Abstract
Preparation of antibiotic A 40926 amide derivatives of formula (I) comprises reacting a compound of formula (III) with an amine of formula (IV) in an inert organic solvent in the presence of a condensing agent. The initial pH of the mixture, measured after diluting a sample of the reaction mixture with 9 volumes of water, is 6.5 - 9.0. NHR3-alk1-(NR4-alk2)p-(NR5- alk3)q-W (IV). R1 = 9-12C alkyl; M = H, ~a-D-mannopyranosyl or 6-O-acetyl-~a-D-mannopyranosyl; Y = -NR2-alk1-(NR3-alk2)p- (NR4-alk3)q-W; R2 = H or 1-4C alkyl; alk1-alk3 = 2-10C alkylene; p, q = 0 or 1; R3, R4 = H or 1-4C alkyl; or R2+R3 = 2-4C alkylene provided that p = 1; or R3+R4 = 2-4C alkylene provided that p, q = 1; W = H, 1-4C alkyl, amino, mono- or di(1-4C)alkylamino or amino (substituted with one or two amino(2-4C)alkyl, (1-4C)alkylamino(2-4C)alkyl or di(1- 4C)alkylamino(2-4C)alkyl); or when p and q = 0, then W forms a piperazino or 4-methylpiperazino ring with the -NR3-alk1- moiety.
