Description
Apremilaszt tartalmú topikális készítményTopical preparation containing apremilast
A találmány tárgya apremilasztot és adott esetben további hatóanyagot tartalmazó topikális készítmény pszoriázis kezelésére. A találmány tárgya továbbá eljárás a találmány tárgya szerinti topikális gyógyszerkészítmény előállítására.The subject of the invention is a topical preparation containing apremilast and optionally an additional active ingredient for the treatment of psoriasis. The subject of the invention is also a method for the preparation of a topical pharmaceutical preparation according to the subject of the invention.
A találmány apremilaszt, kémiai nevén (+)-2-[l.(3-etoxi-4-metoxifenil)-2-metilszulfoniletil]4-acetilaminoizoindolin-l,3-dion hatóanyagot tartalmazó topikális készítményt ismertet. Az apremilaszt PDE-4 inhibitor vegyület, hatásmechanizmusának lényege, hogy blokkolja a ciklikus adenozin-monofoszfát (cAMP) degradálódását a IV. típusú foszfodiészteráz (PDE-4) enzim gátlásán keresztül. A PDE-4 enzimet expresszáló sejtekben ezáltal nő a cAMP szint. Többek között ez a hatásmechanizmus biztosítja gyulladáscsökkentő hatását, mely számos krónikus gyulladásos állapot kezelését teszi lehetővé.The invention describes a topical preparation containing the active ingredient apremilast, whose chemical name is (+)-2-[1.(3-ethoxy-4-methoxyphenyl)-2-methylsulfonylethyl]4-acetylaminoisoindoline-1,3-dione. The essence of the mechanism of action of apremilast PDE-4 inhibitor compound is to block the degradation of cyclic adenosine monophosphate (cAMP) in IV. through the inhibition of the type phosphodiesterase (PDE-4) enzyme. This increases the level of cAMP in cells expressing the PDE-4 enzyme. Among other things, this mechanism of action ensures its anti-inflammatory effect, which enables the treatment of many chronic inflammatory conditions.
Az apremilaszt hatóanyagot és származékait, valamint alkalmazását először a Celgene Corporation gyógyszercég a WO0025777 számú nemzetközi szabadalmi bejelentésében írta le.The active ingredient apremilast and its derivatives, as well as their use, were first described by the pharmaceutical company Celgene Corporation in international patent application No. WO0025777.
Az orális készítményt a WO2013101810A1 nemzetközi szabadalmi bejelentésben ismertetik, melyben leírják a forgalomban lévő filmtabletta összetételét is.The active substance is marketed in the form of film-coated tablets under the brand name Otezla in the indication of psoriasis and psoriatic arthritis. The oral preparation is described in the international patent application WO2013101810A1, in which the composition of the film-coated tablet on the market is also described.
Pszoriázis kezelésére alkalmazott, kizárólag apremilaszt hatóanyagtartalmú krém vagy egyéb topikális készítmény jelenleg nincs kereskedelmi forgalomban, azonban ismert néhány irat, mely apremilaszt hatóanyagtartalmú transzdermális készítményt ismertet. There are currently no creams or other topical preparations used for the treatment of psoriasis that contain only apremilast as an active ingredient, but there are some documents that describe a transdermal preparation containing apremilast as an active ingredient.
A WO2015062611 számú nemzetközi szabadalmi bejelentés a bőr barrier egyensúlyát visszaállítani képes dermatológiai készítményt ismertet, mely apremilaszt hatóanyagot is tartalmazhat. International patent application No. WO2015062611 describes a dermatological preparation capable of restoring the balance of the skin barrier, which may also contain the active ingredient apremilast. The system according to the description contains an aqueous and a lipophilic phase, which is stabilized by the addition of alkyl sulfates and monoglycerides. Unlike the solution of our invention, this system is not water-free and contains other auxiliary materials.
A WO2016198472 számú nemzetközi szabadalmi bejelentés elsősorban aminoguanidin származékot tartalmazó topikális szállítórendszert ír le, melyben adott esetben apremilaszt is szerepel második hatóanyagként. International patent application No. WO2016198472 primarily describes a topical delivery system containing an aminoguanidine derivative, in which apremilast is included as a second active ingredient. The description describes an anhydrous emulsion containing a discontinuous polar phase and a continuous lipid phase, which primarily ensures the absorption of aminoguanidine derivatives. This preparation has only two phases and the polar phase contains auxiliary materials different from our invention.
A CN 105168136 számú kínai szabadalom olyan transzdermális készítményt ismertet, melyben az apremilasztot foszfolipidből (lecitin) és koleszterinből alkotott flexibilis nanoliposzómák segítségével juttatják a bőrbe. A jó biohasznosulással rendelkező rendszer találmányunktól eltérően vizet tartalmaz.Chinese patent number CN 105168136 describes a transdermal preparation in which apremilast is delivered to the skin using flexible nanoliposomes made of phospholipid (lecithin) and cholesterol. Unlike our invention, the system with good bioavailability contains water.
A WO2017168433 számú nemzetközi szabadalmi bejelentés olyan topikális készítményeket ismertet, melyek apremilasztot és hordozóként, illetve felszívódást elősegítő anyagként DMSOt tartalmaznak. Találmányunk tárgya ezen felül a találmány szerinti készítmény alkalmazása pszoriázis, pszoriázis okozta artritisz, dermatítisz, atópiás dermatítisz, akne, dermatomiozítisz, kólitisz ulceróza, Behcet-kór, Crohn-betegség, szarkoidózis, uveitisz, rozácea, lichen plánusz kezelésére.The subject of our invention is also the use of the composition according to the invention to treat psoriasis, psoriatic arthritis, dermatitis, atopic dermatitis, acne, dermatomyositis, ulcerative colitis, Behcet's disease, Crohn's disease, sarcoidosis, uveitis, rosacea, lichen planus.
A pszoriázisos, illetve immunrendszer eredetű bőrgyulladások jellemzője, hogy a bőr egyes rétegei eltérő felületi sajátságokkal rendelkeznek. A pszoriázisos betegek bőre nem intakt, a bőrfelületet alkothatják száraz plakkos, sebes, gyulladt és ép bőrrétegek is. A pszoriázis jellegzetes bőrtünetei a sötétvörös, hámló, bőrfelszínből kiemelkedő papulák a betegség előrehaladtával nagyobb, száraz felületű plakkokká folynak össze. A characteristic of psoriatic or immune-related skin inflammations is that individual layers of the skin have different surface characteristics. The skin of psoriasis patients is not intact, the skin surface can be made up of dry plaque, scarred, inflamed and healthy skin layers. The characteristic skin symptoms of psoriasis are dark red, scaly papules protruding from the skin surface, which merge into larger plaques with a dry surface as the disease progresses. The plaques are separated from the healthy skin surface, their surface is covered by lamellar waxy white peeling, parakeratosis. This layer can be easily scraped off and small bleeding and wounds develop underneath, which we call Auspitz's symptom. This is because the vascular structure of the skin changes in psoriasis plaques.
A single-phase oily or water-based topical preparation is not suitable for the treatment of such different types of skin surfaces. That is why we thought that we would develop a multi-phase, adaptable system, which can also ensure the intake of active ingredients for skin surfaces with different physical properties.
A rendszer kis mennyiségű vizet xerogél formájában képes megkötni, ezáltal szárazon tartani a bőrfelületet. Nagyobb mennyiségű víz hatására, melyet a pszoriázisban szenvedő betegek egyéb terápiák során visznek fel általában a bőrfelületre, in situ emulziós hidrogéllé alakul és a felületet tartósan hidratálja, védi, puhítja olyan módon, hogy a hatóanyagfelszabadulás mechanizmusát eközben gyökeresen nem változtatja meg.The anhydrous gel cream according to the subject of the invention contains 4 phases in one system. The advantage of our preparation is that the phases are able to adhere to skin with different surface characteristics and release the active ingredient they carry. The particles of the dispersion are able to change their active ingredient content along the distribution coefficient and diffuse into the part of the system containing less active ingredient. The system can bind a small amount of water in the form of a xerogel, thereby keeping the skin dry. Under the influence of a larger amount of water, which patients suffering from psoriasis usually apply to the skin during other therapies, it turns into an emulsion hydrogel in situ and permanently hydrates, protects and softens the surface in a way that does not fundamentally change the mechanism of the release of the active ingredient.
Ezek a kopolimerek alkalmasak mind a hidrofil, mind a lipofil fázisban lévő hatóanyag stabilizálására.Surprisingly, we found that the development of a topical preparation with such a complicated operating system can be realized with the help of polyether polysiloxane copolymers, which are currently rarely used in pharmaceutical technology. These copolymers are suitable for stabilizing the active ingredient in both the hydrophilic and lipophilic phases.
Találmányunk szerinti gélkrém 4 fázisa a következőképpen épül fel.The 4 phases of the gel cream according to our invention are structured as follows.
A gélkrém első fázisa PEG szilikon poliéter kopolimer emulgensekkel stabilizált poláris oldószerben oldott apremilasztot tartalmazó gélképzővel kialakított hidrofil fázis. The first phase of the gel cream is a hydrophilic phase formed with a gel former containing apremilast dissolved in a polar solvent stabilized with PEG silicone polyether copolymer emulsifiers. The first phase is a highly hydrophilic colloid phase, in which the mixture of the active ingredient apremilast dissolved in a polar solvent is converted into a hydrogel with a gel former and stabilized with PEG-silicone polyether copolymer emulsifiers. The task of the hydrophilic phase is to penetrate the hydrophilic parts of the skin surface and deliver the active ingredient to these areas.
A PEG-szilikon poliéter kopolimerek polietilén glikollal polimerizált dimetil-polisziloxán vegyületek, más néven dimetikonok. Ezek közül a hidrofil fázis stabilizálására a PEG-10 és PEG-12 dimetikonokat használjuk. A találmány szerinti készítmény hidrofil fázisának stabilizálására a PEG 10 dimetikonok közül a Dow Corning cég TI-6021 W/O Formulation Aid elnevezésű segédanyagát, míg a PEG-12 dimetikonok közül előnyösen a Dow Corning cég TI2011 SPE Speciality Fluid segédanyagát alkalmaztuk. A PEG-12 dimetikonokat jellemzően hatóanyag szolubilizátorként, habstabilizátorként, lágyítóként, hajápolási termékekben a haj gubancosodását gátló segédanyagként, illetve meleg technológiákhoz használnak.PEG-silicone polyether copolymers are dimethylpolysiloxane compounds polymerized with polyethylene glycol, also known as dimethicones. As a gelling agent in the hydrophilic phase and in the fourth phase, any gel-forming acrylic acid polymer, copolymer or their derivatives and mixtures can be used. Polycarbophil derivatives, acrylic acid copolymers cross-linked with divinyl glycol, preferably Lubrizol AA-1 USP Policarbophil can be used as gel formers. The amount of the gel former may be 0.05-20% by weight of the total weight of the gel cream, preferably 1%.
A második, erősen lipofil fázis apremilasztot tartalmazó PEG szilikon poliéter kopolimerrel emulgeált sziloxán polimer emulzió.The second, highly lipophilic phase is a siloxane polymer emulsion emulsified with a PEG silicone polyether copolymer containing apremilast.
A sziloxán polimerek sziloxán kötést [-Si(Me)2-O-] tartalmazó szilíciumorganikus polimerek, más néven szilikonok, vagy polidimetilsziloxán vegyületek, melyek lineáris és térhálós szerkezetűek is lehetnek. A lipofil fázisban alkalmazott sziloxán polimer hordozóanyag bármely szilikon olaj, szilikon viasz vagy szilikon elasztomer közül választható, illetve lehet ezek keveréke. Készítményünk segédanyagaként előnyben részesítjük az egyenes láncú polidimetilsziloxán vegyületeket, 100 est viszkozitású dimetikonokat, előnyösen a Dow Corning cég Silicone Fluid 100 termékét. A polidimetilsziloxán hordozó mennyisége a gélkrém teljes tömegének 0,1-30 tömeg%-a lehet, előnyösen 1 tömeg%-a.Siloxane polymers are organosilicon polymers containing a siloxane bond [-Si(Me)2-O-], also known as silicones, or polydimethylsiloxane compounds, which can have a linear or cross-linked structure. The siloxane polymer carrier material used in the lipophilic phase can be chosen from any silicone oil, silicone wax or silicone elastomer, or it can be a mixture of these. We prefer straight-chain polydimethylsiloxane compounds, dimethicones with a viscosity of 100 est, preferably Dow Corning's Silicone Fluid 100 product as excipients in our preparation. The amount of the polydimethylsiloxane carrier can be 0.1-30% by weight of the total weight of the gel cream, preferably 1% by weight.
Ennek a lipofil fázisnak a készítményen belüli funkciója, hogy a bőrfelület lipofil rétegeibe hatoljon és bejuttassa a hatóanyagot. Ezen felül ez a lipofil réteg felületi védőréteget hoz létre a bőr sérült, nem lipofil területein. The function of this lipophilic phase within the preparation is to penetrate the lipophilic layers of the skin surface and deliver the active ingredient. In addition, this lipophilic layer creates a surface protective layer on damaged, non-lipophilic areas of the skin.
A harmadik, szintén lipofil fázis lipofil kenőcsalapú mátrix, melyet oldásközvetítőkkel, szolubilizáló szerrel és/vagy emulgeálószerrel elegyítve, adott esetben szolubilizáló rendszerrel alakítunk ki.The third, also lipophilic phase, is a lipophilic ointment-based matrix, which is formed by mixing with solubilizers, solubilizers and/or emulsifiers, and if necessary with a solubilizer system.
Ez a fázis képezi a kenőcs alapot, mely hagyományos zsíros krémként viselkedik, a pszoriázisra jellemző plakkos bőrt puhítja, így lehetővé teszi a gélkrém behatolását az alsóbb bőrrétegekbe is.This phase forms the basis of the ointment, which behaves like a traditional greasy cream, softens the plaque skin characteristic of psoriasis, thus allowing the gel cream to penetrate into the lower layers of the skin.
A készítmény kenőcsalapjában szolubilizáló-, illetve emulgeálószerként alkalmazhatunk szorbitán észtereket, polioxietilén-alkil étereket, polioxietilén-szorbitán zsírsav észtereket, polietilén-glikol polipropilén-glikol kopolimereket, koleszterint és származékait, sztearin-, és sztearilalkoholokat és származékaikat.Sorbitan esters, polyoxyethylene-alkyl ethers, polyoxyethylene-sorbitan fatty acid esters, polyethylene-glycol polypropylene-glycol copolymers, cholesterol and its derivatives, stearic and stearyl alcohols and their derivatives can be used as solubilizers and emulsifiers in the lubricant.

Ezen funkció betöltésére használhatunk még a gélkrémben propilén-glikolt, etilén-glikolt, glicerint (előnyösen 100%-os glicerint), glikofurolt, polietilén-glikolt (pl.:bármely PEG származékot PEG100-PEG 1500 közül), propilén-karbonátot, dietil-ftalátot, bármely folyékony alkohol (pl.: etanol, izopropil alkohol, butanol, benzil-alkohol, fenoxietanol) nagy tisztaságú formáját (95%-100% koncentrációban), illetve természetes, félszintetikus vagy szintetikus illóolajokat és komponenseiket, olajsavat, oleil-alkoholt, előnyösen hexilén glikolt. A hexilén glikol mennyisége a készítmény teljes tömegéhez viszonyítva 0,1-60%, előnyösen 5%.In the preparation according to the invention, additional solubilizers can be used, if necessary, which can form a solubilizing system with the above-listed solubilizers. To fulfill this function, we can also use propylene glycol, ethylene glycol, glycerin (preferably 100% glycerin), glycofurol, polyethylene glycol (e.g. any PEG derivative from PEG100-PEG 1500), propylene carbonate, diethyl- phthalates, the highly pure form of any liquid alcohol (e.g.: ethanol, isopropyl alcohol, butanol, benzyl alcohol, phenoxyethanol) (95%-100% concentration), as well as natural, semi-synthetic or synthetic essential oils and their components, oleic acid, oleyl alcohol, preferably hexylene glycol. The amount of hexylene glycol in relation to the total weight of the preparation is 0.1-60%, preferably 5%.
A találmány szerinti készítmény negyedik fázisát polikarbofil gélképzővel alakítjuk ki.The fourth phase of the composition according to the invention is formed with a polycarbophil gel former.
The gel-former is an inactive substance in the dry state, but once it reaches the skin surface, it binds the water there and forms a gel in situ. The hydrogel formed in this way forms a reservoir and ensures a sustained release of the active ingredient.
Találmányunk szerinti rendszer változásra képes a bőrfelületen található víz mennyiségének függvényében. A készítmény alapvetően vízmentes, képes a felületet szárazon tartani. A pszoriázis terápiájában jellemző a hidratálás. The system according to our invention can change depending on the amount of water on the skin surface. The product is essentially water-free and can keep the surface dry. Hydration is typical in psoriasis therapy. Psoriasis patients often use moisturizing creams and solutions. due to which a greater amount of water is present in the layers of their skin surface. Our system can bind this water. It binds a small amount of water in the form of a xerogel, while under the influence of a hundred or two hundred times larger amount of water (applied by hydration during therapy) it turns into an emulsion hydrogel in situ and permanently hydrates, protects and softens the surface. Due to the microemulsion nature of the preparation, this process does not radically change the release mechanism of the active ingredient.
A találmány szerinti topikális készítményben az apremilaszt mennyisége a készítmény teljes tömegére vonatkoztatva 0,3-3 tömeg%.The amount of apremilast in the topical preparation according to the invention is 0.3-3% by weight based on the total weight of the preparation.
Találmányunk szerinti topikális készítmény az apremilaszt mellett adott esetben más hatóanyagot is tartalmazhat. A gélkrém apremilaszt mellett D-vitamin származékot vagy Dvitamin analógot, különösen előnyösen kalcipotriolt tartalmazhat. In addition to apremilast, the topical preparation according to our invention may also contain other active ingredients. A találmány szerinti kombinációs készítmény különösen előnyösen apremilasztot és kalcipotriolt tartalmaz. A kombinációs készítmény másik hatóanyagának mennyisége a készítmény teljes tömegének 0,001-0,01tömeg%-a, előnyösen 0,005 tömeg%-a.Among the vitamin D derivatives or analogues, the following compounds may be included in the combination: calcipotriol, calcitriol, tacalcitol, maxacalcitol, paricalcitol and alfacalcidol. The amount of the other active ingredient of the combination preparation is 0.001-0.01% by weight of the total weight of the preparation, preferably 0.005% by weight.
A találmányunk szerinti gélkrém számára a fent ismertetetett négyfázisú rendszer biztosítja, hogy a hidrofil és a lipofil szövetekbe is képes legyen hatóanyagot juttatni. Ennek a négyfázisú bipoláris diszperziónak a részecskéi a megoszlási hányados mentén képesek a hatóanyagtartalmukat változtatni, tehát a hatóanyag mindig abba az irányba áramlik a rendszeren belül, ahol a hatóanyagleadás nagyobb mértékű.For the gel cream according to our invention, the four-phase system described above ensures that it is able to deliver the active ingredient to both hydrophilic and lipophilic tissues. The particles of this four-phase bipolar dispersion are able to change their active ingredient content along the distribution coefficient, so the active ingredient always flows in the direction in the system where the active ingredient release is greater.
A találmány szerinti gélkrém előállítása során először a hatóanyago(ka)t ultrahang segítségével oldjuk a szerves poláris oldószerben, majd az oldatot elegyítjük a gélképző 30%-val. A kenőcsalap tartalmazza a lipofil hordozót, a gélképző maradékát, a PEG szilikon poliéter kopolimer emulgenseket, a sziloxán polimer hordozót és adott esetben a további oldást elősegítő anyagot, szolubilizálószert. Ezután a hatóanyaggélt belekeverjük a vazelines kenőcsalapba. A gélkrémet állandó kevertetés mellett a krémes állag eléréséig hűtjük, majd turraxolással homogenizáljuk, végül állandó kevertetés mellett tovább hűtjük és homogenizáljuk.During the production of the gel cream according to the invention, the active ingredient(s) are first dissolved in the organic polar solvent with the help of ultrasound, and then the solution is mixed with 30% of the gel former. Next, we prepare the grease trap. The lubricant contains the lipophilic carrier, the remainder of the gel former, the PEG silicone polyether copolymer emulsifiers, the siloxane polymer carrier and, where applicable, the additional dissolution promoting material, solubilizer. The lipophilic lubricant is produced by melting the lipophilic carrier, preferably petroleum jelly, and a solubilizing agent, preferably Span 60, and then suspending the remaining gelling agent in the melt. The melt is cooled with constant stirring until it reaches a thick flow consistency and at this temperature the silicone derivatives are mixed in, preferably Silicone Fluid 100, TI-6021 and TI2011 and, if applicable, the other solubilizer, preferably hexylene glycol. The melt is further cooled with constant stirring. Then mix the active ingredient gel into the Vaseline ointment. The gel cream is cooled with constant stirring until it reaches a creamy consistency, then it is homogenized by turraxing, and finally it is further cooled and homogenized with constant stirring.

Raman mikroszkópos fázis és hatóanyag eloszlás vizsgálatokRaman microscopic phase and active substance distribution studies
A találmány szerinti gélkrém szerkezetének bizonyítására Raman mikroszkópos vizsgálatokat végeztünk. A készítmény különböző fázisait tartalmazó minták spektrumait felvettük és ezek felhasználásával Raman térképezéses vizsgálatokkal bizonyítottuk a hatóanyag elhelyezkedését, mennyiségét az egyes fázisokban, illetve a négy fázis jelenlétét a találmány szerinti készítményben. To prove the structure of the gel cream according to the invention, Raman microscopic tests were performed. We recorded the spectra of the samples containing the different phases of the preparation and using them, we proved the location and quantity of the active ingredient in the individual phases and the presence of the four phases in the preparation according to the invention with Raman mapping tests.
A méréseket Thermo Scientific DXR Raman mikroszkóppal végeztük, a műszerben lévő spektrográfhoz egy 780 nm hullámhosszúságon emittáló lézer fényforrást használtunk, a mintákat 24 mW erősségű fényforrással vizsgáltuk. A spektrumok 1600-600 cm-1 jellemző tartományát vettük figyelembe, a térképezés során felvett spektrumokra ezt illesztettük. Apremilast esetén a térképezés spektrumain az apremilastra jellemző 1765 cm-1 hullámszámnál jelentkező csúcs intenzitását vettük figyelembe, melyhez a 1800-1740 cm-1 hullámszám tartományon lévő alapvonal korrekciót alkalmaztunk. The measurements were performed with a Thermo Scientific DXR Raman microscope, a laser light source emitting at a wavelength of 780 nm was used for the spectrograph in the instrument, the samples were examined with a light source of 24 mW. The characteristic range of the spectra 1600-600 cm-1 was taken into account, and this was fitted to the spectra recorded during the mapping. In the case of apremilast, we took into account the intensity of the peak at the wave number 1765 cm-1 characteristic of apremilast in the mapping spectra, for which a baseline correction in the wave number range 1800-1740 cm-1 was applied.
The effectiveness of the preparation according to our invention was confirmed by an oxazolone-induced contact allergic dermatitis test on mice. This test is accepted as an animal experimental model of psoriasis, given that the T-cells of the immune system play a significant role in the development of atopic dermatitis and psoriasis, which are the common starting points of these two diseases.

A vizsgálatokat Christensen és munkatársai módszere (Christensen AD, Skov S, Haase C.: Local and systemic effects of co-stimulatory blockade using cytotoxic T lymphocyte antigen4-immunoglobulin in dinitrofluorobenzene- and oxazolone-induced contact hypersensitivity in mice. Clin Exp Immunol. A rákövetkező héten 1., 2., 3. The tests were performed according to the method of Christensen et al. (Christensen AD, Skov S, Haase C.: Local and systemic effects of co-stimulatory blockade using cytotoxic T lymphocyte antigen4-immunoglobulin in dinitrofluorobenzene- and oxazolone-induced contact hypersensitivity in mice. Clin Exp Immunol. 2013 Feb;171(2):220-30). For our experiments, we used male BALB/c mice with a body weight of 20-25 g (6-8 animals/group). The animals were anesthetized with 400 mg/kg (10 ml/kg) chloral hydrate administered intraperitoneally. After falling asleep, the back skin of the mice was shaved. Two hours later, the animals were sensitized with 5% oxazolone dissolved in a 4:1 solution of acetone and olive oil (50 μl/animal), which was dripped onto the shaved backs of the mice. After one week, the gel creams to be tested (20 μl/ear) were applied to the inner surface of the right ear of the mice. 30 minutes after the treatment, 1% oxazolone dissolved in a 4:1 ratio solution of acetone and olive oil was dripped onto the inner and outer side of the right ear of the animals using an automatic pipette, and only the solvent was similarly applied to the left ear. In the following week, on days 1, 2, 3 and 7, the thickness of both ears of the animals was measured with a digital caliper. The degree of edema was characterized by the difference in the thickness of the right and left ear. The effect occurring in a one-week process was performed by comparing the areas under the curve (AUC). The statistical evaluation was performed with Dunnett's test after the 24-hour measurement and, in the case of AUC, one-point analysis of variance.
Az oxazolonnal való indukálás után 24 órával külön állatokon mértük a myeloperoxidáz (MPO) enzim aktivitását fül szövetből. A gyulladás helyén beszűrődő neutrofil granulociták mennyiségére lehet következtetni az enzim aktivitásából (Trush, M.A.; Egner, P.A.; Kensler, T.W.: Myeloperoxidase as a biomarker of skin irritation and inflammation. Food and Chemical Toxicology. 1994 Feb 32(2):143-147). Az adatok kiértékelésénél az alap aktivitást és a 60 vagy 120 másodperc utáni aktivitás különbségét vettük, mely értékeket standardizáltunk a minták fehérjetartalmával.24 hours after induction with oxazolone, the activity of myeloperoxidase (MPO) enzyme from ear tissue was measured in separate animals. The amount of neutrophil granulocytes infiltrating the site of inflammation can be inferred from the activity of the enzyme (Trush, M.A.; Egner, P.A.; Kensler, T.W.: Myeloperoxidase as a biomarker of skin irritation and inflammation. Food and Chemical Toxicology. 1994 Feb 32(2):143-147 ). In vivo, MPO forms hypochlorous acid (HOCl) from H2O2 and chloride ions, which is cytotoxic to pathogens. An increase in MPO activity can also be detected in the ear edema test during the development of acute inflammation, the maximum activity of which is at 24 hours (based on a preliminary experiment). The activity (quantity) of MPO was determined colorimetrically at 450 nm after the addition of the substrate (o-Dianizidine dihydrochloride). When evaluating the data, we took the basic activity and the difference between the activity after 60 or 120 seconds, which values were standardized by the protein content of the samples.
A 2. ábrán szemléltetjük a BALB/c egéren oxazolonnal kiváltott kontakt dermatitisz: ödéma nagyságát 24 óra elteltével.Fig. 2 shows the magnitude of the oxazolone-induced contact dermatitis and edema in BALB/c mice after 24 hours.
A 3. ábra mutatja a BALB/c egéren oxazolonnal kiváltott kontakt dermatitisz: 168 órás görbe alatti területetet (AUC).Figure 3 shows oxazolone-induced contact dermatitis in BALB/c mice: area under the curve (AUC) at 168 hours.
A 4. ábrán szemléltetjük a BALB/c egéren oxazolonnal kiváltott kontakt dermatitisz: mieloperoxidáz MPO) szintjét 24 óra elteltével a kontroll értékek %-os arányában.Fig. 4 shows the level of oxazolone-induced contact dermatitis in BALB/c mice: myeloperoxidase MPO) level after 24 hours as a % of the control values.
Based on 2., it can be concluded that the contact dermatitis reaction induced by oxazolone one week after sensitization was statistically significantly inhibited by all 12 samples 24 hours after induction. However, it can also be seen that in the case of treatment with reference samples, the effect is statistically significant, but not dose-proportional.
A 3. ábrán szemléltetett egy hétig fennmaradó hatást igazoló paraméterben (AUC) az 1.-9. példák alapján előállított minták közel azonos, statisztikailag szignifikáns gátlást mutattak, míg a 3 referens minta hatása elmaradt az előző kettőhöz viszonyítva. In the parameter (AUC) confirming the effect remaining for one week illustrated in Figure 3, the 1st-9th the samples produced based on examples showed almost identical, statistically significant inhibition, while the effect of the 3 reference samples was less than the previous two.
A 4. The results of the in vitro myeloperoxidase (MPO) enzyme level measurement performed 24 hours after the induction shown in Figure 4 show that the 1-3. and 7-9 the samples produced based on examples were the most effective, while the reference samples (R1-R-3) did not demonstrably reduce the enzyme activity.
