本発明は、反応装置に関し、特に本発明は核酸の反応を行うための反応装置に関する。

【従来の技術】
近年、生化学、分子生物学の研究、検査で使用される様々な機器の機能、分析を1枚の微少なチップ、すなわちマイクロチップ上に集積させる技術の研究開発が行われている。このようなマイクロチップはμTAS(MicrototalAnalytical System:マイクロトータル・アナリティカル・システム)と称される。μTASは、半導体微細加工技術などを用いてガラスやシリコンなどの基板上にμgあるいはμlのスケールの反応溶液や分析手段を設け、マイクロチップ上で反応・分析を行うものである。このような技術の利点として、試薬、試料、および廃液が少量であること、自動化することにより高速処理が可能であること、システムの小型化が可能であることなどを挙げることができる。

一方、生化学、分子生物学の分野で核酸の増幅方法としてPCR法(Polymerase Chain Reaction:ポリメラーゼ連鎖反応)が一般的に用いられている(特許文献1)。たとえばDNA分析を行うためにはDNAを増幅する必要があるが、このPCR法によれば、増幅しようとするDNA、すなわち鋳型DNAの両端の塩基配列に相補的な塩基配列を有するプライマーおよび耐熱性DNAポリメラーゼを用い、熱変成工程、アニーリング(塩基対形成)工程および伸張反応工程の3段階からなるサイクルを繰り返すことにより、鋳型DNAとほぼ同じDNA断片を増幅することが可能となる。

また、PCR法による核酸の増幅機能をマイクロチップに持たせ、マイクロチップ上でDNA増幅を行うことが提案されている(特許文献2)。図1は、DNA増幅機能を備えた従来のマイクロチップを模式的に示す図である。マイクロチップ1上にはリザーバー2が設けられており、サンプルや反応溶液がリザーバー2に注入される。次に、サンプルや反応溶液は、キャピラリー構造を有する流路5を経て流路5の一部分に設けられた反応部6へ圧力送液される。反応部6には温度調節部3が設けられており、反応部6の温度が制御され、PCR反応が進行する。このようにして得られたPCR産物は圧送され、PCR産物回収部4へ送られる。

しかし、従来のDNA増幅機能を備えたマイクロチップは、サンプルや反応溶液の流路と反応部とを兼ねた単管キャピラリー構造を採用していたため、次のような課題を有していた。

まず、キャピラリーの一部分を反応容器として用いるため、反応容器の容量が極端に少なく、増幅により得られるDNA量が少なかった。また、反応溶液に圧力を付与し、反応部まで送液する必要があるが、この際の細かい制御が困難であった。

また、反応液を反応部で反応させる間、反応部に反応液を保持するためにエアーまたはオイルでシールしなければならないため、エアーまたはオイルを送り、制御する装置が別途必要となり、装置が複雑化していた。

さらに、同一のマイクロチップ上でPCR産物を分析に供することも可能であるが、この場合、分析前にはバッファーを交換する必要がある。しかし、従来のマイクロチップの構造では、バッファーの交換の自動化は困難であった。そのため、当該分析の条件を最適化するためには手作業が必要であった。

【発明が解決しようとする課題】
上記事情に鑑み、本発明の目的は、試料の移動を容易に制御することができる核酸の反応装置を提供することにある。また、本発明の別の目的は、必要充分な量の生成物を得ることができる核酸の反応装置を提供することにある。本発明のさらに別の目的は、試料を確実に反応部に保持し、効率良く反応を行うことができる核酸の反応装置を提供することにある。また、本発明のさらにまた別の目的は、試料を反応部に保持しながら流路中の移動相の置換を行うことができる核酸の反応装置を提供することにある。

【課題を解決するための手段】
本発明によれば、試料導入部および試料回収部を有する主流路と、該主流路の前記試料導入部から前記試料回収部に至る領域から分岐して設けられた副流路と、前記副流路に設けられた反応部と、前記試料導入部、前記試料回収部および前記反応部に、それぞれ独立に電位を与える電位付与手段と、を有することを特徴とする反応装置が提供される。

本発明に係る反応装置には、主流路から分岐して副流路が設けられており、副流路に反応部が設けられている。このような構成とすることにより、主流路とは別個に、副流路および反応部の形状や大きさの設計を行うことが可能となる。このため、主流路をキャピラリー等容量の少ない構成とした場合にも、反応部の容量を大きくすることができる。よって、大量増幅にも適用することが可能である。

また、本発明に係る反応装置では、試料導入部、試料回収部、および反応部のそれぞれに独立に電位を与えることができるため、電位を制御することにより、試料導入部に導入された試料を反応部に移動させ、反応中は試料を反応部に維持し、反応後は生成物を試料回収部に移動させるという一連のステップを、効率よく行うことができる。また、電位を制御することにより、圧力送液等の方法と比較し、微量の試料の送液を確実に制御することができる。また、試料を移動中に混合させることもできるため、効率がよい。さらに、電位を制御して、試料を反応部に維持することができるため、反応部のみを加熱する装置構成とすることが可能となる。したがって、主流路にゲルを用いた分析部など、耐熱性の低い領域が設けられている場合にも、装置全体を加熱する必要がないため、耐熱性の低い領域を保護することができる。

【発明の実施の形態】
本実施形態は、生化学反応を行うマイクロチップに関する。以下、生化学反応がPCR法による核酸の増幅である場合を例に、好ましい実施形態について説明する。

図2は本実施形態に係るマイクロチップ13の構成を示す上面図である。また図3は、図2のマイクロチップ13の基板15およびカバー16の構成を示す斜視図である。図3(a)に示される基板15の上面に図3(b)に示されるカバー16を重ねることにより、図2のマイクロチップ13が得られる。

図3(a)に示すように、基板15には、試料導入部7、試料回収部8、およびこれらを連通させる主流路11が形成されている。また、主流路11から分岐して副流路12が形成されており、副流路12はPCR反応部9に連通する。ここで、後述するように、PCR反応部9は最高95℃まで達するため、基板15には高温に耐え得る材料、たとえばガラス、石英、シリコン、耐熱性プラスチックなどを用いる。基板15の材料への各部材形成方法として、たとえばエッチング、レーザ加工、フォトリソグラフィー等の方法を採用することができる。

なお、図2中に波線で示した領域は、マイクロチップを駆動装置(不図示)にセットした際に、駆動装置に備えられている加熱冷却ユニットが位置する領域を表している。マイクロチップを駆動装置にセットした状態で、PCR反応部9と加熱冷却ユニットとが密着するようになっており、PCR反応部9の温度を制御できるようになっている。また、この際の熱交換が効率良く進行するように、PCR反応部9は図2に示されるような底面積の大きい直方体となっている。

次に、図2を参照して、マイクロチップ13の動作についてDNAの増幅を例として説明する。

まず、試料導入部7よりポリメラーゼ連鎖反応(PCR)用バッファーを注入する。ここで、試料導入部7および試料回収部8は、バッファーのリザーバーとしての機能も兼ね備えている。試料導入部7および試料回収部8に注入されたバッファーの液面がPCR反応部9および主流路11の高さよりも高くなるようにする。また適宜PCR反応部9および主流路11中の気泡を除去する。こうすることにより、試料導入部7、試料回収部8、PCR反応部9および主流路11に液体が均一に行き渡り、以降の処理を円滑、確実に進めることができる。

次に、増幅しようとする領域を有するDNA、増幅しようとする領域の両端に相補的なプライマー、耐熱性DNAポリメラーゼ、および4種の5’デオキシリボヌクレオチド三リン酸(dATP、dGTP、dCTP、dTTP)を所定量ずつ試料導入部7に投入する。

その後、電極10aおよび10dをそれぞれ陰極および陽極として通電すると、マイナスに帯電したDNA、プライマー、耐熱性DNAポリメラーゼおよび4種の5’デオキシリボヌクレオチド三リン酸が電極10dに向かって移動する。このように、図2のマイクロチップではDNAやプライマー等が確実にPCR反応部9へと誘導される。その後、PCR反応部9においてDNA増幅反応を行うが、この間は電極10cを陰極、電極10dを陽極として電流を流す。こうして、DNAやプライマー等がPCR反応部9の外部へと拡散することを防止する。なお、図2の各電極間に形成する電界強度は、たとえば0.1V/cm以上20V/cm以下とする。本実施形態に係る核酸増幅装置では、従来電気泳動等を行う際に流す電流に比べて微弱な電流によって確実にPCR反応部9に試料を保持させることが可能となる。

PCR反応部9でのDNA増幅反応は、加熱冷却ユニットによる温度制御に従って進行する。すなわち、PCR反応部9をたとえば95℃で2分間保つことにより2本鎖DNAを1本鎖に変性させ、次いでPCR反応部9をたとえば50〜60℃付近で2分間保つことにより1本鎖DNAにプライマーをアニーリングさせる。その後、PCR反応部9をたとえば74℃で3分間保つことにより、DNAポリメラーゼによる鎖伸張反応を進行させる。以上を1サイクルとして、これをたとえば30サイクル繰り返す。

DNA増幅反応が終了した後は、電極10bおよび10dをそれぞれ陽極および陰極として電流を流すことにより、PCR反応により得られたPCR産物を試料回収部8へ誘導し、回収する。

また、得られたPCR産物の分析等を行うためにバッファーの交換をする必要がある場合には、PCR反応終了後、電極10cを陰極、電極10dを陽極とし、PCR産物をPCR反応部9内に保持した状態で、交換前のバッファーを圧力送液することにより試料導入部7から試料回収部8へとフラッシュする。その後、交換するバッファーを試料導入部7から注入する。そして、上述の場合と同様にPCR産物を試料回収部8へ誘導することによりバッファーが交換されたPCR産物を得ることができる。

このように、図2のマイクロチップでは、PCR反応部9が従来のキャピラリー内でPCR反応を行うマイクロチップに比べて大きく、またPCR反応部9が良好に温度制御されるため、種々の実験に利用するために十分な量のDNAが得られる。また、主流路11およびPCR反応部9が密閉された構成となっているため、簡易な構成でありながらPCR反応部9に反応液を維持することができる。さらに、PCR産物を試料回収部8に保持させた状態での圧力送液により、容易にバッファー交換を行うことができる。したがって、μTAS等のマイクロ化学反応リアクタに搭載した際にもPCR産物の処理や分析などのステップを最適なバッファー条件で連続的に実施することができる設計となっている。

以上、DNAの増幅を例に図2のマイクロチップの動作について説明したが、増幅対象の核酸がRNAである場合についても、同様に適用可能である。

なお、図2のマイクロチップでは、PCR反応部9を一箇所にのみ設けた場合を示したが、複数とすることもできる。図6は、3つのPCR反応部を含むマイクロチップの構成を示す上面図である。試料導入部7と試料回収部8は主流路21により連通されている。また、副流路22、副流路23、および副流路24にはPCR反応部9a、9bおよび9cが連通している。PCR反応部9a、9bおよび9cには、電極10d、10fおよび10hが設けられている。また、電極10d、10fおよび10hと対向する位置に電極10c、10eおよび10gが設けられている。マイクロチップをこのような構成とした場合、複数の試料を同時にまたは連続して処理することが可能となるため、より一層効率良く核酸を増幅することができる。

なお、本実施形態において、各電極に印加する電圧を制御することによりDNAをキャピラリーの一端に移動させ、これを保持することにより、DNAの保持された一端のみを加熱しても効率よくPCR反応が進行することが、実験例により実証された。以下このような実験例について説明する。

(実験例1)
本実験例では、図2の構成のマイクロチップに先立ち、PCR反応用キャピラリーを用い、これを加熱冷却ユニットに密着させてPCR反応を行った。図4は、本実験例に用いた加熱冷却ユニット101を示す図である。図4の加熱冷却ユニット101の作製は以下のようにして行った。

まず、20mm×20mmのアルミ角材を長さ65mmに切断し、断面の一部を図4のように10m×10mm角削除した。得られたL字型のアルミブロックに、幅a=1.5mm、深さb=2.5mmの短スリット103および長スリット105をそれぞれ形成した。得られた加熱冷却ユニット101は、所定の反応温度に加熱してPCR反応に用いた。加熱冷却ユニット101は3個用意し、それぞれ94℃、44℃、および72℃に設定されたヒートブロック上に固定した。

また、図5は本実験例に用いたPCR反応用キャピラリーを示す図である。図5のPCR反応用キャピラリーは、内径1mmのキャピラリー107に正極111および負極113となる電極109が設けられた電極付キャピラリーチューブである。

以上の実験例より、キャピラリー107に電圧を付与せずに全体を加熱した場合と同様に、キャピラリー107の両端に電圧を印加し、正極側のみを加熱した場合についても効率よくPCR反応が行われることが明らかになった。これは、キャピラリー107の両端に電圧を印加することにより反応成分が正極側に誘導され、誘導された正極側の末端が効率よく加熱されたためであると考えられる。したがって、実験例1を図2の構成のマイクロチップに適用することにより、DNA増幅を効率よく行うマイクロチップが得られる。
