Abstract
Chloro-oleandomycin active against Grampositive micro-organisms and particularly Listeria monocytogenes is prepared by treating the antibiotic oleandomycin with anhydrous hydrogen chloride in an anhydrous organic solvent. Chloro-oleandomycin is separated from unchanged oleandomycin by countercurrent distribution using a solvent system consisting of methyl isobutyl ketone acetone and a phosphate citrate buffer of pH 7.0. Chloro-oleandomycin is recovered from the methyl isobutyl ketone layer. Salts with inorganic and organic acids containing from one to twelve carbon atoms are prepared by adding the appropriate acid, especially under anhydrous conditions, to the above chloro-oleandomycin solution. Specified salts are the mineral acid salts such as the hydrochloride, nitrate and phosphate, and the salts with formic, acetic, butyric, lauric, benzoic, p-nitro benzoic and naphthoic acids. Suitable anhydrous organic solvents are propanol, isopropanol, acetone, methyl ethyl ketone, di-isopropyl ketone, methyl isobutyl ketone, ethyl acetate and propyl acetate. The physical characteristics of chloro-oleandomycin hydrochloride, nitrate and phosphate are described. Specification 779,257 is referred to.ALSO:The antibiotic chloro-oleandomycin (see Group IV (b)) is used orally in aqueous solution or in admixture with the animals food, for the treatment of Listeriosis in animals.
