Claims (1)
- 【特許請求の範囲】 1.cp4/epsps酵素活性を発現するサトウダイコン植物(その子孫を含む)。 1記載の植物。 3.そのゲノムDNAを鋳型としたPCR増幅で、配列番号:18および配列 番号:21の配列により特徴付けられるオリゴヌクレオチドの対を使用したとき 、739bpDNAフラグメントの増幅がもたらされることを特徴とする、請求項 1記載の植物。 4.配列番号:27により特徴付けられるDNAが植物ゲノムの一部を形成す る、請求項3記載の植物。 5.そのゲノムDNAを鋳型としたPCR増幅で、配列番号:20および配列 番号:24の配列により特徴付けられるオリゴヌクレオチドの対を使用したとき 、834bpDNAフラグメントの増幅がもたらされることを特徴とする、請求項 1記載の植物。 6.そのゲノムDNAを鋳型としたPCR増幅で、配列番号:17および配列 番号:22の配列により特徴付けられるオリゴヌクレオチドの対を使用したとき 、1057bpDNAフラグメントの増幅がもたらされることを特徴とする、請求 項1記載の植物。 7.そのゲノムDNΛを鋳型としたPCR増幅で、配列番号:19および配列 番号:25の配列により特徴付けられるオリゴヌクレオチドの対を使用したとき 、1224bpDNAフラグメントの増幅がもたらされることを特徴とする、請求 項1記載の植物。 8.配列番号:1により特徴付けられるDNAが植物ゲノムの一部を形成する 、請求項1記載の植物。 9.該ヌクレオチド配列が非常に反復性のDNA配列を置換する、請求項8記 載の植物。 10.該ヌクレオチド配列に直接結合するゲノムの一部が配列番号:2および 配列番号:3のヌクレオチド配列により特徴付けられる、請求項8記載の植物。 11.配列番号:4により特徴付けられるDNAが植物ゲノムの一部を形成す る、請求項9記載の植物。 12.請求項1から11のいずれかに記載の植物の種子。 13.受託番号NCIMB97087006/2で寄託されている、請求項8記載の植物の種 子。 14.(a)cp4/epspsをコードするDNAを1個含むベクターを保持するAgrob acteraで、インビトロで生育させたサトウダイコン子葉を形質転換する; (b)グリフォセート存在下で芽を再生させる; (c)芽を温室内の土壌に移植する; (d)苗木をグリフォセートで処理する; (e)植物の成長力および葉の退緑を肉眼で等級付する; (g)グリフォセート処理で影響を受けていない正常な成長力および葉の完全に正 常な植物を選択する;そして (h)慣用の育種法を使用して選択した植物を繁殖させる ことを含む、請求項1記載のトランスジェニックサトウダイコンの製造法。 15.cp4/epspsタンパク質をコードするベクターが、配列番号:5に記載の ヌクレオチド配列を有するDNA1個を含むベクターである、請求項14記載の 方法。 16.再生芽のcp4/epspsコードDNAの存在に関してPCRで分析する、請 求項14記載の方法。 17.植物内でのcp4/epspsの発現を可能にする遺伝子でのAgrobacterium介在 形質転換により得られ、右および左T−DNA境界配列の両方を欠く植物である 、グリフォセート耐性サトウダイコン植物(その子孫を含む)。
