製造例1

培養終了後、シャープレス型遠心分離機にて菌体を回収し、菌体を水で2回洗浄した。
得られた菌体を水で菌体濃度10%に調整・懸濁した後、90℃、30分間加熱し菌体内酵素を失活させ、次いで、65℃に冷却した。冷却液のpHを9とし、同温度で60分間処理し、エキスを抽出した。遠心分離機により菌体残渣を除去し、得られた上澄液をpH7に中和し、デキストリンを添加後、スプレードライし、リボ核酸含有酵母エキスを得た。
本酵母エキスは、リボ核酸を27重量%含んでいた。

This yeast extract contained 27% by weight of ribonucleic acid.

実施例2
母豚1頭(LW種)を用い、リボ核酸含有酵母エキスを投与した場合(投与区、2003年9月試験開始)、及び投与しない場合(無投与区、2003年4月試験開始)の試験を行った(於:農事法人組合打戻種豚組合)。
すなわち、母豚の分娩予定日5日前から分娩日までにわたり、表3に示した条件でリボ核酸含有酵母エキスを与え、分娩後の母豚母乳中の5’−ヌクレオチド量、及び離乳時まで(分娩後26日間)の各区毎に下痢した子豚及び虚弱死した子豚の頭数を計測した。なお、本試験においては、下痢及び虚弱治療は実施しなかった。
試験期間中、使用した飼料はウィニーブリード(日本農産工業製、種豚飼育用配合飼料)で、投与区は製造例1で得られたリボ核酸含有酵母エキスを当該飼料に混合して用いた。また、搾乳は分娩直後から開始し、1時間目、2時間目、3時間目、6時間目、分娩3日目までは毎日1回、その後は7日目、14日目、21日目及び28日目に、用指法で搾乳した。
なお、搾乳時に、搾乳しやすいようにホルモン剤(帝国臓器製薬製、アトニン)1mlを、搾乳の10分前に筋肉注射した。
