Abstract
Description
グし細胞増殖状態を確定し種々の成熟段階で正常細胞と異常細胞(白血性骨髄単
球細胞など)を識別する方法、診断と治療の方法、並びにそうした方法に使用す
るリガンドを含めた試薬に関する。本発明は、新たに特性を解明されクローニン
グされた細胞表面受容体p75/AIRM1に対して特異的なリガンド類の同定
、種々の分化段階の骨髄性細胞表面でのp75/AIRM1の発現を示す実験結
果、それに細胞増殖の阻害と細胞アポトーシスの誘発のための該受容体に対応す
るリガンドの使用に、部分的に依拠する。
た特定の転写因子の発現に、依存する。この造血過程は種々の細胞分化段階の認
識を可能にする細胞表面マーカーの逐次発現を特徴とする。たとえばCD34表
面マーカーを発現する細胞は多分化能の造血胚芽細胞を含むが、CD33はそう
した造血胚芽細胞表面には存在しない。他方、CD33は骨髄単球前駆細胞表面
で、並びに骨髄性及び単球細胞表面で発現するが、成熟顆粒球表面では消失する
。
その機能はほとんどわかっていない。ナチュラルキラー(NK)細胞表面で発現
するNK細胞機能調節能をもつ抑制性受容体が本発明者の所属する研究所で同定
され、特性を解明された(Falco他「Journal Experimen
tal Medicine」vol. 90, No. 6,1999年9月2
0日、第793〜802頁)。この受容体はp75/AIRM1 (75 kD
adhesion inhibitory receptor molecu
le 1、75 kDの接着抑制受容体分子1)と命名されたが、その完全なア
ミノ酸配列とそれをコードするヌクレオチド配列はFalco他の図5に示され
ている。同資料は本書で言及する他の諸々の公開資料と共に、参照指示により本
書に組み込まれる。

きわめて有用なことである。p75/AIRM1受容体の同定は、骨髄性細胞の
種々の成熟段階のより厳密な定義を可能にする。これは白血病の正確な類型化に
有用であろうから、その予後及び治療処置に役立つと期待される。

うな抗体がp75/AIRM1受容体分子と相互作用を起こすと、骨髄性細胞が
増殖を停止させられ、またプログラム細胞死すなわちアポトーシスが誘発される
という事実によって立証される。この効果はすでに正常及び白血病骨髄性細胞の
両方で、インビトロで裏付けられており、白血病細胞(たとえば化学療法後の残
存白血病細胞)の一掃を目的とした治療的介入のための重要な分子的土台となる
。
類の使用により正常及び白血病骨髄性細胞を識別する方法、そうしたリガンド類
、及びその診断治療面への使用に関する。
ナル抗体又はその「ヒト化」モノクローナル抗体、あるいは可溶性組換え分子等
のような断片(単鎖Fv、すなわち、scFv抗体分子など)であろう。さらに
他の有用なリガンド類として、p75/AIRM1受容体分子を認識すること及
び/又はそれと特異的に結合することができる他の諸々の分子(たとえば、ペプ
チドや医薬製剤など)がある。
抗体分子の例は免疫グロブリンの各種アイソタイプ又はそのサブクラス、それに
Fab、scFv、Fv、dAb、Fd及び二重特異性抗体などのような抗原結
合ドメインを含む断片である。抗体は様々な方法で修飾することができるので、
用語「抗体分子」は免疫グロブリンの抗原結合ドメインを含む任意のポリペプチ
ドを、天然分子か部分もしくは完全な合成分子かを問わず包含するものとする。
よって、別のポリペプチドと融合させた免疫グロブリン抗原結合ドメインを含む
キメラ分子も包含される。特異的結合メンバーは抗体配列の他に別のアミノ酸を
含めて、たとえば折り畳まれたドメインなどのようなペプチド又はポリペプチド
を形成するように、又は抗原結合能に加えてもう1つの機能的特性を抗体分子に
付与するようにしてもよい。本発明の特異的結合メンバーは検出可能な標識を含
んでもよいし、毒素又は酵素と(たとえばペプチジル結合又はリンカーを介して
)複合体を形成してもよい。本発明では、抗体分子などのようなリガンドをたと
えば抗白血病の見地から、細胞障害作用を増強しうる放射性同位体、毒素、エミ

が大きい場合には、抗体は抗原の特定部分に結合するだけであろうが、抗体が結
合するその特定抗原部分をエピトープという。抗原結合ドメインは1以上の抗体
可変領域(たとえば、VH領域からなるいわゆるFd抗体フラグメント)に由来
しよう。好ましくは、抗原結合ドメインは抗体L鎖可変領域(VL)と抗体H鎖
可変領域(VH)を含む。
分子に対しては有意の結合を示さないであろう。この用語はまた、たとえば多数
の抗原に含まれる特定のエピトープに対して抗原結合ドメインが特異的である場
合にも適用されるが、その場合、抗原結合ドメインを含む特異的結合メンバーは
該エピトープををもつ種々の抗原に結合することができよう。 用語「含む」は一般に「包含する」、すなわち1以上の特徴又は構成要素の存
在を許容するという意味で使用される。
又はそうした結合メンバーをコードする核酸が、一般に本発明に従って使用され
る際の状態をいう。これは、当のリガンド又は核酸が、自然の環境あるいはそれ
らがインビトロ又はインビボの組換えDNA技術により調製される場合にはその
調製される環境(例:細胞培養)で、自然状態で結合している物質、たとえば、
他のポリペプチド又は核酸を含まない又は実質的に含まないことを意味する。リ
ガンド又は核酸は希釈液又はアジュバントと調合されることもあろうが、それで
もなお実際目的上は「単離された」状態にあるとされよう。たとえば、リガンド
は免疫検定でマイクロタイタープレートのコートに使用する場合にはゼラチンま
たは他の担体と混合するのが普通であろうし、診断又は治療に使用する場合には

の抗原結合ドメインを含む。これはマウスモノクローナル抗体であり、当業者は
ごく普通の実験手法により寄託ハイブリドーマ細胞から該抗体のL及びH鎖コー
ド配列を単離することが十分に可能である。これらを組換え発現系にそのまま使
用すれば該抗体を産生させることができよう。さらに、QA79 HV及びLV
可変領域をコードする核酸を使用すればscFv、Fv及びFab抗体などのよ
うな抗体分子をコードする配列を簡単に構築することができるので、当業者は自
由に使える多様な既定技術のうちの任意のものに依拠した組換え発現によりこれ
らの分子を産生させることができる。

が考えられる。該レポーター分子は、検出可能な、そして好ましくは測定可能な
シグナルを直接又は間接に生成しよう。レポーター分子の連結は直接結合でも間
接結合でも、また共有結合(たとえばペプチド結合による)でも非共有結合でも
よい。ペプチド結合による連結は、抗体分子とレポーター分子をコードする遺伝
子融合体の組換え発現の結果でもよい。

加した増殖培地を3日毎に、またその後は毎週、交換した。

により判明した。また、p75/AIRM1はほとんどのCD33+細胞表面で
発現するがCD14+細胞表面では発現しないことが骨髄系細胞集団を対象にし
た解析から判明した。CD33は単離CD34+細胞表面に様々な割合で認めら
れたりまったく認められなかったりしたが、p75/AIRM1は一貫してCD
34+細胞集団中に存在しなかった。これは、骨髄系細胞の発生段階でpd75
/AIRM1のほうがCD33よりも遅く発現することを示唆する。この点は、
CD34+細胞集団をSCFとGM−CSFを添加して培養し、CD33、p7
5/AIRM1両マーカーの発現を調べるという実験により確認された。
る正常及び白血病骨髄性細胞へのプログラム細胞死及び細胞増殖阻害の導入 CD34+臍帯血由来細胞を96平底穴プレートに20,000個/穴の濃度
で塗布し、GM−CSF (100ng/ml)とSCF (50ng/ml)
を添加した完全増殖培地中で培養した。CML(慢性骨髄白血病)細胞を6平底
穴プレートに50,000個/穴の最終濃度で塗布し、GM−CSF (100
ng/ml)だけを添加した完全増殖倍地中で培養した。両細胞型はまた、それ
ぞれ表示の増殖因子を加えたうえで、ヤギ−抗マウス (10μg/ml)をコ
ートした96平底穴プレートにも20,000個/穴の濃度でまいた。

活性化は、正常骨髄性細胞とCML(慢性骨髄白血病)細胞の両方に対して強い
増殖阻害作用を及ぼした。抗p75/AIRM1抗体はまた、(GM−CSFの
存在下で培養すると)高インビトロ増殖速度を示すAML(急性骨髄白血病)の
細胞試料にも強い阻害作用を及ぼすことが判明した。 患者から単離したCML細胞のうち、抗p75/AIRM1による最大の阻害
作用が観察されたのは最高レベルの抗原発現を示す細胞であった。
はアポトーシスによる細胞死過程の開始指標である。アネキシンVの発現は間接
免疫蛍光分析及び細胞蛍光分析に特異的mAb類を使用して評価した。細胞表面
のアネキシンVとの結合、光散乱特性の変化及びヌクレオソームDNAの断片化
はアポトーシスによる細胞死の発生を表示する。
