【発明の属する技術分野】
本発明は、検体中の細胞、微生物、あるいは染色体、核酸などを、抗原抗体反応や核酸ハイブリダイゼーション反応などの生化学反応を利用して分析する技術に関する。

【従来の技術】
血液等の検体の分析装置の多くは、抗原坑体反応を利用した免疫学的な方法、もしくは核酸ハイブリダイゼーションを利用した方法を用いている。こうした分析方法の例を挙げると、被検出物質と特異的に結合する抗体又は抗原などのタンパク質あるいは一本鎖の核酸をプローブとして微粒子、ビーズ、ガラス板などの固相表面に固定し、被検出物質と抗原抗体反応もしくは核酸ハイブリダイゼーションを行なわせる。そして酵素、蛍光性物質、発光性物質などの検知感度の高い標識物質を担持した特異的な相互作用を持つ標識化物質、例えば標識化抗体や標識化抗原又は標識化核酸などを用いて、抗原抗体化合物や二本鎖の核酸を検出して被検物質の有無を検出あるいは被検物質の定量を行うものである。

これらの技術を発展させたものとして、米国特許(USP)5,445,934号公報に、互いに異なる塩基配列を有する多数のDNA(デオキシリボ核酸)プローブを基板上にアレイ状に並べた、いわゆるDNAアレイの記載がある。また、Anal. Biochem.、270(1)、103−111、1999には、多種類のタンパク質をメンブレンフィルター上に並べ、DNAアレイのような構成のタンパク質アレイの作製方法が開示されている。DNAアレイ、タンパク質アレイなどによって一度に極めて多数の項目の検査を行うことが可能になってきている。

また、様々な検体分析方法において、検体による汚染の軽減、反応の効率化、装置の小型化、作業の簡便化などの目的で、内部で必要な反応を行う、使い捨ての生化学反応カートリッジも提案され、特表平11−509094号には、DNAアレイを含む生化学反応カートリッジ内に複数のチャンバーを有し、差圧によって溶液の移動を行い、内部で検体からのDNAの抽出あるいは増幅、又はハイブリダイゼーションなどの反応を可能とした生化学反応カートリッジが開示されている。

そして、上記のような生化学反応カートリッジ内部に外部から溶液を流入する方法としては、外部のシリンジポンプや真空ポンプを利用したものがある。また、生化学反応カートリッジ内部で溶液を移動する方法としては、重力、毛細管現象、電気泳動を利用したものや、そして小型であるため生化学反応カートリッジ内部に配設できるマイクロポンプとして、特許第2832117号には発熱素子を利用したもの、特開2000−274375号には圧電素子を利用したもの、特表平11−509094号にはダイアフラムポンプが開示されている。

【発明が解決しようとする課題】
しかしながら、二次感染、汚染(コンタミ)を防ぐためと、使い勝手の点から、必要な溶液を内蔵した使い捨てのカートリッジを使うことが好ましいが、ポンプを内蔵したカートリッジでは高価になってしまう。

本発明は上述した問題に鑑みてなされたものであり、使い捨てのカートリッジで生化学反応を進める際に、外部のポンプを用いながら、ユーザが検体を注入したあとは溶液を外部に出さずに自動的に生化学反応を進める装置を提供することを目的とする。

以下に、本発明の実施の形態について添付図面を参照して詳細に説明する。

図1に本実施形態における生化学反応カートリッジの外観図を示す。本実施形態の生化学反応カートリッジは、血液等の液体状の検体を注入して後述する処理装置にセットすると生化学反応カートリッジ内部でDNAなどの抽出、増幅が行われ、増幅した検体DNAと生化学反応カートリッジ内部にあるDNAマイクロアレイ上のDNAプローブとのハイブリダイゼーション、ハイブリダイゼーションしなかった蛍光標識付きの検体DNAと蛍光標識の洗浄といった工程が行われる。

生化学反応カートリッジ1の本体はポリメタクリル酸メチル(PMMA)、アクリルニトリル−ブタジエン−スチレン(ABS)、ポリスチレン、ポリカーボネート、ポリエステル、ポリ塩化ビニルなどの透明あるいは半透明のプラスチックで構成されている。生化学反応カートリッジ1内の反応物についての光学的な測定を必要としない場合は、透明な材質でなくとも良い。生化学反応カートリッジ1の上部には、血液等の検体を注射器を用いて注入する際の検体入口2の部分にゴムキャップが固定されている。また、側面には生化学反応カートリッジ1内の溶液を移動するためにノズルを挿入して加圧あるいは減圧を行う複数のノズル入口3が構成され、そこにゴムキャップが固定され、反対側の面も同じ構成になっている。

図2は図1の生化学反応カートリッジの平面断面図である。片側の側面には10個のノズル入口3a〜3jがあり、反対側の側面にも10個のノズル入口3k〜3tがある。各ノズル入口は、空気が流れる空気流路4を介して、溶液を貯蔵する場所あるいは反応を起こす場所であるチャンバー5につながっている。但しノズル入口3n、3p、3q、3sは本実施形態の工程では使わないのでチャンバーにつながっておらず、予備になっている。結局、ノズル入口3a〜3jは流路4a〜4jを介してチャンバー5a〜5jにつながり、反対側のノズル入口3k、3l、3m、3o、3r、3tは、それぞれ流路4k、4l、4m、4o、4r、4tを介してチャンバー5k、5l、5m、5o、5r、5tにつながっている。

そして、チャンバー5a、5b、5c、5kはチャンバー7に、チャンバー5g、5oはチャンバー8に、チャンバー5h、5i、5j、5r、5tはチャンバー9につながっている。さらにチャンバー7は流路10を介してチャンバー8に、チャンバー8は流路11を介してチャンバー9につながっている。流路10にはチャンバー5d、5e、5f、5l、5mがそれぞれ流路6d、6e、6f、6l、6mを介してつながっている。また、チャンバー9の底面は角穴があけられ、そこにガラス基板で作製されたDNAマイクロアレイ12が下側からプローブ面が上になるように貼り付けられてチャンバー9が形成されている。

DNAマイクロアレイ12は、1インチ四方程度のガラス板等の固相表面に、異なる種類のDNAプローブが数十から数十万種高密度に並べてあるものである。このDNAマイクロアレイ12を用いて検体DNAとハイブリダイゼーション反応を行うことによって、一度に数多くの遺伝子の検査ができるという特徴をもつ。また、これらのDNAプローブはマトリクス状に規則正しく並んでおり、それぞれのDNAプローブのアドレス(例えば何行何列という位置)を情報として容易に取り出せるという特徴がある。検査の対象となる遺伝子としては、感染症ウイルス・細菌、疾患関連遺伝子の他、各個人の遺伝子多型等がある。

チャンバー5a、5bには、それぞれ細胞壁をこわすEDTAを含む第一の溶血剤、界面活性剤などのタンパク質変性剤を含む第二の溶血剤が蓄積されている。チャンバー5cにはDNAが吸着するシリカコーティングされた磁性体粒子が蓄積され、チャンバー5l、チャンバー5mには、DNAの抽出の際にDNAの精製を行うために用いる第一、第二の抽出洗浄剤が蓄積されている。チャンバー5dにはDNAを磁性体粒子から溶出する低濃度塩のバッファーからなる溶出液、チャンバー5gには、PCRで必要なプライマー、ポリメラーゼ、dNTP溶液、バッファー、蛍光剤を含むCy−3 dUTPなどの混合液が充填されている。チャンバー5h、5jにはハイブリダイゼーションしなかった蛍光標識付きの検体DNAと蛍光標識とを洗浄するための界面活性剤を含む洗浄剤、チャンバー5iにはDNAマイクロアレイ12を含むチャンバー9内を乾燥させるためのアルコールが蓄積されている。

なお、チャンバー5eは血液のDNA以外のゴミが溜まるチャンバー、チャンバー5fはチャンバー5l、5mの第一、第二の抽出洗浄剤の廃液が溜まるチャンバー、チャンバー5rは第一、第二の洗浄剤の廃液が溜まるチャンバーであり、チャンバー5k、5o、5tは溶液がノズル入り口に流れ込まないために設けたブランクのチャンバーである。

図3は生化学反応カートリッジ1内での溶液の移動や種々の反応を制御する処理装置である。テーブル13は生化学反応カートリッジ1をセットする場所で、テーブル13上には、生化学反応カートリッジ1内で検体からのDNAなどを抽出する際に作動させる電磁石14、検体からのDNAをPCR(Polymerase Chain Reaction)などの方法で増幅させる際に温度制御するためのペルチェ素子15、増幅した検体DNAと生化学反応カートリッジ内部にあるDNAマイクロアレイ上のDNAプローブとのハイブリダイゼーション、ハイブリダイゼーションしなかった検体DNAの洗浄を行う際に温度制御するためのペルチェ素子16が配置され、これらは処理装置全体を制御する制御部17に接続されている。テーブル13の両側(図3の上下)には、電動シリンジポンプ18、19と、それらのポンプにより空気を吐出あるいは吸引を行う出入り口で片側10個ずつある複数のポンプノズル20、21とを有するポンプブロック22、23が配置されている。電動シリンジポンプ18、19とポンプノズル20、21の間には、図示しない複数のよく知られた電動切替バルブが配置され、電動シリンジポンプ18、19と共に制御部17に接続されている。制御部17により、片側10個のうちの1個ずつのポンプノズルを選択的に電動シリンジポンプ18、19に対してオープンになるようにしたり、すべてのポンプノズルをクローズにしたりする制御ができる。また、制御部17は検査者が入力を行う入力部24に接続されている。

本実施形態では血液を検体とし、検査者が血液を注射器を用いて検体入口2のゴムキャップを貫通させて注入すると、血液はチャンバー7に流れ込む。その後、検査者は生化学反応カートリッジ1をテーブル13に置き、図示しないレバーを操作すると、不図示の機構によりポンプブロック22、23は図3の矢印の方向に移動し、ポンプノズル20、21がノズル入口3にゴムキャップを貫通して挿入される。

図2で説明したように、ノズル入口が生化学反応カートリッジの2つのエリアに集中しているので、電動シリンジポンプ、電動切替バルブ、ポンプノズルを内蔵したポンプブロック22、23の形状、配置が単純になる。さらに、両側の側面に集中してそれぞれ複数のノズル入口が設けられているので、必要なチャンバーや流路を確保しながら、ポンプブロック22、23で生化学反応カートリッジを同時に挟み込むという単純な動きで、ポンプノズル20、21を挿入することができ、ポンプブロック22、23の構成も簡単なものになる。そしてノズル入口はすべて同じ高さ、すなわち直線状に配置すると、ノズル入口に接続する流路の高さが全て同じになり、流路の作製が容易になる。

また、図3の処理装置で、複数(n)個の生化学反応カートリッジ用にポンプブロック22、23をn倍に長くした構成にすれば、n個の生化学反応カートリッジを直列に並べて、必要な工程をn個の生化学反応カートリッジに対して同時に行うことができ、極めて簡単な構成で多数の生化学反応カートリッジに対して生化学反応を行わせることができる。

検査者が入力部24で処理開始の命令を入力すると、処理が始まる。図4は本実施形態の処理装置における処理手順を説明するフローチャートである。まずステップS1で、制御部17はノズル入口3aと3kのみをオープンにし、電動シリンジポンプ18から空気を吐出し、電動シリンジポンプ19から空気を吸引してチャンバー5aの第一の溶血剤を血液の入ったチャンバー7に流し込む。このときに、溶血剤の粘性や流路の抵抗によるが、電動シリンジポンプ19からの空気の吸引を、電動シリンジポンプ18からの空気の吐出開始10〜200mS後に開始するように制御すると、流れる溶液の先頭で溶液が飛び出すことがなく、スムーズに溶液が流れる。このように空気の供給の制御のタイミングをずらして、加圧、減圧を制御して溶液をスムーズに流すだけではなく、電動シリンジポンプ19からの空気の吸引を、電動シリンジポンプ18からの空気の吐出開始時からリニアに増加するように制御するなど、細かな制御を行うことにより、さらにスムーズに流すことが可能になる。以下の溶液の移動についても同様である。空気の供給の制御は、シリンジポンプを用いることで容易に実現できたものである。そして、ノズル入口3aと3oのみをオープンにし、電動シリンジポンプ18と19で空気の吐出、吸引を交互に繰り返し、チャンバー7の溶液を流路10に流してその後戻す動作を繰り返して攪拌を行う。あるいは電動シリンジポンプ19から空気の吐出を続けて気泡を発生させて攪拌しても良い。

そして、ステップS4で電磁石14をオンにし、ノズル入口3eと3kのみをオープンにし、電動シリンジポンプ20から空気を吐出し、電動シリンジポンプ19から空気を吸引してチャンバー7の溶液をチャンバー5eに移動する。この移動の際に磁性体粒子とDNAが流路10の電磁石14の上で捕捉される。電動シリンジポンプ18、19の吸引、吐出を交互に繰り返して溶液をチャンバー7、5e間を二回往復させてDNAの捕捉効率を高める。回数をさらに増やすとさらに捕捉効率を高めることができるが、処理時間が余計にかかることになる。

このように、磁性体粒子を利用してDNAを、幅1〜2mm程度、高さ0.2〜1mm程度の小さい流路上で、しかも流れている状態で捕捉するので、極めて効率良く捕捉することができる。捕捉ターゲット物質がRNAあるいはタンパク質の場合も同様である。

次にステップS5で、電磁石14をオフにし、ノズル入口3fと3lのみをオープンにし、電動シリンジポンプ19から空気を吐出し、電動シリンジポンプ17から空気を吸引してチャンバー5lの第一の抽出洗浄液をチャンバー5fに移動する。このとき、ステップS4で捕捉された磁性体粒子とDNAが抽出洗浄液と共に移動して洗浄が行われる。ステップS4と同様にして二回往復した後、電磁石14をオンにし、同様にして二回往復させて磁性体粒子とDNAを流路10の電磁石14の上に回収し、溶液をチャンバー5lに戻しておく。

ステップS6で、ノズル入口3eと3mを用いてチャンバー5mの第二の抽出洗浄液に対して、ステップS5と同じ工程を行ってさらに洗浄する。ステップS7では電磁石14をオンにしたまま、ノズル入口3fと3lのみをオープンにし、電動シリンジポンプ18から空気を吐出し、電動シリンジポンプ19から空気を吸引してチャンバー5dの溶出液をチャンバー8に移動する。このとき、溶出液の作用によって磁性体粒子とDNAが分離し、DNAのみが溶出液と共にチャンバー8に移動し、磁性体粒子は流路10に残る。以上のようにして、DNAの抽出、精製が行われる。抽出洗浄液が入ったチャンバーと洗浄後の廃液が入るチャンバーを用意したので、生化学反応カートリッジ1内でDNAの抽出、精製を行うことが可能になった。

次にステップS8で、ノズル入口3gと3oのみをオープンにし、電動シリンジポンプ17から空気を吐出し、電動シリンジポンプ19から空気を吸引してチャンバー5iのPCR用薬剤をチャンバー8に流し込む。さらに、ノズル入口3gと3tのみをオープンにし、電動シリンジポンプ18と19で空気の吐出、吸引を交互に繰り返し、チャンバー7の溶液を流路11に流してその後戻す動作を繰り返して攪拌を行う。そして、ペルチェ素子15を制御して、チャンバー8内の溶液を96℃の温度に10分保った後、96℃10秒、55℃10秒、72℃1分の工程を30回繰り返し、溶出されたDNAにPCRを行って増幅する。

ステップS9では、ノズル入口3gと3tのみをオープンにし、電動シリンジポンプ18から空気を吐出し、電動シリンジポンプ19から空気を吸引してチャンバー8の溶液をチャンバー9に移動する。さらに、ペルチェ素子16を制御して、チャンバー9内の溶液を45℃で2時間保ってハイブリダイゼーションを行う。このとき、電動シリンジポンプ18と19で空気の吐出、吸引を交互に繰り返し、チャンバー9の溶液を流路6tに移動し、その後戻す動作を繰り返して攪拌を行いながらハイブリダイゼーションを進める。

次にステップS10で、同じ45℃を保ったまま、今度はノズル入口3hと3rのみをオープンにし、電動シリンジポンプ18から空気を吐出し、電動シリンジポンプ19から空気を吸引して、チャンバー9内の溶液をチャンバー5rに移動しながら、チャンバー5hの第一の洗浄液をチャンバー9を通してチャンバー5rに流し込む。電動シリンジポンプ18、19の吸引、吐出を交互に繰り返して溶液をチャンバー5h、9、5r間を二回往復させ、最後にチャンバー5hに戻す。このようにして、ハイブリダイゼーションしなかった蛍光標識付きの検体DNAと蛍光標識とが洗浄される。

【発明の効果】
以上説明したように、本発明によれば、装置側のポンプによりカートリッジ内の空気圧を制御し、カートリッジ内の溶液をそのカートリッジ内のみで移動させて必要な生化学反応を起こさせるので、安価でありながら溶液を外部に出さない使い捨てのカートリッジが可能となり、二次感染、汚染(コンタミ)の恐れがなくなる。さらに、カートリッジが必要な溶液を内蔵しているので、試薬・洗浄液を装置側に準備する必要がなくなり、省力と共に試薬の間違いがなくなるという効果もある。
