本発明は、主としてマイクロアレイの作製のための液滴吐出ヘッド及びその製造方法並びにマイクロアレイの製造方法及び製造装置に関する。

【従来の技術】
遺伝子研究機関、医療、薬品開発の研究機関等においては、DNA解析や免疫学的解析等のために、スライドガラスのような基板上に、数千〜数万種類のDNA(デオキシリボ核酸)やRNA(リボ核酸)、タンパク質などの生体分子溶液をマトリックス状にスポットしたマイクロアレイ(バイオチップサンプル)が用いられる。このようなマイクロアレイを作製する方法としては、接触ピン法、マイクロピペット法、インクジェット法などが知られている。そのうち、特に最近ではインクジェット法が注目を浴びている。これは、インクジェット法が、極少量の液滴を目的の位置に高精度に打つことができること、吐出ヘッドが基板に接触しないこと、基板材料にガラスや合成樹脂、金属、フィルム等任意の材料が使用できること、動作音が低いこと、低コストであることなどの特徴を有しているからである。
インクジェット法は、インクジェットヘッドのアクチュエータ部を、例えば圧電体により変位させることにより、ダイヤフラムのポンプ作用でノズル先端から液滴を吐出させる方法である。

このようなインクジェット法を用いたものとして、例えば特許文献1がある。この方法は、ピペット本体に、ピペット本体の外部から試料を注入する試料注入口と、注入された試料を導入しかつ一時貯留し得るキャビティと、貯留された試料をノズル部の貫通孔を経由して外部に吐出する試料吐出口とを形成してなるとともに、ピペット本体のキャビティ形成位置に対応した部位の外面上に圧電/電歪素子を設けてなり、圧電/電歪素子の駆動によりキャビティ内の体積を変化させ、キャビティ内に貯留された試料の一定量を試料吐出口から吐出させるマイクロピペットを用いるものである。

しかしながら、前記のような従来技術では、複数の基板を用いてそれぞれ独立に、ノズル、試料注入口、キャビティ、および圧電/電歪素子等を構成する必要があるため、ノズル数が少数に限られ、かつ貯留し得る試料(生体分子溶液)の種類および数も少ないものとなる。したがって、数千〜数万種類のマイクロアレイを作製するのに、多数のマイクロピペットが必要になるだけでなく、非常に多くの時間がかかり、コスト高となる問題があった。

本発明は、インクジェット法を利用して、多種類のマイクロアレイを短時間に低コストで作製することができる液滴吐出ヘッド、及びその液滴吐出ヘッドを製造するための効果的な製造方法並びにマイクロアレイを製造するための効果的な製造方法及び製造装置を提供することを目的とする。

【課題を解決するための手段】
本発明に係る液滴吐出ヘッドは、複数の液体貯留部を有する第1の基板と、
前記複数の液体貯留部にそれぞれ独立に連通する複数の流路を有する第2の基板と、
前記複数の流路にそれぞれ独立に連通し、液滴を吐出する複数のノズルを有する一または複数のヘッドチップと、
を備えたことを特徴とする。

このように構成することにより、液体貯留部からヘッドチップのノズルに至るまでそれぞれ独立した一連の吐出径路が構成される。第1の基板にはM行×N列の液体貯留部が配列されるので、多種類のプローブサンプル(生体分子溶液のスポット)をインクジェット法により高精度、高密度に、かつ短時間に作製することができる。すなわち、本発明の液滴吐出ヘッドを使用することにより、高密度のマイクロアレイを短時間に低コストで作製することが可能となる。

本発明において、マイクロアレイとは、ガラス等の基板上に、数千〜数万種類のDNAやRNA、タンパク質などのプローブサンプルが配列されたものである。DNAマイクロアレイ(DNAチップともいう)がその代表的な例である。また、DNA(Deoxyribo Nucleic Acids:デオキシリボ核酸(遺伝物質))とは、塩基、糖(デオキシリボーズ)及びリン酸が結合したもの(ヌクレオチド)がさらに結合し、最終的には二重らせん形となったものである。ここで、塩基はアデニン(A)、グアニン(G)、シトシン(C)、チミン(T)の4種類がある。また、DNAマイクロアレイは、DNAの相補性を利用し、検査、解析に利用するものである。
RNA(ribonuclec acid:リボ核酸)は、リボヌクレオチドが重合した核酸で、リボソームRNA、伝令RNA、転移RNAなどがあり、核のみでなく細胞全体に分布している。塩基はアデニン(A)、グアニン(G)、シトシン(C)、ウラシル(U)の4種類である。また、1本鎖構造である。
タンパク質は、生物細胞に不可欠の主要成分として、20種のL−アミノ酸がペプチド結合することで合成される高分子ポリペプチドの総称であり、様々な分子量、構造、機能をもった生体高分子である。

また、本発明の液滴吐出ヘッドは、前記第1の基板と前記第2の基板と前記ヘッドチップとを互いに接合してなるものである。
これによって、本発明の液滴吐出ヘッドをカートリッジ式に構成することができるので、取り扱いや交換が容易となる。

また、本発明の液滴吐出ヘッドは、前記複数の液体貯留部を前記第1の基板において縦横等間隔に配置してなるものである。
液体貯留部には、例えば、DNA断片を含む溶液が入れられる。そこで、液体貯留部(すなわち、リザーバまたはウェル)をこのように縦横等間隔に配置することにより、例えば縦12個×横8個(合計96個)の配列のように規格寸法に合致したフォーマットの配列とすることができるので、生体分子溶液の注入を従来の自動分注装置やマイクロピペットを使用して、容易に行うことができる。

前述した本発明の液滴吐出ヘッドの製造方法は、複数の液体貯留部を縦横に配列した第1の基板を作製する工程と、
前記複数の液体貯留部にそれぞれ独立に連通するように複数の流路を形成する第2の基板を作製する工程と、
前記第1の基板と前記第2の基板を接合する工程と、
単独で液滴吐出機能を有する一または複数のヘッドチップをそれぞれ独立の吐出径路を有するように前記第2の基板に接合する工程と、を含むものである。
この製造方法により、前述した本発明の液滴吐出ヘッドを高精度に、かつ低コストで製造することができる。また、ヘッドチップは単独で液滴吐出機能を有するものとすることにより製造および品質管理を個別に行うことができるので、液滴吐出ヘッドとしての性能および歩留まりの向上を図ることが可能となる。
また、前記第2の基板を作製する工程において、前記複数の流路の一端部を前記ヘッドチップのノズルピッチと一致するように整列させる。
これにより、ヘッドチップの小型化とマイクロアレイの高密度化が可能である。

本発明に係るマイクロアレイ製造方法は、請求項1乃至5のいずれかに記載の液滴吐出ヘッド、または請求項6に記載の液滴吐出ヘッドの製造方法により製造された液滴吐出ヘッドを用いてマイクロアレイを製造するものである。
また、マイクロアレイの作製に際しては、前記液滴吐出ヘッドのヘッドチップのノズル全部を同時に吐出させる。
したがって、高密度のマイクロアレイを短時間に、低コストで作製することができる。

本発明に係るマイクロアレイ製造装置は、基台と、前記基台上において、直交する方向に相対的に移動するキャリッジおよびテーブルと、前記テーブル上にセットされた一または複数のマイクロアレイ作製用基板とを備え、請求項1乃至5のいずれかに記載の液滴吐出ヘッド、または請求項6に記載の液滴吐出ヘッドの製造方法により製造された液滴吐出ヘッドを前記キャリッジに着脱自在に搭載してなるものである。
したがって、本発明の液滴吐出ヘッドを用いて、高密度のマイクロアレイを短時間に、低コストで作製することができる。また、液滴吐出ヘッドをキャリッジに対して着脱自在とすることにより、液滴吐出ヘッドの交換が容易なため、例えば、DNAチップのプローブサンプルに、蛍光標識されたスポットをハイブリタイズさせる大量の反応試験を効率よく行うことができる。

また、前記キャリッジは、前記液滴吐出ヘッドの装着時、該液滴吐出ヘッドと電気的に接続する構成となっていることが好ましい。これにより、液滴吐出ヘッドの交換が容易になる。
また、前記キャリッジの底面に前記液滴吐出ヘッドのヘッドチップが嵌合する開口部を位置を異にして複数設けたものである。これにより、液体貯留部の配列およびヘッドチップの位置が異なる液滴吐出ヘッドに対しても、キャリッジを共通して使用することができ、別種の液滴吐出ヘッドの着脱、交換が容易となる。

【発明の実施の形態】
以下、本発明の実施の形態を図面により説明する。
実施の形態1.
図1は本発明の実施の形態1による液滴吐出ヘッドの構成図で、断面図で示すものである。図2および図3は液滴吐出ヘッドの平面図と下面図である。
この液滴吐出ヘッド10は、複数の液体貯留部21が形成された第1の基板20と、各液体貯留部21に独立に連通するように第1の基板20との間に複数の流路31が形成された第2の基板30と、各流路31に独立に連通するように複数のノズル(図8参照)が形成された複数のヘッドチップ40とから構成されている。以下、各部の構成について詳細に説明する。

(1)第1の基板(またはリザーバプレート)
第1の基板20は、図1、図2に示すように、ガラスまたはプラスチックからなり、吐出する液体(吐出液体)1として、例えばDNA断片を含む高分子溶液(生体分子溶液)が入れられる複数の液体貯留部21を有する。液体貯留部21はリザーバまたはウェルとして構成されており、したがって、第1の基板20をリザーバプレートと呼んでもよい。

液体貯留部21の個数、配列については特に限定されるものではない。この例では、縦128mm、横86mmの基板20に12×8=96個の液体貯留部21が縦横1cm弱の等間隔(等ピッチ)で格子状に配置されている。このような縦横等間隔(等ピッチ)の配列にすることにより、規格配列寸法(フォーマット)のマイクロタイタープレートに合わせた自動分注装置あるいはマイクロピペットを使用することができ、生体分子溶液を容易に各液体貯留部21に注入することができる。なお、フォーマットされたマイクロタイタープレートのウェル数は縦12個×横8個(計96個)から縦48個×横32個(計1536個)のものなどがあり、これらのフォーマットに合わせて基板20を作製することが好ましい。

液体貯留部21は、特に限定はないが、例えば、有底状の穴21aと、この穴の底部に設けられた小径の貫通孔21bとからなっている。これらの穴21a、21bは機械加工または射出成形等により形成されている。穴21aの大きさは、例えば、直径7mm×深さ10mm程度である。なお、穴21a全体を同じ径の貫通孔で形成してもよいし、あるいは射出成形により、図7に示すように、薄肉の第1の基板20と円筒状の液体貯留部21とを一体に形成してもよい。

(2)第2の基板(またはチャンネルプレート)
図4は第2の基板の平面図、図5は断面図、図6は一部の流路の拡大平面図である。
チャンネルプレートとも呼ばれる第2の基板30は、ガラスまたはプラスチックからなり、第2の基板30の表面には第1の基板20と接合することにより、液体貯留部21にそれぞれ独立に連通する微小な流路31を構成することになる溝31bが形成されている。流路31の一端31aは液体貯留部21の小径の貫通孔21bに連通するように形成されている。また、L字状に屈曲形成された細い溝31bの他端31cは第2の基板30の任意の部位(例えば、基板20の下辺部)に集合して整列させ、さらにヘッドチップ40との接続部となる微小な貫通孔31dに接続することにより、各流路31が構成されている。この例では、縦1列の液体貯留部21の数(12個)に相当する12本分ずつの流路31をそれぞれ一つのグループとしてまとめて整列している。なお、流路31は、通常、ガラスではエッチング、プラスチックは射出成形により形成される。

(3)第1の基板と第2の基板の接合
前記のように構成された第1の基板20と第2の基板30は、好ましくは熱溶着により接合される。すなわち、第1の基板20と第2の基板30を積層状態に接合する。そして熱溶着に際し、流路31の一端31aを液体貯留部21の貫通孔21bに位置合わせして接合する。この熱溶着によって、第2の基板30に形成された各流路31は第1の基板20により被蓋され、各々の液体貯留部21と流路31とがそれぞれ独立に連通する状態となる。なお、接着剤を用いて第1の基板20と第2の基板30を貼り合わせてもよい。この場合、接着剤が流路31の溝31bや貫通孔31d内にはみ出さないように塗布することが肝要である。

(4)ヘッドチップ
ヘッドチップ40は、電気的に接続するだけで単独で液滴吐出機能を有するもの(小型の組立体)である。ヘッドチップ40の構成については図8で詳しく説明する。ここでは、ヘッドチップ40を図1、図3に示すように、第1の基板20に接合または接着された第2の基板30の下面に接着する。そして接着に際し、ヘッドチップ40の流路を第2の基板30における流路31の貫通孔31dに位置合わせして接合する。ヘッドチップ40は、この例では、12個のノズルを有し、8個のヘッドチップ40が第2の基板30の下面下辺部に並べて接合されている。
この接着によって、ヘッドチップ40の各ノズルが、対応する流路31を介して、対応する液体貯留部21と独立に連通する状態となり、それぞれ独立した一連の吐出径路11が構成される。
以上により、この液滴吐出ヘッド10の組立が完了する。

図8は液滴吐出ヘッドにおけるヘッドチップの拡大断面図である。ここでは、一例として静電駆動方式のヘッドチップを示す。
このヘッドチップ40は3枚の基板41、42、43により構成されている。第2の基板30に接合される上側の基板41はガラス基板が用いられ、中間部の基板42にはSi基板を、下側の基板43にはSi基板をそれぞれ用いている。これら3枚の基板のうち基板41、42は陽極接合、基板42、43は接着により相互に接合されている。

上側のガラス基板41には、予め、第2の基板30における流路31の貫通孔31dに連通する貫通孔41aと、アクチュエータ電極41bが形成される。アクチュエータ電極41bは、Si基板42の振動板42cとの間に所定の空隙を確保するために、凹部41c内に形成することが好ましい。このアクチュエータ電極41bとして、ここでは、酸化錫を不純物としてドープした酸化インジウム膜、すなわちITO(Indium Tin Oxide:インジウム酸化第1錫)膜で電極を形成している。ただし、電極41bはこれに限定されるものではない。

中間のSi基板42には、予め、貫通孔41aに連通する貫通孔42aと、この貫通孔42aに流路を介して連通する吐出室42bがエッチングにより形成されている。吐出室42bの底部(ここでは天井壁)が振動板42cとして構成され、アクチュエータ電極41bに所定の空隙でもって対面している。
下側のSi基板43には貫通孔42aと吐出室42bとを結ぶ流路43aと、吐出室42bに連通するノズル43bがエッチングにより形成されている。

前記のように構成された3枚の基板41、42、43を相互に接合した後、切断することによって、一つのヘッドチップ40が作製される。このようにして組み立てられたヘッドチップ40を、前述のように第1の基板20に接合された第2の基板30の下面に、貫通孔31dと貫通孔41aとを一致させて接合することにより、液滴吐出ヘッド10を構成することができる。そしてさらに、アクチュエータ電極41bとSi基板42に発振回路44をフレキシブル回路基板45にて配線することにより、静電駆動方式の液滴吐出ヘッド10が完成する。

第1の基板20の液体貯留部21に吐出液体を注入すると、吐出液体は毛細管力により流路31、吐出室42b等を経由してノズル43bの開口先端まで充填され、ノズル開口先端では表面張力により、いわゆるメニスカスを形成するため、液漏れは生じない。充填が不充分な場合には吸引ポンプを用いる。
その後、発振回路44を駆動することにより、目的のノズル43bから液滴を吐出させることができる。また、ヘッドチップ40内のSi基板42上の電極47を共通電極に構成し、全アクチュエータを同時に駆動することにより、ヘッドチップ40内の全ノズル43bから同時吐出させることができる。

静電駆動方式においては、アクチュエータ電極41bを正に帯電させ、振動板42cを負に帯電させる。このため、振動板42cが電極41b側へ引き寄せられ、吐出室42bの容積が増加する。この状態で発振回路44によりアクチュエータ電極41bへの電荷の供給を止めると、振動板42cは元に戻るため、そのときの圧力により容積変化の差分に相当する一定量の液滴がノズル43bより吐出される。
なお、ここでは、液滴吐出駆動方法を静電気を利用した静電駆動方式で説明したが、圧電体を用いる圧電駆動方式としてもよい。

本実施形態の液滴吐出ヘッド10においては、複数の液体貯留部(またはリザーバ)21がM行×N列で配置されており、かつ、各液体貯留部21からノズル43bに至るまでの一連の吐出径路11はそれぞれ独立に構成されているので、多種類のプローブサンプルをスライドガラス等の固相上にスポット(吐出)することができる。また、第1の基板20上の縦1列分の液体貯留部21が流路31を介して一つのヘッドチップ40に集合させて接続されているので、ヘッドチップ40ごとに同時に吐出させることにより、縦1列分のスポット、例えば12個のスポットを同時に作製することができる。したがって、この場合、8個のヘッドチップ40について8回の同時吐出を行うことにより、96個(96種類)のプローブサンプルのスポットを短時間に作製することができる。しかも、スポットの中心間隔はヘッドチップ40におけるノズル43bのピッチで設定されるため、高密度のマイクロアレイの製造が可能である。

さらに、ヘッドチップ40は第1の基板20、第2の基板30に比べてはるかに小さく、しかも単独で液滴吐出機能を有する構成部品(組立体)であるため、個別に製造、品質管理を行うことができるとともに、ヘッドチップ40自体の低コスト化が可能であることから全体として液滴吐出ヘッド10の低コスト化が可能である。また、第1の基板20および第2の基板30も接合により一体化されるので、接合体(積層組立部品)として個別に品質管理の対象とすることができ、性能の向上、歩留まりの向上を図ることができる。したがって、高品質の液滴吐出ヘッド10を安価に提供することができる。

実施の形態2.
図9は本発明の実施の形態2による液体吐出ヘッドの平面図である。ここでは、2つの配置例(a)、(b)を示している。また、図10は断面図である。
既に明らかなように、第1の基板20に形成される液体貯留部21、および第2の基板30の下面に接合されるヘッドチップ40の配置、個数、大きさ等は使用目的に応じて自由に設計することができる。

図9(a)は、1個のヘッドチップ40を基板の中央部に配置し、液体貯留部21を第1の基板20の左右部分に各々2列で配置する場合である。液体貯留部21の数は例えば1列10個としている。したがって、合計40個(種類)のプローブサンプルを作製することができる。また、前述のように、各液体貯留部21とヘッドチップ40とを結ぶ独立の流路31が第2の基板30に形成され、この第2の基板30は第1の基板20の裏面に接合または接着されている。
このヘッドチップ40は左右2列のノズル配列となっており、ノズル43bは縦方向に所定のピッチPaで配列されている。
したがって、本実施形態の液滴吐出ヘッド10によれば、左右2列のアクチュエータ電極41bを同時駆動することにより、40種類のプローブサンプルを同時に作製することができる。

実施の形態3.
図11は本発明のマイクロアレイ製造装置の概要を示す斜視図、図12は液滴吐出ヘッドを搭載したキャリッジの断面図、図13および図14はキャリッジの平面図と側面図である。
本実施形態のマイクロアレイ製造装置100は、基台110と、基台110上において、水平面内でX方向に移動するキャリッジ120と、水平面内でX軸と直角のY方向に相対的に移動するテーブル130と、キャリッジ120に着脱自在に搭載される前記液滴吐出ヘッド10と、テーブル130にセット(載置)されるスライドガラスや樹脂製等からなるマイクロアレイ作製用基板140と、さらにX軸(キャリッジ)およびY軸(テーブル)の駆動制御手段150とを備えている。また、図示しない吐出制御手段は一般的にPC(パーソナルコンピュータ)と専用回路にて構成されている。

キャリッジ120には、この例では、第1または第2の実施形態で示した液滴吐出ヘッド10が着脱自在に搭載される。キャリッジ120の底面には、図13に示すように、細長い開口部121が設けられており、この開口部121に液滴吐出ヘッド10に設けられた全部のヘッドチップ40が嵌合するようになっている。このとき、図12に示すように、ヘッドチップ40の先端面すなわちノズル開口面43cがキャリッジ120の下面にほぼ一致するように、液滴吐出ヘッド10がキャリッジ120に装着される。したがって、液滴の吐出距離はキャリッジ120の下面とテーブル130上のマイクロアレイ作製用基板140の表面間の空隙間隔Gで設定することができる。

開口部121の近傍には液滴吐出ヘッド10の装着時、各ヘッドチップ40に接続した、フレキシブル回路基板上の接点と接触し電気的な接続をする電極122、123が設けられている。電極122、123は、配線を介してコネクタ124に接続されており、さらに前記吐出制御手段へと接続されている。なお、第2の実施形態の液滴吐出ヘッド10を使用する場合は、図15に示すように、電極122、123は2つでよい。

また、第2の実施形態のように基板の中央部にヘッドチップ40を設けた液滴吐出ヘッド10を使用する場合には、開口部121はキャリッジ120の底面のほぼ中央部に設けられる。したがって、例えば図15に示すように、位置の異なる複数の開口部121と電気的接続のために配線された複数の電極122、123とをキャリッジ底面に設けておけば、多様な形態の液滴吐出ヘッド10に即応することができ、着脱・交換が容易となる。
このように、液滴吐出ヘッド10はキャリッジ120に対して着脱自在なカートリッジ式の構成となっている。さらに、液滴吐出ヘッド10の着脱が容易なように、キャリッジ120の両側面には指先が入るような円弧状の切欠き部125が設けられている。なお、126は液滴吐出ヘッド10を安定に支持するための支持部である。

キャリッジ120はX軸駆動機構によってX方向に走査される。X軸駆動機構は図示していないが、公知のタイミングベルト機構やボールネジ機構等を用い、これにキャリッジ120を結合してパルスモータを駆動することで実施できる。図11では、このようなX軸駆動機構が側面に長穴103を設けたX軸ダクト101内に内蔵されているものである。
テーブル130はY軸駆動機構によってY方向に走査される。このY軸駆動機構は前記X軸駆動機構と同様の構成であり、側面に長穴104を設けたY軸ダクト102内に内蔵されている。
なお、テーブル130上にはマイクロアレイ作製用の基板140が1枚または複数枚セット(載置)される。また、テーブル130は固定式としてもよい。この場合は、キャリッジ120を直交するX、Y両方向に走査可能な構成とする。

本実施形態のマイクロアレイ製造装置では、例えば、第1の実施形態で示した液滴吐出ヘッド10をキャリッジ120に搭載する。各液体貯留部21にはそれぞれ異なる種類の生体分子溶液が注入され、液体貯留部21からヘッドチップ40のノズル43b先端まで充填されている。また、液体貯留部21の配列を縦横等間隔とすることにより、前述のように自動分注装置等を使用して容易に生体分子溶液を注入することができる。
