ホモジニアスルミネセンスエネルギー転移アッセイ
発明の分野
本発明は、検出に時間分割蛍光測定法を用いるエネルギー転移ベースのホモジ
ニアスアッセイの改良に関する。その特定の改良は、特定の型のエネルギー供与
体として用いられるランタニドキレートラベル、所定のアッセイのための最適化
されたエネルギー受容体に関する。ここで、エネルギーが転移する道は最適化さ
れたフィルター及び時間窓を用いて測定され、サンプル由来の全てのおこり得る
干渉を正す方法は、多成分分析のためのアッセイ及び単純化したアッセイプロト
コルの開発を用いる。
発明の背景
本発明の背景を明らかにするために本明細書に用いられる出版物及び他の材料
、特にその実施に関する更なる詳細を供するものは引用により組み込まれる。
バイオアフィニティー又は酵素触媒反応に基づく極めて多種のアッセイが、種
々の生物サンプル(例えば血清、血液、血漿、唾液、尿、便、精漿、汗、分泌液
、羊水、組織ホモジネート、腹水等)、環境の研究(廃水、土壌サンプル)、工
業的工程(工程の溶液、生成物)及び化合物ライブラリー(有機化合物、無機化
合物、天然の産物、生物源の抽出物、生物タンパク質、ペプチド、又はヌクレオ
チド等)におけるサンプルから生物学的に重要な化合物を分析するために開発さ
れている。これらのアッセイのいくつかは特定のバイオアフィニティー認識反応
に依存し、ここでは、(生物学的に結合
する構成物、例えば抗体、天然のホルモン結合タンパク質、レクチン、酵素、レ
セプター、DNA,RNA又はPNAとの)特異的結合アッセイ、又は遺伝的にもしくは
化学的に工作された抗体のような人工的に作られた結合性化合物、モールディド
プラスチックインプリント(分子インプリンティング)等を形成するために一般
に天然の生物結合性構成物が用いられる。これらのアッセイは、一般に、認識、
結合反応及び適切な分離(沈殿及び遠心、ろ過、例えばプラスチック表面、例え
ばコートしたアッセイチューブへのアフィニティー収集、マイクロビーズのスラ
イド、溶媒抽出、ゲルろ過又は他のクロマトグラフィーシステム等のような分離
)後に形成された複合体を定量するためのラベルに依存する。遊離した又は結合
した画分におけるラベルの定量は、一般に未知のサンプルを比較する一セットの
標準を用いて、直接的又は間接的なサンプル中の被検体の計算を可能にする。
これらのアッセイのほとんどにおいて必要とされる分離及び洗浄は、労力を増
やし、遅くし、そして自動化を難しくする。更に、終点測定は速度論的情報(会
合/解離速度)の採取を許容しない。低アフィニティー結合の場合そのアフィニ
ティーが低すぎて、その結合を破壊することなく物理的分離を適用することがで
きない(低アフィニティーレセプター)。特に、スクリーニング(高スループッ
トスクリーニング)のような領域において、自動化をより簡単にし、かつスルー
プットを増加させるであろうより簡単なアッセイ、単純化されたプロトコルにつ
いての一定の要求がある。それは、ホモジニアス又は非分離アッセイで達成する
ことができる。ホモジニアスバイオメディカルアッセイは、一つの均一な相にお
いて行われるアッセイとして定義される。それは、分離した相(例えば試薬を捕
獲する固相)、及び分離を測定前に用いないことを意味する。それ
は、その直接的な監視を可能にする方法において結合アッセイに応答するシグナ
ル生産システムを要求する。先行技術で知られているシステムは、例えば小分子
化合物に適用される蛍光分極アッセイ、酵素モニターイムノアッセイ(Syvaによ
り市販)、種々の蛍光消滅
of Fluorescence in Immunoassays,Wiley,NY,1991を参照のこと)である。単
純化したアッセイ技術の他のカテゴリーは、ホモジニアスアッセイに似て、分離
及び洗浄ステップを避ける非分離アッセイである。この技術の優れた例は、アッ
セイ媒体中の放射性粒子の短距離浸透及び捕獲性試薬でコートされた固体シンチ
レーターに基づくAmershamにより市販されるシンチレーション近接原理である(U
denfriendら(1985)Proc Natl Acad Sci,82,8672及びAnal Biochem,(1987
)161,494)。
今日までに出版されている多数のホモジニアスアッセイデザインにもかかわら
ず、融通性及び感度が優れた分離アッセイのものに適合するであろうアッセイは
ない。その理由は、例えばアッセイが最適化されるホモジニアス対ヘテロジニア
スの異なる方法、低アフィニティー非特異的結合の制御、及びほとんどの存在す
るホモジニアスアッセイ技術の適用性の制限に何倍も関係している。更に、慣用
的なホモジニアス蛍光測定アッセイは、サンプル中の種々の構成物由来のバック
グラウンド干渉に極めて影響を受けやすい。蛍光分極アッセイは、低アフィニテ
ィー非特異的結合(例えばアルブミンに結合するプローブ)及びサンプルの自己
蛍光により妨害される。
時間分割蛍光測定(マイクロ−又はミリ秒範囲での時間域における時間分割)
は、ホモジニアスアッセイのための完全に測定する様式である。なぜならそれは
長い十分な遅延時間(パルス化された励起と放射の記録の開始との間の時間)を
用いる時に有機化合物由来
のバックグラウンド蛍光を全体的に識別することができるからである(報告につ
いて、例えばHemmila(1991);Gudgin Dickinsonら、(1995)J Photochem Photob
iol 27,3)。分離ベースのアッセイに加えて、いくつかのホモジニアス時間分割
蛍光測定アッセイも開示され、特許されている(Mathis(1995)Clin Chem,41,
1391;Selvinら(1994)Proc Natl Acad Sci,USA,91,10024)が、それらには制
限及び欠点がある。本発明の目的は、一般に、長寿命キレートベースの供与体プ
ローブと短寿命(又は非放射性)受容体分子との間の特定のエネルギー転移に基
づく時間分割ホモジニアスアッセイ原理における改良を供することである。
共鳴エネルギー転移
ー(受容体の吸収を伴う供与体の放射)がオーバーラップする条件において2つ
の分子間でおこり、それらは20nm未満の距離である。そのエネルギー転移は、そ
の分子の振動の正確な配向を要求する。
エネルギー転移効率(ΦET)は次式:
ΦET=1/〔1+(r/R0)6〕
(式中、rは供与体分子と受容体分子との距離であり、そしてR0は供与体−受
容体対及びそれらの間の媒体を特徴づける距離パラメータである)
で供される。
蛍光共鳴エネルギー転移(FRET)は、例えば、高分子中の距離を測定するため
に構造研究における顕微鏡ルーラーとして適用されている(Stryer及びHaugland
(1967)Proc Natl Acad Sci,USA,58:719)。共鳴エネルギー転移に加えて、
他のエネルギー転移反応、例えば単純な放射性(供与体により放射される光を受
容体が吸収す
る場合)、衝突エネルギー転移、変換(Dexter(1953)J Chem Physics,21,836
)及び励起子マイグレーション(結晶内)もある。更に、供与体放射は、いくつ
かの供与体のエネルギーレベルへの不活性化効果を有するいくつかの非関連の化
合物で、多数の方法により消光することができる。
Ullmanは、Syva Co.によりFETIA(蛍光エネルギー転移イムノアッセイ)として
市販される抗体認識反応に基づく生物分析アッセイにおけるフォースター型非放
射性エネルギー転移(Ullman,Scharzberg及びRubenstein(1967)J Biol Chem
,251:4172)の適用を最初に開示した(US 3,996,345)。適切なエネルギー供与
体−受容体対の開発はそれらの種々の調査(Ullman及びKhanna(1981)Methods En
zymol,74:28;Khanna及びUllman(1980)Anal Biochem,108,156)に十分に記
載されている。FETIAは、主に、供与体としてキサンテン色素及びフルオレセイ
ンの誘導体を適用する。多数の他のプローブ対もこれまで開発されており、イム
ノアッセイにおいて適用
)。このアッセイには、フルオレセインの実用的な非蛍光誘導体、長寿命遅延蛍
光放射性エオシン(Thakrar及びMiller(1982)Anal Proc,19,329)、長寿命蛍
光ピレン(Morrison(1988)Anal Biochem,174,101)及び非放射性チャコール
がある。FRETはこれまで、基本的な調査及びDNAハイブリダイゼーション(例え
ばMorrisonら(1989)Anal Biochem,183,231;Parkhurstら(1995)Biochemistr
y,34,285)において広範囲に適用されており、他のアッセイは、会合、解離又
は距離を測定することである。
受容体分子の直接の励起の効果を避けるための一時的識別(時間分割)の使用
が、長く励起された状態の供与体としてのピレン、励起のためのパルスレーザー
及び短い減衰時間の受容体を用いて、異
なる減衰時間の有機供与体−受容体対で、Morrison(Morrison(1988)Anal Bio
chem,174,101)により記載される(US 4,822,733)。イムノアッセイに加えて
、時間分割エネルギー転移原理は、受容体としてフルオレセインを用いるホモジ
ニアス溶液ハイブリダイゼーションのために適用される(Morrisonら(1989)II
Ird International Symposium on Quantitative Luminescence Spectrometry in
Biomedical Sciences,Ghent Belgium)。長く励起された状態は、直接励起さ
れたアクセプターの放射が減衰する時間である遅延時間を用いて特定のエネルギ
ー転移を行うことができるという利点を供する。時間分割蛍光測定法において利
用される異なる減衰時間の組合せは、慣用的な短い減衰プローブを用いるFRET技
術に優る明らかな利点を供するであろう。
ルミネセンス(蛍光)のランタニドは、DNA及びタンパク質構造研究のための
道具として長く用いられており(例えばBrittain(1985)Bioinorganic Lumines
cence Spectroscopy,Schulman(ed),Mo
pin(1989)Lanthanide Probes in Life,Chemical and Earth Sciences,Theory
and Practice,Elsevier Science Publisher,Amsterdamを参照のこと)、例え
ばRNA、リボソーム、DNA及びクロマチン、テルモリシン及びトランスフェリン、
カルモジュリン、ATPase、神経膜、赤血球膜タンパク質、リン脂質及びリポソー
ム並びにそ
照のこと)。カルシウム結合タンパク質又はDNAの異なる部位に結合したランタ
ニド間のエネルギー転移は、例えばEuとNdとの間のエネルギー転移を用いてカル
モジュリンとの、イオン結合部位の位置を測定するために微視的ルーラーとして
用いられている(Horrocks及びTingey(1988)Biochemistry,27,413)。TbからE
uへのエネル
ギー転移を測定することによるキレート研究において類似するFRETが用いられて
いる(Brittain(1978)Inorg Chem,17,2762)。
ランタニド及びそれらのキレートは時間分割FRET実験のための優れた候補であ
る。第1に、それらは例外的な長く励起される状態の寿命を有する(Weissman(1
942)Chem.Phys,10,214;Whan及びCrosby(1962)Mol Spectrosc,8,315)。そ
のエネルギー転移は、電気的双極子である遷移間でのみおこる。612〜620nmにお
ける高い蛍光のEuキレートにおける主な遷移(遷移5D0−7F2)は電気的双極
d Choppin(ed)Elsevier Science,Publisher,Amsterdam)、エネルギーを与え
ることができる。しかしながら、590〜595nmにおける放射を作り出す遷移5D0−7
F1は磁気的双極子であり、エネルギーを転移することができない(Dexter,J Che
m Phys 21;836,1953)。更に、ランタニド放射は等方性双極子モーメントを有す
るので、その配向率は不明瞭さが少なくなる(Andoら(1992)Biochem Biophys A
cta,1102,186)。
蛍光ランタニドキレートは、既に1978年から、Stryer,Thomas及びMearesによ
りエネルギー供与体として用いられている。例えば、Tbジピコリネートキレート
は、ロダミンについて6.57nm、エオシンについて4.46nm及びNBDについて4.46nm
の臨界距離(R0)を有すると報告されている。典型的には有効なエネルギー転移
において全体の減衰は上述のシステムにおいて示されるように、2.22msから0.12
msに短くなる。Mearesら(1981,1992)は、エネルギー供与体としてTb−フェニ
ル−EDTAを用いて、酵素が結合したリファマイシンを測定した(Biochemistry 20
;610;Biochemistry 22;6247)。蛍光化合物に加えて、Co(III)及びCo(II)の
ような非ルミネセンス受
容体も報告されている(Cronce及びHorrocks(1992)Biochemistry 31,7963)。E
u及びTbキレートも、クマリン誘導体についてEu及びサリチル酸誘導体についてT
bがエネルギー受容体としてテストされた(Clarkら(1993)Anal Biochem 210,1
)。多数の利用できる非ルミネセンスエネルギー受容体(消光体)はランタニド
キレートについて更なる利点である。それらは、金属イオン、亜硝酸塩(Tanaka
ら(1993)J Photochem Photobiol A:Chem,74,15)、他の常磁性金属イオン及
びそれらのキレート、フリーラジカル(Matkoら(1992)Biochemistry 31,703)
等により消光することができる。
種々の形態のバイオアフィニティーアッセイにおける蛍光ラベルとしてのラン
タニドの使用は多数の報告及び特許(例えば、Hemmi
いる。‘機能し得る’キレートラベルについての初期の考えの例は、例えば次の
特許(US 4,374,120;4,432,907;4,965,211;4,341,957;4,058,732及び4,283,382)
に見い出すことができる。非ルミネセンス標識化キレート(US 4,808,541)及び
解離蛍光エンハンスメント(US 4,565,790)に基づくヘテロジニアスアッセイシ
ステムは最も広い適用を得ている。このシステム及び蛍光形成リガンド及びアッ
セイ後カチオン飽和に基づくシステム(EP-A 354847及びUS 4,772,563)はホモジ
ニアスアッセイに適していない。しかしながら、以下の米国特許:4,761,481;5,
032,677;5,055,578;5,106,957;5,116,989;4,761,481;4,801,722;4,794,191;4,63
7,988;4,670,572;4,837,169及び4,859,777に記載されるような、時間分割FRETア
ッセイに用いることができる特許及び論文に記載される多数の安全な蛍光キレー
トがある。好ましいキレートは、第9歯状(nona-dentate)キレート化リガンド
、例えばテルピリジン(EP-A 403593;US 5,324,825;US 5,202,423;US 5,316,909)
、1又は2の5員
環を有するテルピリジンアナログ(例えばピラゾール、チアゾール、トリアジン)
(米国出願08/548,174及びPCT/FI91/00373)から構成される。極めてよく適し
たキレートは、以下の論文(Takaloら(1994)Bioconjugate Chem,5,278;Mukk
alaら(1993)Helv Chim Acta,76,1361;Remuinnanら(1993)J Chem Soc Perkin T
rans,2,1099;Mukkalaら(1996)Helv Chim Acta,79,295;Takaloら(1996)Hel
v Chim Acta,79)にも言及される。本発明に適した他の長期減衰時間ラベルの
例は、Pd及びptとのホルフィリン誘導体のリン光性複合体である(WO 94/10568)
。
種々の型のホモジニアス結合アッセイにおける長期減衰時間ランタニドの使用
は、例えば混合したリガンドキレート(US 4,587,223)、環境センシティブキレ
ート(EP 324323)及び酵素基質としてのキレート(EP 374221)に基づいても記載さ
れる。
1976年における蛍光共鳴エネルギー転移に基づくホモジニアスアッセイの開発
から、それらはSyvaコーポレーションにより市販される診断において広範囲の適
用を得ている(US 3,996,345)。時間分割蛍光測定システムにおける特定のラベ
ルとしてのランタニドキレートの使用は、最初、1970年代にLeif(Leifら(1976
)Automation of Uterine Cancer Cytology 1,GL Weid,GF Bahar and PH Bart
els(ed)Tutorial of Cytology,Chicago IL)及びWieder(Wieder(1978)Proc
.6th Int.Conf.On Immunofluorescence and related staining techniques,W
Knapp,K Holubar and G Wick(eds),North-Holland,Amsterdam,US 4,058,732
,DE2,628,158)により開示され、後に1980年代に機能する診断システム(US 4,
565,790)が開発されている。他方、蛍光ランタニドは、Stryerらにより1978年
に既に蛍光共鳴エネルギー転移実験のために用いた(Thomasら(1978)Proc Natl
Acad Sci,75,5746;Stryerら、Annual Review
of Biophysics及びBioengineering;Mullins LJ(ed)Annual Reviews,Inc;Palo
Alto,CA,1982,vol 11,p203)。従って、それらのFRETイムノアッセイのため
の長く励起された状態のプローブとしての使用は明らかであるが、なお、特許出
願の対象である。Wieder及びHaleは、Clq及びFRETの使用に基づくアッセイにお
けるエネルギー転移及び消光(EP-A-272320及びWO 87/07955)並びにStavrianopo
ulos(EP-A 242527,1987)を含む種々の技術に基づくホモジニアス時間分割イ
ムノアッセイで特許を得ている。Hoffman La Rocheは、核酸間の相互作用のため
の供与体としてのルマジン型の発色団と受容体としてのルテニウムキレートとを
用いる特定の出願について特許を受けており(EP 439036)、Cis Bio Internati
onalは、フィコビリプロテインのための供与体としてのEu又はTbクリプタートに
基づくアッセイについて出願している。
長い寿命の供与体及び短い寿命の放射性受容体分子の間の特定のエネルギー転
移に基づく時間分割ホモジニアスアッセイの原理は以下の通り要約することがで
きる:エネルギー転移のメカニズムを通して長い減衰時間の供与体の励起された
寿命の間に励起された短時間励起される状態の受容体の放射持続時間は直接励起
された受容体の減衰時間より長い。この事実は、直接励起された受容体からの蛍
光を避けるための一時的な識別(時間分割)の使用を許容する。エネルギー転移
に基づくホモジニアスアッセイにおいて、長寿命の供与体は光パルスで励起され
る(A)(図1)。直接励起された受容体からの干渉を避けるために(B)、適切な
減衰(C)の後、エネルギー転移で励起された受容体の蛍光放射が測定される(
D)。供与体からの長寿命の放射(DE)は、供与体放射がないかごくわずかであ
る波長においてエネルギー転移で励起された受容体(AE)の放射を測定すること
により避けられる(図2)。従って、ホモジニアス
バイオアフィニティーアッセイ(レセプター−リガンド結合、ハイブリダイゼー
ション反応、イムノバインディング、酵素基質結合等)において、供与体−受容
体対の会合又は解離は、エネルギー転移で励起されたアクセプターからのシグナ
ルにおいて各々増加又は減少を測定することにより追跡することができる。
発明の目的及び概要
本発明の目的は、エネルギー供与体で標識された第1の基及びエネルギー受容
体で標識された第2の基を含むルミネセンスエネルギー転移アッセイであって、
該受容体が、短時間の励起状態の寿命のルミネセンス標識又は非ルミネセンス標
識のいずれかであることを特徴とするアッセイを提供することである。前記標識
間の距離の、短くすること又は長くすることの各々から生ずる供与体標識からの
受容体標識へのエネルギー転移における増加又は減少の各々が測定される。本発
明によれば、供与体はルミネセンスランタニドキレートである。
本発明の一態様によれば、そのアッセイは、標識された供与体及び受容体基間
の距離を長くさせる反応に基づく。本発明によれば、供与体ランタニドキレート
は長い励起状態の寿命を有する。
本発明の他の態様によれば、そのアッセイは、標識された供与体及び受容体基
の間の距離を短くし又は長くさせる反応に基づき、前記供与体は、長い励起状態
の寿命を有するルミネセンスランタニドキレートである。本発明のこの態様によ
れば、長い励起状態の寿命のルミネセンスランタニドキレートは、1又は複数の
次の特性:
−高いルミネセンス収率
−1ms又はそれ超の励起状態の寿命、及び
−エネルギー転移のために最適化された放射線分散
を有する。
本発明の更に他の態様によれば、2又はそれ超の被検体が同時に測定され、こ
こで各々の被検体は特定の供与体−受容体対により測定される。
本発明の更に他の態様によれば、受容体のルミネセンス又は非ルミネセンス標
識は、前記第2の群が適切な共有又は非共有結合により結合し又は結合すること
ができる大きな表面を供する固体担体に結合する。
本発明の更に他の態様によれば、受容体は、細胞膜もしくは細胞膜フラグメン
トに固定された親油性化合物であるか、又は受容体は長鎖炭化水素例えば脂肪酸
とコンジュゲートすることにより親油性にされる。
本発明の更に他の態様によれば、エネルギー転移を測定するのに用いられる装
置は、サンプルマトリックスが励起、エネルギー転移又は放射せざるおえないい
ずれかの干渉をモニターしそれらを正すために、励起又は放射の波長における吸
収、異なる持続時間のルミネセンス、散乱及びルミネセンス減衰時間のような多
重のパラメータを同時に又は連続的に追跡することができる。

好ましい実施形態の詳細な記載
本発明は、検出において時間分割蛍光測定法を用いるエネルギー転移に基づく
ホモジニアスアッセイの改良に関する。その特定の改良は、エネルギー供与体と
して用いられる特定の型のランタニドキレート標識、所定のアッセイのために最
適化されたエネルギー受容体に関し、そのエネルギーが転移する道は、最適化さ
れたフィルター及び時間窓を用いて測定され、サンプル由来の全ての可能性ある
干渉を正す方法は、多成分分析のためのアッセイ及び単純化したアッセイプロト
コルの開発を用いる。
用語“第1の基”及び“第2の基”は(これらの群の間の距離が減少する反応
、例えばバイオアフィニティー反応における)バイオアフィニティー認識成分の
ようないずれかの成分、又は分子もしくは基質の一部(例えばその開裂が互いか
ら2つの標識された基を分離させるであろうペプチド分子の遠端)を含むと理解
される。
用語“ルミネセンス”は、蛍光、リン光、化学ルミネセンス、生物ルミネセン
ス及び電気により作られたルミネセンス、光ルミネセンス、放射線ルミネセンス
、音ルミネセンス、熱ルミネセンス及び摩擦ルミネセンスを含む。
解離を測定するアッセイにおける好ましい取合わせは、ルミネセ
ンスの短い減衰時間の受容体及び長い減衰時間のランタニドキレートに基づく供
与体を用い、(受容体の直接の励起から放射される)受容体の直接のルミネセン
スの干渉を避けるために、時間分割蛍光測定法における遅延時間を用いる受容体
分子の放射を追跡することである。受容体分子が過剰であり(時間分割モードで
、それらの干渉はごくわずかである)、結合する試薬の会合がシグナルを増加さ
せる方法でアッセイを作り出すことが要求される。
このようなシステムのために、好ましいキレート標識は高ルミネセンス収率(
εxΦ>2000)、長い励起状態寿命(好ましくは1ms超)及びエネルギー転移の
ために最適化された放射分散を有さなければならない。キレート化されたイオン
の周囲のリガンドフィールドは、例えばEuキレートで、放射の70%超が(610〜62
0nmの範囲で)5D0−7F2においてであり、590nm域ではないようにしなければなら
ない(例えばEuグリプタートの放射(WO 92/01225)並びにテルピリジンのビス−
イミノアセテート誘導体のもの(US 5,324,825;US 5,202,423及びUS 5,316,909
)と比較のこと)。好ましいキレートにおいて、無用な590nmにおける磁気的双
極子遷移及び700nm周辺の放射は抑制される(Li及びSelvin,J Amer Chem Soc 11
7;8132,1995)。本適用のために特に優れたキレートは、高い分子吸収係数(ε)
、極めて長い励起状態寿命及び優れた量子収率(Φ)を有する多重色原性のポリ
カルボキシレートで形成されたEuキレートである(Takaloら、Helv Chim Acta 7
9:789,1996)。Euに加えてTbは、その高いルミネセンスのキレートを用いる場
合、特に有望なエネルギー供与体である。好ましいTbキレートはイミノジアセテ
ート基における結合部位(Mukkalaら、J Alloys Compounds 225;507,1995)又は
光吸収性芳香族構造から十分に単離された結合アームを含むテルピリジンから構
成される。それらの適用のための特に優れたキ
レートは、1又は2のピリジン環がピラゾール(US 08/548,174)又はトリアゾー
ル及びチアゾール環(PCT/FI91/00373)で置換されているテルピリジン誘導体で
ある。Eu及びTbに加えて、Smの作用は、二重又は三重標識ホモジニアスエネルギ
ー転移アッセイを行う可能性を供するだろう。Smは、より高い波長でエネルギー
を与えるという利点を有し、ここで高いルミネセンスキレートの主な放射は643n
mであり、波長スケールを近IRまで続ける機会を与える(近IR放射性蛍光の優れ
たコレクションは異なるソースから市販されるようになっている)。Smの好まし
い安定なキレートは、Savitsky(Savitskyら、SPIE 2388;429,1995)により記
載されるような1,3−ジケトンの多重形態から構成される。かわりの第3の選
択(第3の標識)は、650〜660nmにおいて長い寿命のリン光を放射するリン光性
pt又はPdコプロポルフィリンである(WO 94/10568)。
長い励起状態寿命のランタニドキレートの特に好ましい群は、それらの構造が
スキーム1〜5に開示される化合物1〜12である。そのスキームにおいて、λex c
は励起波長を、τは減衰時間を、そしてεxΦはルミネセンス収量を意味する
。そのスキームに示される値は、タンパク質に結合した化合物から測定される。

会合アッセイのための好ましいアクセプター分子は高い分子吸収係数(好まし
くは100,000超)を有する(出来るだけユニティー(1)に近い量子収量で)高
いルミネセンスである。受容体の吸収は、例えばEuについて611〜620nm、Tbにつ
いて545nm、Smについて643nmにおいて、供与体のエネルギー寄与放射とオーバー
ラップしなければならない。受容体の好ましい特徴は、供与体がいずれの放射を
有さない波長(例えばTbについて574〜575nm、Euについて660〜670nm及びSmにつ
いて675nm周辺)におけるその鋭い放射である。受容体の減衰時間は1μs未満
であるべきである。更に、受容
体は直接的又は間接的に(例えば抗バインダー抗体、レクチン、アビジン等を通
して)、共有結合又は非共有結合で結合パートナーに結合されなければならない
。共有結合でコンジュゲートするプローブとしては、例えばキサンテン色素(ロ
ダミン、テトラメチルロダ
of Fluorescent Probes and Research Chemicals,Molecular Probes 1992-1994
,Mayer及びNeuenhofer,Angew Chem Int Ed Engl 33;1044,1994を参照のこと)
、特定のカルボシアニン(Cy3.18)(Mujumbarら、Bioconjugate Chem 4;105,1993
,Southwickら、Cytometry 11;418,1990)及び特定のポルフィリン(Camman,DE
4,216,696)のものさえ用いることができる。B−フィコエリトリンは、その極
めて高い分子吸収係数(546nmにおいて2,410,00)及び量子収量(0.98)のため、
その大きなサイズ(MW 240,000)にかかわらず、エネルギー転移アッセイに極め
て適する。
Euのための優れた受容体は、611〜620nm域において最大吸収を有するべきであ
る。その波長域のための優れた受容体は、特にIR放射性色素の群から見い出すこ
とができる(報告について、Haugland Molecular Probes,Miller,Spectroscop
y Europe 5;34,1993,Patonay及びAntoine,Anal Chem 63;332A,1991,Southw
ickら、Cytometry 11;418,1990,Mujumbarら、Cytometry 10;11,1989並びにMu
jumbarら、Bioconjugate Chem 4;105,1993,Papkovsky,Appl Fluorescence Te
chnol III;16,1991,Schindele及びRenzoni,J Clin Immunoassay 13;182,19
93を参照のこと)。これらの型の化合物は、フタロシアニン、ホルフィリン、シ
アニン色素(Cy-5,Cy5,18,Cy5,29)(Selvinら、J Amer Chen Soc 116;6029,199
4,Mujumbarら、Bioconjugate Chem 4;105,1993)、コンジュゲートしたキサン
テン(ロダミン800)(Imasakaら、Anal Chem 61;
2285,1989)、スクアラーテ(Chang,EP 176,252)、メチレンブルー、ナイルブル
ー及びオキサジン(Imasakaら、Anal Chem 61;2285,1989)、インドシアニングリ
ーン(Imasakaら、Anal Chem 62;363A,1990)を含む。フィコビリプロテインのう
ちのいくつかは、Eu受容体、特にA-PC,C-PC及びR-PCのための優れた候補である
。Sm又はコプロポルフィリンのための好ましいエネルギー受容体は、IR放射性色
素(上述の報告を参照のこと)。
供与体プローブで標識されたリガンド又は結合試薬と受容体プローブで標識さ
れた結合試薬との間の短い距離を確認するための好ましい方法は、結合性試薬と
直接カップリングさせた活性化プローブ(例えば、受容体で標識したレセプター
タンパク質、抗体又は他の結合タンパク質)を用いることである。他の方法は、
例えば受容体で標識した抗バインダー(例えば抗レセプター)抗体並びにビオチ
ニル化バインダー及び受容体標識(ストレプト)アビジンを用いて、間接的ラベ
リングを用いること、又は供与体標識成分が直接又は間接的に結合するであろう
実際の結合部位の近傍に受容体分子を運ぶために他のバイオアフィニティー反応
を用いることである。
純粋でない膜結合レセプターでの好ましい方法は、親油性膜プローブを用い、
そして受容体分子でそのレセプターの近く又は周囲の膜全てを染色することであ
る。長鎖脂肪族炭化水素鎖に結合した適切なプローブ、例えばオキサカルボシア
ニン(Molecular Probes)がその染色に適している。
各々の特定のアッセイのための結合成分の別個の標識化を避けるであろう更に
他の方法は、固体担体(ポリマー、セラミックス又はガラス等)、例えば高濃度
の受容体分子を含む万能捕獲表面を用いることである。適切な固体担体は、例え
ば1500μmまでの直径を有するビーズもしくは粒子又はいずれかの固体表面であ
り得る。広範
囲の種々のルミネセンスプローブで標識したマイクロビーズは異なるソースから
利用できる。担体に用いる好ましいプローブは、漏出を避けるために水にほとん
ど溶けない疎水性化合物である。これらの標識化に適した種々のプローブは、シ
ンチレーター及びレーザー色素の中から見い出すことができる。高いルミネセン
スのビーズでは、多数の受容体分子が、ビーズコーティングの後に長い距離を補
うことができ、そして蛍光ビーズは実際、エネルギーを受容する表面を供する。
ビーズが、供与体が放射した光のほとんどを吸収することができる場合、単純な
放射性エネルギー転移を適用することができ、ここでそのエネルギー転移は、空
間角の関数であり、10μmビーズでの臨界距離はマイクロメーターの範囲内であ
る。しかしながら、FRETベースのアッセイについて、プラスチックを膜レセプタ
ーを固定するために結合性タンパク質(例えばアグルチニン)で最初にコートす
る場合、長い距離のため無効なエネルギー転移を生じ得る。好ましい取合せは、
これにより、表面活性化ビーズを用いること及び受容体分子とのカップリングの
ための反応性基の部分を用いること、又は(ロダミン標識化アグルチニンのよう
な)受容体で標識した結合表面を用いることもしくは後で受容体で、コートされ
たタンパク質を標識することである。
会合反応のホモジニアスアッセイ(イムノバインディング、レセプターリガン
ド結合、ハイブリダイゼーション反応、酵素−基質結合等)において、結合を測
定するための好ましい方法は、受容体シグナルの増加を追跡することである。受
容体シグナルは、受容体の波長において優れた透過度を有するが、より重要なの
は、供与体の各々の放射ラインのために絶体的に十分にブロックされる、用いる
供与体のために最適化されたフィルターを用いて測定される。そのフィルターは
、(Tbの545又は490nm及びEuの613〜615nmのよう
な)供与体の主な放射ラインから放射するいずれの放射線ももらすべきでない。
更に、エネルギー転移フィルターは、用いる供与体でマイナーな放射ラインのな
い波長領域に位置しなければならない。適切な遅延の使用は受容体の直接的励起
由来の干渉を避ける(最適な遅延は用いる励起パルスの長さに依存するが、少く
とも10倍長いべきである。
エネルギー転移で励起された受容体の減衰は、供与体の減衰及びエネルギー転
移効率の関数である。これにより、(競合的結合アッセイ又は非競合的アッセイ
のような)アッセイの間、全体の減衰は一定でないが、被検体の関数である。特
異的結合が低く、エネルギー転移効率が1%未満である場合の会合アッセイにお
いて、受容体のエネルギー転移放射の減衰時間はほぼ一定であり、供与体の減衰
時間に等しい。このような測定のための遅延及びカウンティング時間は重大では
ない。より高い効率のアッセイのために、その減衰時間は結合により減少し、そ
して短い遅延時間及び合理的な短い計数時間を維持しながら、より急激な応答を
得ることができる。他方、供与体放射を追跡するなら、長い遅延時間を用いてよ
り急激な応答が得られる。なぜならエネルギー転移がおこる時、全体の供与体放
射は減少し、その減衰時間は短くなる。いずれのアッセイにおいても最適な結果
を得るために、アッセイの型、特異的結合の割合及びエネルギー転移効率に従っ
て、計数窓を最適化することが賢明である。
種々の化合物又はサンプルのスクリーニングにおいて、サンプルは、シグナル
減少効果を有し得る。サンプルは、例えば励起光、放射光を吸収し、又は他の型
の消光を引きおこし得る。多重標識カウンター、例えばWallac 1420 VICTORにお
いて、干渉は種々の方法、例えば(1)減衰分析、(2)励起及び放射の波長に
おけるサンプ
ル吸収の同時の光度測定でのモニター、(3)シグナル、供与体及び受容体の同
時の測定、それらの相関関係の計算、及び結合反応のない等量の供与体及び受容
体の対照との比較、又は(4)エネルギー転移又はシグナルの変化に従う結合の
間の速度測定により制御することができる。高効率の結合アッセイでは、減衰時
間の分析は、励起も放射もない色の消滅により影響を受けない独立したパラメー
タである。その減衰の変化は、供与体について又は受容体について追跡すること
ができる(エネルギー転移が増加する時、減衰か減少)。全てのサンプルでの乱
れのない分析を保証するための好ましい方法は、上述の全ての補正法の適当な組
合せを用いることである。
好ましい実施形態によれば、エネルギー転移は、エネルギー転移がない時に受
容体放射測定のために用いられるチャンネルへのクロストーク又は供与体放射を
避ける所定の装置セッティングを用いて時間分割フルオロメーターにより記録さ
れる。
好ましくは、その装置は、励起のいずれもの減衰も自動的に捕正し、そのサン
プルは、アッセイ混合物に希釈されたサンプルの吸光度を同時に追跡され、サン
プル吸収による励起又は放射減衰に従って放射の読み取りを補正され得る。
測定された受容体シグナルは、ゼロキャリブレーターと比較することができる
供与体放射も記録することにより標準化することができ、ここで受容体ルミネセ
ンスに伴う増加のないシグナルの低下が記録され、補正される。
供与体又は受容体の減衰時間を記録して、パラメータを測定する時に用いるこ
とができる。
供与体又は受容体の減衰時間は、サンプル干渉の補正に利用することができる
。
改良されたエネルギー転移アッセイを適用することができるアッ
セイの第2の型は、会合でなく解離を追跡するアッセイである。このようなアッ
セイは、例えば、抗体−抗原複合体が形成され、被検体の添加が新しい平衡形態
を引きおこす(リガンド置換アッセイ)競合的イムノアッセイ(又は他の特異的
結合アッセイ)であり得る。このようなアッセイの他の群は、酵素、例えばペプ
チダーゼ(例えばHIVペプチダーゼ)、プロテアーゼ及び他の酵素(例えばヌク
レオチド加水分解酵素、ヘリカーゼ等)を開裂するアッセイである。解離のホモ
ジニアス測定において、好ましい技術は、供与体の増加した放射を追跡すること
である。
解離に基づくアッセイのための好ましいキレートは、会合ベースのアッセイの
ための上述とかなりの程度、同じである。しかしながら、テルビウムは、しばし
ばより優れた選択である。なぜならそれは、種々の化合物、例えば金属イオン、
窒素化合物、例えばアジド、ニトリド(Tanakaら、J Photochem Photobiol 74;1
5,1993)又はラジカル(スピンラベル、例えばDoxyl,Proxyl又はTempo及び他
の不対電子を含むN−O化合物)で消光することができる。本発明に適した優れ
たTbキレートは上述のものである。
解離測定アッセイのための好ましい受容体は、有効な供与体として可能な限り
消光する優れたエネルギー受容体である。受容体は、ルミネセンスのものであり
得るが、それである必要はない。好ましいFRET受容体は優れた受容体特性(5nm
を超えるR0)を有するが、高いルミネセンスを有する必要はない。上述のルミ
ネセンス化合物に加えて、優れた受容体は、(エリトロシンのような)重原子コ
ンジュゲーションで非ルミネセンスで作られた各々の化合物又は適切な他の置換
基であり得る(例えばKhanna及びUllman,Anal Biochem 108:156,1980,Ullman
及びKhanna,Methods Enzynol 74;28,1981)。立体障害(例えば供与体及び受
容体の両方で標識されたH
IVペプチドでのもの)を避けるために、受容体は小さくなければならず、小さな
分子消光物質(quenchers)、例えばスピンラベルが好ましい。より大きなアクセ
プターの使用は、おそらく、間接的なアプローチ、例えば小さなアフィニティー
ラベル(小さなハプテン、ビオチン)及び実際の解離反応後に行わざるをえない
消光のための別個のステップ(アビジンの受容体標識化抗ハプテンの添加)(終点
検出)の使用を要求する。例えばペプチダーゼのための好ましいアッセイデザイ
ンは、一端が蛍光テリビウムキレートで標識され他端がフリーラジカルで標識さ
れたペプチド基質を用いることである。スピン消光化及びFRET双極子−双極子エ
ネルギー転移の間の選択は、加水分解されていないペプチド内の標識間の実際の
距離による。小さな距離(2nm未満)では、フリーラジカル(スピン)が好まし
い。2つのプローブの距離が5nmを超えるであろうなら、双極子−双極子エネル
ギー転移が好ましい。
会合した形態において消光するようデザインする場合(例えば消光された基質
)、供与体ルミネセンス、及びその減衰時間は解離により増加する。測定におけ
る好ましい遅延時間は長いので、消光した供与体の可能性ある残存ルミネセンス
の検出を避ける。これらのアッセイは、会合アッセイと同様に、時々サンプル干
渉を生ずる傾向があり(例えば色の消滅)、その問題は、上述と同様に解決する
ことができる。
実施例
本発明は、以下の非限定的例により更に詳述される。
実施例1
抗βhCG(ヒト絨毛膜性生殖腺刺激ホルモン)抗体(コードF19-9C1)の蛍光Tbキ
レート化合物6での標識化
エネルギー供与体化合物6を、次の方法で抗βhCG抗体(コードF19-9C1、Wall
ac Oy,Turku,Finland)にコンジュゲートした:抗βhCG抗体(5mg/ml)を4
℃で一晩、50mM炭酸緩衝液pH9.5中30倍モル過剰な化合物6と共にインキュベー
トした。その標識化した抗体を、溶離液としてTris(0.9% NaClを含む50mM Tris
-HCl、pH8)でのゲルろ過(Sephadex G50を重ねたSepharose 6B,0.5×70cm,Ph
armacia,Uppsala,Sweden)により未反応の化合物6から分離した。その標識し
た抗体を分析し、抗体当り4.7分子の化合物6を含むことを見い出した。標識し
た抗体を、0.1%ウシ血清アルブミンを含むTris緩衝液pH7.4中に維持した。
実施例2
抗βhCG(コードM15294)抗体のテトラメチルロダミンイソチオシアネート(TRIT
C)での標識化
エネルギー受容体TRITC(Molecular Probe Inc,Eugene,OR)を以下の方法で
抗βhCG抗体(コードM15294、Wallac Oy)にコンジュゲートした:抗βhCG抗体
(5mg/ml)を、+4℃で一晩、50mM炭酸緩衝液pH9.5中15倍モル過剰なTRITCと
共にインキュベートした。その標識した抗体を、溶離液としてTris(0.9% NaCl
を含む50mM Tris-HCl、pH8)を用いてゲルろ過(Sephadex G50を重ねたSepharose
6B,0.5×70cm,Pharmacia)により未反応のTRITCから分離した。その標識した
抗体を分析し、抗体当り1.7のテトラメチルロダミンを含むことが見い出された
。標識した抗体を0.1%ウシ血清アルブミンを含むTris緩衝液pH7.4中に維持した
。
実施例3
βhCGのホモジニアスエネルギー転移アッセイ
この2部位イムノアッセイにおいて、96ウェルプレートのウェル内の50μlの
βhCG標準を、200μlのTris緩衝液(0.9% NaClを
含む50mM Tris-HCl、pH7.4)中の100ngの化合物6標識化抗体(供与体)及び100n
gのテトラメチルロダミン標識化抗体(受容体)とインキュベートした。用いる
2つの抗βhCG抗体は、βhCGサブユニット上の異なる特定の抗原部位に対して生
ずる。βhCG標準は、血清中の遊離βhCGの測定のための市販のキットからのもの
であった(Wallac Oy)。その反応混合物を、96ウェルプレートシェーカー上で室
温で30分、インキュベートした。その形成された複合体からのテトラメチルロダ
ミン蛍光を、時間分割フルオロメーター(340nmで励起、572nm±3nm、遅延時間5
0μs、窓時間100μs、サイクル時間1ms)、モデル1420 VICTOR(Wallac Oy)
において測定した。βhCGのエネルギー転移アッセイのための標準曲線を図3に
示す。
実施例4
PSA(前立腺特異的抗原)のホモジニアスエネルギー転移アッセイ
PSA上の異なるエピトープを認識する2つの抗体(Wallac Oy)を各々化合物1及
びTRITCで標識した。抗体標識化を、実施例1及び2に記載される通り行った。
この2部位アッセイにおいて、(市販のPSAアッセイキットDELFIA PSAからの)PSA
標準50μlを、200μlのTris緩衝液pH7.4中の100ngの化合物1(供与体)標識
化抗体及び100ngのテトラメチルロダミン標識化抗体(受容体)と一緒にインキ
ュベートした。そのアッセイ混合物を、連続的な振とう下で室温で30分、インキ
ュベートした。その形成された複合体(サンドイッチ)からのテトラメチルロダ
ミン蛍光を、Wallac 1420 VICTOR時間分割フルオロメーターで測定した(340nmで
励起、572nm±3nmで放射、遅延時間100μs、サイクル時間1ms)。ホモジニア
スエネルギー転移アッセイのための標準曲線を図4に示す。
実施例5
受容体としてのテトラメチルロダミン−アビジンでのβhCGのホモジニアスエ
ネルギー転移アッセイ
この2部位イムノアッセイを、実施例3に記載される通り行った。テトラメチ
ルロダミン標識化抗体(受容体)を用いるかわりに、ビオチニル化抗体及びテト
ラメチルロダミン−アビジン(受容体)を用いた。テトラメチルロダミン−アビ
ジン(Molecular Probes)は反応して、ビオチニル化抗体と安定な複合体を形成
した。抗βhCG抗体(コードM15294)をビオチン試薬供給元(Biotinylating Rea
gene 732494,Boehringer Mannheim,Mannheim,Germany)からの説明に従って
ビオチニル化した。形成された複合体(サンドイッチ)からのテトラメチルロダ
ミン蛍光を、Wallac 1420 VICTOR時間分割フルオロメーター(340nmで励起、572n
m±3nmで放射、遅延時間50μs、窓時間100μs、サイクル時間1ms)で測定し
た。ホモジニアスエネルギー転移アッセイについての標準曲線を図5に示す。
実施例6
受容体としてのR−フィコエリトリン−アビジンでのβhCGのホモジニアスエ
ネルギー転移アッセイ
本実験において、テトラメチルロダミン−アビジンのかわりにR−フィコエリ
トリン−アビジン(Molecular Probes,Eugene,OR,USA)を用い、他は、実施例
5の通り行った。形成された複合体(サンドイッチ)からのR−フィコエリトリ
ン蛍光を、Wallac 1240 VICTOR時間分割フルオロメーターで測定した(340nmで励
起、572nm±3nmで放射、遅延時間50μs、窓時間100μs、サイクル時間1ms)
。ホモジニアスエネルギー転移アッセイについての標準曲線を図6に示す。
実施例7
プロゲステロンのホモジニアスエネルギー転移アッセイ
モノクローナル抗プロゲステロン抗体(Wallac Oy)を実施例1に従って化合物
2で標識した。プロゲステロン−ビオチン誘導体(Wallac Oy)を、5/Kaff(Kaff
=用いたプロゲステロン抗体のアフィニティー定数)の濃度で用い、約2倍
超のテトラメチルロダミン−アビジン(Molecular Probes,Inc,OR)を用いた。T
ris緩衝液をインキュベーションに用いた。この競合イムノアッセイにおいて、5
0μlの標準又はサンプルを、100μlの化合物2標識化抗体(1/50000希釈)
及び100μlのプロゲステロン−ビオチン誘導体及びテトラメチルロダミン−ア
ビジン(受容体)とインキュベートした。ビオチニル化プロゲステロンは、標識
された抗プロゲステロン抗体上の結合部位について標準又はサンプルからのプロ
ゲステロンと競合する。その反応混合物を室温で1時間、インキュベートした。
インキュベーションの後、時間分割フルオロメーター(Wallac 1420 VICTOR、34
0nmで励起、572nm±3nmで放射、遅延時間50μs、窓時間100μs、サイクル時
間1ms)で、抗プロゲステロン−抗体−プロゲステロン−ビオチン複合体に結合
したテトラメチルロダミン−アビジンから測定した。その測定されたシグナルは
、サンプル中のプロゲステロン濃度と逆相関している。
実施例8
IL-2のホモジニアスエネルギー転移レセプターアッセイ
純粋な担体のないIL-2(R & D Systems,Minneapolis,MN)(1mg/ml)を、+4
℃において一晩、50mM炭酸緩衝液pH9.2中10倍モル過剰量の化合物2とインキュ
ベートした。その標識したIL-2を、溶離液としてTrisでのゲルろ過(Sephadex G5
0,0.7×48cm,Pharmacia)により未反応のキレートから分離した。その標識した
IL-2を分析し、IL-2分子当り1.4分子の化合物2を含むことが見い出された。IL-
2αレセプターに対する非中和性抗体を、実施例2に従ってテト
ラメチルロダミンイソチオシアネートで標識した。そのホモジニアスレアプター
アッセイにおいて、増加する濃度の非標識化IL-2を、一定量の化合物2標識化IL
-2(供与体)、IL-2αレセプター及びテトラメチルロダミン標識化抗体(受容体
)と同時インキュベートした。Tris緩衝液をこのインキュベーションに用いた。
時間分割フルオロメーター(Wallac 1420 VICTOR、340nmで励起、572nm±3nmで
放射、遅延時間50μs、窓時間100μs、サイクル時間1ms)において、形成さ
れた抗体−レセプターIL-2複合体からの蛍光シグナルを測定することにより、置
換曲線を作成した。
実施例9
化合物6及びテトラメチルロダミンでのカゼインの二重標識化
5ミリグラムのカゼインを0.5mLの0.1M炭酸ナトリウムpH9.2に溶かした。水
中の5.5mMの蛍光化合物6の50マイクロリッターをカゼイン溶液に加え、4℃で
一晩、反応させた。その標識化したタンパク質を、溶離液として水溶液中0.9%
の塩化ナトリウムを用いて、Sephadex G-50カラムを通して精製した。その標識
したカゼインを分析し、カゼイン分子当り1の化合物6分子を含むことを見い出
した。1mL中2ミリグラムの化合物6標識化カゼインを、ジメチルホルムアミド
に溶かした20mMのテトラメチルロダミンイソチオシアネート(TRITC)100μlと
混合した。120μlの1M炭酸ナトリウムpH9.5を加えてpHを調節した後、反応混
合物を4℃で一晩、インキュベートした。その標識したタンパク質を、0.9% Na
Clを含む50mM Tris-HCl pH8で平衡化して流したSephadex G-50カラムを用いて
未反応のTRITCから精製した。化合物6−標識化カゼインについてと同じプロト
コルを用いて、非標識化カゼインもTRITCで標識した。
実施例10
スピンラベルでの化合物6標識化カゼインの標識化
1mLの0.9% NaCl中の2ミリグラムの化合物6標識化カゼイン(実施例9)を
、110μlの1M炭酸ナトリウムpH9.5と混合し、13μlの160mM 3−(2−(2
