La presente invenzione riguarda un procedimento per la preparazione di particelle lavorate a secco che contengono un ingrediente attivo in miscela con un polimero biocompatibile. La presente invenzione riguarda inoltre le particelle lavorate a secco cosi’ ottenute e riguarda composizioni farmaceutiche che contengono tali particelle. In questa descrizione, si usa il termine "ingrediente attivo" per indicare qualsiasi sostanza terapeuticamente attiva oppure qualsiasi miscela che possa venire somministrata vantaggiosamente all’uomo o ad altri animali a scopo di diagnosi, cura, alleviamento di sintomi, trattamento oppure prevenzione di malattie. Il termine "polimero" viene usato per comprendere omopolimeri, copolimeri oppure una loro combinazione. Da ultimo, con l’espressione "particelle lavorate a secco" si possono intendere particelle preparate secondo un procedimento nel quale non si deve sciogliere alcun costituente delle particelle in alcun solvente che debba venire allontanato prima dell’isolamento di dette particelle. Particelle oppure microparticelle contenenti uno o più ingredienti attivi, procedimenti per la loro preparazione e il loro impiego in composizioni farmaceutiche sono ben noti. Quando la preparazione di tali microparticelle comprende la sospensione oppure la dissoluzione di un polimero in un solvente, le microcapsule cosi' ottenute, in generale, contengono (almeno) tracce dei solventi usati nella loro lavorazione; ciò può essere un ostacolo per certi impieghi terapeutici. Quando ia preparazione di tali microparticelle comprende estrusione e/o macinazione, ciò implica la formazione di particelle aventi superfici esterne irregolari; la presenza di un ingrediente attivo sulle superfici esterne e l'irregolarità di dette superfici non consentono un controllo preciso dell'effetto di disaggregazione nel caso di microparticelle destinate a liberare una quantità efficace di ingrediente attivo entro un predeterminato periodo di tempo. When the preparation of such microparticles comprises extrusion and / or grinding, this implies the formation of particles having irregular external surfaces; the presence of an active ingredient on the external surfaces and the irregularity of said surfaces do not allow a precise control of the disaggregation effect in the case of microparticles intended to release an effective quantity of active ingredient within a predetermined period of time.
Sono noti anche alcuni procedimenti per produrre particelle senza l’impiego di solvente e senza l'impiego di tecniche di estrusione e/o di macinazione.
