立体選択的方法 発明の背景
強力なエンドセリンアンタゴニストの商業的に実施可能な不斉合成法の開発中
に、主要な中間体である2−アリールオキシカルボン酸の高度に立体選択的な合
成法を開発することが必要になった。キラル補助部として(R)−パントラクト
ンを使用してDurstとKohにより種々の汎用方法が考案されている[Ko
h,K.; Durst,T. J.Org.Chem.1994,59,46
83参照]。ラセミα−ハロ酸のパントラクトンエステルが種々のフェノキシド
にカップリングされている。この方法で76〜90%のジアステレオ選択性が得
られており、新規に形成された不斉中心は主に「S」立体配置である。キラル補
助部として(S)−乳酸エチルを使用すると「R」立体配置が得られるが、ジア
ステレオ選択性はパントラクトンに比較して有意に低下する(60〜75%)。

率が並である。
本発明は、ピロリジンから誘導されるラクタミドをキラル補助部として使用す
る高度に立体選択的なカップリング反応に関する。発明の要約
本発明は、アリールオキシ基によるα−ハロ酸のアルキル化の立体選択性を増
すために式Iの化合物等のキラル補助部を使用する2−アリールオキシカルボン
酸の立体選択的合成方法に関する。本発明は更に、キラル補助部として有用な式

2)アルキル化誘導体のキラル補助部を水性溶媒混合物中で無機塩基と過酸化物
で加水分解し、この酸の塩:

3)上記の酸の塩を酸性溶液で中和し、酸:
を得る段階を含む。
アルキル化段階はテトラヒドロフラン、トルエン、キシレン又はジメチルホルム
アミド等の有機溶媒中で実施することがで
きる。アルキル化段階では、フェノールのテトラヒドロフラン溶液にt−ブトキ
シドの塩を加えることにより一般に製造されるフェノキシドの塩の溶液を加える
ことが必要である。このアルキル化段階では、カリウム、ナトリウム、リチウム
又はアンモニウム[N(R16)4+、但しR16はH又は(C1−C4)−アルキル
として定義される]等の対イオンをもつフェノキシド塩が有用である。従って、
有機溶媒は多くの場合には混合物であり、例えばテトラヒドロフランとt−ブタ
ノールの混合物である。アルキル化段階に好ましい温度範囲は60℃〜ほぼ室温
(25℃)である。
加水分解段階は、LiOH、KOH、NaOH、KOCH3、NaOCH3、L
iOCH3、KOC2H5、NaOC2H5、LiOC2H5等の無機塩基を使用して
実施することができる。加水分解段階には更に過酸化水素等の過酸化物の存在が
必要である。加水分解段階で有用な水性溶媒混合物はテトラヒドロフラン−水、
トルエン−水、ジメチルホルムアミド−水、又はメタノール、エタノールもしく
はt−ブタノール等の極性有機溶媒である。
中和段階では飽和塩化アンモニウム溶液等の酸性溶液が有用
である。
キラル補助部はアルキル化部位の近傍位置に結合し、求核攻撃方向に影響し得
る容易に脱離可能なキラル基として定義される。本方法で有用なキラル補助部の
例としては、

上記アルキル置換基は、メチル、エチル、イソプロピル、イソブチル、ネオペ
ンチル、イソペンチル等の特定の長さを有する直鎖及び分枝鎖炭化水素を意味す
る。
アルケニル置換基とは、ビニル、アリル及び2−ブテニル等のように各々炭素
−炭素二重結合を含むように修飾された上記アルキル基を意味する。
シクロアルキルとは、3〜8個のメチレン基から構成される環を意味し、各々
他の炭化水素置換基で置換されていてもいなくてもよく、例えばシクロプロピル
、シクロペンチル、シクロヘキシル及び4−メチルシクロヘキシルが挙げられる
。
アルコキシ置換基とは酸素橋を介して結合した上記アルキル基を意味する。
へテロアリールはカルバゾリル、フリル、チエニル、ピロリル、イソチアゾリ
ル、イミダゾリル、イソキサゾリル、チアゾリル、オキサゾリル、ピラゾリル、
ピラジニル、ピリジル、ピリミジル、プリニル又はキノリニルとして定義される
。
ラクタミドキラル補助部はトリメチルアルミニウムとピロリジンを使用してW
einrebアミド化条件下で(R)−又は
(S)−乳酸エチルから合成される[(a)Nahm,S.; Weinreb
,S.M. Tetrahedron Lett.1981,22,3815、
(b)Basha,A.; Lipton,M.; Weinreb,S.M.
Tetrahedron Lett.1977,28,4171参照]。反応
は2時間未満で完了し、エナンチオ純度の検出可能な低下は見られない。生成物
は収率80%で得られ、形成されるラクタミドはそれ以上精製せずに使用できる
。文献記載の方法により市販酸とのDCCカップリング又は市販酸ハロゲン化物
との反応によりラセミα−ハロ酸のラクタミドエステルを調製した[(a)Du

;J.Org.Chem. 1975,40, 3420参照]。
カップリング反応は所望温度(表1)でヨウ化トリカプリルメチルアンモニウ
ムの存在下又は不在下にハロゲン化物のTHF溶液に予め調製したアリールオキ
シドの溶液を加えることにより実施した。反応は従来のエステル助剤を使用する
よりも著し
く早い速度で進行することが認められた。ナトリウム4−メトキシフェノキシド
を用いて−35℃で実施した反応は0.5時間以内に完了し、ジアステレオ選択
性は92%であった(試験番号1)。乳酸エチルを助剤として使用する同様の反
応は0℃で24時間の反応時間を要した。生成物は更に60%と低いジアステレ
オ選択性で得られた。乳酸エチル助剤では常にこのような結果が認められる。ピ
ロリジンラクタミド助剤を用いるカップリング反応はその対応するエステルより
も迅速に進行するので、別のフェノキシド塩も使用できる。

Chiracel ODカラムを使用するHPLCとジアステレオマーメチル
ダブレットの300MHz 1HNMR積分により決定したジアステレオマー。
リチウム4−メトキシフェノキシドは−35℃では非反応性であるが、−15
℃〜周囲温度の範囲の温度ではα−ブロモエステルと容易に反応する(試験番号
3〜6)。温度の効果は観察されず、室温で得られるジアステレオ選択性は−3
5℃でナトリウム塩を用いて得られる値と同等である(試験番号6〜8)。他方
、α−ヨードエステルを用いてカップリング反応を実施すると、ジアステレオ選
択性は有意に増加する(試験番号9及び10)。[α−ヨードエステルはアセト
ン中でα−ブロモエステルをNaIで処理することにより調製した。]周囲温度
と0℃で実施した反応は夫々98:2及び99:1の高いジ
アステレオマー比を示した。
種々のジアステレオマーハロゲン化物と置換フェノールを試験することにより
、反応の一般則を検討した(表2)。
*Supelcosil LCCNカラム又はChiracelMODカラムを
使用するHPLCにより決定したジアステレオマー比(DR)。ジアステレオ選
択性は300MHz 1HNMRでも得られる。**
α−ヨードエステルを使用して実施した反応。
フェノールに関する置換パターンは反応のジアステレオ選択性に最小の影響し
か与えない。立体的にかさ高な2−プロピルフェノールは、かさの小さいフェノ
ールと同様のジアステレオマー比を与える(試験番号1、2〜4)。また、観察
されるジアステレオ選択性はキラル補助部として乳酸エチル又はパントラクトン
を使用して得られるよりも約5〜15%高い。α−ヨードエステルを使用すると
