料の損傷を防止できるようになっている。

【発明が解決しょうとする課題】
しかしながら、上記由来のマイクロリアクタにおいて、試料の送液量が多い場合や試料濃度が低い場合には、試料成分および試薬成分の移動速度を加速させるにはエネルギー的に不十分なため、目的成分を高速・高効率に得ることは難しい。
したがって、本発明の目的は、短時間且つ高効率に目的成分を生成することが可能なマイクロリアクタを提供することである。

【課題を解決するための手段】
上記問題を解決するため、本発明は次のように構成したものである。
請求項1記載の発明は、注入口14と流出口15との間に凹状の混合部17を設け、その混合部17内にナノ材料攪拌子18を配置し、混合部17の領域にあたる上蓋13の上部に混合部17の温度を制御するための冷却部19を埋設させ、混合部17の下部に金属板20を埋設し、その金属板20と混合部18との間に温度センサー挿入口21を設けたものである。
そのようになっているため、図示していない化学合成などで良く使用されている小型攪拌機(スターラー)の回転に伴って、混合部17内のナノ材料攪拌子が回転するために、混合部17内に導入された流体が層流から乱流へと変化するので、試料成分と試薬成分の衝突確率が増し、結果的に反応時間を短くするができる。また、温度センサーによって混合部17内の溶液温度を測定することで、溶液を過剰に加熱する必要がないので、熱的に不安定な試料成分や試薬成分の熱分解を防止することが可能であり、冷却部19で混合部17の上層部を冷却しているので、溶液の気化による気泡の発生も抑制することができる。
請求項2記載の発明は、1本ないしは複数本の単層または複層のカーボンナノチューブから形成されたナノチューブ材料22に、フェライト微粒子(酸化鉄など)、シリカゲル、アルミナなどから選択して用いた磁性粒子23の表面をフルオロシラン、トリメチルクロロシラン、ビニルトリメトキシシラン、γ−メタクリロキシプロピルトリメトキシシランなどから少なくとも1種類のシランカップリング剤24でコーティングし、それをナノチューブ材料22の両側面に固定化したものである。
そのようになっているため、微小空間での流体の混合・攪拌が可能となるとともに、試料成分や試薬成分が磁性微粒子の表面や細孔内部に吸着・残留することがないので、目的成分を高効率に得ることが可能となる。
請求項3記載の発明は、混合部17内の温度を制御する冷却部19は、下側に矩形のペルチェ素子25を配置し、そのペルチェ素子25の上面にナノチューブ集合体26を高密度に配置し、ならにそのナノチューブ集合体25を囲むようにポリピロピレン(PP)、ポリエチレン(PE)、ポリカーボネート(PET)などの高分子樹脂やアルミから形成された枠体27をペルチェ素子25の上面に固定化し、ペルチェ素子25から発生した熱を系外に排出するための冷却ファンを枠体28に設けたものである。
そのようになっているため、ペルチェ素子25から吸収した熱をナノ材料で素早く伝熱するので、アルミ製のヒートシンクに比べて冷却効率が向上するとともに、更なる薄型および軽量化が実現可能となる。

【発明の実施の形態】
本発明の実施形態を図に基づいて説明する。
(第1の実施形態)
図1は、本発明の第1の実施形態を示すマイクロリアクタの側断面図である。
図1に示す100〜500μmの幅および深さの溝が形成されているマイクロ反応本体部12の上面に上蓋13の下面を接合することにより、注入口14、排出口15、微小流路16が形成される。次に、注入口14から送液された試料溶液および試薬溶液を混合部17に導入し、図示していない小型攪拌機の回転に伴いナノ材料攪拌子18が回転し、試料溶液および試薬溶液とが強制的に混合攪拌される。小型攪拌機の中で加熱機能付きのものを用いると、発生した熱が金属板20を介して混合部17に伝熱して混合部17内の溶液を加熱する。
また、温度センサー挿入口21にセットされた白金測温体または熱電素子と上蓋13に埋設された冷却部19で混合部17内の溶液温度を制御する。
この実施の形態のマイクロリアクタは、試料溶液と試薬溶液を高速・高効率に混合して目的成分の生成率を向上させる。また、溶液の過熱による試料および試薬成分の熱分解を防止するとともに、溶液からの気泡の発生を防止することである。
この実施の形態によれば、図示していない小型攪拌機を700〜1500rpmの回転数で回転させることにより、混合部17に導入された二つ以上の流体を強制的に攪拌しているため、流体が層流から乱流へと急速に変化し、試料成分および試薬成分の移動速度が加速されるので、試料成分と試薬成分の衝突確率が大きくなり、最終的に目的成分を短時間に生成できるようになる。尚、反応効率をより一層向上させるには、間欠的に回転方向を切り替える方が好ましい。
また、金属板20には熱伝導性の良い金属材料を用いているため、加熱機能付き小型攪拌機から発生した熱を効率良く混合部17に伝熱できるので、過熱による試料成分および試薬成分の熱分解を防止できるようになる。

(第2の実施形態)
図2は、本発明の第2の実施形態を示すマイクロリアクタの側断面図である。
すなわち、図2に示す1本ないし複数本の単層または複層のカーボンナノチューブからなるナノチューブ材料22の両端面に、フェライト粒子、シリカ、アルミナなどの磁性粒子23の表面層にフルオロシラン、トリメチルクロロシラン、ビニルトリメトキシシラン、γ−メタクリロキシプロピルトリメトキシシランなどから少なくとも1種類のシランカップリング剤24がコーティングされている。

この実施の形態によれば、直径がサブマイクロメールから数十マイクロメートルのシリカ粒子の場合、粒子には多数の亀裂や細孔が存在している上に、粒子内部および表面層に親水性のシラノール基(Si−OH)存在するため、表面を何らかの方法で不活性にしないと、DNA、RNA、タンパク質などの生体分子や官能基にアンモニウムイオン(―NH3+)などを有する有機分子などが細孔内部や表面層のシラノール基と静電気的な相互作用などにより吸着・残留する可能性がある。
そこで、下記の反応式(1)のシランカップリング剤の一種であるトリメトキシフルオロシランによるシリカ粒子表面の化学的処理を例にとって説明する。
表面層のシラノール基(Si−OH)がトリメトキシフルオロシラン(F−Si−(OCH3)3)と反応することにより、安定なシロキサン基(Si−O−Si)となり、表面層はフッ素で覆われるため、粒子の表面は親水性から疎水性へと変化するので、DNA、RNA、タンパク質などの生体分子やアンモニウムイオンを有する有機分子などが粒子の細孔内部および表面層に吸着・残留することを抑制できる。

(第3の実施形態)
図3は、本発明の第3の実施形態を示すマイクロリアクタの側断面図である。すなわち、ペルチェ素子25の下面から吸収した熱は、ナノチューブ集合体26に伝達された後、枠体27に取り付けられた冷却ファン28により系外に排出される。
