Abstract

D 1-Cortisol is prepared by adding solid cortisol to a culture of the micro-organism Corynebacterium simplex and fermenting the culture under aerobic conditions. From 1 gram to 400 grams of cortisol may be used per litre of culture solution. The progress and completion of the fermentation process may be determined by observing microscopically the decrease of crystals of short pillar or plate form (cortisol) and the increase of crystals of needle or long pillar form (D 1-cortisol) in the fermented broth. The D 1-cortisol may be separated by recrystallization of the solid ingredients from an organic solvent such as methanol or acetone. Detailed examples are given.
